Growth Responses of 4 Species to NaCl Concentration in Artificial Soil (NaCl 농도별 토양 처리에 대한 4수종의 생장 반응)

  • In order to inspect growth responses of Fraxinus rhynchophylla Hance, Koelreuteria paniculata Laxmann, Quercus acutissima Carruther and Ulmus parvifolia Jacquin to NaCl treatment, NaCl solution was treated for four months with 0, 25, 50, 100 and 200 mM concentrations, then survival rate, change of relative growth rate, weight, dry weight and pigment content of leaf were investigated. According to NaCl treatment, pH and EC (electrical conductivity) of soil increased, and growth rates of four tree species fell apparently as treatment time became longer. U. parvifolia had the highest survival rate with 15% in the 200mM treatment, and the other three species withered in the treatment. Relative growth rate, weight and dry weight decreased when NaCl treatment time grew longer. The total chlorophyll declined after it rose to 60 days, and the total chlorophyll and carotenoid of the all species according to NaCl treatment did not change very much. With the result from anlayzing growth responses of four tree species to oxidative stress which occurs during NaCl treatment, U. parvifolia has the highest tolerance, followed by K. paniculata, F. rhynchophylla and Q. acutissima.

A Numerical Study for Stability of Tunnel in Jointed Rock Using Barton-Bandis Model (BB절리모델을 활용한 절리암반속 터널안정성의 수치해석적 연구)

  • For the pertinent use of NMT method, both characteristics of joints (JRC, JCS and ${\phi}_r$) and characteristics of rock mass (Q-Value) must be investigated carefully. The main objective of the study presented is to investigate how sensitive the predicted behaviour of an underground excavation is to various realistic assumptions about some input parameter for the jointed rock mass. Joint pattern in the tunnel is predicted by statistical approach (chi-square test). In this paper, sensitivity studies involving in joint characteristics were carried out. The parametric studies involving change in Barton-Bandis joint model have shown that JCS is relatively insensitive to JRC and ${\phi}_r$. An increase in JRC value may not, according to the Barton-Bandis model, necessarily lead to a decrease in displacement. The importance of dilation in predicting the behaviour of a rock mass around an excavation is emphasized from a comparison of the Barton-Bandis joint behaviour model with the Mohr-Coulomb model. The Barton-Bandis model predicted higher stress, which allow for the build-up of stress caused by dilatant behaviour.

Heat Shock Protein 90 Gene Expression in Juvenile Sea Cucumber Apostichopus japonicus (Echinodermata; Holothuroidea) according to Releasing Methods (어린 돌기해삼 Apostichopus japonicus (Echinodermata; Holothuroidea) 방류 방법에 따른 열충격단백질90 유전자의 발현 분석)

  • Sea cucumber, Aposticopus japonicus, is a major invertebrate species in the coastal regions of Korea. To evaluate the short-term stress levels according to the releasing methods, this study investigated the gene expression profiles of heat shock protein 90 (HSP90) by real-time quantitative polymerase chain reaction. When the juvenile sea cucumbers were packed in the vinyl bag with oxygen followed by transportation for 30 min or air-exposed for 1 h, the HSP90 gene expression levels in the experimental groups were significantly increased compared to those of the control groups (transported group, p=0.001; air-exposed group, p=0.032). The experimental group at 6 h post-release by seed-spreading method and at 2~6 h post-release by underwater hose-releasing method on board a fishing boat showed that the levels of HSP90 gene expression were not statistically significant but decreased slightly compared to the control group (seed-spreading group, p=0.069; hose-releasing group, p=0.093). On the other hand, the HSP90 gene expression showed an increasing pattern as the time passed (~6 h) after underwater release of juvenile sea cucumbers by divers (p=0.061). These results suggest that HSP90 gene expression can be used to investigate short-term stress response and effective releasing methods of juvenile sea cucumbers.

An Updated Pooled Analysis of Glutathione S-transferase Genotype Polymorphisms and Risk of Adult Gliomas

  • Objective: Glutathione S-transferases (GSTs) are multifunctional enzymes that play a crucial role in the detoxification of both the endogenous products of oxidative stress and exogenous carcinogens. Recent studies investigating the association between genetic polymorphisms in GSTs and the risk of adult brain tumors have reported conflicting results. The rationale of this pooled analysis was to determine whether the presence of a GST variant increases adult glioma susceptibility by combining data from multiple studies. Methods: In our meta-analysis, 12 studies were identified by a search of the MEDLINE, HIGHWIRE, SCIENCEDIRECT and EMBASE databases. Of those 12, 11 evaluated GSTM1, nine evaluated GSTT1 and seven evaluated GSTP1 Ile105Val. Between-study heterogeneity was assessed using ${\chi}^2$-based Q statistic and the $I^2$ statistic. Crude odds ratios (ORs) with corresponding 95% confidence intervals (CIs) were used to estimate the association between GSTM1, GSTT1 and GSTP1 polymorphisms and the risk of adult gliomas. Results: The quantitative synthesis showed no significant evidence to indicate an association exists between the presence of a GSTM1, GSTT1 or GSTP1 Ile105Val haplotype polymorphism and the risk of adult gliomas (OR, 1.008, 1.246, 1.061 respectively; 95% CI, 0.901-1.129, 0.963-1.611, 0.653-1.724 respectively). Conclusions: Overall, this study did not suggest any strong relationship between GST variants or related enzyme polymorphisms and an increased risk of adult gliomas. Some caveats include absence of specific raw information on ethnic groups or smoking history on glioma cases in published articles; therefore, well-designed studies with a clear stratified analysis on potential confounding factors are needed to confirm these results.

Assessment of Heavy Metal Effects on the Freshwater Microalga, Chlorella vulgaris, by Chlorophyll Fluorescence Analysis (엽록소형광분석을 이용한 담수산 클로렐라(Chlorella vulgaris)에 미치는 중금속의 영향 평가)

  • The response of the freshwater microalga, Chlorella vulgaris, to heavy metal stress was examined based on chlorophyll fluorescence analysis to assess the toxic effects of heavy metals in freshwater ecosystems. When toxic effects were analyzed using regular chlorophyll fluorescence analysis, photosystem II activity($F_v/F_m$) decreased significantly when exposed to $Cu^{2+}$ and $Hg^{2+}$ for 12 h, and decreased in the order of $Hg^{2+}>Cu^{2+}>Cd^{2+}>Ni^{2+}$ when exposed for 24h. The effective photochemical quantum yield(${\phi}{\prime}_{PSII}$), chlorophyll fluorescence decrease ratio($R_{Fd}$), minimal fluorescence yield($F_o$), and non-photochemical quenching(NPQ), but not photochemical quenching(qP), responded sensitively to $Hg^{2+}$, $Cu^{2+}$, and $Cd^{2+}$. These results suggest that $F_v/F_m$, as well as ${\phi}{\prime}_{PSII}$, $R_{Fd}$, $F_o$, and NPQ could be used to assess the effects of heavy metal ions in freshwater ecosystems. However, because many types of heavy metal ions and toxic compounds co-occur under natural conditions, it is difficult to assess heavy metal toxicity in freshwater ecosystems. When Chlorella was exposed to heavy metal ions for 12 or 24h, $F_v/F_m$ and maximal fluorescence yield($F_m$) changed in response to $Hg^{2+}$ and $Cu^{2+}$ based on image analysis. However, assessing quantitatively the toxic effects of several heavy metal ions is challenging.

  • A new approach for antimicrobial is based on the overproduction of reactive nitrogen species (RNS), especially; nitric oxide (NO) and peroxinitrite (ONOO-) are important factors to deactivate the bacteria. Recently, non-thermal atmospheric pressure plasma jet (APPJ) has been frequently used in the field of microbial sterilization through the generation of different kinds of RNS/ROS species. However, in previous study we showed APPJ has combine effects ROS/RNS on bacterial sterilization. It is not still clear whether this bacterial killing effect has been done through ROS or RNS. We need to further investigate separate effect of ROS and RNS on bacterial sterilization. Hence, in this work, we have enhanced NO production, especially; by applying a 1% of HNO3 vapour to the N2 based APPJ. In comparison with nitrogen plasma with inclusion of water vapour plasma, it has been shown that nitrogen plasma with inclusion of 1% of HNO3 vapour has higher efficiency in killing the E. coli through the high production of NO. We also investigate the enhancement of NO species both in atmosphere by emission spectrum and inside the solution by ultraviolet absorption spectroscopy. Moreover, qPCR analysis of oxidative stress mRNA shows higher gene expression. It is noted that 1% of HNO3 vapour plasma generates high amount of NO for killing bacteria.

Development of Evaluation Technique of High Temperature Creep Characteristics by Small Punch-Creep Test Method (I) - Boiler Superheater Tube - (SP-Creep 시험에 의한 고온 크리프 특성 평가 기술 개발(I) - 보일러 과열기 튜브 -)

  • In this study, a small punch creep(SP-Creep) test using miniaturized specimen(10${\times}$10${\times}$0.5mm) is described to develop the new creep test method for high temperature structural materials. The SP-Creep test is applied to 2.25Cr-lMo(STBA24) steel which is widely used as boiler tube material. The test temperatures applied for the creep deformation of miniaturized specimens are between 550∼600$^{\circ}C$. The SP-Creep curves depend definitely on applied load and creep temperature, and show the three stages of creep behavior like in conventional uniaxial tensile creep curves. The load exponent of miniaturized specimen decreases with increasing test temperature, and its behavior is similar to stress exponent behavior of uniaxial creep test. The creep activation energy obtained from the relationship between SP-Creep rate and test temperature decreases as the applied load increases. A predicting equation or SP-Creep rate for 2.25Cr-lMo steel is suggested. and a good agreement between experimental and calculated data has been found.

In silico characterisation, homology modelling and structure-based functional annotation of blunt snout bream (Megalobrama amblycephala) Hsp70 and Hsc70 proteins

  • Background: Heat shock proteins play an important role in protection from stress stimuli and metabolic insults in almost all organisms. Methods: In this study, computational tools were used to deeply analyse the physicochemical characteristics and, using homology modelling, reliably predict the tertiary structure of the blunt snout bream (Ma-) Hsp70 and Hsc70 proteins. Derived three-dimensional models were then used to predict the function of the proteins. Results: Previously published predictions regarding the protein length, molecular weight, theoretical isoelectric point and total number of positive and negative residues were corroborated. Among the new findings are: the extinction coefficient (33725/33350 and 35090/34840 - Ma-Hsp70/ Ma-Hsc70, respectively), instability index (33.68/35.56 - both stable), aliphatic index (83.44/80.23 - both very stable), half-life estimates (both relatively stable), grand average of hydropathicity (-0.431/-0.473 - both hydrophilic) and amino acid composition (alanine-lysine-glycine/glycine-lysine-aspartic acid were the most abundant, no disulphide bonds, the N-terminal of both proteins was methionine). Homology modelling was performed by SWISS-MODEL program and the proposed model was evaluated as highly reliable based on PROCHECK's Ramachandran plot, ERRAT, PROVE, Verify 3D, ProQ and ProSA analyses. Conclusions: The research revealed a high structural similarity to Hsp70 and Hsc70 proteins from several taxonomically distant animal species, corroborating a remarkably high level of evolutionary conservation among the members of this protein family. Functional annotation based on structural similarity provides a reliable additional indirect evidence for a high level of functional conservation of these two genes/proteins in blunt snout bream, but it is not sensitive enough to functionally distinguish the two isoforms.

New Isolated Zero Voltage Switching PWM Boost Converter (새로운 절연된 영전압 스위칭 PWM 부스트 컨버터)

  • In this paper, an isolated ZVS-PWM boost converter is proposed for single stage line conversion. For power factor correction, we used the half bridge topology at the primary side of isolation transformer permitting switching devices to operate under ZVS by using circuit parastics and operating at a fixed duty ratio near 50%. Thus the relatively continuous input current distortion and small size input filter are also achievable. The ZVS-PWM boost operation of the proposed converter can be achieved by using the boost inductor $L_f$, main switch $Q_3$, and simple auxiliary circuit at the secondary side of isolation transformer. The secondary side circuit differ from a conventional PWM boost converter by introduction a simple auxiliary circuit. The auxiliary circuit is actived only during a short switching transition time to create the ZVS condition for the main switch as that of the ZVT-PWM boost converter. With a single stage, it is possible to achieve a sinusoidal line current at unity power factor as well as the isolated 48V DC output. Comparing to the two stage schemes, overall effiency of the proposed converter is highly improved due to the effective ZVS of all devices as well as single stage power conversion. Thus, it can be operated at high switching frequency allowing use of small size input filter. Minimum voltage and current stress make it high power application possible.

Sildenafil Ameliorates Advanced Glycation End Products-Induced Mitochondrial Dysfunction in HT-22 Hippocampal Neuronal Cells

  • Objective : Accumulation of advanced glycation end-products (AGE) and mitochondrial glycation is importantly implicated in the pathological changes of the brain associated with diabetic complications, Alzheimer disease, and aging. The present study was undertaken to determine whether sildenafil, a type 5 phosphodiesterase type (PDE-5) inhibitor, has beneficial effect on neuronal cells challenged with AGE-induced oxidative stress to preserve their mitochondrial functional integrity. Methods : HT-22 hippocampal neuronal cells were exposed to AGE and changes in the mitochondrial functional parameters were determined. Pretreatment of cells with sildenafil effectively ameliorated these AGE-induced deterioration of mitochondrial functional integrity. Results : AGE-treated cells lost their mitochondrial functional integrity which was estimated by their MTT reduction ability and intracellular ATP concentration. These cells exhibited stimulated generation of reactive oxygen species (ROS), disruption of mitochondrial membrane potential, induction of mitochondrial permeability transition, and release of the cytochrome C, activation of the caspase-3 accompanied by apoptosis. Western blot analyses and qRT-PCR demonstrated that sildenafil increased the expression level of the heme oxygenase-1 (HO-1). CoPP and bilirubin, an inducer of HO-1 and a metabolic product of HO-1, respectively, provided a similar protective effects. On the contrary, the HO-1 inhibitor ZnPP IX blocked the effect of sildenafil. Transfection with HO-1 siRNA significantly reduced the protective effect of sildenafil on the loss of MTT reduction ability and MPT induction in AGE-treated cells. Conclusion : Taken together, our results suggested that sildenafil provides beneficial effect to protect the HT-22 hippocampal neuronal cells against AGE-induced deterioration of mitochondrial integrity, and upregulation of HO-1 is involved in the underlying mechanism.
