Isolation of ginsenosides Rb1, Rb2, Rc Rd, Re, Rf and Rg1 from cinseng root by high performance liquid chromatography

  • Ginsenosides Rb1, Rb2, Rc, Rd, Re, Rf and Rg1 were effectively isolated from ginseng root by preparative liquid chromatography (LC) on two PrepPAK-500/c18 cartridges in series and semipreparative LC on a .mu. Bondapak cabohydrate analysis column, a .mu.Bondapak C18 column or a .mu. Porasil column. The identities of the isolated ginsenosides were confirmed by analytical high-performance liquid chromatography (HPLC) and infrared spectrophotometry. By this method large scale isolation of pure ginsenosides was efficiently accomplished.

Simulation of Pre-burner Performance in Liquid-fueled Rocket Engine for Satellite Launch Vehicle (위성 발사체 액체 로켓 엔진의 Pre-burner 성능 모사)

  • In-house code which simulates the performance of pre-burner in staged-combustion liquid-fueled rocket engine was developed and demonstrated. Chemical reaction of pre-burner was modeled based on analytic algorithm of CEA (Chemical Equilibrium with Applications) and gas dynamics model was incorporated with it. Comparison results between in-house code and CEA have shown small relative errors except at very high or very low O/F ratio. Also the performance curve obtained by in-house code revealed that the calculated values follow the overall trend of real engine (RD-8) performance quite well and at steady-state operation, the deviation became very small.

  • A system design has been performed for a vacuum thrust 88 ton staged combustion cycle rocket engine. Previous research has been used to estimate the performance of the engine components. And the algorithm has been proposed to evaluate the converged engine system performance. The present methodolgy has been verified by comparing the published data for RD-180. The present work adopts the most of the previous KSLV-II engine heritage for both performance improvement and cost competitiveness. The combustion pressure has been decided as 12MPa considering manufacturing difficulty, cost and performance improvement, and as a result the vacuum specific impulse has increased by 23.4s.

Comparative Study on the Efficiency Test Using Remodeled Bubble Generating Pump for Aquarium Fish and Established Air Sampling Pump (관상어용 기포발생기를 개조한 시료채취 장치와 기존의 시료채취기와의 성능 비교)

  • This study is designed to compare the performance of established samplers (personal air sampler and MiniVOL portable air sampler) commonly used in the air environment or work environment with that of the sampler made by remodeling the air bubble generator for aquarium fishes. Sampling method used in this study is the filter collection method for PM10 and total suspended particles (TSP), the liquid collection method for sulfur dioxide ($SO_2$) and nitrogen dioxide ($NO_2$), and the solid collection method for toluene, respectively. There is not a significant difference in the average concentration of TSP between the Gilian personal air sampler (1st, $0.316{\pm}0.095$; 2nd $0.191{\pm}0.090$; 3rd, $0.185{\pm}0.073mg/m^3$) and the remodeled sampler (1st, $0.317{\pm}0.106$, 2nd $0.201{\pm}0.050$; 3rd $0.189{\pm}0.081mg/m^3$). There are also not significant differences in the average concentration of PM10 among the Gilian personal air sampler ($0.058{\pm}0.006mg/m^3$), the remodeled sampler ($0.052{\pm}0.008mg/m^3$) and the MiniVOL portable air sampler ($0.054{\pm}0.007mg/m^3$). The average concentration of the SO2 by the established sampler and the remodeled one is $3.79{\pm}0.21ppb$ and $3.45{\pm}0.15ppb$, respectively. In addition, there are not sigmficant differences in the average concentration of the NO2 between the Gilian personal air sampler (1st, $0.325{\pm}0.068$; 2nd $0.341{\pm}0.206$; 3rd, $2.971{\pm}0.078{\mu}g/m^3$) and the remodeled sampler (1st, $0.300{\pm}0.062$; 2nd $0.332{\pm}0.144$, 3rd, $2.968{\pm}0.085{\mu}g/m^3$). There are not significant differences in the average concentration of toluene between the Gilian personal air sampler (1st, $0.499{\pm}0.072$; 2nd $0.598{\pm}0.112$; 3rd $2.284{\pm}0.077{\mu}g/m^3$) and the remodeled sampler (1st, $0.463{\pm}0.133$; 2nd $0.603{\pm}0.082$; 3rd $2.353{\pm}0.115{\mu}g/m^3$). From these results, we can conclude that the performance of the remodeled sampler is not different from that of established samplers. There is possibility that the remodeled sampler can be used as a alternative device for Gilian personal air sampler in area and personal air sampling.

Simultaneous Analysis Method for Polar and Non-polar Ginsenosides in Cultivated Wild Ginseng by Reversed-phase HPLC-CAD (HPLC-CAD에 의한 산양삼의 극성 및 비극성 ginsenoside 동시 분석)

  • Cultivated wild ginseng is a widely used dietary supplement and medicinal herb. The aim of this study was to optimize the ginseng using high performance liquid chromatography (HPLC)- charged aerosol detection (CAD) for ginsenoside analysis. CAD measures the physical property of an analyte and responds to almost all non-volatile species, independent of their nature, spectral properties, or particle size. It has become widely employed in pharmaceutical analysis. The cultivated wild ginseng extracts were analyzed for compositions of ginsenosides Rb1, Rd, Rg1, Rf, Re, and Rh1. The optimal analysis condition was set up from an experiment using a gradient. Ten grams of cultivated wild ginseng were extracted with 95% EtOH 100 ml for 24 hr at 80℃. The contents of the 6six major ginsenosides in the cultivated wild ginseng extract were Rb1 (5.48±0.12 mg/g), Rd (5.33±0.14 mg/g), Rg1 (12.80± 0.05 mg/g), Rf (19.08±0.68 mg/g), Re (19.87±0.05 mg/g), and Rh1 (16.47±0.16 mg/g), respectively. HPLC showed that the protopanaxatriol group (Rg1, Rf, Re, Rh1) had more content than the protopanaxadiol group (Rb1, Rd) in cultivated wild ginseng extract. In summary, the ginsenosides were identified with HPLC-CAD analysis, and their presence and quantity imply the importance of quality control, as well as the pharmacological activity of the ginseng root.

Effects of Cold Stress on Broiler Performance and Ascites Susceptibility

  • The present study was conducted to determine the effect of cold stress on broiler performance and ascites susceptibility. Male chicks were obtained from a commercial strain of broiler breeders. The trial was divided into two treatments (control and cold stress groups). Ascites was induced in broiler chickens in the trial by exposing the chickens to low temperature (Ta) and by supplying a pelleted diet. The two experimental treatments consisted of: 1) Control group, $33.3^{\circ}C$ the $1^{st}$ wk, $30.2^{\circ}C$ the $2^{nd}$ wk, and $27.5^{\circ}C$ the $3^{rd}$ wk. 2) Cold stress group, $29.0^{\circ}C$ the $1^{st}$ wk, $26.4^{\circ}C$ the $2^{nd}$ wk, and $23.1^{\circ}C$ the $3^{rd}$ wk. From the end of the $3^{rd}$ wk all broilers were reared to 6 wk of age at a constant temperature of $21^{\circ}C$. There was significant difference in live BW during wk 1 to 5. The control group was consistently the heaviest; however, at 6 wk of age, both groups weighed the same. Body weight gain up to 3 wk was significantly decreased by cold stress. During wk 3 and 6 the chicks in the cold stress group had greater BW gain compared with the chicks in the control group. There were significant differences in mortality due to ascites between the groups. During wk 3 and 6 the cold stress group exhibited the most ascites mortality (9.52%) when compared with the control group (1.90%). At 5 wk of age cold stress condition caused significant changes in packed cell volume (PCV), hemoglobin (Hb) and red blood cell counts (RBC). Right ventricle weight was significantly heavier in the cold stress group than the control. There were also significant differences in right ventricle/total ventricle (RV/TV) ratios at 5 wk. the right ventricle/total ventricle ratios in the cold stress group was higher (0.25) than the control group (0.20). It was concluded that, fast growth and cold temperatures are the primary triggers for ascites during commercial broiler production.

Determination of Ginseng Saponins by Reversed-Phase High Performance Liquid Chromatography (역상 고속액체크로마토그라피를 이용한 홍삼 사포닌의 정량)

  • Major saponins in ginseng were analysed using reverse phase high performance liquid chromatography with binary mobile phase gradient control system instead of normal phase column. The optimum condition were as following : reverse phase column; ${\mu}{\beta}ondapak\;C_{18}$ column (Waters, $3.9mm{\times}300\;mm,\;5{\mu}m$), methyl cyanaide/water binary mobile phase gradient control system, solvent flow rate; 1.5 ml/min, and UV($203{\mu}m$ ) detector. The complete separation of ginsenoside $Rb_1,\;Rb_2,\;Rc,\;Rd,\;Re,\;Rf\;and\;Rg_1$ was achieved within 55 min. The Regression coefficients of the calibration curves for seven ginsenosides were 0.99.

  • In this work, we have designed and manufactured radar target simulation equipment for the performance analysis of synthetic aperture radar(SAR) systems. First, we have explained the function and performance specification of the target simulation equipment and point target scenario generation for validation of the SAR system. In addition, we have developed a simple and accurate calibration method for the time delay of the SAR system using the manufactured target simulation equipment. We have analyzed the point target impulse response function of the SAR image acquired using the SAR system and the target simulation equipment. It was observed that the measured peak to side lobe ratio(=-13.25 dB) and resolution(=0.49 m) are in good agreement with the corresponding theoretical values.

A Five-Phase Induction Motor Speed Control System Excluding Effects of 3rd Current Harmonics Component

  • In this paper an effective five-phase induction motor (IM) and its drive methods are proposed. Due to the additional degrees of freedom, the five-phase IM drive presents unique characteristics for enhancing the torque producing capability of the motor. Also the five-phase motor drives possess many other advantages when compared to traditional three-phase motor drives. Some of these advantages include, reducing the amplitude and increasing the frequency of the torque pulsation, reducing the amplitude of the current without increasing the voltage per phase and increasing the reliability. In order to maximize the torque per ampere, the proposed motor has concentrated winding, the produced back electromotive force (EMF) is almost trapezoidal, and the motor is supplied with the combined sinusoidal plus the third harmonic of the currents. For demonstrating the superior performance of the proposed five-phase IM, the motors are also analyzed on the synchronously rotating reference frame. To supply trapezoidal current waveform and to exclude the effect of the $3^{rd}$ harmonic current, a new control stratagem is proposed. The proposed control method is based on direct torque control (DTC) and rotor flux oriented control (RFOC) of the five-phase IM drives. It is able to reduce the acoustical noise, the torque, the flux, the current, and the speed pulsations during the steady state. The DTC transient merits are preserved, while a better quality steady-state performance is produced in the five phase motor drive for a wide speed range. Experimental results clearly demonstrated a more dynamic steady state performance with the proposed control system.

Preparative separation of minor saponins from Panax notoginseng leaves using biotransformation, macroporous resins, and preparative high-performance liquid chromatography

  • Background: Ginsenosides with less sugar moieties may exhibit the better adsorptive capacity and more pharmacological activities. Methods: An efficient method for the separation of four minor saponins, including gypenoside XVII, notoginsenoside Fe, ginsenoside Rd2, and notoginsenoside Fd, from Panax notoginseng leaves (PNL) was established using biotransformation, macroporous resins, and subsequent preparative high-performance liquid chromatography. Results: The dried PNL powder was immersed in the distilled water at $50^{\circ}C$ for 30 min for converting the major saponins, ginsenosides Rb1, Rc, Rb2, and Rb3, to minor saponins, gypenoside XVII, notoginsenoside Fe, ginsenoside Rd2, and notoginsenoside Fd, respectively, by the enzymes present in PNL. The adsorption characteristics of these minor saponins on five types of macroporous resins, D-101, DA-201, DM-301, X-5, and S-8, were evaluated and compared. Among them, D-101 was selected due to the best adsorption and desorption properties. Under the optimized conditions, the fraction containing the four target saponins was separated by D-101 resin. Subsequently, the target minor saponins were individually separated and purified by preparative high-performance liquid chromatography with a reversed-phase column. Conclusion: Our study provides a simple and efficient method for the preparation of these four minor saponins from PNL, which will be potential for industrial applications.
