Linkage Map Construction and Molecular Genetic Approach in Capsicum spp.

  • We have constructed a molecular linkage map of chili pepper (Capsicum spp.) in an interspecific (C. annuum cv. TF68 x C. chinense cv. Habanero) F$_2$ population of 107 plants with 150 RFLP and 430 AFLP markers. The resulting linkage map consists of 11 large (206-60.3 cM) and 5 small (32.6- 10.3 cM) linkage groups cover-ing 1,320 cM with an average map distance between framework markers of 7.5 cM. Most (80%) of the RFLP markers were pepper-derived clones and these markers were evenly distributed across the genome. By using 30 primer combinations, 444 AFLP markers were generated in the F$_2$population. The majority of the AFLP markers clustered in each linkage group, although PstI/MseI markers were more evenly distributed than Eco RI/MseI markers within the linkage groups. Genes for biosynthesis of carotenoids and capsaicinoids were mapped on our linkage map. This map will provide the basis of studying secondary metabolites in pepper.

Phylogenetic Study of Trichaptum Species Based on the RFLP Analysis of Mitochondrial DNA

  • Eight strains of Trichaptun (Polyporaceae), two strains from each species of T. abietinum, T. biforme, T. fusco-violaceum, and T. laricinum were examined to see their phylogenetic relationship by digesting mitochondrial DNAs with EcoRV, Hind III, XbaI, and PstI, and then analyzing fragmentation patterns with the methods of Nei and Li. T. abietinum, T. biforme, and T. laricinum developed an independent phylogenetic lineage, respectively, but T. fusco-violaceum FP-133997-sp showed a close relationship with two strains of T. bioforme, and T. fusco-violaceum HHB-4016-sp barely grouped with those of T. laricinum. Based on the results of the RFLP analysis of mtDNA, it is concluded that T. fusco-violaceum is under way to differentiation into two different subgroups.

  • Inoculation of human fecal cysts to suckling Mongolian gerbils, two Giardia lamblia isolates, Kl and K2, were established as axenic cultures. Using this in vitro culture, both two Giardia isolates were grouped by using two genetic analysis. With genetic analysis of SSS-rDNA sequences, both K1 and K2 were found as members of hopkins'group 1, despite some nucleotide differences noticed in K1 (5 differences/292 bases.). The other genetic study used PCI-RFLP of the tim (triose phosphate isomerase) Nash's group 2 can bot be a separate group, but a part of Hopkins' group 1.

  • Bone mineral density (BMD) is influenced by genetic and environmental factors. Among genetic study; calcitonin receptor (CTR) gene is a good candidate influencing the inter-individual difference in BMD because CTR is involved in calcium and bone metabolism. Thus, we investigated the distribution of C1377T polymorphism in the CTR gene among male Korean elite athletic and control groups, respectively and also an association with BMD in lumbar spine and femoral neck. Our results suggested that this polymorphism of CTR gene was not significantly associated with lumbar spine or femoral neck BMDs in the both groups, respectively. However, we found that there was the racial difference in genotype distribution of this polymorphism between Caucasian and Asian populations. Though we could not detect the significant association between C1377T polymorphism of CTR gene and lumbar spine or femoral neck BMDs, further studies using other ethnic groups are necessary to clarify the precise role in BMD of CTR gene.

Genetic Characterization based on Partial 28S rRNA Gene Sequence of Korean Two Scallops (한국산 가리비 2종의 28S rRNA 유전자 염기서열에 의한 유전적 특성)

  • 한국산 가리비, 큰가리비(Patinopecten yessoensis)와 주문진가리비(Chlamys swifti), 2종에 대한 28S ribosomal RNA 유전자의 PCR- 산물을 이용 RFLP 및 염기서열을 밝히고, 이미 보고된 2과 3종의 염기서열과 상동성을 비교 분석하였다. 그 결과 28S rRNA유전자를 이용하여 7가지 제한효소를 처리한 PCR-RFLP의 종간 차이에서 Taq I 제한효소에서만 차이를 볼 수 있었다. 한편 두종간에 28S rRNA유전자의 D1 부위의 염기서열에서 231개 부위 중 14군데에서 변이를 보였다.

Molecular identification of Anisakid worm third stage larvae isolated from masou salmon Oncorhynchus masou (시마연어, Oncorhynchus masou에서 분리된 아니사키스 속 선충 3기 유충의 분자생물학적 방법을 이용한 동정)

  • Anisakid nematodes third stage larvae were isolated from the muscles of masou salmon (Oncorhynchus masou). Fish were purchased from Jumunjin fishery market in Gangneung. Four Anisakid third stage larvae were isolated from 4 fish. Molecular identification of the isolated worms was conducted by PCR-RFLP analysis of ribosomal DNA internal transcribed spacer region and direct sequencing of mitochondrial DNA cox2 gene. As results, all the tested individual worms were identified as Anisakis simplex (sensu stricto). This is the first report of molecular detection of anisakid worms in salmonid fishes in Korea.
