Bacterial Root Rot and Bacterial Leaf Blght of Ficus spp. by Pseudomonas cichorii and P. viridiiflava in Korea (Pseudomonas cichorii와 P.viridiflava에 의한 Ficus 속 식물의 세균성뿌리썩음병 및 세균성잎마름병)

  • Nine samples of soft rotten roots and blighted leaves of Ficus spp. plants were collected from the vinyl-houses in Taejeon, Seongnam, Suweon and Yangjae in 1988 and pathogenic bactea were isolated from them Results of the studies on morphological, cultural, physiologucal and pathological characteristics indicated that the bacteria from Ficus retusa were Pseudomonas cichorii, from Ficus retusa \`Golden leaf\` and Ficus benjamina were P.viridiflava. These are the first description of bacteria which caused the diseases on Ficus spp. in Korea. We proposed to name the disease of Ficus retusa by P. cichorii as \"bacterial root rot of Ficus retusa\" and Ficus retusa(Golden leaf) and Ficus benjamina by P. viridiflava as \"bacterial leaf blight of Ficus retusa (Golden leaf)\", \"bacterial blight of Ficus benjamina\", respectively.

First Report of Pseudomonas viridiflava Causing Leaf Spot of Cucumber in Korea (Pseudomonas viridiflava에 의한 오이 점무늬병의 발생 보고)

  • A severe disease with leaf spots and necrotic symptoms was observed in cucumber (Cucumis sativus L.) seedlings in April 2018 at a nursery in Kimjae, Korea (35o 47'09.8"N 127o 2'24.3"E). The infected plants initially showed spots on water-soaked cotyledons which, at later stages, enlarged and spread to the leaves, which the lesions becoming dry and chlorotic. The symptomatic samples were collected from cucumber and the isolates were cultured on LB agar. The representative bacterial strain selected for identification showed fluorescent on King's medium B, was potato rot-positive, levan and arginine dihydrolase-negative, oxidase-negative and tobacco hypersensitivity-positive in LOPAT group 2 as determined by LOPAT tests. A pathogenicity test was carried out on a 3-week-old cucumber. After 3 days of inoculation, leaf spots and necrotic symptoms appeared on the cucumber, similar to the originally infected plants. The infecting bacterial strain was identified as Pseudomonas viridiflava, by 16S rDNA sequence analysis. This is the first report of leaf spot diseases on cucumber caused by P. viridiflava.

  • Erwinia carotovora subsp. carotovora, Pseudomonas viridiflava and Pseudomonas marginalis pv. marginalis were tested for their pathogenicity to 35 kinds of domestic flowering plants. Among them, the following domestic flowering plants showed clear symptoms. 1. Erwinia carotovora subsp. carotovora : (Needle inoculation). Carnation(Dianthus catgophylius L.), madagascar periwinkle(Vinca rosea L.), flower gentle(Amaranthus tricolor L.), snapdragon(Antirrhinum majus L.), chrysanthemum(Chrysanthemum morifolium Ram.), mexiacan ageratum(Ageratum houstonianum Mill), china aster(Callistephus chinensis), youth and old age(Zinnia elegans Jacq.), common nasturtium(Tropaeorum majus L.), scarlet sage(Salvia splendens F.), dahlia(Dahia hybrida), pot marigold(Calendula officinalis L.), begonia treevine(Cissus dicolor Blume), cosmos(Cosmos bipinnatus Cav.), globe amaranth(Gomphrena globosa L.), black eyed susan(Thumbergia alata Bojer), common gypsophila(Gypsophila elegans Bieb.), ghent gladiolus(Gladiolus gandavensis Van.), indian shot(Canna orchiodes Bailey), iris(Iris nertschinskia Lodd), cyclamen primula(Dodecatheon meadia L.), scarlet kafir lily(Clivia miniata Regel.), flowering cabbage(Brassica oleracea L.). (Spray inoculation). Carnation, madagascar periwinkle, flower gentle, snapdragon, common nasturtium, ghent gladiolus, indian shot, cyclamen primula. 2. Pseudomonas viridiflava : (Needle inoculation). Carnation, madagascar periwinkle, snapdragon, chrysanthemum, cockscomb, mexican ageratum, china aster, common nasturtium, common petunia(Petunia hybrida Vilm), pot marigold, begonia treevine, cosmos, black eyed susan, common gypsophila, ghent gladiolus, indian shot, cyclamen primula, scarlet kafir-lily, flowering cabbage. (Spray inoculation). Common nasturtium, ghent gladiolus, indian shot, cyclamen primula. 3. Pseudomonas marginalis pv. marginalis : (Needle inoculation). Carnation, madagascar periwinkle, flower gentle, snapdragon, cockscomb, mexican ageratum, youth and old age, common nasturitium, common petunia, sweet william(Dianthus barbatus L.), pot marigold, begonia treevine, cosmos, common gypsophila, ghent gladiolus, indian shot, iris, cyclamen primula, scarlet kafir-Lily, flowering cabbage. (Spray inoculation). Common nasturtium, ghent gladiolus, indian shot, cyclamen primula.

  • The pathogenic bacteria isolated from the rotted fruits and tubers of red pepper, tomato, eggplant and potato which were collected from farms of Chungnam province and the markets of Seoul, Pusan, Taejon, Chonju, Iri, Weonju, Jecheon, Chungju and Cheonan in Korea. These isolates were divided into ninety-eight isolates of Enoinia sp. and seven isolates of Pseudomonas spp. on the basis of their taxonomic characteristics. Among seven isolates of Pseudomonas spp., six isolates were identified as P. viridiflava and two were P. marginalis and ninety-eight isolates of Erwinia sp. as E. carotovora. subsp. carotovora on the basis of bacteriological properties.

  • Twenty-six cabbage rot samples were collected from markets and main cabbage-growing areas in Korea and isolates of pathogenic bacteria were obtained from the collection. According to their bacteriological characteristics and pathogenicity, the majority of rot bacteria were identified as Erwinia carotovora subsp. carotovora. Pseudomollas cichorii. P. viridiflava and Xanthomonas campestris pv. campestri. Among these four species, E. carotovora subsp. carotovora and X. campestris pv. campestris were found to cause serious damage to cabbage production throughout Korea. Two species of the causal bacteria were sometimes isolates from a single cabbage rot sample and it suggest that the mixed infection of the two bacteria be occurred under field condition.
