Screening of Genetic Polymorphisms of CYP3A4 and CYP3A5 Genes

  • Given the CYP3A4 and CYP3A5's impact on the efficacy of drugs, the genetic backgrounds of individuals and populations are regarded as an important factor to be considered in the prescription of personalized medicine. However, genetic studies with Korean population are relatively scarce compared to those with other populations. In this study, we aimed to identify CYP3A4/5 polymorphisms and compare the genotype distributions among five ethnicities. To identify CYP3A4/5 SNPs, we first performed direct sequencing with 288 DNA samples which consisted of 96 Koreans, 48 European-Americans, 48 African-Americans, 48 Han Chinese, and 48 Japanese. The direct sequencing identified 15 novel SNPs, as well as 42 known polymorphisms. We defined the genotype distributions, and compared the allele frequencies among five ethnicities. The results showed that minor allele frequencies of Korean population were similar with those of the Japanese and Han Chinese populations, whereas there were distinct differences from European-Americans or African-Americans. Among the pharmacogenetic markers, frequencies of $CYP3A4^*1B$ (rs2740574) and $CYP3A5^*3C$ (rs776742) in Asian groups were different from those in other populations. In addition, minor allele frequency of $CYP3A4^*18$ (rs28371759) was the highest in Korean population. Additional in silico analysis predicted that two novel non-synonymous SNPs in CYP3A5 (+27256C>T, P389S and +31546T>G, I488S) could alter protein structure. The frequency distributions of the identified polymorphisms in the present study may contribute to the expansion of pharmacogenetic knowledge.

포장 방법에 따른 즉석섭취형 함박스테이크의 냉장저장 중 품질 변화 (Quality changes in ready-to-eat hamburg steak depending on the packaging methods during chilled storage)

  • 본 연구에서는 포장방법에 따른 함박스테이크의 냉장저장 중 품질변화를 조사하였다. 대조구는 polylactide(PLA) tray를 이용한 함기포장(AC)이었으며, 처리구로는 진공포장(VAP)과 탈산소제 봉입포장(OS)이 적용되었다. 총균수는 대조구와 처리구 모두 저장기간의 경과에 따라 유의적으로 증가하였으며, 특히 AC구에서의 총균수 증가가 다른 처리구들에 비해 두드러진 것으로 나타났다. 또한 저장기간 중 TBA와 VBN값은 AC구에서 가장 빠른 증가를 보였으며 그 다음으로는 OS, 그리고 VAP포장구의 순이었다. 관능평가 결과 VAP구와 OS구 시료들은 모든 관능평가 항목에서 12일째까지 5.0점 이상의 평가를 받은 반면, AC구는 모든 항목에서 5.0 미만으로 이미 상품성을 상실한 것으로 확인되었다. 이러한 결과들은 VAP나 OS처리를 통하여 포장 내 산소를 제거함으로써 호기적 미생물의 생장과 산화작용의 억제로 인한 지방산패나 단백질부패속도가 지연된 것에 기인한 것으로 판단된다.

지구온난화에 따른 국내 기후변화와 젖소 착유우의 생산효율에 미치는 영향 평가 : 모델 시뮬레이션을 이용한 접근 (Climate Change by Global Warming and Its Effects on Production Efficiency of Lactating Dairy Cows in Korea : a Simulation Modeling Approach)

  • 지난 25년간 지구온난화로 인한 국내 기후변화 양상은 지역 간에 차이가 있어, 고온지역은 평균온도의 증가를 보이지는 않았으나, 최저기온의 지속적인 상승과 그에 따른 열대야 발생을 확인할 수 있었다. 또한 그로 인한 젖소의 고온스트레스 발생, 섭취량의 감소 및 생산성 저하가 예상된다. 저온지역의 경우에는 여름철 평균온도 및 최저온도의 상승으로 인해 연평균기온이 유의적으로 상승한 반면, 겨울철에는 오히려 최저기온의 지속적인 하강이 관찰됨에 따라 동물의 저온스트레스가 가중될 것으로 예상된다. 착유우가 저온스트레스 상태에 있을 때는 에너지요구량과 건물섭취량이 증가하여 생산 효율이 떨어지며, 사료효율의 감소로 경제성은 감소되고 우유 생산비는 증가한다. 특히, 극심한 저온스트레스 또는 사료, 음수 및 우사바닥의 결빙은 섭취량 감소를 야기하며 이에 따른 생산성의 저하는 더욱 가중될 것으로 예상된다. 결론적으로 지구온난화로 인한 기후변화에 따른 지역별 영향은 다르며, 온도 스트레스에 의한 낙농우의 생산성 저하를 최소화하고, 동물의 복지와 건강을 증진시키기 위해서는 지역별 기후변화 특성에 맞춘 사양기술의 개발이 필요하다.

Estimation of Nutritive Value of Whole Crop Rice Silage and Its Effect on Milk Production Performance by Dairy Cows

  • The nutritive value and utilization of whole crop rice silage (WCRS), Hamasari, at yellow mature stage was determined by three studies. In first study, chemical composition, in vivo digestibility and metabolizable energy (ME) content of WCRS was determined by Holstein steers. WCRS contains 6.23% CP, its digestibility is 48.4% and estimated TDN is 56.4%. Its ME content was 1.91 Mcal/kg DM. Gross energy (GE) retention (% of GE intake) in steers is only 22.7% most of which was lost through feces (44.7% of GE intake). It takes 81 minutes to chew a kg of WCRS by steers. In another study, the effect of Hamasari at yellow mature stage at three stages of lactation (early, mid and late lactation) and two levels of concentrate (40 or 60%) on voluntary intake, ME content and ME intake, milk yield and composition using lactating Holstein dairy cows were investigated. Total intake increased with the concentrate level in early and mid lactation, but was similar irrespective of concentrate level in late lactation. WCRS intake was higher with 40% concentrate level than with 60% concentrate. ME intake by cows increased with the concentrate level and WCRS in early lactating cows with 40% concentrate can support only 90% of the ME requirement. Milk production in accordance with ME intake increased with the increase in concentrate level in early and mid lactating cows but was similar in late lactating cows irrespective of concentrate level. Fat and protein percent of milk in mid and late lactating cows were higher with for 60% concentrate than 40%, but reverse was in early lactating cows. Solids-not-fat was higher with for 60% concentrate than 40% concentrate. Finally in situ degradability of botanical fractions such as leaf, stem, head and whole WCRS, Hamasari at yellow mature stage was incubated from 0 to 96 h in Holstein steers to determine DM and N degradability characteristics of botanical fractions and whole WCRS. Both DM and N solubility, rate of degradation and effective degradability of leaf of silage was lower, but slowly degradable fraction was higher compared to stem and head. Solubility of DM and N of stem was higher than other fractions. The 48 h degradability, effective degradability and rate of degradation of leaf were always lower than stem or head. In conclusion, voluntary intake of silage ranged from 5 to 12 kg/d and was higher with low levels of concentrate, but milk yield was higher with high levels of concentrate. Fat corrected milk yield ranged from 19 to 37 kg per day. For consistency of milk, early lactating cows should not be allowed more than 40% whole crop rice silage in the diet, but late lactating cows may be allowed 60% whole crop rice silage.

Effects of Post-harvest Storage Duration and Variety on Nutrient Digestibility and Energy Content Wheat in Finishing Pigs

  • This study was conducted to investigate the effects of post-harvest storage duration and wheat variety on the digestibility and energy content of new season wheat fed to finishing pigs. Two wheat varieties (Shi and Zhong) were harvested in 2013 and stored in the warehouse of the Fengning Pig Experimental Base at China Agricultural University for 3, 6, 9, or 12 mo. For each storage period, 12 barrows were placed in metabolism crates and allotted to diets containing 1 of the 2 wheat varieties in a randomized complete block design. The experimental diets contained 97.34% wheat and 2.66% of a vitamin and trace mineral premix. With an extension of storage duration from 3 mo to 12 mo, the gross energy (GE) and crude protein (CP) of the wheat decreased by 2.0% and 12.01%, respectively, while the concentration of neutral detergent fiber (NDF), acid detergent fiber (ADF) and starch content increased by 30.26%, 19.08%, and 2.46%, respectively. Total non-starch polysaccharide, total arabinose, total xylose and total mannose contents decreased by 46.27%, 45.80%, 41.71%, and 75.66%, respectively. However, there were no significant differences in the chemical composition between the two wheat varieties with the exception of ADF which was approximately 13.37% lower in Shi. With an extension of storage duration from 3 mo to 12 mo, the digestible energy (DE), metabolizable energy (ME) content and the apparent total tract digestibility of GE, CP, dry matter, organic matter, ether extract, ADF and metabolizability of energy in wheat decreased linearly (p<0.01) by 5.74%, 7.60%, 3.75%, 3.88%, 3.50%, 2.47%, 26.22%, 27.62%, and 3.94%, respectively. But the digestibility of NDF changed quadratically (p<0.01). There was an interaction between wheat variety and storage time for CP digestibility (p<0.05), such that the CP digestibility of variety Zhong was stable during 9 mo of storage, while the CP digestibility of variety Shi decreased (p<0.05). In conclusion, the GE, DE, and ME of wheat was stable during the first 3 to 6 mo of post-harvest storage, and decreased during the following 6 to 12 mo of storage under the conditions of this study.

Differences in Physicochemical and Nutritional Properties of Breast and Thigh Meat from Crossbred Chickens, Commercial Broilers, and Spent Hens

  • The objective of this study was to compare the physicochemical and nutritional properties of breast and thigh meat from commercial Chinese crossbred chickens (817 Crossbred chicken, 817C), imported commercial broilers (Arbor Acres broiler, AAB), and commercial spent hens (Hyline Brown, HLB). The crossbred chickens, commercial broilers and spent hens were slaughtered at their typical market ages of 45 d, 40 d, and 560 d, respectively. The results revealed that several different characteristic features for the three breeds. The meat of the 817C was darker than that of the other two genotypes. The 817C were also characterized by higher protein, lower intramuscular fat, and better texture attributes (cooking loss, pressing loss and Warner-Bratzler shear force [WBSF]) compared with AAB and HLB. The meat of the spent hens (i.e. HLB) was higher in WBSF and total collagen content than meat of the crossbred chickens and imported broilers. Furthermore, correlation analysis and principal component analysis revealed that there was a clear relationship among physicochemical properties of chicken meats. With regard to nutritional properties, it was found that 817C and HLB exhibited higher contents of essential amino acids and essential/non-essential amino acid ratios. In addition, 817C were noted to have highest content of microelements whereas AAB have highest content of potassium. Besides, 817C birds had particularly higher proportions of desirable fatty acids, essential fatty acids, polyunsaturated/saturated and (18:0+18:1)/16:0 ratios. The present study also revealed that there were significant differences on breast meat and thigh meat for the physicochemical and nutritional properties, regardless of chicken breeds. In conclusion, meat of crossbred chickens has some unique features and exhibited more advantages over commercial broilers and spent hens. Therefore, the current investigation would provide valuable information for the chicken meat product processing, and influence the consumption of different chicken meat.

Genetic and Economic Analysis for the Relationship between Udder Health and Milk Production Traits in Friesian Cows

  • A total of 4,752 monthly lactation records of Friesian cows during the period from 2000 to 2005 were used to estimate genetic parameters and to determine the effect of udder health on milk production traits. Three milk production traits were studied: 305-day milk yield (305-dMY), 305-day fat yield (305-dFY) and 305-day protein yield (305-dPY). Four udder health traits were studied: somatic cell count (SCC), mastitis (MAST), udder health status (UDHS) with 10 categories and udder quarter infection (UDQI) with 7 categories. Mixed model least square analysis was used to estimate the fixed effects of month and year of calving and parity (P) on different studied traits. Sire and dam within sire were included in the model as random effects. Data were analyzed using Multi-trait Derivative Free Restricted Maximum Likelihood methodology (MTDFREML) to estimate genetic parameters. Unadjusted means of 305-dMY, 305-dFY, 305-dPY and SCC were 3,936, 121, 90 kg and 453,000 cells/ml, respectively. Increasing SCC from 300,000 to 2,000,000 cells/ml increased UDQI from 5.51 to 23.2%. Losses in monthly and lactationally milk yields per cow ranged from 17 to 93 and from 135 to 991 kg, respectively. The corresponding losses in monthly and lactationally milk yields return per cow at the same level of SCC ranged from 29.8 to 163 and from 236 to 1,734 Egyptian pounds, respectively. Heritability estimates of 305-dMY, 305-dFY, 305-dPY, SCC, MAST, UDHS, UDQI were 0.31${\pm}$0.4, 0.33${\pm}$0.03, 0.35${\pm}$0.05, 0.23${\pm}$0.02, 0.14${\pm}$0.02, 0.13${\pm}$0.03, and 0.09${\pm}$0.01, respectively. All milk production traits showed slightly unfavorable negative phenotypic and genetic correlations with SCC, MAST, UDHS and UDQI. There were positive and high genetic correlations between SCC and each of MAST (0.85${\pm}$0.7), UDHS (0.87${\pm}$0.10) and UDQI (0.77${\pm}$0.06) and between MAST and each of UDHS (0.91${\pm}$0.11) and UDQI (0.83${\pm}$0.07). It could be concluded that the economic losses from mastitis and high SCC are considerable. The high genetic correlation between SCC and clinical mastitis (CM) suggest that the selection for lower SCC would help to reduce or eliminate the undesirable correlated responses of clinical mastitis associated with selection for increasing milk yield. Additionally, it is recommended also that if direct information on under health traits is not available, measures of SCC can be inclusion in a selection criteria to improve the income from dairy cows.

Effects of Exogenous Enzymes on Ruminal Fermentation and Degradability of Alfalfa Hay and Rice Straw

  • This study was conducted to evaluate the use of exogenous enzymes as a potential means of improving the ruminal digestion (i.e., degradability) of alfalfa hay and rice straw. Twenty six enzyme-additives were examined in terms of protein concentration and enzymic activities on model substrates. The exogenous enzymes contained ranges of endoglucanase, xylanase, ${\beta}$-glucanase, ${\alpha}$-amylase, and protease activities. Six of the enzyme additives were chosen for further investigation. The enzyme additives and a control without enzyme were applied to mature quality alfalfa hay substrate and subsequently incubated in rumen batch cultures. Five of the enzyme additives (CE2, CE13, CE14, CE19, and CE24) increased total gas production (GP) at 48 h of incubation compared to the control (p<0.05). The two additives (CE14 and CE24) having the greatest positive effects on alfalfa hay dry matter, neutral detergent fibre (NDF) and acid detergent fibre (ADF) degradability were further characterized for their ability to enhance degradation of low quality forages. The treatments CE14, CE24, a 50:50 combination of CE14 and CE24 (CE14+24), and control (no enzyme) were applied to mature alfalfa hay and rice straw. For alfalfa hay, application of the two enzyme additives, alone and in combination, increased GP compared to the control at 48 h fermentation (p<0.05), whereas only CE14 and CE14+24 treatments improved GP from rice straw (p<0.05). Rumen fluid volatile fatty acid concentrations throughout the incubation of rice straw were analyzed. Acetate concentration was slightly lower (p<0.05) for CE14${\times}$CE24 compared to the control, although individually, CE14 and CE24 acetate concentrations were not different from the control. Increases (p<0.05) in alfalfa hay NDF degradability measured at 12 and 48 h of incubation occurred only for CE14 (at 12 h) and for CE14+24 (at 12 and 48 h). Similarly, ADF degradability increased (p<0.05) with CE14 and CE14+24. As for rice straw, increased DM degradability was observed at 12 and 48 h of incubation for all enzyme treatments with an exception for CE14 at 12 h. The degradability of NDF was improved by all the enzyme treatments at either incubation time, while ADF degradability was only enhanced at 48 h. Overall, the enzymes led to enhanced digestion of mature alfalfa and there was evidence of improved digestibility of rice straw, an even lower quality forage.

Carcass Characteristics and Meat Quality of Korean Native Ducks and Commercial Meat-type Ducks Raised under Same Feeding and Rearing Conditions

  • This study was conducted to compare carcass characteristics and physico-chemical meat quality in two different genotype ducks raised under identical feeding and rearing conditions. A total of ninety 1-d-old Korean native ducks (KND, n = 45) and commercial meat-type ducks (Grimaud, n = 45) were fed same experimental diets during 56 d and 42 d, respectively to obtain similar slaughter weights. The experimental diet for starter period contained 20% crude protein (CP) and 2,900 kcal nitrogen corrected true metabolizable energy (TMEn)/kg of diet and that for grower period contained 17% CP and 3,050 TMEn/kg of diet. Average daily gain and feed efficiency of KND were inferior to those of commercial meat-type ducks (p<0.05). Carcass weight was not different between two genetically different ducks, but carcass yield of KND was significantly higher (p<0.05) than that of commercial meat-type ducks. There were no significant differences in cooking loss and pH of breast meat between two genetically different ducks, but water holding capacity of KND was significantly higher than that of commercial meat-type ducks. The linoleic acid and total polyunsaturated fatty acid of breast meat from KND were significantly higher (p<0.05) than the corresponding part from commercial meat-type ducks. Significant differences were detected in water holding capacity and the content of linoleic acid and polyunsaturated fatty acid, which were significantly higher in KND, whereas growth performance tended to be superior in commercial ducks. At the market weight, the meat from KND was judged to have better qualities with regard to higher water holding capacity and greater content of polyunsaturated fatty acid compare with meat from commercial meat-type duck.

Evaluation of Genome Based Estimated Breeding Values for Meat Quality in a Berkshire Population Using High Density Single Nucleotide Polymorphism Chips

  • The accuracy of genomic estimated breeding values (GEBV) was evaluated for sixteen meat quality traits in a Berkshire population (n = 1,191) that was collected from Dasan breeding farm, Namwon, Korea. The animals were genotyped with the Illumina porcine 62 K single nucleotide polymorphism (SNP) bead chips, in which a set of 36,605 SNPs were available after quality control tests. Two methods were applied to evaluate GEBV accuracies, i.e. genome based linear unbiased prediction method (GBLUP) and Bayes B, using ASREML 3.0 and Gensel 4.0 software, respectively. The traits composed different sets of training (both genotypes and phenotypes) and testing (genotypes only) data. Under the GBLUP model, the GEBV accuracies for the training data ranged from $0.42{\pm}0.08$ for collagen to $0.75{\pm}0.02$ for water holding capacity with an average of $0.65{\pm}0.04$ across all the traits. Under the Bayes B model, the GEBV accuracy ranged from $0.10{\pm}0.14$ for National Pork Producers Council (NPCC) marbling score to $0.76{\pm}0.04$ for drip loss, with an average of $0.49{\pm}0.10$. For the testing samples, the GEBV accuracy had an average of $0.46{\pm}0.10$ under the GBLUP model, ranging from $0.20{\pm}0.18$ for protein to $0.65{\pm}0.06$ for drip loss. Under the Bayes B model, the GEBV accuracy ranged from $0.04{\pm}0.09$ for NPCC marbling score to $0.72{\pm}0.05$ for drip loss with an average of $0.38{\pm}0.13$. The GEBV accuracy increased with the size of the training data and heritability. In general, the GEBV accuracies under the Bayes B model were lower than under the GBLUP model, especially when the training sample size was small. Our results suggest that a much greater training sample size is needed to get better GEBV accuracies for the testing samples.
