Study on CsRCI2D and CsRCI2H for improvement of abiotic stress tolerance in Camelina sativa L.

  • Oilseed crop Camelina (Camelina sativa L.) is a suitable for biodiesel production that has high adaptability under low-nutrient condition like marginal land and requires low-input cost for cultivation. Enhanced abiotic stress tolerance of Camelina is very important for oil production under the wide range of different climate. CsRCI2s (Rare Cold Inducible 2) are related proteins in various abiotic stresses that predicted to localized at plasma membrane (PM) and endoplasmic reticulum (ER). These proteins are consist of eight-family that can be divided into tail (CsRCI2D/E/F/G) and no-tail (CsRCI2A/B/E/H) type of C-terminal. However, it is still less understood the function of C-terminal tail. In this study, CsRCI2D/H genes were cloned through gateway cloning system that used pCB302-3 as destination vector. And we used agrobacterium-mediated transformation system for generation of overexpression (OX) transformants. Overexpression of target gene was confirmed using RT-PCR and segregation ratio on selection media. We analyzed physiological response in media and soil under abiotic stresses using CsRCI2D and CsRCI2H overexpression plant. To compare abiotic stresses tolerance, wild type and CsRCI2D/H OX line seeds were sown on agar plate treated with various NaCl and mannitol concentration for 7 days. In the test of growth rate under abiotic stress on media, CsRCI2H OX line showed similar to NaCl and mannitol stress. In the other hand, CsRCI2D OX line showed to be improved stress tolerance that especially increased in 200mM NaCl but was similar on mannitol media. In greenhouse, WT and CsRCI2D/H OX lines for physiological analysis and productivity under abiotic stresses were treated 100, 150, 200mM NaCl. Then it was measured various parameters such as leaf width and length, plant height, total seed weight, flower number, seed number. CsRCI2H OX line in greenhouse did not show any changes in physiological parameters but CsRCI2D OX line was improved both physiological response and productivity under NaCl stress. Among physiological parameters of CsRCI2D OX line under NaCl stress, leaf length and width were observed shorter than WT but it were slightly longer than WT in 200mM NaCl stress. Furthermore, total seed weight of CsRCI2D OX line under stress displayed to decrease than WT in normal condition, but it was gradually raised with increasing NaCl stress then more than WT relatively. These results suggested CsRCI2D might be contribute to improve abiotic stress tolerance. However, function of CsRCI2H is need to more detail study. In conclusion, overexpression of CsRCI2s family can generate various environmental stress tolerance plant and may improve crop productivity for bio-energy production.

Infection Characteristics of Korean Trichinella Isolate to Some Kind of Experimental Animals

  • Present study was performed to investigate the host-parasite relationship of the Korean Trichinella isolate (KTI). In the experiment to observe the infectivity of KTI to several kinds of animals, the reproductive capacity index (RCT) was highest in cats, and that in mice, hamsters and rats was followe4 in descending order. However, birds, i.e. wild goose and chicken, did not infect with KTI. The number of larvae per a gram of muscle (LPG: 377) was highest in the tongue of cats experimentally infected with KTI larvae. LPG in the diaphragm, anterior leg, back, posterior leg and abdominal muscles were 313, 246, 234, 225 and 170 respectively. Muscle larvae recovered at 55 days after infection were revealed the highest infectivity (RCI: 137.2) in mice. RCI was comparatively low in the mice infected with less than 25 day-old and more than 300 day-old larvae. In the experiment to observe the susceptibility of KTI by the mouse strain, ICR (RCI: 137.2), C57BL/6 (RCI: 108.8), DBA/2 (RCI: 107.1), C3H (RCI: 98.7), BALB/c (RCI: 96.9), FVB (RCI: 96.1) and B6C3F1 (RCI: 85.3) were very susceptible. However, BDF1 (RCI: 57.7) and CBA (RCI. 57.1) were revealed the moderate susceptibility, and B6CBAF1 (RCI: 23.1) was shown the lowest. The infection sites of adults were posteriorly transferred in the small intestine of experimental mice according to the infection periods of muscle larvae. The infection characteristics of KTI observed in this study may be useful as the basic data in the advanced studies, furthermore in the study of other Trichinella isolates.

Expression of CsRCI2s by NaCl stress reduces water and sodium ion permeation through CsPIP2;1 in Camelina sativa L.

  • Camelina (Camelina sativa L.) is a potential bio-energy crop that has short life cycle about 90 days and contains high amount of unsaturated fatty acid which is adequate to bio-diesel production. Enhancing environmental stress tolerance is a main issue to increase not only crop productivity but also big mass production. CsRCI2s (Rare Cold Inducible 2) are cold and salt stress related protein that localized at plasma membrane (PM) and assume to be membrane potential regulation factor. These proteins can be divide into C-terminal tail (CsRCI2D/E/F/G) or no-tail group (CsRCI2A/B/C/H). However, function of CsRCI2s are less understood. In this study, physiological responses and functional characterization of CsRCI2s of Camelina under salt stress were analyzed. Full-length CsRCI2s (A/B/E/F) and CsPIP2;1 sequences were confirmed from Camelina genome browser. Physiological investigations were carried out using one- or four-week-old Camelina under NaCl stress with dose and time dependent manner. Transcriptional changes of CsRCI2A/B/E/F and CsPIP2;1 were determined using qRT-PCR in one-week-old Camelina seedlings treated with NaCl. Translational changes of CsRCI2E and CsPIP2;1 were confirmed with western-blot using the antibodies. Water transport activity and membrane potential measurement were observed by cRNA injected Xenopus laevis oocyte. As results, root growth rate and physiological parameters such as stomatal conductance, chlorophyll fluorescence, and electrolyte leakage showed significant inhibition in 100 and 150 mM NaCl. Transcriptional level of CsPIP2;1 did not changed but CsRCI2s were significantly increased by NaCl concentration, however, no-tail type CsRCI2A and CsRCI2B increased earlier than tail type CsRCI2E and CsRCI2F. Translational changes of CsPIP2;1 was constitutively maintained under NaCl stress. But, accumulation of CsRCI2E significantly increased by NaCl stress. CsPIP2;1 and CsRCI2A/B/E/F co-expressed Xenopus laevis oocyte showed decreased water transport activity as 61.84, 60.30, 62.91 and 76.51 % at CsRCI2A, CsRCI2B, CsRCI2E and CsRCI2F co-expression when compare with single expression of CsPIP2;1, respectively. Moreover, oocyte membrane potential was significantly hyperpolarized by co-expression of CsRCI2s. However, higher hyperpolarized level was observed in tail-type CsRCI2E and CsRCI2F than others, especially, CsRCI2E showed highest level. It means transport of $Na^+$ ion into cell is negatively regulated by expression of CsRCI2s, and, function of C-terminal tail is might be related with $Na^+$ ion influx. In conclusion, accumulation of NaCl-induced CsRCI2 proteins are related with $Na^+$ ion exclusion and prevent water loss by CsPIP2;1 under NaCl stress.

Clinical Significance of Immature Reticulocyte Fraction and Reticulocyte Cellular Indices in Pediatric Anemia Patients (망상적혈구 지수 및 미성숙망상적혈구 분획의 소아 빈혈에서의 임상적 의의)

  • Purpose : Flow cytometric automated reticulocyte analysis is a superior method to manual reticulocyte counting, with respect to precision and sensitivity. Furthermore, flow cytometric analysis is able to measure immature reticulocyte fraction(IRF) and reticulocyte cellular indices(RCI : cell hemoglobin content : CHr, mean cell volume : MCVr, cell hemoglobin concentration mean : CHCMr, distribytion width : RDWr, HDWr, CHDWr). In this study, we investigated the mean values and clinical significances of IRF and RCI in healthy children and pediatric anemia patients. Methods : IRF and RCI were measured with an automated blood cell analyzer, ADVIA 120(Bayer, USA) using oxazine 750 dye, in 57 healthy children and 61 children with anemia. The anemia group consisted of 27 iron deficiency anemia(IDA) patients and 34 patients with anemia associated with acute infection(AAI). We compared the mean values of IRF and RCI in the control group classified according to age, between anemia groups and the control group, and between the IDA group and the AAI group. Results : For the normal control group, the mean values of IRF, CHr, MCVr and HDWr were higher in neonates when compared to older children. The mean values of IRF and RDWr were significantly higher, and the mean values of CHr and CHCMr were significantly lower in the IDA group when compared to the control group. The mean value of IRF was significantly higher, and the mean value of CHDWr was significantly lower in the AAI group when compared to the control group. The mean values of IRF, CHr and CHCMr were significantly lower in the IDA group when compared to the AAI group. Conclusion : We could determine the normal mean values of IRF and RCI in healthy children classified according to age for understanding of hematopoietic response differences according to age. The evaluation of IRF and RCI by automated reticulocyte analyzer seemed to be accurate and clinically useful for the early diagnosis of anemia and the differentiation of IDA from AAI.

Establishment of Resilient Infrastructures for the Mitigation of an Urban Water Problem: 2. Robustness Assessment of Structural Alternatives for the Problems of Water Pollution (도시 물 문제 저감을 위한 회복탄력적 사회기반시설 구축: 2. 수질오염 문제 구조적 대안의 내구성 평가)

  • This study evaluated structural alternatives for managing water quality problems by reinterpreting and then applying the robustness-cost index (RCI) for urban flood problems. Cases of endogenous hormone pollution in treated sewage and proliferation of protozoa in intake-water were chosen as representative examples because they have different types of regulation standards for the treatment. Current facilities and structural alternatives with robustness indices (RIs) greater than unity were determined to be robust. The RI was combined with the cost index (CI) to obtain the RCI values. For the endogenous hormone pollution in treated sewage, a human-oriented estrogen $17{\beta}$-estradiol was selected as a target pollutant. The RI and RCI values for a structural alternative, extension of the current sewage treatment facility for advanced treatment, were greater than the values for the current practice of conventional activated sludge process. For the intake-water pollution by protozoa, UV and ozone disinfection facilities were evaluated for inactivation of Cryptosporidium parvum. The RCI values for ozone disinfection were greater than those for UV disinfection. Based on the results and the logics involved in the calculation of RCI for water quality issues we studied, we proposed procedures for establishing and implementing structural alternatives for the restoration from and prevention of outbreaks of water quality problems.

Reid Colposcopic Index Evaluation: Comparison of General and Oncologic Gynecologists

  • The Reid colposcopic index (RCI) helps physicians for interpret the results of colposcopic examination. To compare the accuracy of RCI in colposcopic evaluation between general and oncologic gynecologists, this prospective trial was conducted by invited women over 20 years of age who were scheduled for a colposcopy at Chiang Mai University Hospital between August, 2008 and May, 2014 to participate. Pregnant patients or those having a history of hysterectomy or conization were excluded. During the colposcopy, all patients were simultaneously evaluated by general and oncologic gynecologists utilizing the RCI. Further management with either a biopsy or LEEP in each patient was dependent on the decision of the attending oncologic gynecologist. The accuracy of the RCI in diagnosing HSIL or more was calculated by the comparison with the final histology. Finally, 135 patients were recruited into this study. The sensitivity, specificity, PPV, NPV, and accuracy of RCI in diagnosing HSIL or more in general gynecologists were 45.2%, 80.7%, 41.1%, 83.2% and 72.6% while in the oncologic gynecologists were 51.6%, 85.6%, 51.6%, 85.6% and 77.8%, respectively. The difference in accuracy between evaluator groups was not significant (p-value=0.28). Of 3 patients with invasive cervical cancer, all were undetected by the general gynecologists using RCI while only 1 invasive cervical cancer was missed via RCI by the oncologic gynecologists. We conclude that RCI could be used by general gynecologists in provincial hospitals with major concerns about missing invasive cervical cancer. A short training period regarding colposcopy might help to resolve this problem.

  • Current cities encounter various types of water problems due to rapid urbanization and climate change. The increasing significance of urban water problems calls for the establishment of resilient alternatives to prevent and minimize social loss that results from these phenomena. As a background research for establishing resilient infrastructures for the mitigation of urban water problems, we evaluated the robustness of structural alternatives for urban flood as a representative case. Combining the robustness index (RI) and the cost index (CI), we suggested the robustness-cost index (RCI) as an indicator of the robustness of structural alternatives, and applied the index to assess the existing infrastructures and structural alternatives (i.e., sewer network expansion, additional storage tank construction, and green roof construction) at a site prone to floods located around Gangnam-station, Seoul, Korea. At a rainfall intensity frequency range of 2 to 20 years, the usage of a storage tank and a green roof showed relatively high RCI value, with a variation of an alternative showing greater RCI between the two depending on the size of design rainfall. For a rainfall intensity frequency of 30 years, installing a storage tank with some green roofing was the most resilient alternative based on the RCI value. We proposed strategies for establishing resilient infrastructures for the mitigation of urban floods by evaluating the robustness of existing infrastructures and selecting optimal structural alternatives with the consideration of scales of design disaster.

Quality Characteristics and Antioxidant Activities of Aged Black Chestnut Inner Shells (숙성 흑율피의 품질 특성 및 항산화성)

  • The study was conducted to compare quality characteristics and antioxidant activities of aged black chestnut inner shells (ACI) and raw chestnut inner shells (RCI). RCI were aged for 15 days at high humidity and temperature. As storage time progressed, the pH level decreased while acidity increased. The sugar concentration ($^{\circ}Brix$) and reducing sugar concentration (%) increased during storage, whereas L (lightness) decreased. Compared with RCI, L value of ACI was darker than that of RCI after 9 days of storage. In the Hunter color system, L and a (redness) values of ACI were higher than those of RCI, whereas there was no visible difference in degree of aging. Although total phenol contents increased with degree of aging, phenol contents of ACI increased more than those of RCI after 9 days of aging. $IC_{50}$ values for 1,1-diphenyl-2-picrylhydrazyl and hydroxyl radical scavenging activities decreased with degree of aging. Based on these results, it is suggested that aged black chestnut inner shells were appropriate in terms of both physicochemical and antioxidative activities.
