Screening of High Temperature-Tolerant Oleaginous Diatoms

  • Screening suitable strains with high temperature adaptability is of great importance for reducing the cost of temperature control in microalgae cultivation, especially in summer. To obtain high temperature-tolerant diatoms, water samples were collected in summer from 7 different regions of China across the Northeast, North and East. A total of 731 water samples was collected and from them 131 diatom strains were isolated and identified based on the 18S rRNA sequences. Forty-nine strains out of the 131 diatoms could survive at 30℃, and 6 strains with relatively high biomass and lipid content at high temperature were selected and were found to be able to grow at 35℃. Cyclotella sp. HB162 had the highest dry biomass of 0.46 g/l and relatively high triacylglycerol (TAG) content of 237.4 mg/g dry biomass. The highest TAG content of 246.4 mg/g dry biomass was obtained in Fistulifera sp. HB236, while Nitzschia palea HB170 had high dry biomass (0.33 g/l) but relatively low TAG content (105.9 mg/g dry biomass). N. palea HB170 and Fistulifera sp. HB236 presented relatively stable growth rates and lipid yields under fluctuating temperatures ranging from 28 to 35℃, while Cyclotella HB162 maintained high lipid yield at temperatures below 25℃. The percentage of saturated fatty acids and monounsaturated fatty acids in all the 6 strains was 84-91% in total lipids and 90-94% in TAGs, which makes them the ideal feedstock for biodiesel.

  • This is a convergence study to investigate the factors affecting the job stress of new nurses. The subjects of this study were 170 nurses and collected data by self-reported questionnaire. Data were analyzed by descriptive statistics, indepentent t-test, ANOVA, correlation and stepwise multiple regression analysis. The results of this research were the explanatory power was 57.2% with emotional labor, interpersonal relationship and turnover intention on job stress. As a result, it was confirmed that strengthening emotional labor management ability, interpersonal relationship ability and turnover intention was important to reduce job stress. It is necessary to study repeatedly the predictors of job stress through objective measurement.

  • The crystal and molecular structure of thiosinamine, $H_2NCSNHCH_2CHCH_2$, has been determined by X-ray diffraction method. The crystals are monoclinic, space group $P2_1/a$ with four molecules in a unit cell of dimensions, a = 9.819(3), b = 8.553(3), c = 9.170(2)${\AA}$, ${\beta}$ = 127.3(1)$^{\circ}$, and z = 4. Intensity data for 814 reflections were collected with a Rigaku-Denki automatic four circle diffractometer. The structure was solved by direct and Fourier methods. Refinements were carried out by full matrix least-squares method to a final R value of 0.046. The thiourea unit is planar, and the bond lengths and angles in that unit agree well with those in the compounds which contain a thiourea moiety. The molecules are linked together by the two patterns of N-H${\cdots}$S hydrogen bonds along the b-axis.

  • College of Pharmacy, Ewha womans University, Seoul, 120-750, Korea 2,3,7,8-Tetrachlorodibenzo-p-dioxin (TCDD) is the most potent halogenated aromatic hydrocarbon congener that induces expression of several genes including CYP1A1. Exposure to TCDD results in many toxic actions such as carcinogenesis, hepatotoxicity, immune suppression, and reproductive and developmental toxicity. Dramatic differences in dioxin toxicity have been observed between the sexes of some animal species, suggesting hormonal modulation of dioxin action. Many studies have been reported and propose several mechanisms of anti-estrogenic effects of TCDD. In contrast, the effect of estrogen on the regulation of CYP1A1 are not clear at present. There are several reports showing conflicting results. It seems that induction/inhibition of CYP1A1 may be dependent on cell-type and concentration. The purpose of this study was to investigate the regulation of TCDD-induced CYP1A1 gene expression by estradiol and its metabolites. We examined whether estradiol and its metabolites altered TCDD-mediated induction of CYP1A1 enzyme activity. 17 ${\beta}$ estradiol and 16 ${\alpha}$ estriol at non cytotoxic concentrations caused a significant concentration dependent decline of TCDD-induced EROD activity To determine whether reduced EROD activity reflected altered CYP1A1 mRNA expression, we measured CYP1A1 mRNA level by RT-PCR. And to examine whether estradiol and its metabolites have effects on TCDD-induced CYP1A1 gene expression at the transcription level, we also peformed transient transfection with an AhR responsive reporter plasmid containing the 5' flanking region of the human CYP1A1 gene to examine whether estradiol and its metabolites have effects on TCDD-induced CYP1A1 gene expression at the transcription level.

Current and voltage loading tests off resistive SFCL

  • We have performed the current and voltage loading tests of resistive superconducting fault current limiters (SFCLS) based on $YBa_2$$Cu_3$$O_{7}$(YBCO) films with the diameter of 2 inch. The SFCL consists of meander-type YBCO stripes covered with 200 nm Au layer grown in situ for current shunt and heat dispersion at hot spots. The minimum quench current of an SFCL unit was about 25 Apeak. Seven SFCL units were connected in parallel fur the current load ing tests at power source of 100 $V_{rms}$ $/2,000A_{rms}$. This SFCL units had maximum limiting current of 170 Apeak during the fault instant and then successfully controlled the fault current below 100 Apeak within 1~2 msec after short circuit. Increased short current also reduced the quench completion time with little change of current limiting characterization. We connected six SFCL units in series fur the voltage loading tests at power source of $1,200 V_{rms}$/170 $A_{rms}$ at this time. The shunt resistors were inserted into each SFCL unit to eliminate power imbalance originated from serial connection of SFCL units. Each SFCL unit was quenched simultaneously during the fault condition. The current increased up to 40 $A_{peak}$ and decreased to 14 $A_{peak}$ after 3 cycles. Quench was completed within 1 msec after the fault. We confirmed operating characteristics of 140 kVA($120 A_{rms}$ $\times$ 1,200 $V_{rms}$) SFCL and presented the manufacturing possibility of 3.3 kV SFCL using 4 inch YBCO films.BCO films.lms.

Sulfonylurea therapy in a patient with insulin treated neonatal diabetes due to mutation in Kir6.2 (Kir6.2 유전자변이에 의해 발생한 신생아 당뇨병 1례)

  • Permanent neonatal diabetes(PND) is a rare form of diabetes characterized by insulin-requiring hyperglycemia that is diagnosed within the first 3 months of life. In most cases, the causes are not known. Recently, mutations in the gene KCNJ11 encoding the Kir6.2 subunit of the ATP-sensitive K+ charmel have been described in patients with PND. We report a child with PND due to a lysine-to-arginine substitution at position 170(K170R) of gene encoding Kir6.2 Our patient was diagnosed at 7 weeks of age and had been treated with subcutaneous insulin for 6.5 years. Recently, our patient has been changed from subcutaneous insulin to oral glibenclamide therapy at a daily dose of 7.5 mg 3 times a day(0.9 mg/kg/day) at the age of 6.5 years. Before glibenclamide therapy, c-peptide level was 0.1 ng/ml(normal 1.0-3.5 ng/ml) and hemoglobin HbA1c level was 7.8%(normal <6%). After 6 days of treatment, her c-peptide and insulin levels were 2.3 ng/ml and $9.6{\mu}U/ml$(normal $5-25{\mu}U/ml$), respectively. After 1 month later, the insulin and c-peptide levels were in the nonnal range without any episodes of hyper- or hypoglycemia. This case demonstrated that oral sulfonylurea may be the treatment of choice in PND patients with KCNJ11 mutation even at a young age.

Preparation and C-V characteristics of $Y_2O_3-StabilzedZrO_2$ Thin Films by PE MO CVD (플라즈마 화학 증착법에 의한 $Y_2O_3-StabilzedZrO_2$박막의 제조와 Capacitance-Voltage특성)

  • Yttria-stabilized zirconia(YSZ) films were prepared onto p-type (100) silicon wafer by a plasma-enhanced metallorganic chemical vapor deposition(PE MO CVD) processing involving the application of vapor mixture of tri(2.2.6.6-tetramethyl-3, 5-heptanate) yttrium$[Y(DPM)_3]$, zirconiumtriflouracethyla cetonate$(Zr(tfacac)_4$ and oxygen gas. The x-ray diffraction(XRD) and fourier transform infrared spectra(FT1R) results showed that the deposited YSZ films had a single cubic phase. $Y_2O_3$ content of YSZ film was analyzed by PIXE(partic1e induced x-ray emission). The experimental results by PIXE revealed that 12.lmol%, 20.4mol% and 31.6mol% $Y_2O_3$ could be obtained as the $Y(DPM)_3$ bubbling temperature varied at $160^{\circ}C, 165^{\circ}C$ and $170^{\circ}C$ respectively. The increase of $Y(DPM)_3$ bubbling temperature caused shifting flat band voltage to have a negative value.

  • Compression set (CS) and weight loss by thermal degradation of the ACM rubber composite sample prepared for an automotive part were measured simultaneously at several given temperatures of $150^{\circ}C$, $160^{\circ}C$, $170^{\circ}C$, and $180^{\circ}C$. Using the relationship between them, thermal life of the sample could be predicted at a given operating temperature by applying Toops method which is based on the analysis of non-isothermal TGA thermograms. Conversion by weight loss showed a linear relationship with CS changes, exhibiting 4.2% at CS 40%. Activation energy of thermal degradation was calculated as 120.2 kJ/mol at 4.2% of weight loss from Flynn-Wall-Ozawa analysis. When the expected life was set as time to reach CS 40% at $120^{\circ}C$, the life time of the sample was calculated as 9,700 hrs when Toops method was applied.

Survey on the Wearing and Fit of Dress Shirts for Middle-Aged Men -Focused on Difference according to Stature Group- (중년 남성의 드레스셔츠 착용 실태와 맞음새 조사 - 키 집단에 따른 차이를 중심으로 -)

  • The purpose of this study was to investigate the wearing & fit of ready-made and online custom dress shirts by surveying middle-aged men aged 40-54 years in order to collect information necessary for making good fit dress shirts. Also, we analyzed whether there were significant differences in the actual wearing of dress shirts according to the stature of middle-aged men. The stature was classified according to KS K 0050 as $R(157{\leq}x<170)$ and $T(170{\leq}x<182)$. The 23 questionnaire items were divided into three categories: 'General purchasing behavior and wearing', 'Fit satisfaction of ready-made dress shirts' and 'Wearing and fit satisfaction of online custom dress shirts'. Through the results of the study, we could understand the actual wearing condition of ready-made and online custom dress shirts. Also, differences in stature groups were found. The significant differences according to the stature groups were found in the interest in fashion, the purchase criteria, the limitation of the shirt size selection due to stature and body shape, shirt size satisfaction, repair experience, dissatisfaction with purchase and the experience of purchasing online custom dress shirts. Suggestions should be made to improve dissatisfaction with stature groups in the ready-made dress shirts. In addition, the study confirmed the consumer demand for the online custom dress shirts. In order to activate the online custom market, the online custom dress shirts company needs to reduce consumer dissatisfaction and improve size suitability.

  • Mechanical surface treatment is an excellent approach widely used to modulate and improve the performance and service life of bearings, gears, and frictional joints. The main purpose of this study is to investigate and compare the effect of ultrasonic nanocrystal surface modification (UNSM) and wonder process craft (WPC) on the surface and tribological properties of SUJ2 bearing steel. The surface roughness and hardness of the untreated and treated (UNSM- and WPC-treated) specimens were measured and compared. Their tribological properties were evaluated using a micro-tribometer under grease-lubricated and dry conditions against itself. Surface hardness measurement results revealed that both the UNSM- and WPC-treated specimens had a higher hardness than that of the untreated specimen. The surface roughness of the untreated specimen was reduced after UNSM and WPC treatments. Abrasive wear mode was observed on the surface of the specimens worn under grease-lubricated conditions, while adhesive wear mode was found on the surface of the specimens worn in dry conditions. According to the tribological test results, the friction coefficient and wear rate of the untreated specimens were reduced by the application of both the UNSM and WPC treatments under grease-lubricated and dry conditions.
