Selective Cytotoxicity Platinum (II) Complex Containing Carrier Ligand of cis-1,2-Diaminocyclohexane (Cis-Diaminocyclohexan을 배위자로 하는 배금(II)착체의 선택적 세포독성)

  • The use of cisplatin is limited by severe side effects such as renal toxicity. Our platinum-base drug discovery is aimed at developing drugs capable of diminishing toxicity and improving antitumor activity. We synthesized new Pt (II) complex analogue [Pt (cis-DACH)(DPPP)]. 2NO$_3$ (PC) containing cis-1,2-diaminocyclohexane as a carrier ligand and 1,3-bis(diphenylphosphino) propane as a leaving group. Furthermore, nitrate was added to improved the solubility. In this study, its structure was determined and its antitumor activity against SKOV-3 and NIH-OVCAR-3 human ovarian adenocarcinoma, and in vitro cytotoxicity was determined against primary cultured rabbit kidney proximal tubular and renal cortical cells of human kidney using colorimetric MTT assay. PC demonstrated acceptable antitumor activity against SKOV-3 and NIH-OVCAR-3 human ovarian adenocarcinoma and significant activity as compared with that of cisplatin. The toxicity of PC was found quite less than that of cisplatin using MTT and $^3$H-thymidine uptake tests in rabbit proximal tubular cells and human kidney cortical cells. PC was used for human cortical tissue in 7 weeks hitoculture by the glucose-consumption tests. We determined that the new platinum drug has lower nephrotoxicity than cisplatin. Based on these results, this novel platinum (II) complex compound (PC) represent a valuable lead in the development of a new anticancer chemotherapeutic agent capable of improving antitumor activity and low nephrotoxicity.

  • Platinum (Pt) nanoparticles were prepared by the liquid-phase reduction of tetraammineplatinum (II) chloride $([Pt(NH_3)_4]Cl_2)$ using Nafion as a stabilizer under various conditions of the Nation phase. This method is novel in its use of electrostatic interactions between the Pt complex ions and sulfonic groups in the hydrated Nation molecules. The synthesized Pt nanoparticles of the recast film system had a cubic shape. In the case of the Nation solution system, the Pt nanoparticles mainly had a spherical shape. The shapes and sizes of the Pt nanoparticles were strongly influenced by the Nation phase.

A Study on Catalytic Activity of Oxygen Reduction Reaction and Performance of PEMFC using Pt/C Synthesized by Modified Polyol (수정된 폴리올법으로 합성된 Pt/C를 이용한 산소환원반응성 및 고분자전해질 연료전지 성능 연구)

  • In this research, we investigate electrical performance and electrochemical properties of carbon supported Pt (Pt/C) that is synthesized by polyol method. With the Polyol_Pt/C that is adopted for oxygen reduction reaction (ORR) as cathode of proton exchange membrane fuel cells (PEMFCs), their catalytic activity and ORR performance and electrical performance are estimated and compared with commercial Pt/C(Johnson Mattey) catalyst. Their electrochemically active surface (EAS) area are measured by cyclic voltammetry (CV), respectively. On the other hand, regarding ORR activity and electrical performance of the catalysts, (i) linear sweeping voltammetry by rotating disk electrode and (ii) PEMFC single cell tests are used. The CV measurement demonstrate EAS of Polyol_Pt/C is compared with commercial JM_Pt/C. In case of Polyol_Pt/C, its half-wave potential, kinetic current density are excellent. Based on data obtained by half-cell test, when PEMFC single cell tests are carried out, current density measured at 0.6V and maximum power density of the PEMFC single cell employing Polyol_Pt/C are better than those employing commercial Pt/C. Conclusively, Polyol_Pt/C synthesized by modified polyol process shows better ORR catalytic activity and PEMFC performance than other catalysts.

Fusion anti-cancer drugs of cisplatin analogue and fatty acids for multi-targeted cancer treatment (시스플라틴과 지방산을 결합한 퓨전 항암제)

  • Cispatin has become one of the most widely used anticancer drugs for decades. One of the drawback of cisplatin (II) complex is that it not only targets cancerous cells but also normal cells causing several serious side effects in patients. We have synthesized Pt(IV) complex that are needed to have the ability to kill target cells selectively in a short time before drug resistance develops. By introducing PDK inhibitor, butyric acid and valproic acid, on Pt complex, two fusion anti-cancer agents 3 and 4 have been synthesized and characterized their structures by nmr and mass spectrometer. MTT assay was performed with $Pt(IV)-Bu_2$ 3 and $Pt(IV)-Val_2$ 4 against MCF-7 cell line. As a result, cisplatin, Pt(IV) complexes 3 and 4 were treated, cell viabilities at $50{\mu}M$ cencentration were decreased to 39%, 54% and 84% respectively.

Potential Energy Curves and Geometrical Structure Variations for [MX4]2- : M=Ni(II), Pd(II), Pt(II); X=Cl-, Br-) Dissociating into ([MX3]- + X-) : Ab Initio Study

  • Potential energy curves and internuclear (M-X) distance variations for dissociation reactions of $[MX_4]^{2-}$ into ($[MX_3]^-$ + $X^-$) have been calculated using ab initio Hartree-Fock (HF), second order M$\ddot{o}$ller-Plesset perturbation (MP2), and Density Functional Theory (DFT) methods with a triple zeta plus polarization (TZP) basis set. The equilibrium geometrical structures of $[MX_4]^{2-}$ are optimized to tetrahedral geometry for $[NiX_4]^{2-}$ and square planar geometry for ($[PdX_4]^{2-}$ and $[PtX_4]^{2-}$). The bond (M-X) distances of $[NiCl_4]^{2-}$, $[NiBr_4]^{2-}$, $[PdCl_4]^{2-}$, $[PdBr_4]^{2-}$, $[PtCl_4]^{2-}$, and $[PtBr_4]^{2-}$ at the DFT level are 2.258, 2.332, 2.351, 2.476, 2.367, and 2.493 $\AA$, respectively. The dissociation energies for the bond dissociation of ($[MX_3]^-$${\cdot}{\cdot}{\cdot}$$X^-$) at the DFT level are found to be 4.73 eV for $[NiCl_4]^{2-}$, 4.89 eV for $[NiBr_4]^{2-}$, 4.93 eV for $[PdCl_4]^{2-}$, 5.57 eV for $[PdBr_4]^{2-}$, 5.44 eV for $[PtCl_4]^{2-}$, and 5.87 eV for $[PtBr_4]^{2-}$. As the (M${\cdot}{\cdot}{\cdot}$X) distance of ($[MX_3]^-$${\cdot}{\cdot}{\cdot}$$X^-$) increases, the distance variation (Rt) of trans (M-X) bond at the trans-position is shorter than those (Rc) of two cis (M-X) bonds at the cisposition. Simultaneously the atomic charge variation of trans-X atom is more positive than those of equilibrium $[MX_4]^{2-}$ structures, while the variation of leaving X group is more positive.

  • Optimal preparation guidelines of a cathode catalyst layer by non-precious metal catalysts were evaluated based on electrochemical performance in single-chamber microbial fuel cells (MFCs). Experiments for catalyst loading rate revealed that iron(II) phthalocyanine (FePc) can be a promising alternative, comparable to platinum (Pt) and cobalt tetramethoxyphenylporphyrin (CoTMPP), including effects of substrate concentration. Results showed that using an optimal FePc loading of $1mg/cm^2$ was equivalent to a Pt loading of $0.35mg/cm^2$ on the basis of maximum power density. Given higher loading rates or substrate concentrations, FePc proved to be a better alternative for Pt than CoTMPP. Under the optimal loading rate, it was further revealed that 40 wt% of FePc to carbon support allowed for the best power generation. These results suggest that proper control of the non-precious metal catalyst layer and substrate concentration are highly interrelated, and reveal how those combinations promote the economic power generation of single-chamber MFCs.

  • Two new benzimidazolyl-containing complexes have been synthesized by reactions of $Cu^{II}$ salts and 1,2-bis(benzimidazolyl) benzene ($H_2bbbz$) with two different dicarboxylate ligands. When phthalate acid ($H_2pt$) was employed as secondary ligand, a 0D molecular complex Cu$(H_2bbbz)(pt){\cdot}(H_2pt)$ (1)was furnished and when the secondary ligand was instead by a linear bridging ligand of terephthalic acid ($H_2tp$) a 1D zipper-like coordination polymer $[Cu(H_2bbbz)(tp){\cdot}2(C_2H_5OH){\cdot}H_2O]_n$ (2) was obtained, suggesting the structure-direction effect of the secondary dicarbxylate ligand. The preliminary investigation on the spectral properties of the complexes was also presented.

  • Coordination isomers of cis-(N-N)Pt(Glu) prepared by reaction of cis-(N-N)Pt($SO_4$) (N-N=2$NH_3$, ethylenediamine(en),(R,R)-1,2-diaminocyclohexane (DACH), N,N,N',N'-tetramethylethylenediamine (TMEDA)) with barium glutamate in water have been monitored and characterized by $^1H-NMR$, $^{13}C-NMR$, IR, and mass spectra. The reaction at room temperature affords the mixture of O,O'-and N, ${\alpha}$ O-chelated platinum(II) complexes. The O,O'-chelate initially formed isomerized to N,${\alpha}$O-chelate on standing for a long time or increasing temperature. The ratio of the two isomers at room temperature depends on the nature of the nitrogen donor coligand (N-N).

  • The aim of this study was to evaluate capability of pertussis toxin (PT) to active mouse macrophages. The investigations were undertaken to determine whether the role played by this toxin required the A-protomer of the toxin to ADP-ribosylate a guanine nucleotide binding protein (a class I activity) or was dependent on the binding of B-oligomer of the toxin to the surface of target cells (a Class II activity). The results of these experiments have established that the mechanism of macrophage activation with PT seems to be dependent upon a Class II activity of the toxin.

  • Objectives : The purpose of this study is to evaluate flexor-extensor muscle strength of trunk according to Sasang constitution. Methods : This study was carried out with the data from comprehensive medical testing. People were performed Trunk Extension Flexion(TEF) Program of CYBEX NORM System and QSCC(Questionnaire of Sasang Constitution Classification) II test. And then we made three groups according to Sasang constitution and analyzed isokinetic strength on the TEF. Results : 1. A significant difference(Taeumin>Soeumin>Soyangin) was found in flexor muscle peak torque(PT) of trunk according to Sasang constitution(P<0.05). 2. A significant difference was not found in flexor muscle peak torque%body weight (PT%BW) of trunk according to Sasang constitution. 3. A significant difference(Taeumin>Soeumin>Soyangin) was found in extensor muscle peak torque(PT) of trunk according to Sasang constitution(P<0.05). 4. A significant difference(Soeumin>Taeumin>Soyangin) was found in extensor muscle peak torque%body weight(PT%BW) of trunk according to Sasang constitution(P<0.05). Conclusions : There is a significant difference in isokinetic strength on the TEF according to Sasang constitution.
