Comparative RFLP Analysis of Chromosome 2M of Aegilops comosa Sibth et Sm. Relative to Wheat (T. aestivum L.)

  • Based on the co-linearity in the Triticeae, comparative RFLP analysis of 2M chromosome of Ae. comosa Sibth et Sm. was performed with 2MS and 2M additional lines of Triticum aestivum L. cv. Chinese Spring. Among the wheat RFLP probes conserved in the short arms of wheat chromosome 2, those above psr912 were located on the long arms of 2M in Aegilops comosa. The rest probes on the short arm and all the probe sequences on the long arm of group 2 chromosome in wheat were conserved on the equivalent chromosomal position in Aegilops comosa. So, it is apparent that some chromosomal segment from the short arm had been transferred to long arm while reconstructing 2M chromosome relative to wheat chromosomes. The break-point was located between psr912 and psr131 of the short arm. This rearrangement of chromosome 2M might be a molecular evidence of the M genome speciation from an ancestral type.

  • The present study has been performed to develop a PCR technology to identify human immunoglobulin(Ig) allotypes with restriction fragment length polymorphism(RFLP) using a probe. Genomic DNA were ampilified with PCR tecnology using primers from peripheral blood lymphocytes of 10 periodontal patiens, whose Ig allotypes have been pre-determined by serological tecnique using heagglutination technique. The result indicated that the RFLP patterns could successfully differentiate the Ig allotypes, which suggests that this technology can be developed as a tool useful for population genetics studies.

Association between Genetic Variation in the Human Factor Ⅶ Gene and Essential Hypertension in Korean Population

  • In view of the effect of factor Ⅶ as a risk factor for essential hypertension, we investigated the length (I/D) polymorphism at position 323 promoter region and exon 8-Msp I RFLP of the human factor Ⅶ gene in the Korean patients with essential hypertension and normal controls. There were no significant differences in the allele, genotype and haplotype frequencies of these polymorphisms between normotensive and essential hypertensive subjects. The significant linkage disequilibrium was however, detected between two polymorphic sites. The Msp I RFLP and I/D polymorphism were also significantly associated with plasma triglyceride (TG) levels. Therefore, our results suggest that the significant association between two genetic variations in the human factor Ⅶ gene and plasma TG level may reflect the potential role of human factor Ⅶ gene as one of the genetic components for cardiovascular risk.

Phylogenetic Analysis of Trichaptum Based on the RFLP of PCR-Amplified DNAs

  • To infer phylogenetic relationships between species of Trichaptum (Polyporaceae), RFLP analyses of PCR-amplified DNAs were accomplished. Regions coding for ITSs of nuclear SSU rRNA genes and for mitochondrial SSU rRNA genes from thirteen strains of four Trichaptum species (T. abietinum, T. biforme, T. fusco-violaceum, and T. laricinum) were amplified and digested with eight restriction enzymes. All the fragmentation patterns were characterized and coded as 0/1 for the absence/presence of fragments. A phylogenetic tree based on the combined data sets was constructed using the Dollo parsimony method. While every two strains of T. abietinum, T. biforme, T. fusco-violaceum, and T. laricinum formed an independent group, the other strains of T. abietimum and T. fusco-violaceum made mixed groupings among compared strains. It is inferred that T. abietinum and T. fusco-violaceum have more variations, possibly geographic or physiological ones, than other species in the genus.

  • To investigate the possibility of horizontal gene transfer between agricultural microorganisms and soil microorganisms in the environment, Bacillus subtilis KB producing iturin and the PGPR recombinant strain Pseudomonas fluorescens MX1 were used as model microorganisms. The soil samples of cucumber or tomato plants cultivated in pots and the greenhouse for a six month period were investigated by PCR, real-time PCR, Southern hybridization, and terminal restriction fragment length polymorphism (T-RFLP) fingerprinting. Our data from Southern blotting and T-RFLP patterns suggest that the model bacteria do not give significant impacts on the other bacteria in the pots and greenhouse during cultivation.

PCR-mediated RFLP to Identify 'Bangpoong', a Crude Drug (유전자 감식에 의한 방풍(防風)의 감별)

  • Bangpoong (防風) is a popular crude drug used to expel wind from the body surface (祛風解表) to remove dampness (勝濕). And to relieve pain (正痛) and spasm (正痙). In China and Japan, roots of Saposhnikovia divaricata (Turcz.) Schischk. Is used as Bangpoong. However, the roots of Peucedanum japonicum Thunb. Or Glehnia littoralis (A. Gray) Fr. Schmidt ex Miquel, being called Sikbangpoong (植防風) and Wonbangpoong (元防風) respectively are used instead of Bangpoong in Korea. The ITS regions of nuclear ribosomal DNA were analyzed to determine original plants and to design a molecular identification method for the crude drugs used as Bangpoong in Korea and China. It is demonstrated that RFLP analysis via PCR has the great potential as a novel tool to test crude drugs for the quality control and standardization.

PAHs로 오염된 광양만 퇴적토의 미생물 군집분석

  • 연안에 가까운 광양만 퇴적토를 대상으로 하여 PAHs 오염도와 말단제한절편 다형성 분석방법(T-RFLP method)을 이용하여 미생물 군집구조를 조사하였다. 조사된 10개 정점의 PAHs오염도는 80$\pm$20~5,690$\pm$580 (평균 1,499$\pm$1,716) ng/g dry wt.였으며 폐수종말처리장이 위치하는 하천 입구에 위치한 정점 (Wl)과 광양제철소 우수배출구 입구에 위치한 정점에서 상대적으로 높은 농도를 보였다. T-RFLP방법으로 조사한 미생물 군집은 10개 정점이 4개의 그룹으로 구분되었으며 정점 Wl에서 다른 정점과 다른 독특한 군집구조를 형성하였다. PAHs 농후배양시료에서의 미생물군집구조 변화와 비교해 볼 때 조사대상지역 퇴적토의 미생물 군집구조는 PAHs의 오염에 부분적으로는 영향을 받지만 입도, 유기물 등과 같은 환경요인이 군집구조를 결정하는 주된 요인인 것으로 생각된다.

  • Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) assay was used to detect and identify four fish viruses, fish iridovirus, viral hemorrhagic septicaemia virus (VHSV), viral nervous necrosis virus (VNNV), hirame rhabdovirus (HRV). Four viruses were detected by PCR with each specific primers. Identification of iridovirus was achieved by digesting the PCR amplified fragment with a restriction enzyme ApaⅠ. It was possible to distinguish positive from false positive PCR amplicons of VHSV by RFLP of PstⅠ or HindⅢ restriction enzymes. VNNV was identified using RFLP of BamHⅠrestriction enzyme and HRV was identified by XbaⅠ restriction enzyme. This approach can be used for more rapid, simple and specific diagnosis of fish viral diseases.

  • Genetic relatedness of medically important Exophiala species such as E. dermatitidis, E. mansonii, and three E. jeanselmei varieties: jeanselmei, lecanii-corni, and heteromorpha was examined using PCR-RFLP(restriction fragment length polymorphism) of ribosomal DNA, M-13, $(GTG)_5$ and nucleotide sequences of ribosomal ITS(internal transcribed space) II regions. Three E. jeanselmei varieties showing distinct band patterns for each DNA markers as well as different nucleotide sequences of ribosomal ITS II regions could be considered as a separate species. E. dermatitidis and E. mansonii demonstrated the identical band patterns of RFLP of ribosomal DNA, M-13, and $(GTG)_5$ markers. However, nucleotides sequences of ribosomal ITS II region were different between these two species.

Genetic Polymorphisms of t-PA and PAI-1 Genes in the Korean Population

  • Abnormalities in fibrinolysis system is associated with risk of hypertension. In this report, the Alu repeat insertion/deletion (I/D) polymorphism of tissue plasminogen activator (t-PA) and the Hind III RFLP of plasminogen activator inhibitor-1 (PAI-1) genes were investigated in 115 normotensives and 83 patients with hypertension, and their association with anthropometrical data and plasma biochemical parameters were analyzed. There were no significant differences in the gene frequencies of the two candidate genes between normotensives and hypertensives, respectively. Our results indicate lack of associations between the two polymorph isms in t-PA and PAI-1 genes and risk of hypertension in the population under study. However, the Hind III RFLP of PAI-1 gene was significantly associated with plasma glucose level, suggesting its role in glucose metabolism. It needs to be tested whether this RFLP of PAI-1 gene is associated with insulin resistance syndrome or non-insulin dependent diabetes mellitus (NIDDM) in the Korean population.
