Distribution characteristics of chemical oxygen demand and Escherichia coli based on pollutant sources at Gwangyang Bay of South Sea in Korea (남해 광양만에서 오염원에 따른 화학적 산소요구량과 대장균의 해역별 분포특성)

  • This study aimed to understand seasonal and geographical characteristic of chlorophyll-${\alpha}$ (chl-${\alpha}$), COD (chemical oxygen demand) and Escherichia coli at Gwangyang Bay during the period from February 2010 to November 2012. The bay is divided into three different zones based on the pollutant levels and geographical characteristics. During the study periods, water temperature, salinity, Chl. ${\alpha}$, and chemical oxygen demand (COD) varied in the range of $4.68-28.63^{\circ}C$, 1.94-33.84 psu, 0.31-35.10 ${\mu}gL^{-1}$, and 0.70-13.35 $mgL^{-1}$, respectively. Total chl-a concentration were high at the zone I, which can be characterized as a semi-enclosed eutrophic area, and it were low at the zone III, which is influenced by low nutrients of surface warm water current from offshore of the bay. The high concentration of COD was observed at inner bay during the four seasons and the water quality level was kept to be bad condition during spring season at the zone II, which is influenced by Seomjin River water. The highest colony form of E. coli was recorded to be 3550 $cfuL^{-1}$ during summer at station 1 (zone I), whereas it was relatively kept low level during all seasons at the zone III. As a result, the E. coli was correlated with water temperature (r=0.31 p<0.05) and salinity (r=-0.55 p<0.05), implying that those parameters have play an important crucial role in proliferation of E. coli. Consequently, our results indicated that the E. coli can be significantly promoted within pollutant sources including the high nutrients supplied by rive discharge during spring and summer rainy seasons in semi-enclosed area of Gwangyang Bay.

Monitoring for Microbiological Quality of Rice Cakes Manufactured by Small-Scale Business in Korea (소규모 가공경영체 떡류의 생산과정에 따른 미생물학적 품질조사를 위한 모니터링)

  • The purpose of this study was to evaluate the microbial contamination level of Korean traditional rice cakes (Garaetteok, Injeolmi, Gyeongdan), as well as manufacturing environment of small-sized businesses in Korea. The contamination levels of total aerobic bacteria, coliforms, and Bacillus cereus in raw materials were 3.76-4.48, 2.21-4.14, and 1.02-1.15 log CFU/g respectively. On the other hand, Escherichia coli was not found. It has been found that the contamination level of total aerobic bacteria, coliforms, and B. cereus in the raw material decreased after the washing process, but it increased again during the soaking and grinding process. However, after the steaming stage, the contamination level increased again during the molding and cooling process, suggesting the need to take cautions in managing cooling water and molded rice cakes in the process. These results suggest that the safe management of cooling water and taking cautions in the drying process after steaming of rice cakes are necessary for controlling cross-contamination. No E. coli was detected during the manufacturing process involving all tested rice cakes. The microbial contamination level of manufacturing environment such as rice grinder and rice cake forming machine was high. Therefore, in terms of food safety strategy, it is necessary to consider introducing systematic cleansing and disinfection procedure to processing equipment and environment for the sake of reducing microbiological risks.

Quality characteristics of takju by different method of rice pre-treated (밑술 담금 방법을 달리한 탁주의 품질 특성)

  • In this study, we analyzed three rice pre-treatment methods for the preparation of mit-sool (first mashing), with godubap (steamed rice), beombeok (undercooked porridge), and juk (porridge). The results can provide basic data for the development of takju with improved quality. In terms of the base alcohol, godubap produced the highest amount of alcohol (8.1%) after two days, followed by beombeok and juk. After two days of fermentation, soluble solids were measured at 15.3, 15.1, and 1.4°Brix in beombeok, juk, and godubap, respectively, The glucose content of godubap did not change significantly; however, the maltose content increased. In beombeok, maltose decreased from 16.48 mg/mL on the initial day 0 to 1.27 mg/mL on day 9. In juk, glucose and maltose increased from 14.05 and 11.49 mg/mL on day 0 to 31.39 and 42.53 mg/mL on day 2, respectively. Succinic acid levels increased in the godubap and beombeok mit-sool. However, succinic acid was not detected in godubap and juk during the initial stage of fermentation, and lactic acid was not detected in beombeok mit-sool. During the fermentation stage of deot-sool (second mashing), the highest alcohol content of takju made with godubap alcohol was 18.0% and the lowest alcohol content was 15.1% in beombeok on day 9. The total acidity was as high as 0.54% for beombeok, 0.41% for juk, and 0.39% for godubap. In the flavor analysis, ethyl caprate and ethyl caprylate, which were high on day 0 in godubap, decreased upon the completion of fermentation on day 9. Banana-flavored isoamyl alcohol content increased. For juk, ethyl caprate and ethyl caprylate, which were high on day 0, decreased on day 9. Flavor components, such as ethyl palmitate and ethyl linoleate were increased.

Spatiotemporal and Longitudinal Variability of Hydro-meteorology, Basic Water Quality and Dominant Algal Assemblages in the Eight Weir Pools of Regulated River(Nakdong) (낙동강 8개 보에서 기상수문·기초수질 및 우점조류의 시공간 종적 변동성)

  • The eutrophication and algal blooms by harmful cyanobacteria (CyanoHAs) and freshwater redtide (FRT) that severely experiencing in typical regulated weir system of the Nakdong River are one of the most rapidly expanding water quality problems in Korea and worldwide. To compare with the factors of rainfall, hydrology, and dominant algae, this study explored spatiotemporal variability of the major water environmental factors by weekly intervals in eight weir pools of the Nakdong River from January 2013 to July 2017. There was a distinct difference in rainfall distribution between upstream and downstream regions. Outflow discharge using small-scale hydropower generation, overflow and fish-ways accounted for 37.4%, 60.1% and 2.5%, respectively. Excluding the flood season, the outflow was mainly due to the hydropower release through year-round. These have been associated with the drawdown of water level, water exchange rate, and the significant impact on change of dominant algae. The mean concentration (maximum value) of chlorophyll-a was $17.6mg\;m^{-3}$ ($98.2mg\;m^{-3}$) in the SAJ~GAJ and $29.6mg\;m^{-3}$ ($193.6mg\;m^{-3}$) in the DAS~HAA weir pools reaches, respectively. It has increased significantly in the downstream part where the influence of treated wastewater effluents (TWEs) is high. Indeed, very high values (>50 or $>100mg\;m^{-3}$) of chlorophyll-a concentration were observed at low flow rates and water levels. Algal assemblages that caused the blooms of CyanoHAs and FRT were the cyanobacteria Microcystis and the diatom Stephanodiscus populations, respectively. In conclusion, appropriate hydrological management practices in terms of each weir pool may need to be developed.

Effect of Corn Silage and Soybean Silage Mixture on Rumen Fermentation Characteristics In Vitro, and Growth Performance and Meat Grade of Hanwoo Steers (옥수수 사일리지와 대두 사일리지의 혼합급여가 In Vitro 반추위 발효성상 및 거세한우의 성장과 육질등급에 미치는 영향)

  • The present study was conducted to examine the effect of soybean silage as a crude protein supplement for corn silage in the diet of Hanwoo steers. The first experiment was conducted to evaluate the effect of replacing corn silage with soybean silage at different levels on rumen fermentation characteristics in vitro. Commercially-purchased corn silage was replaced with 0, 4, 8, or 12% of soybean silage. Half gram of the substrate was added to 50 mL of buffer and rumen fluid from Hanwoo cows, and then incubated at 39℃ for 0, 3, 6, 12, 24, and 48 h. At 24 h, the pH of the control (corn silage only) was lower (p<0.05) than that of soybean-supplemented silages, and the pH numerically increased along with increasing proportions of soybean silage. Other rumen parameters, including gas production, ammonia nitrogen, and total volatile fatty acids, were variable. However, they tended to increase with increasing proportions of soybean silage. In the second experiment, 60 Hanwoo steers were allocated to one of three dietary treatments, namely, CON (concentrate with Italian ryegrass), CS (concentrate with corn silage), CS4% (concentrate with corn silage and 4% of soybean silage). Animals were offered experimental diets for 110 days during the growing period and then finished with typified beef diets that were commercially available to evaluate the effect of soybean silage on animal performance and meat quality. With the soybean silage, the weight gain and feed efficiency of the animal were more significant than those of the other treatments during the growing period (p<0.05). However, the dietary treatments had little effect on meat quality except for meat color. In conclusion, corn silage mixed with soybean silage even at a lower level provided a greater ruminal environment and animal performances, particularly with increased carcass weight and feed efficiency during growing period.

The Effect of Single and Mixed Microbial Inoculation on the in situ Fiber Digestibility and Silage of Rice Straw Contaminated Mycotoxins (단일 및 복합 미생물 접종이 곰팡이독소 오염 볏짚의 사일리지 및 In situ 섬유소 소화율에 미치는 영향)

  • This study was conducted to evaluate the efficacy of adding the microbial inoculants to silage for reducing mycotoxins in rice straw silage. When a single agent of L. plantarum and a mixed agent of L. plantarum and S. cerevisiae were added in rice straw silage contaminated mycotoxins, it had an effect on silage fermentation and fiber degradation as well as mycotoxin reduction. Among the mycotoxins, only ochratoxin A and zearalenone were found in the test sample. Ochratoxin A and zearalenone showed a decreasing trend with the addition of silage inoculants compared to the control groups (38.11±2.22 and 633.67±50.30 ㎍/kg), and there was a significant difference at the mixed agents; 27.78±2.28 and 392.72±25.04 ㎍/kg, respectively (p<0.05). The pH was lower in the single agent and the mixed agent compared to the control (p<0.05). The concentration of lactic acid was higher in the single agent (11.73±0.31 mM) than in the control group (8.18±0.93 mM), and the highest concentration was 16.01±0.88 mM in the mixed agent (p<0.05). Acetic acid and propionic acid were found to be significantly lowered with the addition of silage inoculants (p<0.05). Total VFA was also lower at the addition of silage inoculants than the control group (p<0.05). The rumen in situ dry matter degradation of NDF and ADF was maintained at the highest levels of the mixed agent during the culture period, followed by the single agent and the control group at the lowest level. NDF and ADF degradation showed a significant difference at all time points after 12 and 24 hours of culture, respectively (p<0.05). The study results showed that the silage inoculants had the positive effects on quality increasing of rice straw silage; fermentative charateristics, fiber degradation and mycotoxins reduction. Ochratoxin A and zearalenone were greater reduction by adding bacterial inoculants of silage. Therefore it is considered that L. plantarum and S. cerevisiae will improve the quality and stability with remediation of mycotoxin in silage.

A survey on status of quality and risk assessment in dentifrices and mouthwashes (치약제 및 구중청량제의 품질 실태 조사 및 안전성 평가)

  • The quality of the products was investigated by analyzing fluorine content, pH, preservatives and tar colors in 31 dentifrice products (6 items for children) and 15 mouthwash products (2 items for children) marketed. It was intended to provide correct information to consumers by checking whether the standards and product indications match. As a result of measuring the fluoride concentration, 26 dentifrice and 15 mouthwash products contained from 48 to 1,472 ppm and from 85 to 225 ppm, respectively. Fluorine detection rates of dentifrice and mouthwash products were 83.9 and 83.3 %, respectively showing similar levels. Of the 41 fluoride-detected dentifrice and mouthwash products, 40 were 90.7~109.8 % of the displayed amount and suitable for the fluorine content standard of 90.0 to 110.0 %, but one dentifrice was found to be inappropriate at 36.3 % of the content indicated on the product. The pH of the dentifrice was 5.1~9.4, and the mouthwash was 4.2~6.2, which met all standards. As a result of simultaneous analysis of the concentration of six preservatives, benzoic acid was detected the most in 15 cases with a 30.6 % detection rate, sorbic acid was detected in 9 cases (detection rate of 18.4 %), and all four types of methyl p-hydroxybenzoate, ethyl p-hydroxybenzoate, propyl p-hydroxybenzoate, butyl p-hydroxybenzoate were not detected. As a result of analyzing the concentration of 10 types of tar colors, six types including red40, yellow4, yellow5, yellow203, green3, and blue1 were detected in a total of 9 cases (2 dentifrices and 7 mouthwashes) with blue1 being the most frequently detected. Detected fluorine concentration, added preservatives and tar colors were consistent with the product markings and it was well written on product packaging. The detected preservatives and tar colors were at a safe level due to low risk compared to Acceptable Daily Intake.

Efficacy and Safety Evaluation of an Air Sterilizer Equipped With an Electrolytic Salt Catalyst for the Removal of Indoor Microbial Pollutants (염촉매 전기분해 공기살균기의 효능 평가)

  • Recently, there has been increasing interest in enhancing the indoor air quality, particularly in response to the growing utilization of public facilities. The focus of this study was on assessing the efficacy and safety of an air sterilizer equipped with electrolytic salt catalysts. To that end, we evaluated the antimicrobial activity of the vapor spraying from the air sterilizer and its cytotoxicity in condensed form on human cell lines (HaCaT, BEAS-2B, and THP-1). Against the test organisms, which comprised five bacterial strains (Staphylococcus aureus, Bacillus subtilis, Escherichia coli, Pseudomonas aeruginosa, Salmonella typhimurium) and one fungal strain (Candida albicans), the air sterilizer exhibited relatively high antimicrobial activities ranging from 10.89 to 73.98% following 1 and 3 hr of vapor spraying, which were notably time-dependent. Importantly, cytotoxicity assessments on human cells indicated no significant harmful effect even at a 1.0% concentration. Comprehensive safety evaluations included morphological observations, gene expression (Bcl-2, Bax) tests, and FACS analysis of intracellular ROS levels. Consistent with previous cytotoxicity findings, these estimates demonstrated no significant changes, highlighting the air sterilizer's safety and antimicrobial activities. In a simulated 20-hr operation within an indoor environment, the air sterilizer not only showed an 89.4% removal of total bacteria but also a 100.0% removal of Escherichia sp. and fungi. This research outlines the potential of the developed electrolytic salt catalyst air sterilizer to effectively remove indoor microbial pollutants without compromising human safety, underscoring the solution that it offers for improving indoor air quality.

Quality characteristics of different parts of garlic sprouts produced by smart farms during growth (스마트팜 생산 새싹마늘의 부위별 및 생육 기간에 따른 품질 특성)

  • Garlic sprouts can provide data on functional and food processing materials. This study compared the leaves, bulbs, and roots of garlic sprouts grown on smart farms during two growth periods (20 and 25 days). In addition, data for garlic bulbs grown in open fields were presented as reference materials. All garlic sprouts' total free sugar content decreased as the growth period increased. All plant parts' total organic acid content decreased as the growth period progressed, except for the root section. Potassium, phosphorus, and sulfur content increased during growth in all parts of the garlic sprouts. Alliin content decreased in all parts of the plant over time, whereas thiosulfinate content increased in the roots but decreased in the leaves and bulbs. Total polyphenol content increased in all parts of the plant during the growth period, except for the bulb, whereas the flavonoid content did not change significantly over time. The 2,2-diphenyl-1-picrylhydrazy (DPPH) and 2,2'-azinobis (3-ethylben-zothiazoline 6-sulfonate) (ABTS) free radical scavenging activities, as well as the superoxide dismutase (SOD)-like activity of garlic sprouts were 37.45-65.47%, 59.12-89.81%, and 89.52-98.59%, respectively. These activities tend to decrease during the growth period. Here, we showed that garlic sprouts have higher levels of functional substances and physiological activities than general garlic sprouts. It was also determined that a growth period of 20 days was suitable for garlic sprouts. Data for research on functional and food-processing materials can be obtained by analyzing garlic sprouts produced by smart farms.

Effects of Nutrient Levels and Feeding Regimen of a.m. and p.m. Diets on Laying Hen Performances and Feed Cost (산란계에 대한 오전용 사료와 오후용 사료의 영양수준 및 급여방법이 산란능력과 사료비에 미치는 영향)

  • Two experiments were conducted to study the effects of nutrient level and feeding method of split diets for a.m. and p.m. on laying hen performance, feed cost and eggshell quality. In experiment 1, 384 ISA Brown layers of 30∼38wk of age were assigned to four treatments which comprise of three replicates each containing 32 birds. The control(C) was fed a conventional single diet throughout the day and split diet groups(T1, T2 and T3) were offered high energy/protein-low Ca diets, and low energy/protein-high Ca diets in a.m.(04:00∼15:00) and p.m.(15:00∼21:00), respectively. In the split diet groups, daily ME and CP consumption, and feed cost were significantly reduced(P<0.05) compared to the C, while the hen-day egg production, average egg weight and daily feed intake were not different among treatments. Due to the reduced daily ME and CP intakes and feed cost, the conversions of feed, ME, CP and feed cost required per day and per kg egg mass were also significantly improved(P<0.05) in the split diet groups. Eggshell qualities (egg specific gravity, egg breaking strength and eggshell thickness) were improved(P<0.05) by split diet feeding. As the Ca level of the p.m. diet increased. In Experiment 2, 384 ISA Brown layers of 50∼58 wk of age were used in three treatments and each treatment was represented by four replicates each containing 32 birds. The control(C) was fed a conventional single diet throughout the day and split diet group(T1) was offered high energy/protein-low Ca diets, and low energy/protein-high Ca diets in a.m.(04:00∼l5:00) and p.m.(15:00∼21:00), respectively. T2 group was fed the diet mixed (50:50) with the a.m. diets in mash and p.m. diet in pellet used T1 group. In T1 and T2 groups, daily feed intake and average egg weight were significantly reduced(P<0.05) compared to the C, while the hen-day egg production was not influenced by the feeding system. Daily ME and CP consumption, and feed cost were reduced(p.0.05), and the conversions of ME, CP and feed cost required per egg were also significantly improved(P<0.05) in T1 and T2, while the conversions of feed, ME, CP and feed cost required per kg egg mass were not different to the C. Eggshell qualities of T1 and T2 were improved(P<0.05) compared to the others. It was concluded the feed and nutrients consumption, feed cost per day or per kg egg mass could be reduced by introducing split diets for a.m. and p.m. and the feeding method of mixed diet of split diets were also convenient and effective for sparing feed cost and improvement of eggshell quality.
