Real-Time RT-PCR for Validation of Reovirus Type 3 Safety During the Manufacture of Mammalian Cell Culture-Derived Biopharmaceuticals (세포배양 유래 생물의약품 생산 공정에서 Reovirus Type 3 안전성 검증을 위한 Real-Time RT-PCR)

  • Validation of viral safety is essential in ensuring the safety of mammalian cell culture-derived biopharmaceuticals, because numerous adventitious viruses have been contaminated during the manufacture of the products. Mammalian cells are highly susceptible to Reovirus type 3 (Reo-3), and there are several reports of Reo-3 contamination during the manufacture of biopharmaceuticals. In order to establish the validation system for the Reo-3 safety, a real-time RT-PCR method was developed for quantitative detection of Reo-3 in cell lines, raw materials, manufacturing processes, and final products as well as Reo-3 clearance validation. Specific primers for amplification of Reo-3 RNA was selected, and Reo-3 RNA was quantified by use of SYBR Green I. The sensitivity of the assay was calculated to be $3.2{\times}10^0\;TCID_{50}/ml$. The real-time RT-PCR method was proven to be reproducible and very specific to Reo-3. The established real-time RT-PCR assay was successfully applied to the validation of Chinese hamster ovary (CHO) cell artificially infected with Reo-3. Reo-3 RNA could be quantified in CHO cell as well as culture supernatant. When the real-time RT-PCR assay was applied to the validation of virus removal during a virus filtration process, the result was similar to that of virus infectivity assay. Therefore, it was concluded that this rapid, specific, sensitive, and robust assay could replace infectivity assay for detection and clearance validation of Reo-3.

  • Viral safety is an important prerequisite for clinical preparations of mammalian cell culture-derived biopharmaceuticals, because numerous adventitious viruses have been contaminated during the manufacturing process. In particular, Chinese hamster ovary (CHO) cells are highly susceptible to several RNA viruses including reovirus (Reo), bovine viral diarrhea virus (BVDV), and bovine parainfluenza virus (BPIV) and there have been reports of such viral contaminations. Therefore, viral detection during the CHO cell process is necessary to ensure the safety of biopharmaceuticals against viruses. In this study, a multiplex reverse transcription (RT)-PCR assay was developed and subsequently evaluated for its effectiveness as a means to simultaneously detect Reo, BVDV, and BPIV during the manufacture of cell culture-derived biopharmaceuticals. Specific primers for Reo, BVDV, and BPIV were selected, and a multiplex RT-PCR was optimized. The sensitivity of the assay for simultaneous amplification of all viral target RNAs was $7.76{\times}10^2\;TCID_{50}/ml$ for Reo, $7.44{\times}10^1\;TCID_{50}/ml$ for BVDV, and $6.75{\times}10^1\;TCID_{50}/ml$ for BPIV. The multiplex RT-PCR was proven to be very specific to Reo, BVDV, and BPIV and was subsequently applied to the validation of CHO cells artificially infected with each virus. It could detect each viral RNA from CHO cells as well as culture supernatants. Therefore, it was concluded that the multiplex RT-PCR assay can be applied to detection of the adventitious viruses during the manufacture of cell culture-derived biopharmaceuticals.

Evaluation of the Availability of Implant placement using Ridge Expansion Osteotomy (REO) (치조능확장골절단술을 이용한 임프란트 식립술의 유용성 평가)

  • Purpose : Ridge expansion osteotomy (REO) has been introduced when it is necessary to expand narrow crestal ridge with simultaneous implant placement. This study has designed to evaluate the clinical availability of REO. Materials and methods : Subject were patients who had visited Seoul National University Bundang Hospital from July. 2003 to December, 2005 for implant placement using REO by one surgeon. Intraoperative and postoperative complication, failure of initial osseointegration and marginal bone resorption were estimated using electronic medical record and periapical radiography. Twenty?three patients, 8 males and 15 females, mean age 51, ranged 18 to 72, were treated for mean 26 months, ranged from 16 months to 46 months. Results : Mean diameter and length of implants placed at upper anterior, were 3.72mm and 13.32mm each other. Guided bone regeneration and ridge splitting were accompanied in this study. Five cases of cortical bone fracture, three cases of crestal bone loss more than 2mm, 2cases of gingival recession, and 2 cases of infection were noted, but there were no implants removed because of disintegration. Success rate of implant was 91.7%, even if survival rate of implant was 100%. In addition, there were no statistical significance between the success rate of REO and bone graft(p>0.05). Conclusion : Based on the results of the present study, it can be concluded that REO technique is reliable for implant placement at atrophic ridge with adequate height compared to bone graft and other osteotomies for ridge expansion, but care should be taken of esthetic problem such as gingival recession because of crestal bone resorption from trauma by osteotome.

Interaction Effect of Temperature and Moisture Content on the Oil Expression of Perilla Seed (들깨종자의 압착착유에 미치는 온도와 수분함량의 상호작용 효과)

  • In order to elucidate the interaction effect between temperature and moisture content on the oil expression of perilla seed, recovery of expressed oil (REO) and volumetric strain of pressed cake (VSPC) of both roasted and unroasted perilla seeds were observed at different temperatures of 30, 40, 50 and $60^{\circ}C$, and different moisture contents of 2.5, 4.5, 6.5 and 8.5% (w.b). And duration of press was 11 min and applied pressure was 50 MPa. At the low temperature REO and VSPC of roasted and unroasted perilla seed increased in high moisture content and at the high temperature those increased in low moisture content. But REO and VSPC at 8.5% moisture content were decreased without relation to temperature. From the analysis of variance between expression factors and REO and VSPC, temperature and moisture contents showed high significance. Also the interaction effect between temperature and moisture content was higher than temperature. In our experimental conditions, the highest interaction effect between expression factors was observed in the range of $2.5{\sim}4.5%$ of moisture content in all temperatures. The maximum REO of unroasted perilla seeds was observed as 84.4% at 2.5% of moisture content and $60^{\circ}C$, and that of roasted one was as 84.3% at 6.5% of moisture content and $30^{\circ}C$.

Rare Metal Contents and Their Implications of Seabed Mineral Resources Explored by Korea (한국이 탐사 중인 해저광물자원의 희유금속 함량과 의미)

  • Seabed mineral resources explored by Korea are categorized into major three types of deposit; manganese nodule, manganese crust and polymetallic sulfides. Pt displays high enrichment factors (400, ore/crust ratios) in manganese nodule. Rare earth oxide content in manganese nodule ranges from 0.037 to 0.302 REO % with mean value of 0.12 REO %. Both of Te and Pt are enriched elements in manganese crust, displaying enrichment factors of 10800 and 150, respectively. Rare earth oxide's contents of manganese crust are slightly higher than manganese nodule's (0.013~0.387 REO %, average = 0.18 REO %). Se and In are outstanding rare metals from seabed polymetallic sulfides, showing enrichment factors of 1300 and 110, respectively. Au (0.8~26.3 g/t) and Ag (0.9~348.0 g/t) are another enriched elements in polymetallic sulfides. The main concern at exploiting seabed mineral resource will be a securing rare metals for high-technology industries and rare metals from subsea mineral deposits will add economic values to commodity candidates such like Co, Ni and Cu.

Effect of Temperature and Pressure on the Oil Expression of Perilla Seed (온도와 압력이 들깨종자의 압착착유에 미치는 영향)

  • In order to elucidate the temperature and pressure effect on the oil expression of perilla seed, recovery of expressed oil (REO) and volumetric strain of both roasted and unroasted perilla seeds were observed at different temperature, pressure and for different periods of press. In this experiment, moisture content of perilla seed was adjusted to 2.5% and temperature used were 30, 40, 50 and $60^{\circ}C$. Pressure applied were 10, 30, 50 and 70 MPa, and periods of press were 5, 7, 9 and 11 min. As temperature and pressure were increased or periods of press was lengthened, REO and volumetric strain of pressed cake were increased. Maximum REO of unroasted perilla seeds were found to be 85.59% and those of roasted perilla seeds be 85.30%, at 70 MPa, $60^{\circ}C$, and for 11 min. Viscosity of expressed oil were exponentially dependent on temperature and REO were increased as viscosity was decreased. From statistical analysis between effects of expression factors and REO and volumetric strain of pressed cake, importance of their effects was decreased in the order of pressure, temperature, $temperature{\times}pressure$ and periods of press. The multiple regression equation between REO(Y) and temperature (T), pressure (P), and periods of press (D) were as follows; $Y=7.95+36.85P+1.12T^2-0.55TP-5.08P^2\;r^2=0.97$ for unroasted perilla seed (p<0.01), $Y=4.50T+39.23P+0.83T^2-1.71P-5.07P^2\;r^2=0.99$ for roasted perilla seed (p<0.01).

  • Ridge expansion osteotomy(REO) technique is a simple and more conservation method to widen a narrow alveolar ridge in the maxilla. This method is superior to drilling method in soft and narrow maxillary alveolar ridge and allows the surgeon to widen the ridge in routine office procedure. Therefore, it is the treatment of choice to implant the maxilla with narrow alveolar ridge. This article presents clinical cases and discusses the advantages, rationale and surgical protocol of REO technique.

  • Ridge expansion osteotomy(REO) procedure is used to widen the narrow ridge in locations that are too thin to permit the use of standard implant drills. The objective of this technique is to maintain, if possible, all of the existing maxillary bone by pushing the bone aside with minimal trauma. The author has used tis procedure on five patients who had narrow ridge in their soft maxillary bone. Fifteen implants were placed in sites needing ridge expansion using REO. The satisfactory results were obtained. The REO procedure is thought to be a safer and more conservative method of widening a narrow ridge. And this procedure is heatless, offers excellent tactile sensitivity, control, visibility, and takes advantage of available bone.

Optimization of Batch Expression of Sesame Oil (참기름의 회분식 착유의 최적화)

  • In order to optimize the batch expression of sesame oil, recovery of expressed oil(REO) from roasted and unroasted sesame seeds were observed at different temperature, pressure, pressing duration and moisture content, and relatinship between REO and effects of expression factors were analysed. REO was high at 2.5~4.5% moisture content, 30~$50^{\circ}C$ and 30~50MPa, and decreased abruptly with increasing moisture content above 4.5%. The optimum temperature, pressure, pressing duration and moisture content were $40.1^{\circ}C$, 54.4MPa, 21.7min and 1.3% for unroasted seeds and $44.4^{\circ}C$, 37.8MPa, 14.4min and 2.52% for roasted seeds, respectively. REO in optimum condition was 84.6% in unroasted seed and 81.7% in roasted seed. From the statistic analysis between effects of expression factors and REO, importance of their effects was decreased in the order of moisture content, pressure, temperature and pressing duration. And also interaction effects were high in $pressure{\times}moisture$ content, $temperature{\times}moisture$ content and temperature pressure. The multiple regression equation between REO(Y) and temperature(T), pressure(P), moisture content(M), and pressing duration(D) were as follows ; $Y=18.20$ $35.66P$$24.52M-4.45P^{2}-1.20TM-4.02PM-6.62M^{2},\;r^{2}=0.89$, for unrosated sesame seed, $Y=117.93$$16.40P-58.61M-2.75P^{2}$$1.79TM-1.65PM$$7.16M^{2},\;r^{2}$$=0.91$ for roasted sesame seed.

Impingement heat transfer within 1 row of circular water jets : Part 1-Effects of nozzle configuration (1열 원형 충돌수분류군에 의한 열전달의 실험적 연구 (제1보, 노즐형상의 영향))

  • Experiments were carried out to obtain the effects of nozzle configuration and jet to jet spacing on the heat transfer characteristics of single line of circular water jets impinging on a constant heat flux plane surface. The nozzle configurations are Cone type, Reverse cone type and Vertical circular type, and the nozzle arrays are single jet(nozzle dia. 8 mm), 1 row of 3 jets and 1 row of 5 jets. Jet velocities ranging from 3m/s to 8m/s were investigated for the nozzle to target plate spacing of 80 mm. For the Cone and Reverse cone type nozzle arrays, the average Nusselt number of 1 row of 5 jets was larger than that of 1 row of 3 jets at Re$_{D}$<45000, but that of 1 row of 3 jets was larger than that of 1 row of 5 jets at $Reo\le45000$. For the Vertical circular type nozzle, however, the average Nusselt number of 1 row of 3 jets was larger than that of 1 row of 5 jets at all jet velocities. In the condition of fixed mass flow rates, the maximum heat transfer augmentation was obtained for 1 row of 5 jets and was over 2 times larger than that of the single jet for all nozzle configurations. The nozzle configurations that produce the maximum average Nusselt number are as follows: For 1 row of 3 jets, the Vertical circular type at $Reo\le45000$ and the Reverse cone type at $Reo\le45000$. But, they are the Reverse cone type at Re$_{D}$<55000 and the Vertical circular type at$Reo\le55000$ for 1 row of 5 jets.
