  • Phylogenetic signal present in the mitochondrial 16S ribosomal RNA gene (16S rDNA) and the nuclear large subunit ribosomal RNA gene (28S rDNA) was explored to assess their utility in resolving family level relationships of the superfamily Tephritoidea. These two genes were chosen because they appear to evolve at different rates, and might contribute to resolve both shallow and deeper phylogenetic branches within a highly diversified group. For the 16S rDNA data set, the number of aligned sites was 1,258 bp, but 1,204 bp were used for analysis after excluding sites of ambiguous alignment. Among these 1,204 sites, 662 sites were variable and 450 sites were informative for parsimony analysis. For the 28S rDNA data set, the number of aligned sites was 1,102 bp, but 1,000 bp were used for analysis after excluding sites of ambiguous alignment. Among these 1000 sites, 235 sites were variable and 95 sites were informative for parsimony analysis. Our analyses suggest that: (1) while 16S rDNA is useful for resolving more recent phylogenetic divergences, 28S rDNA can be used to define much deeper phylogenetic branches; (2) the combined analysis of the 16S and 28S rDNAs enhances the overall resolution without losing phylogenetic signal from either single gene analysis; and (3) additional genes that evolve at intermediate rates between the 16S and 28S rDNAs are needed to further resolve relationships among the tephritoid families.

  • From a cluster of structural rRNA genes which has previsouly been cloned (Hwang and Kim, in submission; J. Microbiol. Biotechnol.), a 1.0-kb Eco RI fragment of DNA which shows significant homology to the 25S and rRNA s of Tricholoma matsutake was used for sequence analysis. Nucleotide sequence was bidirectionally determined using delection series of the DNA fragment. Comparing the resultant 1016-base sequence with sequences in the database, both the 3'end of 25S-rRNA gene and 5S rRNA gene were searched. The 5S rRNA gene is 118-bp in length and is located 158-bp downstream of 3'end of the 25S rRNA gene. IGSI and IGS2 (partial) sequences are also contained in the fragment. Multiple alignment of the 5S rRNA sequences was carried out with 5S rRNA sequences from some members of the subdivision Basidiomycotina obtained from the database. Polygenetic analysis with distance matrix established by Kimura's 2-parameter method and phylogenetic tree by UPGMA method proposed that T. matsutake is closely related to efibulobasidium allbescens. Secondary structure of 5S rRNA was also hypothesized to show similar topology with its generally accepted eukaryotic counterpart.

  • The diversity of cyanobacteria in Sri Lanka was studied in different water reservoirs, paddy fields, brackish water and tsunami affected areas using light microcopy, 16S rRNA sequences, followed by phylogenetic analysis. Based on light microscopy, 24 genera were identified from environmental samples belonging to the orders Chroococcales, Oscillatoriales, Pleurocapsales and Nostocales. In cultures, 33 genera were identified from all five cyanobacterial orders, including Stigonematales. Based on 16S rRNA gene sequences and their morphology, two isolates were identified up to species level, 72 to genus level, one isolate up to family and 11 up to order level. Twelve isolates couldn't be assigned to any taxonomic level. The results of 16S rRNA gene sequences along with the phylogenetic analysis indicated that some cyanobacterial isolates could be accommodated to genus or order level. The 16S rRNA sequence analysis data in this study confirmed that order Nostocales and order Pleurocapsales cyanobacteria are monophyletic while orders Chroococcales, Oscillatoriales and Stigonematales cyanobacteria are polyphyletic. Polyphasic approach including the combination of light microscopy, cultures and the analysis of 16S rRNA gene sequences provide a promising approach to ascertain the diversity of cyanobacteria in different habitats.

  • The purpose of this study was to analyze the financial status of Korean elderly households to make a financial plan for retirement preparation. The sample obtained from 1997 KHPS, consisted of 326 households of 55-64 aged and 210 households of 65 and over aged. Statistics employed for the analysis were frequencies, percentile, univariate analysis and logistic analysis. And Lorenz curve and Gini coefficient were used to compare the financial status. The results could be summarized as follows. First, total income, total expenditure, total debt and net worth of 55-64 aged group were higher than those of 65 and over group, while total asset of 55-64 aged group was similar to 65 and over group's Second, total expediture to total imcome ratio(R1) and total asset to total debt ratio(R4) of 55-64 aged group were higher than 65 and over group's, but net worth to minimum cost of living ratio(R2) and financial asset to monthly expenditure ratio(R3) of 65 and over group were higher than 55-64 aged group's . Third, R4's Gini coefficient was the highest and R1's Gini coefficient was the lowest among all Gini coefficients. Also, R1's and R4's Gini coefficients of 55-64 aged group were greater than 65 and over group's. ourth, variables affecting R1's stability of 55-64 aged group were household head's age, educatonal attainment and family type, while those of 65 and over group were household head's age, past economic experience and the number of children. There was no significant variable affecting R2's stability of 55-64 aged group, but there was only household head's pst economic experience affecting those of 65 and over group. The variable contributing to R3's stability of 55-64 aged group was residence while the variables of 65 and over group were household head's educational attainment, past economic experience and the number of children. The variables contributing to R4's stability of 55-64 aged group were the number of children and residence, and the number of children was one variable of contributing to R4's stability of 65 and over aged group.

  • A theoretical cycle analysis has been performed for a basic heat pump, charged with non-azeotropic refrigerant mixtures, R22/R114 and R13B1/R152a. At first, a procedure is introduced to calculate thermodynamic properties simply and correctly, and the advantages of using refrigerant mixtures are discussed through the cycle analysis. It is shown that by using refrigerant mixtures in the heat pump, several improvements can be made. In comparison with conventional pure refrigerants, the application of refrigerant mixtures results in high reliabilities caused by the extension of the application limit, energetic improvements, and a continuous capacity control. From generalizing various results, the optimum compositions in refrigerant mixtures are also determined. The 30%/70% and 40%/60% compositions are selected for R22/R114 and R13B1/R152a, respectively.

  • Most of the pile's vertical static load tests in construction sites are the proof load tests, which is difficult to accurately estimate the ultimate bearing capacity and analyze the reliability of piles. Therefore, a reliability analysis method based on the proof load-settlement (Q-s) data is proposed in this study. In this proposed method, a simple ultimate limit state function based on the hyperbolic model is established, where the random variables of reliability analysis include the model factor of the ultimate bearing capacity and the fitting parameters of the hyperbolic model. The model factor M = RuR / RuP is calculated based on the available destructive Q-s data, where the real value of the ultimate bearing capacity (RuR) is obtained by the complete destructive Q-s data; the predicted value of the ultimate bearing capacity (RuP) is obtained by the proof Q-s data, a part of the available destructive Q-s data, that before the predetermined load determined by the pile test report. The results demonstrate that the proposed method can easy and effectively perform the reliability analysis based on the proof Q-s data.

  • This study has demonstrated the molecular variation of 5S rRNA genes in 15 Allium subgenus Rhizirideum and 1 Allium subg. Allium. For cloning of the 5S rRNA genes, PCR products were obtained from amplification with oligonucleotide primers which were derived from the conserved coding region of 5S rRNA genes. These amplified PCR products were cloned and identified by FISH and sequence analysis. The 5S rRNA loci were primarily located on chromosomes 5 and/or 7 in diploid species and various chromosomes in alloploid species. The size of the coding region of 5S rRNA genes was 120 bp in all the species and the sequences were highly conserved within Allium species. The sizes of nontranscribed spacer (NTS) region were varied from 194 bp (A. dektiude-fustykisum, 2n=16) to 483 bp (A. sativum). Two kinds of NTS regions were observed in A. victorialis var. platyphyllum a diploid, A. wakegi an amphihaploid, A. sacculiferum, A. grayi, A. deltoide-fistulosum and A. wenescens all allotetraploids, while most diploid species showed only one NTS region. The species containing two components of NTS region were grouped with different diploid species in a phylogenetic tree analysis using the sequences of 5S rRNA genes and adjacent non-coding regions.

  • The region containing 18S rRNA gene, ITS 1 and part of the 5.8S rRNA gene of the Atractylodes japonica Koidz was amplified by PCR and the product cloned in a pBluescript SK II plasmid. DNA sequence of the cloned DNA was determined and submitted to the GenBank (accession number EU678363). Phylogenetic analysis of the ITS 1 DNA showed close similarity with the other plant species of the family Compositae. The extract of the plant materials of five different members of the family Compositae was analyzed by HPLC to detect atractylon. Extract of the A. japonica Koidz showed presence of significant amount of atractylon. However, noticeable amount of atractylon was not detected by the same analyses from the extracts of the other plants belonging to the family Compositae including Artemisia capillaris, Chrysantemum zawadskii, Eclipta prostrata or Taraxacum platycarpum.
