  • The bacterial secondary production was measured at 6 sites of Lake Soyang in October, 1989 by $^{3}$H-thymidine incorporation rate. Verfication for the method of bacterial secondary production measurement showed that $^{3}$H-thymidine incorporated into DNA, RNA and protein by average percentage of 38.45, 42.27 and 20.07%, respectively. THe more increased incoporated $^{3}$H-thymidine, the more increasde DNA fraction, but protein fraction was generally low. Incorporation of rate of /usp 3/H-thymidine. $^{3}$H-leucine into protein correlated with protein fraction of incorporated $^{3}$H-thymidine. Conversion factors were calculated as follows; $1.83*10 ^{20}$ cells/moles of thymidine incorporated/hr and 1.69*10$^{22}$ cells/moles of leucine incorporated/hr.

in vitro에서 B(a)P이 착상전 마우스 배자에 미치는 독성학적 영향에 관한 연구 (Toxicological Effects of B(a)P on Preimplantation Mouse Embryos in Vitro)

  • Effects of B(a)P on preimplantation mouse embryos in vitro were studied. Preimplantation mouse embryos were exposed to a concentration of 0.3, 1, 3 and 10 $\mu$M B(a)P for 72 hrs. The toxicological effects of B(a)P were evaluated by morphological observation of embryos up to the blastocyst stage, and by measuring DNA, RNA and protein synthesis by radioactive precursor incorporation. At 1 $\mu$M B(a)P did not affect preimplantation development but interfered with hatching and ICM formation. Suppressing effect of ICM formation was dose dependent. At the eight cell stage, the developmental rate was decreased at above 3 $\mu$M of B(a)P. At the blastocyst stage, attachment and trophoblast outgrowth were diminished at the 10 $\mu$M of B(a)P and ICM formation was decreased at 1 $\mu$M of B(a)P. Inner cell number of blastocyst was decreased dose dependently. So, number of ICM was one of the most sensitive and toxicological end point. The RNA incorporation rate of 0.1 $\mu ^3$H-uridine was dosedependent and the protein incroporation of 0.5 $\mu Ci ^{35}$S-methionine showed a significant decrease after 48 hrs. But the DNA incorporation rate of methyl-$^3$H thymidine was not affected. Our results suggested that B(a)P did not affect the DNA replication but transcription was inhibited by dose dependent manner. There delay of development during the blastocyst stage was mainly due to the inhibition of RNA synthesis followed by protein synthesis.

  • In this study, we demonstrate that the biological unnatural amino acid incorporation method can be utilized in vivo to synthesize an alkyne-terminated telechelic protein, Synthesis of terminally-functionalized polymers such as telechelic polymers is recognized to be important, since they can be employed usefully in many areas of biology and material science, such as drug delivery, colloidal dispersion, surface modification, and formation of polymer network. The introduction of alkyne groups into polymeric material is particularly interesting since the alkyne group can be a linker to combine other materials using click chemistry. To synthesize the telechelic recombinant protein, we attempted to incorporate the L-homopropargylglycine into the recombinant GroES fragment by expressing the recombinant gene encoding Met at the codons for both N- and C-terminals of the protein in the Met auxotrophic E. coli via Hpg supplementation. The Hpg incorporation rate was investigated and the incorporation was confirmed by MALDI-TOF analysis of the telcchelic recombinant protein.

$^{13}C$ NMR Studies of Metabolic Pathways Regulated by HSP104 in Saccharomyces cerevisiae

  • HSP104 protein in Saccharomyces cerevisiae is known to provide thermotolerance when induced by various kinds of stresses, such as a mild heat shock, ethanol, and hypoxia. It helps cells survive at an otherwise lethal temperature. Mechanisms by which HSP104 protein works are yet to be elucidated. In order to understand a molecular basis of thermotolerance due to HSP104 protein induced by a mild heat shock, studies on respiratory pathways were carried out in the wild type as well as in the hsp104 deleted mutant. Especially the degree of 13C-acetate incorporation into glutamate-C4 was examined for both strains using 13C-13C homonuclear spin coupling measurements, since glutamate is in a rapid equilibrium with α-ketoglutarate in the TCA cycle. In addition, the temperature effects on the rate of 13C incorporation are compared with or without HSP104 protein expressed. Finally, the inhibitory effect of HSP104 on the respiration pathway was confirmed by the measurements of oxygen consumption rates for both strains.

미성숙 쥐 자궁에서 Tamoxifen의 Antiestrogen 효과에 관한 연구 : II. Ribonucleic Acid 및 단백질 합성능력에 관하여 (Study on Antiestrogenic Effects of Tamoxifen in Immature Rat Uterus: II. Effects on Synthesis of Ribonucleic Acid and Protein)

  • The present study has been carried out to elucidate the antiestrogenic effects of tamoxifen on RNA and protein synthesis in uteri of immature rats. Immature female Sprague-Dawley rats were allocated into 4 groups and injected with $5{\mu}g$ of estradiol-$17{\beta}$, $50{\mu}g$ of tamoxifen, a combination of both, or vehicle only subcutaneously three times with an interval of 24 hours respectively. The specific activities of $^3H$-uridine incorporation into uterine RNA and those of $^3H$-leucine incorporation into uterine protein were measured before and 1, 3, 6, 12, 24, 48 and 72 hours after the above treatments. The results obtained were summarized as follows; 1. Tamoxifen itself increased RNA synthesis an hour after treatment(169.18% of control), but it's specific activity was reduced to control level after 3 hours. Tamoxifen inhibited significantly (p<0.01) the activity of RNA synthesis of estradiol-$17{\beta}$. 2. The increasing rate of protein synthesis was lower in tamoxifen treated group than that in estradiol-$17{\beta}$ treated group. While the rate was steadily increased up to 357.4% of control by estradiol-$17{\beta}$ in 72 hours, tamoxifen itself failed to increase the rate after 24 hours and significantly (p<0.01) inhibited the activity of estradiol-$17{\beta}$(-167.4%).

  • This study was performed to increase the availability of Sedum sarmentosum (Dolnamul) and to improve the nutraceutical value of rice cakes (sulgitteok). The contents of crude protein, mineral, dietary fiber, water holding capacity, and hardness significantly and directly increased with lyophilized sedum powder (SP). Pore ratio and expansion rate decreased in samples containing more than 10% SP compared to the control. In a sensory evaluation, a positive correlation was detected between overall acceptability and taste ($R^2$=0.99, p<0.01), and color ($R^2$=0.72, p<0.05). Total polyphenol contents of the SP-treated groups were significantly elevated, accompanied by an increase in radical scavenging ability estimated by the 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay. Replacing 10% of the rice powder with SP efficiently improved the antioxidant and nutritional values of sulgitteok as well as its the sensory quality.

Conjugated Linoleic Acid as a Key Regulator of Performance, Lipid Metabolism, Development, Stress and Immune Functions, and Gene Expression in Chickens

  • It has been well documented from animal and human studies that conjugated linoleic acid (CLA) has numerous beneficial effects on health. In chickens, CLA exerts many effects on performance ranging from egg quality and yolk lipids to meat quality. Although there are several CLA isomers available, not all CLA isomers have the same incorporation rates into egg yolk: cis-9,trans-11 and trans-10,cis-12 CLA isomers are more favorably deposited into egg yolk than other isomers investigated, but of the two isomers, the former has a higher incorporation rate than the latter. CLA alters the amounts and profiles of lipids in plasma, muscles and liver. Furthermore, increased liver weight was reported in chickens fed dietary CLA. As observed in egg yolk, marked reduction in intramuscular lipids as well as increased protein content was observed in different studies, leading to elevation in protein-to-fat ratio. Inconsistency exists for parameters such as body weight gain, feed intake, feed conversion ratio, egg production rate and mortality, depending upon experimental conditions. One setback is that hard-cooked yolks from CLA-consuming hens have higher firmness as refrigeration time and CLA are increased, perhaps owing to alterations in physico-chemistry of yolk. Another is that CLA can be detrimental to hatchability when provided to breeders: eggs from these breeders have impaired development in embryonic and neonatal stages, and have increased and decreased amounts of saturated fatty acids and monounsaturated fatty acids (MUFAs), respectively. Thus, both problems can be fully resolved if dietary sources rich in MUFAs are provided together with CLA. Emerging evidence suggests that CLA exerts a critical impact on stress and immune functions as it can completely nullify some of the adverse effects produced by immune challenges and reduce mortality in a dose-dependent manner. Finally, CLA is a key regulator of genes that may be responsible for lipid metabolism in chickens. CLA down-regulates both expression of the gene encoding stearoyl-CoA desaturase-1 and its protein activity in the chicken liver while up-regulating mRNA of sterol regulatory element-binding protein-l.

간에서의 RNA, 단백질 생합성에 미치는 인삼성분의 생화학적 연구 (Biochemical Studies of Ginseng Saponin on RNA and Protein Biosynthesis in the Rat Liver)

  • 조사포닌을 복강내 투여하면 1) 핵내의 RNA polymerase의 활성도, 2) 핵내의 RNA합성, 3) 세포질의 RNA합성,4) 세포질내의 폴리리보좀 함량, 5) in vitro 상태에서의 쥐간의 polysome과 micro-some으로의 아미노산 유입율, 6) 방사능 표지된 아미노산의 혈청 단백질로의 유입율이 증가하였음을 과거에 보고 한 바 있으며 또한 4주간 조사포닌을 투여한 쥐에서 적출한 간세포를 전자 현미경으로 조사한 결과, 조면 소포체가 상당히 증가하였으며 초원심분리기로서 막에 결합한 ribosome에서의 polysome함량의 증가를 확인하였다. 최근 streptozotocin으로 유도한 단백질 결핍성 당뇨병 쥐에 $Rb_2$를 계속적으로 주사한 결과 blood urea nitrogen과 간내의 urea 농도가 현저히 감소하였으며 혈청내의 총단백질과 알부민의 농도가 대조군의 수치에 비하여 증가한 반면 간내의 RNA와 총 ribosome, 막에 결합된 ribosome의 함량이 증가하였다. 또한 $Rb_2$투여로 혈청내의 총단백질로의 방사능 표지 전구물질의 유입량이 증가하였으며 당뇨쥐에서의 질소균형을 개선시켰다. 이러한 실험적 결과에 근거하여 인삼 사포닌은 대사를 촉진시키고 RNA, 단백질 합성 등을 촉진하는 것으로 생각된다.

  • The ability of $^{67}Ga$, administered carrier free as the citrate complex, to localize in human and animal tumors to an extent sufficient to permit visualization of the lesion by scanning is well established. However, neither the mechanism of $^{67}Ga$ uptake by tumors or inflammatory cells nor its relationship to cell type or to the biochemical status of the cell is yet understood. Author investigated the uptake of $^{67}Ga-citrate$ using subcellular tissue fractionation of rat livers treated with $CCl_4$ associated with the $^3H-thymidine$ incorporation rate to detect subcellular localization of $^{67}Ga$ and it's relationship in DNA synthesis. Large amounts of $^{67}Ga$ associated with the soluble portion of tissue homogenate rather than with isolated cell organelles and not related nuclei residue in the regenerating period after hepatocellular injury caused by $CCl_4$. The elevated uptake of $^{67}Ga$ in the livers of $CCl_4$ treated rats was also inhibited when protein synthesis was stopped by cyclohexamide. Thus protein and the soluble portion of issue homogenates seems to play an important role in the elevated uptake of $^{67}Ga$ in liver injury induced by $CCl_4$ treated rats.
