Crab Water Extrct Ameliorates Reflux Esophagitis in Rats (역류성 식도염에 대한 방해(螃蟹) 열수추출물의 보호효과)

  • Objectives: In this study, we aim to demonstrate an effect of crab water extract (CWE) on reflux esophagitis (RE) using lipopolysaccharide (LPS)-induced Raw 264.7 cell and rat model. Methods: To investigate on LPS-induced Raw 264.7 cell, CWE was co-treated with LPS. CWE suppressed Nitric Oxide (NO) production which increased by LPS treatment. Also, CWE showed no cytotoxicity at the concentrations range from 500 ㎍/㎖ to 2000 ㎍/㎖. Next, to investigate the protective effects of CWE on RE rat model, eighteen rats were divided in to three groups: sham group, reflux esophagitis group, and reflux esophagitis pre-treated with 100 mg/kg CWE 1 h before surgery. RE was induced by a pylorus and forestomach ligation operation and all rats were sacrificed after 4 h 30 min from surgery. Results: In gross examination, the CWE administration attenuated esophageal mucosal injury upon histological evaluation of reflux esophagus of rats. The CWE downregulated the expression levels of proteins related to inflammation, such as COX-2 and TNF-α in the esophagus tissue. In addition, the CWE suppressed the NF-κB and IκB-α activation. Conclusions: Based on these findings, we concluded that CWE could possess protective effect against damage to the esophagus due to reflux esophagitis.

Ginsenoside Re inhibits pacemaker potentials via adenosine triphosphate-sensitive potassium channels and the cyclic guanosine monophosphate/nitric oxide-dependent pathway in cultured interstitial cells of Cajal from mouse small intestine

  • Background: Ginseng belongs to the genus Panax. Its main active ingredients are the ginsenosides. Interstitial cells of Cajal (ICCs) are the pacemaker cells of the gastrointestinal (GI) tract. To understand the effects of ginsenoside Re (GRe) on GI motility, the authors investigated its effects on the pacemaker activity of ICCs of the murine small intestine. Methods: Interstitial cells of Cajal were dissociated from mouse small intestines by enzymatic digestion. The whole-cell patch clamp configuration was used to record pacemaker potentials in cultured ICCs. Changes in cyclic guanosine monophosphate (cGMP) content induced by GRe were investigated. Results: Ginsenoside Re ($20-40{\mu}M$) decreased the amplitude and frequency of ICC pacemaker activity in a concentration-dependent manner. This action was blocked by guanosine 50-[${\beta}-thio$]diphosphate [a guanosine-5'-triphosphate (GTP)-binding protein inhibitor] and by glibenclamide [an adenosine triphosphate (ATP)-sensitive $K^{+}$ channel blocker]. To study the GRe-induced signaling pathway in ICCs, the effects of 1H-[1,2,4]oxadiazolo[4,3-a]quinoxalin-1-one (a guanylate cyclase inhibitor) and RP-8-CPT-cGMPS (a protein kinase G inhibitor) were examined. Both inhibitors blocked the inhibitory effect of GRe on ICC pacemaker activity. L-NG-nitroarginine methyl ester ($100{\mu}M$), which is a nonselective nitric oxide synthase (NOS) inhibitor, blocked the effects of GRe on ICC pacemaker activity and GRe-stimulated cGMP production in ICCs. Conclusion: In cultured murine ICCs, GRe inhibits the pacemaker activity of ICCs via the ATP-sensitive potassium ($K^{+}$) channel and the cGMP/NO-dependent pathway. Ginsenoside Re may be a basis for developing novel spasmolytic agents to prevent or alleviate GI motility dysfunction.

Optimization of microwave-assisted extraction process of Hordeum vulgare L. by response surface methodology (반응표면분석법을 이용한 새싹보리 마이크로웨이브 추출공정의 최적화)

  • This study attempted to find optimum extract range of active ingredient for barley sprouts (Hordeum vulgare L.). Extracts from Hordeum vulgare L. were made by microwave extraction method and total polyphenol content (TPC), total flavonoid content (TFC), DPPH radical scavenging activity (DPPH) were measured with extract of Hordeum vulgare L.. Response surface methodology (RSM) was applied to a extraction process, and central composite design (CCD) was also used for this process to examine the optimum condition. Independent variables ($X_n$) are concentration of ethanol ($X_1$: 0, 25, 50, 75, 100%), microwave power ($X_2$: 60, 120, 180, 240, 300 W), extraction time ($X_3$: 4, 8, 12, 16, 20 min). Dependent variables ($Y_n$) are TPC ($Y_1$), TFC ($Y_2$), DPPH radical scavenging ($Y_3$). It is formed by sixteen conditions to extract. The $R^2$ value of dependent variables is ranged from 0.90 to 0.97 (p<0.05). Experiments values within the optimal range (40% of ethanol concentration, 120 W of microwave power, 18 min of extraction time) were 3.74 mg GAE/g (TPC), 3.00 mg RE/g (TFC), 35.43% (DPPH), respectively. Under the optimized conditions, predicted value showed no significant difference comparing with the experimental values.

Antiviral activity of ginsenosides against coxsackievirus B3, enterovirus 71, and human rhinovirus 3

  • Background: Ginsenosides are the major components responsible for the biochemical and pharmacological actions of ginseng, and have been shown to have various biological activities. In this study, we investigated the antiviral activities of seven ginsenosides [protopanaxatriol (PT) type: Re, Rf, and Rg2; protopanaxadiol (PD) type: Rb1, Rb2, Rc, and Rd)] against coxsackievirus B3 (CVB3), enterovirus 71 (EV71), and human rhinovirus 3 (HRV3). Methods: Assays of antiviral activity and cytotoxicity were evaluated by the sulforhodamine B method using the cytopathic effect (CPE) reduction assay. Results: The antiviral assays demonstrated that, of the seven ginsenosides, the PT-type ginsenosides (Re, Rf, and Rg2) possess significant antiviral activities against CVB3 and HRV3 at a concentration of $100{\mu}g/mL$. Among the PT-type ginsenosides, only ginsenoside Rg2 showed significant anti-EV71 activity with no cytotoxicity to cells at $100{\mu}g/mL$. The PD-type ginsenosides (Rb1, Rb2, Rc, and Rd), by contrast, did not show any significant antiviral activity against CVB3, EV71, and HRV3, and exhibited cytotoxic effects to virus-infected cells. Notably, the antiviral efficacies of PT-type ginsenosides were comparable to those of ribavirin, a commonly used antiviral drug. Conclusion: Collectively, our findings suggest that the ginsenosides Re, Rf, and Rg2 have the potential to be effective in the treatment of CVB3, EV71, and HRV3 infection.

Relation Between the Yield and the Cutting Length of Mulberry Branch in Autumn (추잠기의 중간벌채정도와 수량과의 관계)

  • This work was carried out to know the relation between the yield and the cutting length of mulberry branches in autumn. The results obtained are summarized as fellows, 1. Most of the experimented mulberry trees grew evenly, of which branches showed 171cm to 177cm in average length. 2. No difference could be seen in autumn yield among the treatments, however, the shorter branches were remained, the higher in ratio of re-sprouting bud. 3. The spring yield of cutting mode in 100cm and 200cm remained was as much as non-treatment, but dropped remarkably in below 80cm remained. The high productivity in the 100cm or above renamed mode accounts for the low ratio of no-sprouting bud and accelerating the growth of the shoot, which consequently become much of shoot yield per 1m of branch. By cutting branches, however, the ratio of amount of leaves per shoot became low. 4. The cutting mode in 100cm and 120cm remained can yield as much as non-treatment even in the total yields of spring and autumn. but 100cm mode seems to be more advanceable in autumn owing to a little sparing labor for picking leaves. Thus, the most desirable cutting mode in autumn may be in 100cm remained, which usually has been done at the part of one third or half of the whole branches.

Effect of Rice Fermented using Poria cocos (a Wood-decay Fungus) Mycelium on Fermentation of Doenjang (Soybean Paste) (복령(Poria cocos) 균사체 발효쌀의 첨가가 된장의 품질에 미치는 영향)

  • We investigated changes in doenjang quality upon addition of rice fermented with Poria cocos mycelium. Sensory evaluation showed that rice addition to 15% (w/w) was optimal. The content of free amino acids was 1,899.17 mg% in doenjang fermented for 90 days with P. cocos mycelium. The rice contained seven essential amino acids: leucine, valine, phenylalanine, isoleucine, lysine, threonine, and methionine. The levels of essential amino acids increased after fermentation with P. cocos mycelium. Vitamin A (7.47 RE/100 g), Vitamin D (0.45 mg/100 g), and Vitamin E (5.73 mg/100 g) were detected in the experimental preparation but not in the control. In terms of electron-donating ability, the highest scores were 73.8% (in a water extract) and 76% (in an ethanolic extract) of doenjang fermented with P. cocos mycelium. Nitrite-scavenging ability was higher in the ethanolic extract than in the water extract of rice fermented with P. cocos mycelium.

Purification and Properties of Polygalacturonase from Ganoderma lucidum (Ganoderma lucidum이 생산하는 Polygalacturonase의 정제 및 특성)

  • The properties of polygalacturonase by Ganoderma lucidum in liquid culture were investigated. The enzyme was composed of an endo- and an exo-polygalacturonase. The endo- and exo-polygalacturonase were purified approximately 56 and 9.2-fold, respectively, through ammonium sulfate fractionation, gel filtration on Biogel P-100, anion exchange chromatography on DEAE-cellulose, gel chromatography on Sephadex G-150 and re-gel chromatography on Sephadex G-150. The endo- and exo-polygalacturonase had higher affinity for apple pectin than for citrus pectin or pectic acid. The Km values of the endo- and exo-polygalacturonase for apple pectin, determined on the Lineweaver-Burk plot, were 1.44 and 10.6 mg $ml^{-1}$ for apple pectin, respectively. Purified endo-polygalacturonase was found to be homogeneous electrophoretically and had a molecular weight of 54,000 estimated on SDS polyacrylamide gel. The optimal pH for the activity of the enzymes was 4.0. The endo- and exo-polygalacturonase were stable in the pH range of 4.0 to 6.0 and 3.5 to 5.5, respectively. The optimal temperatures of the endo- and exo-polygalacturonase were 40 and $60^{\circ}C$, respectively. The exo-polygalacturonase was more resistant to heat than the endo-polygalacturonase, requiring heating for 40 min at $80^{\circ}C$ for complete inactivation. The activity of the endo-polygalacturonase was increased by $Ca^{++}$ and $Mn^{++}\;ions$, while that of the exo-polygalacturonase was increased by $Ca^{++}\;ion$ only, and was not affected by $Mn^{++}\;ion$.

Hearing Ability of Bambooleaf wrasse Pseudolabrus japonicus caught in the coast of Jeju (제주 연안에서 어획된 황놀래기의 청각 능력)

  • In order to improve the availability of underwater sound by the fundamental data on the hearing ability, the auditory thresholds for the bambooleaf wrasse pseudolabrus japonicus were determined at 80Hz, 100Hz, 200Hz, 300Hz, 500Hz and 800Hz by heartbeat conditioning method using pure tones coupled with a delayed electric shock. The audible range of the bambooleaf wrasse extended from 80Hz to 800Hz with the best sensitivity around 100Hz and 200Hz. In addition, the auditory thresholds over 300Hz increased rapidly. The mean auditory thresholds of the bambooleaf wrasse at the test frequencies, 80Hz, 100Hz, 200Hz, 300Hz, 500Hz and 800Hz were 100dB, 95.1dB, 94.8dB, 109dB, 121dB and 125dB, respectively. Auditory critical ratios for the bambooleaf wrasse were measured using masking stimuli with the spectrum level range of about 70, 74, 78dB (0dB re $1{\mu}Pa/\sqrt{Hz}$). According to white noise level, the auditory thresholds increased as compared with thresholds in a quiet background noise. The Auditory masking by the white noise spectrum level was stared over about 60dB within 80~300Hz. Critical ratios to be measured at frequencies from 80Hz to 300Hz were minimum 33dB and maximum 39dB.

Shear and Bond Strength of Activated Hwangtoh Concrete Beam (활성 황토 콘크리트 보의 전단 및 부착 강도)

  • As a eco-friendly material, Hwangtoh (red clay) has been studied for complete or partial replacement of portland cement. Most of existing studies focused on the material properties of the Hwangtoh concrete including the compressive strength, drying shringkage, and creep. In the present study, the shear strength of the beams made with the Hwangtoh concrete was tested. Further, bond strength of tension re-bars embedded in the Hwangtoh concrete was tested. One of the concrete tested consisted of activated Hwangtoh replacing 20% of the cement. The other consisted 100% activated. Hwangtoh replacing all the cement. The beam specimens were tested under two point static loading. The test result showed that the shear strength of activated Hwangtoh concrete beams replacing 20% and 100% of cement was equivalent to that of the ordinary portland cement concrete beam. However, the bond strength of activated Hwangtoh concrete replacing 100% of the cement was less than that of the ordinary portland cement concrete.

Percutaneous Sacroplasty : Effectiveness and Long-Term Outcome Predictors

  • Objective : To evaluate the effectiveness and long-term outcome predictors of percutaneous sacroplasty (PSP). Methods : This single-center study assessed 40 patients with sacral insufficiency fractures using the short-axis technique under C-arm flat-panel detector computed tomography (CT). Two radiologists reviewed the patients' magnetic resonance and CT images to obtain imaging findings before PSP and determine technical success, respectively. The short-term outcomes were visual analog scale score changes and opioid usage reductions. Long-term outcomes were determined using telephone interviews and the North American Spine Society (NASS) patient-satisfaction index at least one year after PSP. Results : Technical success was achieved without any significant complications in 39 patients (97.5%). Telephone interviews were possible with 12 patients and failed in 10 patients; death was confirmed in 18 patients. Fifteen patients (50%) re-visited the hospital and received conservative treatment, including spinal injections. Nine patients reported positive satisfaction (NASS patient-satisfaction index 1 or 2), while the negative satisfaction group (NASS patient-satisfaction index 3 or 4, n=3) showed a higher incidence of compression fractures at the thoracolumbar spine level (66.7% vs. 22.2%) and previous spinal injection history (66.7% vs. 33.3%). The poor response group also showed higher incidences of facet joint arthrosis (100% vs. 55.6%), central canal stenosis (100% vs. 22.2%), neural foraminal stenosis (33.3% vs. 22.2%), scoliosis (100% vs. 33.3%), and sagittal malalignment (100% vs. 44.4%). Conclusion : PSP was effective for sacral insufficiency fractures and showed good long-term outcomes. Combined compression fractures in the thoracolumbar spine and degenerative lumbar pathologies could be possible poor outcome predictors.
