Production of Laccase and Bioremediation of Pentachlorophenol by Wood-Degrading Fungus Trichophyton sp. LKY-7 immobilized in Ca-Alginate Beads

  • The lignin-degrading fungus Trichophyton sp. LKY-7 was immobilized in ca-alginate beads for laccase production and PCP remediation. The immobilized Trichopphyton sp. LKY-7 enabled the repeated use of this fungus for laccase production and produced high amount of laccase throughout 5 cycles incubation. As a laccase inducer oak wood meal(Quercus variabilis) seemed to be effective laccase inducer for Trichophyton sp. LKY-7, and the optimum addition amount was 1%(W/W) in glucose-peptone medium. Biotransformation of pentachlorophenol by immobilized Trichophyton sp. LKY-7 reached an efficency of up to 90% without toxic inhibition. Immobilized Trichophyton sp. LKY-7 might thus be applicable for semicontinuous laccase production and bioremediation to serve inoculum for reactor system.

Impact of Alkali Pretreatment to Enzymatic Hydrolysis of Cork Oak (Quercus Variabilis) (알칼리 전처리가 굴참나무의 효소 당화에 미치는 영향)

  • Dissolving part of xylan and lignin in lignocellulosic biomass by base can be used as pretreatment technique. Cork oak was pretreated with sodium hydroxide solution and the pretreatment effects were evaluated with two critical factors - NaOH concentration and pretreatment temperature. Some of xylan and lignin were removed by base pretreatment. At $90^{\circ}C$ and 13% NaOH pretreatment, 22.0% of lignin and 78.8% of xylan removed by base treatment. Enzymatic hydrolysis of cork oak which was pretreated at higher temperature or concentration was further improved. After pretreatment of cork oak with 13% NaOH at $90^{\circ}C$, the conversion rate of cellulose to fermentable sugars were reached up to 91.3%. At ethanol fermentation with enzymatic hydrolysate from different pretreatment conditions, all enzymatic saccharification liquids were well fermented by Saccharomyces cerevisiae.

  • 본 연구에서는 소나무와 굴참나무의 생엽을 대상으로 기후변화에 따른 연소특성을 분석하고자 월별 기온 및 습도 등과 같은 기상조건별 연소특성을 비교하는 연구를 수행하였다. 소나무의 경우, 연중 내내 생엽의 채취가 가능하지만 굴참나무의 경우에는 생엽 채취가 가능한 6월부터 10월까지를 실험 대상으로 선정하였다. 기상조건은 각 시료별 생엽을 채취한 일자의 기상청 발표 자료를 적용하였으며, 연소특성분석은 콘칼로리미터, 연기밀도시험기, 발화점시험기 등과 같은 장비들을 활용하였다. 분석결과로는 열방출량을 비롯하여, 연기방출량, 함수율, 발화점 등을 측정하여 비교하였다.

  • The forest vegetation of Mt. Naejang, southwestern Korea were classified into ten associations in four alliances of one order by Z-M school method as follows: Quercetalia serrato-mongolicae ord. nov. Rhododendro-Pinion densiflorae all. nov. Rhododendro mucronulati-Pinetum densiflorae Kim et Yim 1986 Acero-Quercion mongolicae all. nov. Rhododendro-Quercetum mongolicae assoc. nov. Carpinion laxiflorae Kim et Yim 1986 Quercetum variabilis Kim et Yim 1986 Carpinetum laxiflorae Kim et Yim 1986 Carpinetum tschonoskii Kim et Yim 1986 Daphniphyllum macropodum community Quercus aliena-Carpinus tschonoskii community Corno-Zelkovion serratae all. nov. Corno-Linderetum erythrocarpae assoc. nov. Torreyo-Zelkovetum serratae assoc. nov. Acero-Zelkovetum serratae assoc. nov. On the other hadn four groups and six subgroups were divided by the species association, and their groups and subgroups showed the coincidence with the alliances and associations, phytosociological units.

The Vegetational Characteristics of Bongamsa Forest Genetic Resources Reserve Area in Mt. Heuyang (희양산 봉암사 산림유전자원보호구역 산림군락구조 특성)

  • The purpose of this study was to research the vegetation structure of the Bongamsa Forest Genetic Resourses Resreve Area in Mt. Heuyang, Mungyeong, Gyeongsangbuk-do. For doing this, ninety-two plots($100m^2/plot$) were set up and investigated, and then Classification analysis and Ordination analysis were carried out. As a result, the vegetation of this area is divided to nine communities; Quercus mongolica community, Quercus variabilis community, Pinus densiflora community, Pinus densiflora-Quercus serrata community, Pinus densiflora-Quercus mongolica community, Quercus serrata community, Decideous broad leaf community, Pinus koraiensis community, Larix kaempferi community. To compare between the communities, statistical analyses were conducted with topographical condition and the results of the vegetational investigation of each community. In altitude, slope, the number of species, the number of individuals in canopy and the number of individuals of understory layer, the mean averages among the communities were different in statistically significance. Then we analysed the vegetation community structure with Importance Percentage of each stratum. The oak tree communities were expected to keep or expand the actual communities because oak trees are spread widely in canopy and understory layers. But the pine tree dominant communities were expected to be succeeded to oak tree communities in the future because of the wide expansion of oak trees.

Flora and Vegetation of Southwest Chuncheon Area (춘천 남서부지역의 식물상과 식생)

  • The flora and vegatation of the Southwest Chuncheon area were investigated from March, 2006 to September, 2007. The vascular plants were summarized as 914 taxa; 111 families, 448 genera, 766 species, 2 subspecies, 119 varieties, and 27 forma. Among the investigated 914 taxa, 33 Korean endemic, four endangered, and 26 rare plants were included. Based on the list of the species of specially designated plants by the Ministry of Environment, 152 taxa were recorded in the investigated area. The naturalized plants were 46 taxa, and life form spectra was H-R5-D1-e type. 914 taxa listed consists of 308 taxa(33.7%) of edible plants, 276 taxa(30.2%) of medicinal plants, 148 taxa(16.2%) of pasture plants, 127 taxa(13.9%) of ornamental plants, 36 taxa(3.9%) of timber plants, 25 taxa(2.7%) of fiber plants and 4 taxa(0.4%) of industrial plants. The vegetation of Southwest Chuncheon area were classified five communities as Betula costata-Acer mono, Quercus mongolica, Quercus mongolica-Quercus variabilis, Quercus mongolica-Quercus dentata, and Pinus densiflora.

  • The study was carried out to investigate the flora and community classfication of forest vegetation in Mt. Yumyeong from March. 2002 to October. 2003. As a result of that. the vascular plants were 98 families, 331 genera, 570 species, 1 subspecies, 81 varieties, 4 formae, 2 hybrid, 658 taxa in all. Korean endemic plants were 10 families, 14 genera, 13 species, 2 variaties, 1 hybrid, 16 taxa in all; rare and endangered plants were 9 families, 10 genera, 11 species, 11 taxa in all: naturalized plants were 12 families, 22 genera, 28 species, 28 taxa in all. The forest vegetation was classified into 1 order, 1 alliance, 6 communities; Rhododendro - Quercetalia mongolicae (Kim, 1990) Lindero - Quercion mongolicae (Kim, 1990) A. Carex humiljs var. nana - Pinus densiflora Community B. Oplismenus undulatifoljus - Pinus koraiensis Community C. Fraxinius rhynchophylla - Quercus mongolica Community D. Cornus controversa - Quercus mongolica Community E. Quercus variabilis - Quercus mongolica Commnuty F. Lespedeza bicolor - Quercus mongolica Community.

Vegetation Composition and Structure of Mt. - Kumbong, Uiseong-Gun, Korea (의성 금봉산일대 산림식생의 조성 및 구조)

  • Vegetation composition and structure of forest community around Mt. Kumbong, Uiseong-Gun, Korea, were studied using a qualitative and a quantitative approach. Vegetation types and various ecological characteristics including constancy classes, relative coverage, species diversity and interspecific similarity of the major component species were analyzed. Seventy relatives were sampled in $100 m^2$ plots. Forest communities were grouped into mountain vegetation and lower-valley vegetation. The former were divided into 3 communities (Quercus mongolica, Quercus variabilis and Quercus dentata communities). The latter was an Acer mono community. Vegetation units were classified into ten groups. Woody vegetation coverage was $50\~65\%$. The lower-valley vegetation type showed from $2.06\pm0.25\;to\;2.31\pm0.26$ in the species diversity. The species showing interspecific similarity over 0.5 with Quercus mongolica appeared to be Pinus densiflora, Quercus dentata and Tilia amurensis in the tree and subtree layer, and Lindera obtusiloba, Fraxinus sieboldiana, Carex humilis and Carex siderostica in the shrub and herb layer, The species showing higher interspecific similarity with Acer mono were Cornus controversa and Ulmus davidiana in the tree and subtree layer, and Philadelphus schrenchkii and Aristolochia manshuriensis in the shrub and herb layer.

Syntaxonomical and Synecological Description on the Forest Vegetation of Juwangsan National Park, South Korea (주왕산국립공원 삼림식생의 군락분류와 군락생태)

  • The forest vegetation of Juwangsan National Park, which is famous for its towering scenic valleys, was syntaxonomically described. The study adopted the $Z{\ddot{u}}rich$-Montpellier School's method emphasizing a matching between species composition and habitat conditions. A combined cover degree and the r-NCD (relative net contribution degree) were used to determine a performance of 265 plant species listed-up in a total of 52 phytosociological $relev{\acute{e}}s$. Nine plant communities were classified through a series of table manipulations, and their distribution and actual homotoneity($H_{act}$) were analyzed. Syntaxa described were Carex gifuensis-Quercus mongolica community, Athyrium yokoscense-Quercus mongolica communiy, Arisaema amurense-Quercus serrata community, Lespedeza maximowiczii var. tomentella-Quercus variabilis community, Tilia rufa-Quercus dentata community, Carex ciliatomarginata-Carpinus laxiflora community, Aristolochia manshuriensis-Zelkova serrata community, Onoclea orientalis-Fraxinus mandshurica community, and Carex humilis var. nana-Pinus densiflora community. A zonal distribution was reviewed and the altitude of about 700 m was the transition zone between the cool-temperate central montane zone (Lindero-Quercenion mongolicae region) and southern submontane zone (Callicarpo-Quercenion serratae region). Only 19 taxa were associated with r-NCD 10% or more, most of which were tree species occurring in the Lindero-Quercenion and some of which was a member of open forests. Species composition of forest vegetation was much less homogeneous, showing the lowest $H_{act}$. Nearly natural forests and/or secondary forests in the Juwangsan National Park were defined as a regional vegetation type, which reflects much stronger continental climate in the Daegu regional bioclimatic subdistrict, rhyolitic tuff predominant, and wildfire interference.

Effects of Molecular Weight of Polyethylene Glycol on the Dimensional Stabilization of Wood (Polyethylene Glycol의 분자량(分子量)이 목재(木材)의 치수 안정화(安定化)에 미치는 영향(影響))

  • This study was carried out in order to prevent the devaluation of wood itself and wood products causing by anisotropy, hygroscopicity, shrinkage and swelling - properties that wood itself only have, in order to improve utility of wood, by emphasizing the natural beautiful figures of wood, to develop the dimensional stabilization techniques of wood with PEG that it is a cheap, non-toxic and the impregnation treatment is not difficult, on the effects of PEG molecular weights (200, 400, 600, 1000, 1500, 2000, 4000, 6000) and species (Pinus densiflora S. et Z., Larix leptolepis Gordon., Cryptomeria japonica D. Don., Cornus controversa Hemsl., Quercus variabilis Blume., Prunus sargentii Rehder.). The results were as follows; 1) PEG loading showed the maximum value (137.22%, Pinus densiflora, in PEG 400), the others showed that relatively slow decrease. The lower specific gravity, the more polymer loading. 2) Bulking coefficient didn't particularly show the correlation with specific gravity, for the most part, indicated the maximum values in PEG 600, except that the bulking coefficient of Quercus variabilis distributed between the range of 12-18% in PEG 400-2000. In general, the bulking coefficient of hardwood was higher than that of softwood. 3) Although there was more or less an exception according to species, volumetric swelling reduction was the greatest in PEG 400. That is, its value of Cryptomeria japonica was the greatest value with 95.0%, the others indicated more than 80% except for Prunus sargentii, while volumetric swelling reduction was decreased less than 70% as the molecular weight increase more than 1000. 4) The relative effectiveness of hardwood with high specific gravity was outstandingly higher than softwood. In general, the relative effectiveness of low molecular weight PEG was superior to those of high molecular weight PEG except that Quercus variabilis showed more than 1.6 to the total molecular weight range, while it was no significant difference as the molecular weight increase more than 4000. 5) According to the analysis of the results mentioned above, the dimensional stabilization of hardwood was more effective than softwood. Although volumetric swelling reduction was the greatest at a molecular weight of 400. In the view of polymer loading, bulking coefficiency reduction of swelling and relative effectiveness, it is desirable to use the mixture of PEG of molecular weight in the range of 200-1500. To practical use, it is recommended to study about the effects on the mixed ratio on the bulking coefficient, reduction of swelling and relative effectiveness.
