채소류 모잘록병균에 길항하는 Bacillus ehimensis YJ-37의 대량배양 최적조건 (Optimization of Large Scale Culture Conditions of Bacillus ehimensis YJ-37 Antagonistic to Vegetables Damping-off Fungi)

  • 본 연구는 채소류 모잘록병균인 R. solani AG4에 길항하는 미생물 B. ehimensis YJ-37의 500$m\ell$ 플라스크 배양, 5 $\ell$ jar fermentor 배양, 2,000 $\ell$ 대형탱크의 배양 조건 및 길항물질 생성능을 조사하였다. 500$m\ell$ 플라스크 배양에서의 길항물질 최적생산조건은 1.5% starch, 0.6% peptone, 0.3% $Na_2$HP $O_4$, 0.15% MgC1$_2$, pH 8.0, 배양온도 32$^{\circ}C$, 배양시간 54시간으로 나타났고, 이때 미생물의 수는 1.42$\times$$10^{ 8}$ cfu/$m\ell$21g-DCW/$\ell$), 항진균 활성은 13.9mm로 나타났다. 5$\ell$ jar fermenter 배양에서의 길항물질 최적생산조건은 플라스크배양에서의 최적 생산조건에서 공기주입량과 교반속도를 변화시켜 조사한 결과, 공기주입량 2 vvm, 교반속도 200 rpm, 배양시간 48시간 이후 미생물의 수와 균체의 량은 2.06 $\times$ $10^{8}$ cfu/$m\ell$ 및 30g-DCW/$\ell$, 항진균 활성은 13.4 mm로 나타났다. 2,000 $\ell$ 대형탱크 배양에서는 교반속도 200 rpm, 산소주입량 30 vvm으로 고정하고 10일간 배양하여 미생물의 생육 및 길항물질 생성능을 조사한 결과는 미생물의 수와 균체의 량은 0.81 $\times$ $10^{8}$ cfu/$m\ell$와 12g-DCW/$\ell$, 항진균 활성은 8.6mm로 나타나 5$\ell$ jar fermenter 배양에 배해 균의 수는 1/10로 줄었으며 항진균 활성도 64%로 낮게 나타났다.다.

  • 오이검은별무늬병균인 Cladosporium cucumerinum KACC 40576에 대하여 높은 항진균 활성을 나타내는 KMU-13은 Norway Lillehammer 근교 토양시료에서 분리하였으며, 형태학적, 배양 및 생화학적인 특성을 조사한 결과와 16S rDNA 염기서열을 분석한 결과, B. subtilis로 판단되었다. B. subtilis KMU-13의 항진균 물질의 생산을 위한 최적조건은 LB broth를 기본배지로 하여 탄소원으로 0.5% maltose와 질소원으로 0.5% bactopeptone을 첨가하였을 때 가장 높은 항진균 활성을 나타내었으며, 배양조건은 $30^{\circ}C$, 180rpm, 48시간이였고, 배양초기 pH는 6.0으로 조사되었다. TLC에서 Rf값이 0.64로 확인된 밴드에서만 항진균 활성을 가지는 것으로 조사되었으며 잿빛곰팡이병, 고추 탄저병, 참외 탄저병, 수박 등의 박과작물의 덩굴마름병, 토마토 시들음병, 글라디올러스 마른썩음병, 토마토 질병, 수박 덩굴쪼김병, 수박 질병, 사과나무 역병, 벼 잎집무늬마름병, 참외 줄기썩음병, 그리고 고추 균핵병에 대하여 양호한 항진균 활성을 나타내는 것으로 조사되었다.

Plant Growth-Promoting Potential of Endophytic Bacteria Isolated from Roots of Coastal Sand Dune Plants

  • Endophytic bacteria associated with the roots of coastal sand dune plants were isolated, taxonomically characterized, and tested for their plant growth-promoting activities. Ninety-one endophytic bacterial isolates were collected and assigned to 17 different genera of 6 major bacterial phyla based on partial 16S rDNA sequence analyses. Gammaproteobacteria represented the majority of the isolates (65.9%), and members of Pseudomonas constituted 49.5% of the total isolates. When testing for antagonism towards plant pathogenic fungi, 25 strains were antagonistic towards Rhizoctonia solani, 57 strains were antagonistic towards Pythium ultimum, 53 strains were antagonistic towards Fusarium oxysporum, and 41 strains were antagonistic towards Botrytis cinerea. Seven strains were shown to produce indole acetic acid (IAA), 33 to produce siderophores, 23 to produce protease, 37 to produce pectinase, and 38 to produce chitinase. The broadest spectra of activities were observed among the Pseudomonas strains, indicating outstanding plant growth-promoting potential. The isolates from C. kobomugi and M. sibirica also exhibited good plant growth-promoting potential. The correlations among individual plant growth-promoting activities were examined using phi coefficients, and the resulting data indicated that the production of protease, pectinase, chitinase, and siderophores was highly related.

Ophiostomatoid Fungi in Pine Wilt Disease and Oak Wilt Disease in Korea

  • Pinewood nematode (PWN, Bursaphelenchus xylophilus) is a serious pathogenic worm that quickly dry pine trees to death. Recently, PWN has been devastating huge amounts of conifer trees in Korea. As a first step to explore the association and ecological roles of fungi in PWN life cycle in Korea, in this study we first isolated and indentified fungi from PWN-infested Korean pine and Japanese black pine wood sampled in Jinju, Sacheon, Pocheon, Chuncheon, Gwangju, and Hoengseong in Korea. A total of 144 fungal isolates were obtained from Japanese black pine wood and 264 fungal isolates from Korean pine wood. Their morphology and nucleotide sequences of the ITS rDNA and ♌-tubulin gene were examined for species identification. Ophiostoma ips, Botrytis anthophila, Penicillium sp., Hypocrea lixii, Trichoderma atroviride, O. galeiforme, Fusarium proliferatum were identified from Japanese black pine wood. Leptographium koreanum, L. pini-densiflorae, Ophiostoma ips, Penicillium raistrick, Trichoderma sp. were isolated from Korean pine wood. O. ips and L. koreanum were the major species on the two different PWN-infected pine tree. The cultivation of PWN on fungal mat of the identified species did some enhance PWN reproduction. The ambrosia beetle, Platypus koryoensis, is a serious pest of oak trees in Korea. In this study we investigated filamentous fungi present in the body of the beetle. Fourteen genera of filamentous fungi belonging to Ascomycota and Basidiomycota were isolated. All the obtained genera were isolated in the mitosporic state. The identified fungi were classified in 11 distinct orders including the Ascomycota (Eurotiales, Hypocreales, Microascales, Ophiostomatales, Pleosporales, and Sordiales) and Basidiomycota (Agaricales, Corticiales, Polyporales, and Russulales Xylariales). Within Ascomycota, 13 species were found. Meanwhile five species were found within Basidiomycota. The results showed the presence of diverse fungi in P. koryoensis. Among the isolated fungi, some were able to produce wood degrading enzymes. Further fungal isolation was performed with P. koryoensis infested Quercus mongolica trees sampled at Kumdan mountain in Hanam-Si, Gyeonggi province from June of 2009 to June of 2010. Penicillin spp. and Trichoderma spp. were the major species of mold fungi group. Pichia guilliermondii was the major species of mold yeast group. Raffaelea quercus-mongolicae was also isolated, but its isolation frequency was not high. Other species identified were Ambrosiella xylebori, Fusarium solani, Cryphonectria nitschke, Chaetomium globosum, and Gliocladium viride, Candida kashinagacola, C. maritima, C. vanderkliftii, Saccharomycopsis crataegensis.

Characterization of a Phenazine and Hexanoyl Homoserine Lactone Producing Pseudomonas aurantiaca Strain PB-St2, Isolated from Sugarcane Stem

  • A novel strain of fluorescent pseudomonad (PB-St2) was isolated from surface-sterilized stems of sugarcane grown in Pakistan. The bacterium was identified as Pseudomonas aurantiaca on the basis of 16S rRNA gene sequence analysis and results from physiological and biochemical characteristics carried out with API50 CH and QTS 24 bacterial identification kits. Assays using substrate-specific media for enzymes revealed lipase and protease activities but cellulase, chitinase, or pectinase were not detected. The bacterium was unable to solubilize phosphate or produce indole acetic acid. However, it did produce HCN, siderophores, and homoserine lactones. In dual culture assays on agar, the bacterium showed antifungal activity against an important pathogen of sugarcane in Pakistan, namely Colletotrichum falcatum, as well as for pathogenic isolates of Fusarium oxysporium and F. lateritium but not against F. solani. The antifungal metabolites were identified using thin-layer chromatography, UV spectra, and MALDI-TOFF spectra and shown to be phenazine-1-carboxylic acid (PCA), 2-hydroxyphenazine (2-OH-PHZ), and N-hexanoyl homoserine lactone (HHL) (assessed using only TLC data). The capacity of this bacterium to produce HCN and 2-OH-PHZ, as well as to inhibit the growth of C. falcatum, has not been previously reported.

종이변색균류의 배양적 특성 및 화학적 방법에 의한 변색제거 (Cultural characteristics of fungal species associated with deterioration or foxing of paper and chemical removal)

  • The annals of Joseon dynasty, especially the volumes of King SeJong(1418-1450 A.D.), were heavily deteriorated by fungi. Investigations on the deteriorating and foxing fungi were carried out. Fungal structures on the beeswax, which were coated on the both side of Han-Ji, were suspected to be involved in the deterioration, and were observed by SEM. Isolation and culturing of these fungi were tried by scrubing swab samples and placing on the artificial media. Culture-independent approaches were used to identify the fungal strains associated with damages of beeswax and foxing of the paper by the analyses based on DNA sequences data from the specific ITS region of rDNA regions. In addition, well-known paper staining fungi(PSF), i.e., Aspergillus terreus var. terreus, Fusarium oxysporum, Chaetomium globosum, Cladosporium cladosporioides, and Alternaria solani, were compared in the mycelial growth and stain on beeswax and papers under different environmental conditions (temperature, light, moisture, etc). Fungal strains isolated from the air samples in the storage room and shelves were identified as Irpex sp., Arthrinium sacchari, Cladosporium tenuissimum, Aspergillus sclerotiorum, Sistotrema brinkmannii, and Hypoxylon bovei var. microsporum The isolated strains were compared in growth and stain patterns on beeswax and papers(Han-Ji, Hwa-Ji, and Yang-Ji) whether these can cause damage or foxing on the annals or not.

Burkholderia pyrrocinia LA101 선발 (Selection of Burkholderia pyrrocinia LA101)

  • The purpose of this study was to investigate eco-friendly measures to manage major diseases which cause heavy economic damages to ginseng. Morphological, physicochemical, and molecular biological species identification was carried out after isolating useful antagonistic bacteria from ginseng fields. In addition, optimal conditions for mass culture were established, and he efficacy of the bacteria in the prevention of the diseases was verified in the field. The results showed that about 150 bacteria were extracted from 150 ginseng fields in the whole county. Among them, B. pyrrocinia LA101 was finally selected, which had a strong antagonistic potency against Alternaria panax, Botrytis cinerea, Rhizoctonia solani, and Cylindrocarpon destructans on agar media. The B. pyrrocinia LA101 is a baculiform gram-negative bacterium identified as Burkholderia pyrrocinia according to results from an API(Analytical Profile Index) kit, 16S rRNA, and gyrase gene sequencing analysis. It was donated to the microbe bank of the Agricultural Genetic Resources Center at the National Academy of Agriculture Science under the Rural Development Administration on September 28, 2011 (Donation No. KACC91663P). A patent for the mass culture technology was granted in August 2012 (Patent No. 10-1175532).

An Endophytic Nodulisporium sp. from Central America Producing Volatile Organic Compounds with Both Biological and Fuel Potential

  • A Nodulisporium sp. (Hypoxylon sp.) has been isolated as an endophyte of Thelypteris angustifolia (Broadleaf Leaf Maiden Fern) in a rainforest region of Central America. It has been identified both on the basis of its morphological characteristics and by scanning electron microscopy as well as ITS sequence analysis. The endophyte produces volatile organic compounds (VOCs) that have both fuel (mycodiesel) and use for biological control of plant disease. When grown on potato dextrose agar, the organism uniquely produces a series of ketones, including acetone; 2-pentanone; 3-hexanone, 4-methyl; 3-hexanone, 2,4-dimethyl; 2-hexanone, 4-methyl, and 5-hepten, 2-one and these account for about 25% of the total VOCs. The most abundant identified VOC was 1,8 cineole, which is commonly detected in this group of organisms. Other prominent VOCs produced by this endophyte include 1-butanol, 2-methyl, and phenylethanol alcohol. Moreover, of interest was the presence of cyclohexane, propyl, which is a common ingredient of diesel fuel. Furthermore, the VOCs of this isolate of Nodulisporium sp. were selectively active against a number of plant pathogens, and upon a 24 h exposure caused death to Phytophthora palmivora, Rhizoctonia solani, and Sclerotinia sclerotiorum and 100% inhibition to Phytophthora cinnamomi with only slight to no inhibition of the other pathogens that were tested. From this work, it is becoming increasingly apparent that each isolate of this endophytic Nodulisporium spp., including the Daldina sp. and Hypoxylon spp. teleomorphs, seems to produce its own unique set of VOCs.

Antioxidant and Antifungal Activity of Endophytic Fungi Associated with Agarwood Trees

  • Several species of Aquilaria and Gyrinops are native to Indonesia and well known as agarwood-producing trees with a high economic value. Their bioactive compounds have a wide spectrum of uses, such as in medicine and cosmetics. These genera have undergone extensive search for novel bioactive compounds. The purpose of this study was to isolate, identify, and characterize the endophytic fungi community associated with Aquilaria malaccensis, A. microcarpa, Gyrinops versteegii, and A. crassna trees and investigate their bioactive properties as antioxidant agents and antagonists. A total of 50 fungi were successfully isolated from different tissues of the four species of agarwood-producing trees. Two isolates exhibited strong antioxidant activity, namely, Apodus oryzae (R2MC3A, $IC_{50}$ 60.92 mg/mL) and Diaporthe sp. (P1DS1[C], $IC_{50}$ 76.65 mg/mL). Two isolates, Pestalotiopsis theae (P3BS3[B]) and Curvularia sp. (P2CD3A), showed >75% antifungal activity against pathogenic Fusarium solani. The results revealed that endophytic fungi associated with the studied agarwood-producing trees had potential antioxidant and antifungal activities for further applications in biotechnology.

취나물 점무늬병을 억제하는 Paenibacillus polymyxa JE201의 생물학적 특성 (Biological Characterization of Paenibacillus polymyxa JE201 with Antifungal Activity Against Fungal Leaf Spot Disease of Aster scaber)

  • 취나물 재배 시 문제가 되는 점무늬병(Septoria sp.)에 대해 항균활성을 나타내는 세균 P. polymyxa JE201를 분리하였다. JE201 균주를 이용하여 점무늬병 방제효과 포장검정을 시행한 결과, 대조구로 사용한 세레나데 맥스와 비슷한 정도의 방제효과를 나타낼 정도로 효과가 있었으며, 다른 식물 병원성 진균에 대한 항균활성 스펙트럼을 조사한 결과, 10종 중 9종에 대해서도 균사생장억제력이 크게 나타나는 것으로 보아, 앞으로 추가적인 연구를 통해 다른 병에도 적용할 수 있을 것으로 기대된다.
