Antineoplastic Effect of Several Herbal Medicine Mixtures on SNU-80 Anaplastic Thyroid Carcinoma Cell Line (수종 한약 복합물의 역형성갑상선암세포 SNU-80에 대한 항암효과)

  • Objectives: The purpose of this study was to investigate the antineoplastic effect of several herbal medicine mixtures (compositions of Astragalus membranaceu, Angelica gigas, Trichosanthes kirilowii, Panax ginseng, Rhus verniciflua Stokes) on the SNU-80 anaplastic thyroid carcinoma cell line. Methods: MTT assay was used to examine whether our herbal medicine mixtures decreased cell growth rate of SNU-80. Wound healing assay and Transwell invasion assay was performed to investigate whether our herbal medicine mixtures affect the migration and invasion of anaplastic cancer cells, SNU-80. ELISA assay was performed to know if our herbal medicine mixtures suppressed the expression of pro-invasive molecules, such as vascular endothelial growth factor (VEGF) and matrix metalloproteinase-2 (MMP-2) secreted from SNU-80. Results: MTT assay demonstrated that A. membranaceus:A. gigas:T. kirilowii=1:1:1 or 3:1:1, A. membranaceus:A. gigas :T. kirilowii:P. ginseng=1:1:1:1 or 3:1:1:1, A. membranaceus:A. gigas:T. kirilowii:P. ginseng:R. verniciflua Stokes=1:1:1:1:1 or 3:1:1:1:1 strongly suppressed the growth of SNU-80. Wound healing assay demonstrated that A. membranaceus:A. gigas=3:1, A. membranaceus:A. gigas:T. kirilowii=1:1:1 or 3:1:1, A. membranaceus:A. gigas:T. kirilowii:P. ginseng=1:1:1:1 or 3:1:1:1, A. membranaceus:A. gigas:T. kirilowii:P. ginseng:R. verniciflua Stokes=1:1:1:1:1 or 3:1:1:1:1 inhibited the migration of SNU-80. Also, A. membranaceus:A. gigas=1:1, A. membranaceus:A. gigas:T. kirilowii=1:1:1 or 3:1:1, A. membranaceus :A. gigas:T. kirilowii:P. ginseng=1:1:1:1 or 3:1:1:1, A. membranaceus:A. gigas:T. kirilowii:P. ginseng:R. verniciflua Stokes =1:1:1:1:1 or 3:1:1:1:1 suppressed the expression of MMP-2. Conclusions: The results obtained in this study suggest that several herbal medicine mixtures suppresse the growth and inhibit the migration and invasion of SNU-80, which is anaplastic thyroid cancer cells. Especially, A. membranaceus:A. gigas: T. kirilowii=1:1:1 mixture had a stronger anti-cancer effect.

Changes Observed in Doenjang (Soybean Paste) Containing Fermented-Rhus verniciflua Extract During Aging (발효옻 추출물 첨가 된장의 숙성에 따른 변화)

  • Fermented-Rhus verniciflua (FRV) extract is increasingly used in fermented soy products, fermented vinegars, and certain alcoholic beverages. In this study, we investigated the effects of FRV extract on the physicochemical properties of doenjang (soybean paste). Addition of FRV extract to doenjang resulted in a 28.2-45.4% increase in the amino acid content and a 1.3- to 1.5-fold increase in the concentrations of glutamic acid, which imparts a savory flavor to doenjang. The concentration of biogenic amine (BA) of the sample containing the extract was 5.3-52.6% lower than that of the control. The major components of BA included tyramine (55.1-74.6%), followed by putrescine, spermidine, tryptamine, and cadaverine, in decreasing concentrations. The organic acid concentration of the sample containing the extract was 1.2-1.3-fold higher than that of the control. The total free sugar concentration was 163.4 mg/100 g in the control and 206.6-276.8 mg/100 g in the supplemented sample, showing a 1.3- to 1.9-fold increase as the addition of the extract.

Effect of Hot Water Extracts of Roasted Rhus vemiciflua Stokes on Antioxidant Activity and Cytotoxicity (볶음 처리한 옻나무 열수추출물의 항산화성 및 암세포 성장억제효과)

  • The inhibitory effects of hot water extracts of Rhus verniciflua Stokes pith and peel roasted at 170, 200 and $220^{\circ}C$ on lipid peroxidation, formation of DPPH free radicals and growth of four human cancer cells such as HepG2 (liver cancer), SNU-1 (stomach cancer), MCF-7 (breast cancer) and Widr (colon cancer) were examined. The antioxidant activities and growth inhibitory effects on cancer cells of hot water extracts of peel were higher than those of pith, and the activities were dose-dependent. The roasting temperature showing the highest antioxidant activities and growth inhibitory effects on cancer cells was in the range of $170\~200^{\circ}C$ The lipid peroxidation and formation of DPPH free radicals of hot water extracts of roasted pith and peel were inhibited to 50.9, $56.5\%\;and\;79.0,\;78.4\%$ at the concentration of $500\mu g/mL$, respectively. The growth inhibitory effects of roasted pith and peel on cancer cells were in the order of Widr (41.5, $36.0\%$) > HepG2 (61.5, $44.0\%$) > MCF-7 (92.0, $69.2\%$)> SNU-1 (100, $100\%$) cells at the concentration of $1,000\mu g/mL$ as compared with the control, respectively. These results suggest that roasted Rhus verniciflua Stokes could be an useful natural medicinal plant for colon cancer.

Effects of Feeding Rhus verniciflua Extract on Egg Quality and Performance of Laying Hens (옻나무 추출액 급여가 산란계의 생산성 및 계란 품질에 미치는 영향)

  • This experiment was conducted to investigate the effects of drinking of Rhus tree-extract on laying performance and egg quality in hens. Four hundred eighty, 55-wk-old ISA brown, laying hens were divided into six groups, control, Rhus tree-extract 500 ppm, 1,000 ppm, 2,000 ppm, 3,000 ppm and 5,000 ppm. The hens were fed a supplemented drink containing Rhus tree-extract for 12 weeks. Egg production and egg mass increased by drinking Rhus tree-extract (p<0.05) and the feed conversion ratio also improved in Rhus tree-extract groups. Cecal numbers of Lactobacillus spp., E. coli and Salmonella were not different in treatments. Availability of protein and ash improved in the Rhus extract groups. The eggshell breaking strength and egg shell thickness were significantly increased in Rhus tree-extract 3,000 ppm and Rhus tree-extract 2,000 ppm groups compared to the other groups. Also, egg yolk color and Haugh unit were significantly improved by the dietary Rhus tree-extract (p<0.05).

Effect of Theaflavin and Thearubigin from Microbial Fermented Teas on Urushiol Allergic Contact Dermatitis (미생물 발효차의 Theaflavin.Thearubigin에 의한 옻 알레르기성 접촉피부염에 미치는 영향)

  • 본 연구는 옻(Rhus verniciflua)에 대한 국산 미생물 발효차 폴리 페놀물질 인 theaflavin(TF)과 thearubigin(TR)의 항알레르기 효과를 조사하였다. HPLC, column chromatography 및 TLC로 이용하여, 한국 원주산, 일본산 및 중국산 락커(lacquer)로부터 아주 강한 알레르기를 유발하는 long chain alkylphenol을 함유하는 crude urushiol 성분을 분리하였다. 고분자 이중 및 삼중불포화물질의 함량은 중국산 라커의 crude urushiol에 가장 많이 포함되어 있었다. 중국산 라커의 crude urushiol에서 유발되는 접촉성 알레르기 피부염 발생은 5%농도에서 한국산이나 일본산 라커의 시료보다 높았으며, TF 및 TR를 처리한 모든 농도에서 접촉성 알레르기 피부염의 유의적인 감소를 나타내었다. 이들 결과로부터 발효차의 폴리페놀성 TF와 TR은 옻의 접촉성 피부염에 대한 유의적인 항알레르기 효과를 나타내었다.

Two New Caffeoyl Threonate Esters from the Leaves of Toxicodendron vernicifluum

  • Toxicodendron vernicifluum, also called as Rhus verniciflua is a deciduous tree belonging to Anacardiaceae family. Two new caffeoyl threonate esters, rhuseols A (1) and B (2), together with 5-O-(E)-caffeoylquinic acid methyl ester (3) were isolated from the leaves of T. vernicifluum. The structures of isolated compounds were established by using 1D and 2D NMR in combination with HR-ESI-MS. Compounds 1 - 3 showed DPPH radical scavenging effects with IC50 values of 47.9, 107.8 and 15.4 μM, respectively. Taken together, these compounds might contribute to the antioxidant properties of the leaves of T. vernicifluum, which will be useful for various oxidative stress mediated diseases.

  • 현재 우리의 식습관은 식물성 식습관을 벗어나 동물성 식품의 이용 비중이 점차 증가하고 있으며, 따라서 육가공품의 소비가 증가하고 있는 추세이다. 대부분의 육가공품이 장기간 보존을 함에 따라 여기에 보존료를 첨가하고 있어 위생상 문제점을 야기하고 있다. 이에 항산화 능력이 뛰어난 옻을 이용한 육가공 제품을 제조하여 제품의 품질과 밀접한 관련성이 있는 지질의 안정성을 분석하였다. 일반돈육, 옻 추출물을 사료에 첨가하여 사육한 돈육, 일반 돈육에 옻 추출물을 2%, 4% 및 8% 첨가시켜 각각 육제품을 제조하여 4$^{\circ}C$에서 60일간 저장하였다.   • This study was carried out to investigate the effect of dietary RVS on the WHC, color stability, and lipid oxidation of Hanwoo (Korean cattle) beef during cold storage after thawing, RVS fed Hanwoo (Korean cattle) beef was more effective in increasing WHC, color stability and delaying lipid oxidation than that without RVS in diet. And 4% RVS fed Hanwoo (Korean cattle) beef showed the greatest effects.

  • Methanolic aqueous extracts of 100 plants were screened for antioxidative activity using Fenton's reagen/ethyl linoeate system and free radical scavenging activity using DPPH free radical generating system. The results suggest that at least six plants including Eugenia caryophyllata, Alpinia offiicinarum, Rhus verniciflua, Curcuma longa, Rheum palmatvm and Evodia officinalis may be the potential sources of antioxidant, But only one plant, Cornus officinalis, may be the potential source of free radical scavenger from natural plants.
