칡추출물이 알코올을 급여한 흰쥐의 뇌조직에 미치는 영향 (The Effect of Puerariae thubergiana Bentham Extract on Brain Tissue in Alcohol-Treated Rats)

  • 칡추출물이 알코올성 뇌손상에 미치는 영향 을 구명하기 위해 알코올을 투여한 흰쥐에게 갈화와 갈근을 수준별 (I;1.2 g/kg B.W., II;2.4 g/kg B.W.) 로 5주간 급여한 후 \ulcorner 열수추출물이 알코올 대사와 유리기 생성 및 제거효소 활성에 미치는 영향을 관찰하였다. ADH 활성응ㄴ 갈화 및 갈근추출물 급여시 에탄올만 투 여한 대조군에 비하여 유의적으로 감소한 반면, ALDH 활성은 갈근추출물 급여군에서 유의 적으로 증가하였는데 1수준군의 증가정도가 현저하였다. P450 함량과 AD, AH 활성은 대조 군에 비하여 칡 열수추출물 급여시 감소되는 경향이었는데 갈근 급여군의 감소효과가 현저 하게 나타났다. AO와 XO 활성은 갈화 및 갈근추출물 급여군이 대조군에 비하여 감소되었 는데 특히 갈근추출물 1수준군에서 현저하게 나타났다. SOD 활성은 칡 열수추출물 급여시 증가하였으며 CAT와 GSH-Px 활성은 에탄올 투여로 증가된 활성이 갈근 열수추출믈 I 수 준 급여시 유의적으로 증가되었다. 이상의 결과에서 갈화 및 갈근 열수추출물 급여는 뇌조 직 중의 에탄올 대사효소계의 활성을 촉진시켰으며, 유리기제거 효소의 활성을 억제하고 항 산화효소계를 활성화하여 에탄올 투여에 인한 뇌조직의 산화적 스트레스를 완화시킬 수 있 을 것으로 사료된다.

갈근(葛根) 추출물이 cisplatin에 의해 유도된 rat mesangial cell의 apoptosis에 미치는 영향 (Effects of Puerariae Radix extract on Cisplatin-Induced Apoptosis of Rat Mesangial Cells)

  • Puerariae radix (PR) is a popular natural herb and a traditional food in Asia, which has antithrombotic and anti-allergic properties and stimulates estrogenic activity. One of the major side effects of cisplatin is nephrotoxicity, leading to acute renal failure. Recent study has suggested a role of ROS and p53 in renal cell injury by cisplatin. We studied that protective effects of PR on cisplatin-induced apoptosis in rat mesangial cell. Rat mesangial cell was preincubated with PR (50, 100, 150 and 200 ${\mu}g/m{\ell}$) for 12 hr and then treated with 30 ${\mu}M$ cisplatin for 24 hr. Protective effect of PR on cisplatin-induced apoptosis in ECV304 cells was determined using MTT assay, FDA-PI staining, flow cytometric analysis, caspase-3 activity assay, ROS assay and western blot. Our results showed that PR inhibited in cisplatin-induced apoptosis and ROS production in ECV304 cells. Moreover, PR reduced ERK, p38 and JNK activation that increased in cisplatin-treated rat mesangial cell. Furthermore, activation of p53 by cisplatin in rat mesangial cell was inhibited by PR treatment. These results suggest that protective effect of PR on cisplatin-induced apoptosis in rat mesangial cell may be associated with reduction of ERK, p38, JNK, p53 activation.

갈근 추출물이 사염화탄소로 유발된 흰쥐의 간 손상에 대한 보호효과 (Protective Effects of Extract of Puerariae radix on Hepatic injury Induced by Carbon Tetrachloride In Rat.)

  • Dawley 계 숫컷 흰쥐(200~210g)를 정상군, $CCl_4$-대조군, $CCl_4$-실험군으로 나누어 실험군은 갈근 에탄올 추출물을 1,000mg/kg,b.w의 용량으로 1일1회7일간 경구 투여 후 사염화탄소를 0.6mg/kg,b.w의 용량으로 복강내 주사 후 다음날 개복하여 혈청내의 Alanine aminotransferase(ALT), Aspartate aminotransferase(AST), Alkaline phosphatase(ALP), Glutamyltranspeptidase(${\gamma}$-GT), Lactate dehydro-genase(LDH)의 활성도와 중성지방, 콜레스테롤 함량을 측정한 결과 갈근 추출물 투여군에서 $CCl_4$-대조군과 비교하여 유의적인 감소(p<0.05)를 나타내었으며, HDL-콜레스테롤은 유의적인 증가(P0.05)를 나타내었다. 간조직중의 지질과산화 함량과 Catalase(CAT), Glutathione peroxiddase(GSH-Px)의 항산화 효소 활성은 유의적인 감소를 나타내었다. 반면에 간 조직중의 glutathione 함량은 유의적인 증가를 나타내었다. 이 실험 결과 갈근 에탄올 추출물은 사염화탄소 투여에 의한 간 손상을 억제하는 보호물질과 항산화 물질을 함유하고 있음을 알 수 있었다.

뇌해마의 장기양 조직배양을 이용한 한약물의 뇌신경세포손상 보호효능 연구 (Neuroprotective Effects of Medicinal Herbs in Organotypic Hippocampal Slice Cultures)

  • Objectives : For the screening of neuroprotective effects of medicinal herbs, the complex system of animal models suffer some disadvantages in controlling critical parameters such as blood pressure and body temperature. Additionally, application of drugs to the appropriate brain area sometimes is difficult, due to poor permeability though the blood brain barrier, and so potential protective effects might be masked. Methods : Organotypic hippocampal slice culture (OHSC) method has the advantages of being relatively easy to prepare and of maintaining the general structure, including tissue integrity and the connections between cells. Drugs can easily be applied and neuronal damage can easily be quantified by using tissues and culture media. This study demonstrates neuroprotective effects of Puerariae radix (葛根, PR), Salviae miltiorrhizae radix (丹蔘, SR), Rhei rhizoma (大黃, RR), and Bupleuri radix (柴胡, BR). These were screenedand compared to MK-801, antagonist of NMDA receptors, by using OHSC of 1 week-old Sprague-Dawley rats. Oxygen/glucose deprivation (OGD) were conducted in an anaerobic chamber $(85%\;N_2,\;10%\;CO_2\;and\;5%\;H_2)$ in a deoxygenated glucose-free medium for 60 minutes. Water extracts of each herbs were treated to culture media with $5\;{\mu}g/ml$ for 48 hours. Results : Neuronal cell death in the cultures was monitored by densitometric measurements of the cellular uptake of propidium iodide (PI). PI fluorescence images were obtained at 48 hours after the OGD and medicinal herb treatment. Also TUNEL-positive cells in the CAI and DG regions and LDH concentrations in culture media were measured at 48 hours after the OGD. According to measured data, MK-801, PR, SR and BR demonstrated significant neuroprotective effect against excessive neuronal cell death and apoptosis induced by the OGD insult. Especially, PR revealed similar neuroprotective effect to MK-801 and RR demonstrated weak neuroprotective effect. Conclusions : These results suggest that OHSC can be a suitable method for screening of neuroprotective effects of medicinal herbs. 갈근이 고지방·고탄수화물식이와 저용량 streptozotocin-유도 당뇨병 마우스에 미치는 효능 연구 (Effect of the root extract of Pueraria thunbergiana Bentham on high fat/high sucrose diet and single low-dose streptozotocin-induced diabetic mice)

  • Objectives : This study was performed to investigate the effect of root extract of Pueraria thunbergiana Bentham (Puerariae Radix, PR) in diabetic mice as similar as emaciation-thirst disease in Oriental medicine. Methods : C57BL/6 mice were fed high fat (HF) and high sucrose (HS) for 8 weeks, and then administrated with 90 mg/kg body weight (bw) of streptozotocin (STZ) for induction of diabetes which is similar to the middle emaciation stage. After 5 days, blood glucose levels were measured, and selected the mice with ranges above $250mg/d{\ell}$. PR water extract was administrated orally once a day for 4 weeks with high fat and high sucrose. The levels of glucose, insulin, total cholesterol, triglyceride, ${\gamma}glutamyl$ transpeptidase (${\gamma}GTP$), glutamic oxaloacetic transaminase (GOT) and glutamate pyruvate transaminase (GPT) were analysed in the serum. Also, observed their histological changes by hematoxylin and eosin (H&E) of different organs, lung, heart, pancreas, stomach, liver, and kidney. Results : PR extract significantly decreased the levels of serum glucose and insulin in diabetic mice. PR extract significantly increased the levels of triglyceride, total cholesterol, GOT and GPT in diabetic mice. In H&E stain, PR extract inhibited the histopathological changes of lung (as a channel of the upper emaciation stage in the channel-tropism theory), pancreas (as a channel of the middle emaciation stage) and kidney (as a channel of the lower emaciation stage) in diabetic damage. Conclusions : PR extract has an anti-diabetic effect in HF/HS and low-dose STZ-induced diabetic mice. This result suggests that PR follows the channel-tropism theory in the emaciation-thirst disease through the protection of lung, pancreas and kidney.
