Expression, Purification and NMR Studies on MC4R-TM2 Mutant

  • Melanocortin-4 receptor (MC4R) subtype is associated with obese humans. Especially, in a patient with severe early-onset obesity, novel heterozygous mutation in the MC4R gene was detected, resulting in an exchange of aspartic acid to asparagine in $90^{th}$ amino acid residue located in the predicted second trans-membrane domain (TM2). Mutations in the melanocortin-4 receptor (MC4R) gene are the most frequent monogenic causes of severe obesity which have been described as heterozygous with loss of function. In order to compare structure difference between MC4R wild type (MC4R-TM2-wt) and mutant (MC4R-TM2-D90N), we designed both MC4R-TM2-wt and MC4R-TM2-D90N construct in pET 21b vector. In this study, we optimized high-yield purification procedure for recombinant TM2-D90N. Eluted recombinant protein was resolubilized under urea condition for thrombin cleavage reaction and we conducted the high-performance liquid chromatography (HPLC) with reverse phase column under 1% acetonitrile, 0.01% TFA buffer solution. The molecular size of purified target peptide was confirmed by Tricine-SDS page analysis. To characterize MC4R-TM2-D90N, we have performed $^{15}N$-isotope labeling of peptide using M9 media and purified labeled target peptide for hetero-nuclear NMR spectroscopy.

Theory and Practices of Water Pollution Control by Wetland - a Case Study of Reed Wetland in Baiyangdian Lake

  • Wetland is an important eco-system on the earth and can effectively control agricultural non-point source pollution. Reed is a typical wetland plant for land/inland water ecotone in north China. The studies indicated that reed had a underground rooted-stem, which formed a 'high speeded-connecting vessels' i.e. reed root channel (RRC) in Baiyangdian lake of Hebei province. It spread predominantly along horizontal direction underground and are mainly distributed at 18-75 cm. The results of field work from healthy reed-wetland (HRW) and regarded reed-wetland (RRW) showed that the reed, averagely, in HRW is 4.2 m height, 1 cm diameter, 65/m2 density; in RRW is 2.4 m height, 3 mm diameter, 86/m2 density. These results indicated the regradation of the function of RRC in RRW. The results of laboratory work of sewage purification from reed soil column (RSC) $(0\~100cm)$ and wheat soil column (WSC) showed that the efficiency of purification to sewage, in RSC, is high than in WSC, especially for phosphorus. The efficiency of purification, in RSC, is $92.6\%$ for total phosphorus, $43.5\%$ for total nitrogen, $54.1\%$ fur COD, respectively; in WSC, is $86.0\%$ for total phosphorus, $241.3\%$ for total nitrogen, $29.8\%$ for COD, respectively.

Development of the Smart Autonomous Moving Air Purifier System (스마트 자율주행 공기청정기 시스템 개발)

  • Recently, since fine dust has become a serious social problem, air purifiers are in the spotlight as a countermeasure against this. Therefore, in this paper, we conducted R&D on the Smart Autonomous Moving Air Purifier System. The developed Smart Autonomous Moving Air Purifier can improve the limitations of the standard used area of existing air purifiers and perform an air purification function efficiently. In addition, we developed App and Web-based programs together for convenient use of Smart Autonomous Moving Air Purifier. Easily operate three air purification modes (Selection mode, Autonomous highest zone mode, Autonomous instant purification mode) through the App and conveniently monitor statistical values (Recent data, Total data, Warning) anywhere through the Web. And, we showed through test that the proposed Smart Autonomous Moving Air Purifier is more efficient than existing air purifiers.

A Study on Particulate Matter Reduction Effects of Vegetation Bio-Filters by Airflow Volume (공조풍량별 식생바이오필터의 입자상 오염물질 저감효과 연구)

  • As the influence of fine dust on society spreads gradually, the public's interest in indoor air is increasingly rising. Air-purifying plants are drawing keen attention due to their natural purifying function enabled by plant physiology. However, as their fine dust reduction mechanism is limited to adsorption only, vegetation bio-filters that optimize purification effects through integration with air-conditioning systems is rising as an alternative. In accordance with the relevant standard test methods, this study looked into the fine dust reduction assessment method by air-conditioning airflow volume that can be used for the industrial spread of vegetation bio-filters. In the case of PM10 at 300 ㎍/m3, it was in the order of EG-B(3,500CMH, 29 min.) < EG-A (2,500CMH, 37 min.) < CG(0CMH, 64 min.) for reaching the maintenance level (100 ㎍/m3) of publicly used facilities. For reaching the WHO Guideline(50 ㎍/m3) requirement, it was in the order of EG-B (51 min.) < EG-A (160 min.) < CG (170 min.). In the case of PM2.5, it was in the order of EG-B (26 min.) < EG-A (33 min.) < CG (57 min.) for reaching the maintenance level (50 ㎍/m3) of publicly used facilities. It was in the order of EG-B (48 min) < EG-A (140 min) < CG (158 min) for reaching the WHO Guideline (25 ㎍/m3) requirement. The findings from the analysis showed that fine dust can be reduced most efficiently when the system is operated at 3,500CMH level. The limitation of this study is that due to the absence of a way of assessing the stress of plants in vegetation bio-filters, generating optimal air-conditioning air flow of the relevant system and economics analysis against the existing facility-type air purification system have been clarified, which should be explored further though follow-up studies.

Characteristics and Function Assessment of Inland Wetlands in Chungnam Province (충청남도 내륙습지 특성 및 기능평가)

  • This study was surveyed from May to October, 2008 in order to classify type distribution and evaluate the function of inland wetland as a ecological axis in Chungnam province. Assessment was done by modified-RAM (Rapid Assessment Method). RAM is consisted of total 8 functions and divided into high, moderate, low. The conservation grade of RAM is divided into 4 grades; absolute conservation, conservation, improvement and restoration. Throughout survey on total 13 wetlands of Lacustrine, Palustrine wetland which are distributed in Chungnam province, their function was assessed. As result, the 2 wetlands were judged as absolute conservation grade by assessment of 8 functional contents, and 7 sites were improvement wetlands and 4 sites were conservation wetlands. The function of wetlands assessed as conservation grade showed high in water quality protection and improvement. Also, showed high in vegetation diversity, wildlife habitat and aesthetic recreation. Meanwhile, showed low in Water quality purification, Shoreline/Stream Bank Protection. Of wetlands evaluated as conservation grade, Jeong-juk Ji and Dun-ri reservoir were assessed as absolute conservative area. These wetlands are essential to be managed continuously as a area having high ecological value. Farther, these wetlands will be done as a axis of ecological network related to 'Kumbuk jeongmaek' ecosystem.

An Analysis of the Customer Demand for Digital Convergence of the Furniture in the Living Space of Ubiquitous Home (유비쿼터스 주택 거실 가구의 디지털 컨버젼스화를 위한 소비자 요구 분석)

  • The purpose of this study was to analyze the customer demand for design development of digital convergence furniture in the living space of the ubiquitous home. This study used the individual survey method and was collected from July 1 to 20, 2008. The frequency and percentage were used for data analysis by SPSS window's 12. The results of the study were as follows: The first, a lot of survey targets were 30-40 years old, university graduates, over 50,000,000 won in annual income and 30-40 pyung in house size. The second, what types of activity were carried out in the living room were acquisition of knowledge>entertainment activity>communication activity>household chores. The third, what types of furniture were used in the living room were A/V furniture>table>sofa>computer table>bookself. The forth, customers preferred to picture telephone service in sociality, home-theater service in entertainment and home appliances remote control service in information service. The fifth, the digital functions which can integrate with the living roon furniture, in the item for A/V furniture, was front door control function which recognizes visitors with the screen. In the item for table was function to search internet and for sofa was self purification function. Finally, the digital convergence furniture is not only for one activity but also for various types of activities that can comprehensively function to materialize the concept of comfortable furniture in the living space.

Production and characterization of a monoclonal anti-glutathione-S-transferase(GST) antibody

  • Analysis of protein is often frustrated by the inability to isolate large amounts of purified protein from a native source. To overcome this problem, fusion protein expression systems such as pGEX system have been widely used. Using pGEX system, the desired protein could be easily obtained in a large amount in E. coli, and then the fusion protein could be used for the study of the function of the given protein. To analyze and purify the GST fusion protein, anti-GST antibody could be used as one of the system of choice. However, the production and characterization of monoclonal anti-GST antibody has not been studied extensively yet. To produce monoclonal anti-GST antibody, GST was purified from E. coli transformed with pGEX-cs, one of the pGEX system and was used as an antigen. The monoclonal antibody was produced by fusion of the immunized spleen cells with SP2-0 myeloma cells. The antibody was characterized by ELISA, western blotting, etc. The monoclonal antibody produced in this study (mAb-GSTA) showed strong and specific immunoreactivity against not only GST but also GST-fusion proteins. Also, mAb-GSTA was successfully used for the immunoaffinity purification of the GST ${\beta}$-Rc.-third intracellular-loop fusion protein. The results of the present study suggest that mAb-GSTA may be used for the identification and purification of GST fusion proteins.

  • The objective of the present study was to investigate cloning, expression, and functions of the recombinant protein, Siva1. Siva1 gene was synthesized by RT-PCR from HCT116 cells. Plasmids were cleaved with the restriction endonuclease, BamH1/Sal1 and products were connected to pQE30, which underwent cleavage by BamH1/Sal1. The recombinant plasmid, pQE30-Siva1, was identified after digestion with restriction endonucleases followed by transformation into E. coli M15. Expression of Siva1 was induced by IPTG and identified by SDS-PAGE following purification with affinity chromatography. The results showed that size of Siva1 was 12 kDa, consistent with the molecular weight of the His-Siva1 fusion protein. Functional test demonstrated that Siva1 significantly inhibited the invasion and migration of HCT116 cells. It may thus find clinical application for control of cancers.

A Study on the Synthesis and Refining of Isothiazoline Derivatives (Isothiazoline 유도체의 합성 및 정제에 관한 연구)

  • Isothiazoline derivatives is widely used to food, medical drug and industrial goods, cosmetics etcs, and it makes to restrain and to sterilize a breeding of microbe as a preservative and a sterilizing agent. It differs with the raw material of paraoxybenzoic acid derivatives or imidazolydinyl urea to be in use at present, on the efficacy and effect, and has various characteristics. This synthesis makes 3,3'-dithiodipropionic chloride to add a thionyl chloride in 3,3'-dithiodipropionic acid, and 3,3'-dithiodipropionic methyl amide makes to synthesize in a reflux reaction the mono methyl amine to 3,3'-dithiodipropionic chloride. And last synthesis becomes to make chlorination-cyclization molecule doing a reflux reaction in the temperature of $90{\sim}100^{\circ}C$ to mix excessively thionyl chloride and ethylene dichloride to 3,3'-dithiodipropionic methyl amide. The last synthesis material has got in the mixture of 5-chloro-2-methyl-4-isothiazoline-3-one and 2-methyl-4-isothiazoline-3-one, and it is so-called isothiazoline derivatives. The purification of isothiazoline derivatives makes to fuse in ethyl acetate, and makes to decolorize and to deodorize in recrystallization. This experiment has been in synthesis and purification of isothiazoline derivatives, and has tried to measure on the antisepsis and sterilization function of microbe according to pH or content change.

Separation and Purification of Soybean $\beta$-amylase Isozymes (대두 $\beta$-amylase Isozyme의 분리 및 정제)

  • The soybean $\beta$-amylase ($\alpha$-1, 4-glucan maltohydrolase, EC 3.2.1.2) is composed of seven isozymes(I', I, II, III, IV, V and VI), and isozyme II and IV are the main components among these. The Purification of $\beta$-amylase isozymes from soybean whey were performed by ammonium sulfate fractionation, CM-Sephadex C-50 column chromatography, DEAE-Sephadex chromatography and Gel filtration. The resulted purity of $\beta$-amylase was throughly confirmed by electrophoresis, and then determined its isoelectric point and molecular weight. The results obtained were as follows, 1. Five active fractions of soybean p-amylase were derived on CM-Sephadex C-50 column chromatography. 2. Seven active bands of p-amylase isozymes were detected by isoelectric focusing gel electrophoresis, and their isoelectric points(I' to VI) were 5.07, 5.15, 5.25, 5.40, 5.55, 5.70 and 5.93, respectively. 3. Isozyme II and IV were main components of soybean $\beta$-amylase. 4. The molecular weights of both isozyme II and IV were determined to be 56,000 daltons by the result of SDS polyacrylamide gel electrophoresis. 5. Km values of main isozyme II & IV for amylopectin were determined to be 2.25 mg/ml, which suggest the same function of each isozyme.
