  • Citri Reticulatae Viride Pericarpium(CRVP) is being used to regulate the flow of Q(氣) However, the mechanism of it's pharmacological actions is not well understood. The purpose of this research was to investigate effects of CRVP contractil response of isolated on abdominal and femoral artery in rabbits and renal artery in pigs. 1. Abdominal artery was relaxed by CRVP in a dose-dependent manner. 2. Femoral artery was relaxed by CRVP in a dose-dependent manner 3. Pretreatment with methylene blue and indomethacin did not inhibited CRVP induced relax in abdominal artery and femoral artery. 4. Renal artery was relaxed by $H_{2}O$ fraction in a dose-dependent manner, 5. Pretreatment with regitine inhibited $H_{2}O$ fraction(CRVP) induced relax in renal artery in a dose-dependent manner. 6. Renal artery was not relaxed by hexane fraction(CRVP) in a dose-dependent manner. 7. Pretreatment with regitine$(10^{-7}M)$ was relaxed by methylene chloride(MC)(CRVP) and $H_{2}O$ fraction in a $0.5{\mu}g/m{\ell}$.

Solitary Cysticercus Granuloma of the Brainstem - Case Report - (뇌간에 발생한 단발성 뇌낭미충증 - 증례보고 -)

  • Isolated brainstem invlovement of neurocysticercosis is rare. We present the clinical and radiological features of a patient with a solitary cysticercus granuloma in the brainstem confirmed by MRI and CSF ELISA test. Good response was obtained from conservative treatment, using praziquantel. We believe that patients from regions where cysticercosis is endemic, who have no systemic disease, who present with nonprogressive symptoms and signs of brainstem involvement, and display a solitary ring-enhancing mass in the brainstem measuring less than 20mm on neuroimaging may be managed conservatively.

  • Using of masonry infill as partitions, in flat slab frame buildings is a common practice in many parts of the world. The infill is, generally, not considered in the design and the buildings are designed as bare frames. More of fundamental information in the effect of masomary infill on the seismic performance of RC building frames is in great demand for structural engineers. Therefore the main aim of this research is to evaluate the seismic performance of such buildings without (bare frame) and with various systems of the masonary infill. For this purpose, thirteen three dimensional models are chosen and analyzed by SAP2000 program. In this study the stress strain relation model proposed by Crisafulli for the hysteric behaviour of masonary subjected to cyclic loading is used. The results show that the nonstructural masonary infill can impart significant increase global strength and stiffness of such building frames and can enhance the seismic behaviour of flat slab frame building to large extent depending on infill wall system. As a result great deal of insight has been obtained on seismic response of such flat slab buildings which enable the structural engineer to determine the optimum position of infill wall between the columns.

  • This study was carried out to investigate the effects of electrical stimulation on the change Tyrosine-Hydroxylase immunoreactive(TH-IR) cells in the Central gray and retrorubral field of the male SD rats. 9 healthy and normal rats were divided into three groups, 3 SD rat in each group. The one group has been stimulated by electroacupuncture(EA, 2Hz) for 30min and the other group by EA for 1hr 30min and control group has been stimulated. TH-IR cells were found in the Central gray(CG) and Retrorubral field(RF). The numbers of TH-IR cells of CG and RF were significantly increased after 30 min (CG $6.2{\pm}0.83$, RF $1.4{\pm}0.55$)as compared with control group(CG $24{\pm}3.16$, RF $6.4{\pm}0.55$) and were also significantly increased after 1hr 30min(CG $75.6{\pm}4.51$, RF $18.8{\pm}0.89$) than 30min. These results show that TH is related into CG and RF in response to electrical stimulation.

  • GroEL is well known as a molecular chaperone. In order to determine the dynamic stress response of the Escherichia coli groE promoter, a groE-lacZ operon fusion in the chromosome was constructed. Stress leading to ${\sigma}^{32}$ synthesis induces transcription from E. coli groE promoter, since the promoter is ${\sigma}^{32}-regulated$. When the strain was stressed with ethanol, phenol, and sodium chloride, clear inductions of ${\beta}-galactosidase$ were observed. Two types of simultaneous stresses of sodium chloride and phenol induced the enze much more than either of the two alone, suggesting that stress was an additive. The combined stress resulted in the highest induction of the enzyme in this system. The groE-lacZ fusion strain developed in this study can conveniently be used to detect other harmful pollutants in the environment. Stress treatment of cells containing recombinant proteins, which need GroEl, by ethanol, phenol, or sodium chloride, might have a tendency to increase their biological activities.

Estimating the Economic Value of Ecotourism in Anmyeondo Tidal Wetlands Using the Contingent Valuation Method (조건부가치측정법을 이용한 안면도갯벌의 생태관광에 대한 경제적 가치추정)

  • The paper is to estimate the economic value of ecotourism in Anmyeondo tidal wetlands using a double-bounded dichotomous choice (DBDC) model of the Contingent Valuation Method (CVM). For the reliability and the validity of CVM a survey was conducted for 511 visitors by personal interview method in Anmyeondo during August 2000, and respondents were asked for maximum Willingness-To-Pay (WTP) to buy a ticket fer the tidal flat's ecotourism. Overall, respondents answered that they would be willing to pay 55,879 won for a ticket under the study. All equations and results estimated are statistically significant at the 1% or 5% level. For the aggregation of WTP estimates, the sample values are extended to the total annual number of visitors in Anmyeondo area during 1998. A conservative estimate, which considers the calibration factor (0.5) recommended by NOAA (1994) and a positive response rate (83.17%), is 73 billion won.

Asymmetrical Distribution of P2Y Nucleotide Receptors in Rabbit Inner Medullary Collecting Duct Cells

  • We cultured the rabbit inner medullary collecting duct (IMCD) cells as monolayers on collagen-coated membrane filters, and investigated distribution of the P2Y receptors by analyzing nucleotide-induced short circuit current $(I_{sc})$ responses. Exposure to different nucleotides of either the apical or basolateral surface of cell monolayers stimulated $I_{sc}.$ Dose-response relationship and cross-desensitization studies suggested that at least 3 distinct P2Y receptors are expressed asymmetrically on the apical and basolateral membranes. A $P2Y_2-like$ receptor, which responds to UTP and ATP, is expressed on both the apical and basolateral membranes. In addition, a uracil nucleotide receptor, which responds to UDP and UTP, but not ATP, is expressed predominantly on the apical membrane. In contrast, a $P2Y_1-like$ receptor, which responds to ADP and 2-methylthio-ATP, is expressed predominantly on the basolateral membrane. These nucleotides stimulated intracellular cAMP production with an asymmetrical profile, which was comparable to that in the stimulation of $I_{sc}.$ Our results suggest that the adenine and uracil nucleotides can interact with different P2Y nucleotide receptors that are expressed asymmetrically on the apical and basolateral membranes of the rabbit IMCD cells, and that both cAMP- and $Ca^{2+}-dependent$ signaling mechanisms underlie the stimulation of $I_{sc}$.

Bile Acid Modulation of Gastroinstinal Smooth Muscle Contraction and Ionic Currents

  • We have examined whether bile acids can affect the electrical and mechanical activities of circular smooth muscle of canine colon and ileum, using isometric tension measurement or patch clamp technique. It was found that a dilution of canine bile $(0.03{\sim}2%\;by\;volume)$ enhanced or inhibited the amplitude of spontaneous contractions. An individual component of bile, deoxycholic acid (DCA) enhanced the frequency and amplitude of the spontaneous contractile activity at $10^{-6}\;M,$ while DCA at $10^{-4}\;M$ inhibited the contraction. Similarly, the response to cholic acid was excitatory at $10^{-5}\;M$ and inhibitory at $3{\times}10^{-4}\;M.$ Taurocholic acid at $10^{-4}\;M$ enhanced the amplitude of muscle contraction. Electrically, canine bile at 1% reversibly depolarized the colonic myocytes under current clamp mode. Bile acids also elicited non-selective cation currents under voltage clamp studies, where $K^+$ currents were blocked and the $Cl^-$ gradient was adjusted so that $E_{Cl}^-$ was equal to -70 mV, a holding potential. The non-selective cation current might explain the depolarization caused by bile acids in intact muscles. Furthermore, the bile acid regulation of electrical and mechanical activities of intestinal smooth muscle may explain some of the pathophysiological conditions accompanying defects in bile reabsorption.

p38 MAPK and $NF-_{\kappa}B$ are Required for LPS-Induced RANTES Production in Immortalized Murine Microglia (BV-2)

  • Using murine immortalized microglial cells (BV-2), we examined the regulation of RANTES production stimulated by lipopolysaccharide (LPS), focusing on the role of mitogen-activated protein kinase (MAPK) and nuclear factor $(NF)-{\kappa}B.$ The result showed that RANTES (regulated upon activation of normal T cell expressed and secreted) was induced at the mRNA and protein levels in a dose- and time-dependent manner in response to LPS. From investigations of second messenger pathways involved in regulating the secretion of RANTES, we found that LPS induced phosphorylation of extracellular signal-regulated kinase (Erk), p38 MAPK and c-Jun-N-terminal kinase (JNK), and activated $(NF)-{\kappa}B.$ To determine whether this MAPK phosphorylation is involved in LPS-stimulated RANTES production, we used specific inhibitors for p38 MAPK and Erk, SB 203580 and PD 98059, respectively. LPS-induced RANTES production was reduced approximately 80% at $25\;{\mu}M$ of SB 203580 treatment. But PD 98059 did not affect RANTES production. Pyrrolidine-dithiocarbamate (PDTC), $(NF)-{\kappa}B$ inhibitor, reduced RANTES secretion. These results suggest that LPS-induced RANTES production in microglial cells (BV-2) is mainly mediated by the coordination of p38 MAPK and $(NF)-{\kappa}B$ cascade.

Systemic Administration of the Potassium Channel Activator in the Polystyrene Latex Bead-Induced Cerebral Vasospasm (Polystyrene Latex Bead에 의한 뇌혈관연축 모델에서 K+ 통로활성제의 전신투여)

  • Objectives : It has been reported that the presence of a pharmacologically inactive foreign substance, polystyrene latex bead, in subarachnoid space activates a non-specific immunological response and elicits arterial narrowing. Recently the activation of potassium($K^+$) channels may be of benefit in relieving cerebral vasospasm. The present study examined the effects of systemic administration of a ATP-sensitive $K^+$ channel activator, cromakalim, on the polystyrene latex bead-induced cerebral vasospasm. Methods : The spasm models similar to that caused by subarachnoid blood injection were created by injection of bead into rabbit cisterna magna. Intravenous injections of cromakalim were administered twice daily(bid) 30 minutes after induction of vasospasm. Animals were killed by perfusion-fixation 2 days after vasospasm. Basilar arteries were removed and sectioned, and the luminal cross-sectional areas were measured. Results : Injection of bead elicited an arterial constriction, reducing arterial diameter to 33.3% of resting tone. Cromakalim inhibited bead-induced constriction at a dose of 0.3mg/kg(Mann-Whitney test, p<0.01). Conclusion : These results support the concept that the cellular events triggered by inactivation of ATP-sensitive $K^+$ channels are responsible for the pathogenesis of vasospasm. The findings also indicate that cromakalim represents a potential therapeutic agents for the treatment of cerebral vasospasm.
