음곡 상응부위에 시술한 방기 약침이 LPS로 유도된 흰쥐의 염증성 신손상에 미치는 영향 (The Effect of Stephaniae tetrandrae Radix Herbal-acupuncture at KI10 on LPS Induced Nephritis in Rats)

  • Objectives : This study aimed to evaluate the effects of Stephaniae tetrandrae Radix Herbal-acupuncture(SR-HA) at KI10(Umgok) on nephritis induced by lipopolysaccharide(LPS) in rats. Methods : Male SD rats (n=32) were allocated to four groups; normal, LPS, saline, and SR-HA. The rats in the LPS, saline, and SR-HA groups were injected intra-peritoneally with LPS (2 mg/kg) to induce nephritis. The rats in the saline and SR-HA groups were treated with saline injection and SR-HA respectively at KI10 three times a week. To evaluate the effects of SR-HA at KI10 on nephritis in rats, the authors evaluated the numbers of WBC and neutrophils in blood, and the levels of creatinine, TNF-${\alpha}$, CINC-1 in serum, creatinine, total protein in urine and renal MPO. Results : SR-HA at KI10 significantly inhibited the increase of WBC and neutrophils in blood, TNF-${\alpha}$, CINC-1 in serum, creatine, total protein in urine and MPO in kidney of LPS-stimulated rats. Conclusions : SR-HA at KI10 may have therapeutic effects on nephritis in LPS-stimulated rats.

The Effect of Boron Supplementation on Bone Strength in Ovariectomized Rats Fed with Diets Containing Different Calcium Levels

  • The effect of calcium and boron supplementation on bone strength was determined in growing and ovariectomized (OVX) Sparague-Dawley rats. Rats were divided into 9 groups and fed diet with different intake levels of calcium and boron for 4 weeks. About fifty percentages of rats in each group were OVX and the others were sham-operated. The rats were fed same diets after operation for 8 weeks. The feed intake, body weight gain, and FER were significantly higher in OVX rats than those in sham-operated ones. Serum osteocalcin, bone formation biomarker, was significantly increased with increment in calcium and boron intakes. Serum estradiol was lower in OVX rats than in sham-operated ones. Bone mineral density of femur was significantly lower in OVX rats than in other group. The breaking forces of bones were not significantly different among the groups. The urinary excretion of deoxypyridinoline, osteolytic marker was significantly increased with increment in calcium intake and ovariectomy. The urinary calcium excretion was significantly increased with increment in calcium intake, but decreased with increment in boron intake. According to theses results, the boron supplementation resulted in higher serum osteocalcin and lower urinary calcium excretion. Therefore, it could be suggested that the boron supplementation may be complementary and useful to calcium nutrition for bone health.

In vivo Pharmacological Evaluation of Newly Synthesized Nonpeptidic $AT_1$ Receptor Antagonists in Rats

  • This study was conducted to characterize the in vivo pharmacology of KR-30988, KR-30992 and losartan, new AT antagonists, given as i.v. cumulative doses, in two antimal models of high renin, conscious renal artery-ligated hypertensinve rats (RHRs) and nomotensive rats anesthetized with urethane (90 mg/kg, i.p.) and .alpha.-chloralose (90 mg/kg, i.p.), with a special emphasis on the phamacological characterization of the latter model. In conscious RHRs, KR-30988, KR-30992, losartan and captopril caused a dose-dependent decrease in blood pressure, their relative potencise ($ED_{20}$) being 0.057, 0.028, 0164 and 0.018 mg/kg, i.v., repectively. In anesthetized rats, 2 hours after anesthesia, plasma renin activity was increased from 7.31 tp 34.07 ng/ml/h, the level approximately 1.5 times greater than the highest level in RHRs. In anesthtized rats, the $ED_{20}$s for all four compounds were 0.004 mg/kg i.v., respectively. By comparison, $ED_{20}$sfrom anesthetized rats were 4 to 5 times smaller than those from conscious RHRs, with a good correlation (.gamma. = 0.999) noted between thetized rats to the hypotensive activity of the compounds and the same order of potencies intwo models. These results suggest that, in addition to PHRs, the normotensive rats anesthetized as above can serve as a suitable model for the rapid phamacological evaluation of $AT_1$ receptor antagonists.

Changes of CA1 Excitability in Rats after Prenatal Methylazoxymethanol Treatment

  • Experimentally induced cortical disorganization exhibits many anatomical features which are characteristic of cortical malformations in children with early-onset epilepsy. We used an immunocytochemical technique and extracellular field potential recordings from the dorsal hippocampus to determine whether the excitability of the CA1 pyramidal cells was enhanced in rats with exnerimentallv induced hippocampal dysplasia. Compared with control rats, the MAM-treated rats displayed a decrease of paired pulse inhibition. When $GABA_A$ receptor antagonists were blocked with $10{\mu}M$ bicuculline the amplitude of the second population spike of the MAM-treated of rats was similar to that of the first population spike, as was in the control rats. The MAM-treated rats had fewer somatostatin and parvalbumin-immunoreactive neurons than the control rats. These results suggest that the enhanced neuronal responsiveness of the in vivo recording of the CA1 in this animal model may involve a reduction of CA1 inhibition.

Effects of Eurycoma longifolia Jack in Maintaining Mating Behavior of Sexually Experienced Castrated Male Rats

  • The effects of Eurycoma longifolia Jack were studied in maintaining mating behavior of sexually experienced castrated male rats after dosing them with 500 mg/kg daily of E. longifolia Jack for 10 days prior to test and later continued for two weeks where the rats were then castrated. The similar dosage was then continued for 12 weeks post-castration. However, $400\;{\mu}g/day$ of testosterone was administered subcutaneously on the day of castration and lasted for 6 weeks post-castration but later raised to $800\;{\mu}g/day$ until 12 week post-castration. Tests were conducted weekly from 2-6 weeks and 8-12 weeks post-castration. Results showed that all the experimental male rats exhibited mating behavior before castration. Further results also indicated that E. longifolia Jack successfully maintained mating behavior but less than precastration level from 2-6 weeks and later increased from 8-12 weeks post-castration. Similarly, $400\;{\mu}g/day$ of testosterone was effective in maintaining mating behavior from 2-6 weeks post-castration. However, $800\;{\mu}g/day$ of testosterone managed to return the male rats to the precastration level with all male rats exhibited mating behavior from 8-12 weeks post-castration. Further results also indicated that testosterone significantly increased the penis weights (p < 0.05) as compared to the E. longifolia Jack. In conclusion, this study shows that E. longifolia Jack continued to maintain mating behavior of sexually experienced castrated male rats, giving further evidence of the folkuse of this plant as aphrodisiac.

황련뿌리 추출물성분이 흰쥐의 지질대사에 미치는 효과 (Effects of Extracts of Coptis japonica on Lipid Metabolism in Rats)

  • Coptis rhizoma was extracted with methanol or hot water. Methanol extracts had higher berberine content than water extracts, whereas the contents of Ca, Mg, Zn and K were higher in the water extracts. Rats were fed on the purified diets rich in 0.12% cholesterol only or supplemented with 0.5% of each extracts for four weeks. The concentration of total serum cholesterol was significantly lower in the rats given the extracts, irrespective of the extractants. Although the concentration of serum HDL-cholesterol was comparable among the groups, the ratio of HDL-/total-cholesterol was higher in rats given the extracts. The concentration of serum free fatty acids and ketone body were significantly elevated in rats given the extracts. On the other hand, the extracts supplements significantly decreased the contents of liver cholesterol and triacylglycerol. The amount of fecal bile acids was markedly higher in rats given the extracts, especially in those given water extracts. The results show an effective hypocholesterolemic action of Coptis rhizoma and it seems that the lower concentration of serum and liver cholesterol is resulted from the increased fecal bile acids in rats.

국내 야생 집쥐(Rattus norvigicus)의 유행성 출혈열바이러스 감염증의 역학조사 (Epidemiological survey on Hantaan-related virus infection of wild rats(Rattus norvigicus) in Korea)

  • Sixty eight wild rats were caught from Seoul, Kyonggi, Kangwon, Cholla, and Kyongsang provinces in Korea. All of them were confirmed in the same species, Rattus norvigicus. All of sera from wild rats were examined by indirect immunofluorescent test against Hantaan-related virus. Detection of anti-body to Hantaan-related virus were 4 out of 68 rats(5.9%), including 2 of 8(25.0%) rats in Kangwon and 2 of 20 rats(10.0%) in Kyongsang province. No difference of the sero-positive rates between sex of rats tested to Hantaan-related virus was recognized. According to age, four rats with antibodies(1:320~640) to Hantaan-related virus belonged one subadult, one young adult, one middle aged adult, and one old adult, respectively.

Influence of Long-term Supplementation with Korean Red Ginseng on in vivo Antioxidant Capacities in Rats

  • Effects of ginseng on in vivo antioxidant capacities with age were studied in rats. All rats were reared in the conventional system. Ginseng-treated rats were supplied with ginseng water extracts (25 mg/kg/day) continuously from 6 weeks of age to spontaneous death. None of the rats showed any discernible adverse effects of treatment with ginseng-containing water. There was no significant difference in body weight (BW) gains with age between treated and control groups. However, ginseng extracts did cause a decrease in the level of serum low density lipoprotein (LDL)-cholesterol, glucose, and thiobarbituric acid reactive substances (TBARS) in the treated rats. The activities of superoxide dismutase (SOD), catalase, and glutathione peroxidase in liver cytosol decreased with age in the control group. However, these enzyme activities were well maintained in the ginseng-treated rats and, especially, catalase and glutathione peroxidase activities were consistently higher than in control rats. The levels of total sulfhydryl group (T-SH) and glutathione reductase (GR) were unchanged, and glutathione-s-transferase (GST) activity gradually decreased with age in both groups. There were no differences in T-SH, GR, or GST between the control and treatment groups. These results indicate that long-term administration of ginseng retards age-related deterioration in some biochemical parameters such as cholesterol, glucose, and lactate dehydrogenase in serum and it has an enhancing effect on antioxidant capacity in the liver.

Study of moxibustion stimulation in the ovariectomized rat model

  • In order to examine the effectiveness of moxibustion stimulation for climacteric disturbance, we administered moxibustion stimulation to ovariectomized (OVX) rats, and compared the tail surface temperature, laboratory parameters, and the level of locomotor activity with those of untreated rats. Twenty-four female Wistar rats (8 weeks old, 160 - 180 g) were divided into three groups. The rats in the OVX-M group underwent moxibustion stimulation after ovariectomy. The rats in the OVX-C group underwent ovariectomy but did not receive moxibustion stimulation. The rats in the Normal group received neither ovariectomy nor moxibustion stimulation. The level of locomotor activity was determined by a metabolism measuring system. The tail surface temperature was significantly lower in the OVX rats before moxibustion stimulation than in the Normal group. In the OVX rats before moxibustion stimulation, there was no clear difference in the level of locomotor activity between the active and resting phases, and the pattern of locomotor activity was irregular. After moxibustion stimulation, the tail surface temperature of the OVX-M group did not significantly differ from that of the Normal group, and the pattern of locomotor activity of the OVX-M group became diphasic with clear active and resting phases, similar to that observed in the Normal group. These results demonstrate that moxibustion stimulation is effective for the treatment of climacteric disturbance.

인삼사포닌이 정상및 Alloxan투여 흰쥐에서 수종의 대사물질및 효소활성에 미치는 영향 (Effects of Ginseng Saponin on the Metabolites and Enzymes of Normal and Alloxan-intoxicated Rats)

  • In order to evaluate the mode of action of ginseng saponin. metabolic changes in the plasma of normal and aloxan-intoxicated rats were compared Normal groups were administered only ginseng saponin 0, 5, 50mg/kg for 16 days, but alloxan-intoxicated groups were administered alloxan 25mg/kg for 3 days in addition to ginseng saponin. (1) No significant change in the concentration of glucose, cholesterol, and triglyceride (TG) was observed in normal rats but great inhibitory effect, except elevation of TG, was observed in alloxan-intoxicated rats. However, blood urea nitrogen was elevated in both normal and alloxan- intoxicated rats by administration of ginseng saponin 50mg/kg, and it was considered to be due to the impaired kidney function caused by overdose toxicity. (2) In normal rats, COT and ALP actIn,its were not changed by administration of ginseng saponin but GPT activity was decreased significantly. In alloxan-intoxicated rats, ginseng saponin exerted inhibitory action on the elevation of COT, CPT and ALP activity. But administration of ginseng saponin Smghg was much more effective than administration of 50mg/kg. (3) There fore, we concluded that ginseng saponin has the adaptogenic activity showing little effect on normal metabolism but great preventive action on alloxan-intoxicated rats.
