Apparent Ileal Digestibility of Nutrient in Plant Protein Feedstuffs for Finishing Pigs

  • Five barrows (average initial body weight 58.6 kg) were used to determine the apparent ileal digestibilities of amino acids, DM, N and energy in various soybean meal, rapeseed meal and coconut meal in finishing pigs. Dietary treatments included 1) KSBM (Korean soybean meal), 2) CSBM (Chinese soybean meal), 3) ASBM (Argentine soybean meal), 4) RSM (Rapeseed meal), and 5) CNM (Coconut meal). The diets were corn starch-based and formulated so that each protein source provided the same amount of total ME (3,490 kcal/kg), CP (15.70%), lysine (1.00%), Ca (0.80%) and P (0.60%). Protein content of the KSBM was higher than the CSBM and ASBM, with all values similar to those expected, and protein content of the CNM was lower than that of the SBM preparation and RSM. Apparent ileal digestibilities of histidine, lysine, threonine, alanine, asparatic acid, cystine, glutamic acid and serine were greater for the KSBM, CSBM, ASBM and RSM than for the CNM (p<0.05). Also, the apparent ileal digestibilities of methionine, leucine, phenylalanine, valine and tyrosine were greater for the KSBM than for the CSBM, ASBM, RSM and CNM (p<0.05). Overall, the apparent ileal digestibilities of total essential amino acids were greater for the KSBM than for the CSBM, ASBM, RSM and CNM (p<0.05), and the apparent ileal digestibilities of total non essential amino acids were greater for the KSBM, CSBM, ASBM and RSM than for the CNM (p<0.05). No difference (p>0.05) in apparent digestibility of DM at the small intestine was observed among the treatments. However, the apparent digestibility of DM at the total tract was greater for the KSBM than for the CSBM, ASBM, RSM and CNM (p<0.05). Also, apparent digestibilities of N and digestible energy at the small intestine and total tract were greater for the KSBM than for the RSM and CNM (p<0.05). In conclusion, nutrient digestibility values of SBM preparations and RSM were relatively high compared to CNM.

Evaluation Effects of Spray-dried Egg Protein Containing Specific Egg Yolk Antibodies as a Substitute for Spray-dried Plasma Protein or Antibiotics in Weaned Pigs

  • In Exp. 1, a total of 36 pigs (6.55$\pm$0.10 kg average initial body weight and 21 d average age) were used in a 14 d growth study to determine the effects of replacing spray-dried plasma protein (SDPP) with spray-dried egg protein containing specific egg yolk antibody (SDEP) on growth performance and nutrient digestibility in weaned pigs. The pigs were blocked by weight and assigned to treatments based on sex. There were three pigs per pen and four pens per treatment. Dietary treatments were 0, 3, or 6% SDEP and contained 6, 3, or 0% SDPP, respectively. Through the entire experimental period, average daily gain (ADG), average daily feed intake (ADFI), and gain/feed tended to decrease as the concentration of SDEP increased in the diets. However, there were no significant differences among the treatments (p>0.05). As the addition of SDEP in the diets increased, apparent digestibilities of dry matter (DM) and nitrogen (N) were decreased without significant (p>0.05). For Exp. 2, 36 pigs (2.63$\pm$0.04 kg average initial body weight and 10 d average age) were used in a 14 d growth study to determine the effects of antibiotic replacement with SDEP on growth performance and nutrient digestibility in early-weaned pigs. The pigs were blocked by weight and assigned to treatments based on sex. There were three pigs per pen and four pens per treatment. Dietary treatments included 1) ANTIBIOTIC (corn-dried whey-soybean meal based diet+0.08% antibiotics, 4 mg of tiamuline hydrogen fumarate; 10 mg of sulfadimidine per kg of complete diet), 2) SDEP0.1 (corndried whey-SBM based diet+0.1% SDEP), and 3) SDEP0.2 (corn-dried whey-SBM based diet+0.2% SDEP). ADG and gain/feed of pigs fed the SDEP0.2 diet were higher than for pigs fed the ANTIBIOTIC diet without significant (p>0.05). Pigs fed the diet with SDEP0.2 tended to have increased apparent digestibilities of DM and N compared to pigs fed the ANTIBIOTIC diet without significant (p>0.05). In conclusion, the dietary SDEP seemed to be partial replacing the SDPP portion of high nutrient dense diet for weaned pigs. Also, dietary SDEP seemed to be approximately 0.2% or more when the pigs fed the antibiotic-free diet for early-weaned pigs.

A PAS-Containing Histidine Kinase is Required for Conidiation, Appressorium Formation, and Disease Development in the Rice Blast Fungus, Magnaporthe oryzae

  • Rice blast disease, caused by the ascomycete fungus Magnaporthe oryzae, is one of the most important diseases in rice production. PAS (period circadian protein, aryl hydrocarbon receptor nuclear translocator protein, single-minded protein) domains are known to be involved in signal transduction pathways, but their functional roles have not been well studied in fungi. In this study, targeted gene deletion was carried out to investigate the functional roles of the PAS-containing gene MoPAS1 (MGG_02665) in M. oryzae. The deletion mutant ΔMopas1 exhibited easily wettable mycelia, reduced conidiation, and defects in appressorium formation and disease development compared to the wild type and complemented transformant. Exogenous cAMP restored appressorium formation in ΔMopas1, but the shape of the restored appressorium was irregular, indicating that MoPAS1 is involved in sensing the hydrophobic surface. To examine the expression and localization of MoPAS1 in M. oryzae during appressorium development and plant infection, we constructed a MoPAS1:GFP fusion construct. MoPAS1:GFP was observed in conidia and germ tubes at 0 and 2 h post-infection (hpi) on hydrophobic cover slips. By 8 hpi, most of the GFP signal was observed in the appressoria. During invasive growth in host cells, MoPAS1:GFP was found to be fully expressed in not only the appressoria but also invasive hyphae, suggesting that MoPAS may contribute to disease development in host cells. These results expand our knowledge of the roles of PAS-containing regulatory genes in the plant-pathogenic fungus M. oryzae.

Mitochondrial Genome Sequences of Spirometra erinaceieuropaei and S. decipiens (Cestoidea: Diphyllobothriidae)

  • The present study was performed to compare the mitochondrial genomes between 2 Spirometra tapeworms, Spirometra erinaceieuropaei and Spirometra decipiens (Cestoidea: Diphyllobothriidae), which larval stages are important etiological agents of sparganosis in humans. For each species, the full mitochondrial genome was amplified in 8 overlapping fragments using total genomic DNA purified from a single worm as the template. The mitochondrial genomes were 13,643 bp (S. erinaceieuropaei) and 13,641 bp (S. decipiens) in length and contained 36 genes; 12 protein-coding genes, 2 ribosomal RNA (rRNA, small and large subunits), and 22 transfer RNAs (tRNAs). The 12 protein-coding genes constituted 10,083 bp (S. erinaceieuropaei) and 10,086 bp (S. decipiens) of their respective mitochondrial genomes. The tRNA genes, ranging in length from 56 to 70 bp, were identified based on putative secondary structures such as the typical cloverleaf shape. A total of 23 intergenic sequences, varying from 1 to 204 bp in size, were interspersed in S. erinaceieuropaei (total, 504 bp) and S. decipiens (total, 496 bp) mtDNA. The 12 protein-coding genes of S. erinaceieuropaei and S. decipiens differed by 12.4%, whereas the overall difference in mtDNA sequence between S. erinaceieuropaei and S. decipiens was 12.9%. Thus, from the standpoint of the mitochondrial genome, S. decipiens represents a valid species that can be distinguished from S. erinaceieuropaei.

  • AMP-activated protein kinase sucrose non-fermenting 1 (Snf1) is a representative regulator of energy status that maintains cellular energy homeostasis. In addition, Snf1 is involved in the mediation of environmental stress such as salt stress. Snf1 regulates metabolic enzymes such as acetyl-CoA carboxylase, indicating a possible role for Snf1 in metabolic regulation. In this article, we performed nuclear magnetic resonance (NMR) spectroscopy to profile the metabolic changes induced by Snf1 under environmental stress. According to our NMR data, we suggest that Snf1 plays a role in regulating cellular concentrations of a variety of metabolites during environmental stress responses.

Complete Sequence of the Mitochondrial Genome of Spirometra ranarum: Comparison with S. erinaceieuropaei and S. decipiens

  • This study was undertaken to determine the complete mitochondrial DNA sequence and structure of the mitochondrial genome of Spirometra ranarum, and to compare it with those of S. erinaceieuropaei and S. decipiens. The aim of this study was to provide information of the species level taxonomy of Spirometra spp. using the mitochondrial genomes of 3 Spirometra tapeworms. The S. ranarum isolate originated from Myanmar. The mitochondrial genome sequence of S. ranarum was compared with that of S. erinaceieuropaei (GenBank no. KJ599680) and S. decipiens (GenBank no. KJ599679). The complete mtDNA sequence of S. ranarum comprised 13,644 bp. The S. ranarum mt genome contained 36 genes comprising 12 protein-coding genes, 22 tRNAs and 2 rRNAs. The mt genome lacked the atp8 gene, as found for other cestodes. All genes in the S. ranarum mitochondrial genome are transcribed in the same direction and arranged in the same relative position with respect to gene loci as found for S. erinaceieuropaei and S. decipiens mt genomes. The overall nucleotide sequence divergence of 12 protein-coding genes between S. ranarum and S. decipiens differed by 1.5%, and 100% sequence similarity was found in the cox2 and nad6 genes, while the DNA sequence divergence of the cox1, nad1, and nad4 genes of S. ranarum and S. decipiens was 2.2%, 2.1%, and 2.6%, respectively.

Effect of Grinding on Color and Chemical Composition of Pork Sausages by Near Infrared Spectrophotometric Analyses

  • Near Infrared spectroscopy was applied to the samples of processed pork to see the effect of grinding on chemical components analyses. Data from conventional chemical analyses of moisture, fat, protein, NaCl were put into calibration model by NIR of reflectance mode. The other properties observed were pH and color parameters ($L^*,\;a^*,\;b^*$). Spectral ranges of 400~2500 nm and 400~1100 nm were compared for color parameters. Spectral ranges of 400~2500 nm and 1100~2500 nm were compared for chemical components and pH. Different spectral ranges caused little changes in the coefficients of determination or standard errors. $R^{2,}s$ of calibration models for color parameters were in the range of 0.97 to 1.00. $R^{2,}s$ of calibration models of intact sausages for moisture, protein, fat, NaCl and pH were 0.98, 0.89, 0.95, 0.73 and 0.77, respectively using spectra at 1100~2500 nm. $R^{2,}s$ of calibration models of ground sausages for moisture, protein, fat, NaCl and pH were 0.97, 0.91, 0.97, 0.42 and 0.56, respectively using spectra at 1100~2500 nm.

Enzymatic Activities of Allergen Extracts from Three Species of Dust Mites and Cockroaches Commonly Found in Korean Home

  • Allergen extracts from dust mites and cockroaches commonly found in Korean homes were used to evaluate their enzymatic activity as they are believed to influence allergenicity. Allergen extracts were prepared from 3 dust mite species (Dermatophagoides farinae, D. pteronyssinus, and Tyrophagus putrescentiae) and 3 cockroach species (Blattella germanica, Periplaneta americana, and P. fuliginosa) maintained in the Korea National Arthropods of Medical Importance Resource Bank. Proteins were extracted in PBS after homogenization using liquid nitrogen. The activities of various enzymes were investigated using the API Zym system. No significant difference in phosphatase, lipase, or glycosidase activity was observed among the 6 allergen extracts, but much difference was observed in protease activity. Protease activity was assessed in more detail by gelatin zymography and the EnzChek assay. Extract from T. putrescentiae showed the highest protease activity, followed by those of the cockroach extracts. Extracts from D. farinae and D. pteronyssinus showed only weak protease activity. Gelatinolytic activity was detected mainly in a 30-kDa protein in D. farinae, a 28-kDa protein in D. pteronyssinus, a > 26-kDa protein in T. putrescentiae, a > 20-kDa protein in B. germanica, and a > 23-kDa protein in P. americana and P. fuliginosa. The information on various enzymatic activities obtained in this study may be useful for future studies. In particular, the strong protease activity found in cockroach extracts could contribute to sensitization to cockroach allergens, which is known to be associated with the development of asthma.

The CsSTE50 Adaptor Protein in Mitogen-Activated Protein Kinase Cascades Is Essential for Pepper Anthracnose Disease of Colletotrichum scovillei

  • Anthracnose, caused by the ascomycete fungus Colletotrichum scovillei, is a destructive disease in pepper. The fungus germinates and develops an infection structure called an appressorium on the plant surface. Several signaling cascades, including cAMP-mediated signaling and mitogen-activated protein kinase (MAPK) cascades, are involved in fungal development and pathogenicity in plant pathogenic fungi, but this has not been well studied in the fruit-infecting fungus C. scovillei. Ste50 is an adaptor protein interacting with multiple upstream components to activate the MAPK cascades. Here, we characterized the CsSTE50 gene of C. scovillei, a homolog of Magnaporthe oryzae MST50 that functions in MAPK cascades, by gene knockout. The knockout mutant ΔCsste50 had pleiotropic phenotypes in development and pathogenicity. Compared with the wild-type, the mutants grew faster and produced more conidia on regular agar but were more sensitive to osmotic stress. On artificial and plant surfaces, the conidia of the mutant showed significantly reduced germination and failed to form appressoria. The mutant was completely non-pathogenic on pepper fruits with or without wounds, indicating that pre-penetration and invasive growth were both defective in the mutant. Our results show that the adaptor protein CsSTE50 plays a role in vegetative growth, conidiation, germination, appressorium formation, and pathogenicity in C. scovillei.

Effects of Plant Protein Source Containing Multienzyme on Performance and Milk Characteristics in Sow (포유모돈에 있어 복합효소제 함유 식물성 단백질 공급원이 생산성과 돈유성상에 미치는 영향)

  • A total of thirty sows(Landrace×Yorkshire) were used to determine the effects of plant protein source containing multienzyme on performance, nutrients digestibility and milk characteristics. A feeding trial was conducted for 21 days from parturition to weaning. Experimental diets were supplied for 1 week before the parturition day and throughout the experimental period. Dietary treatments included: 1) Control(CON; basal diet), 2) CGLT(included corn gluten) and 3) FSPM(included fermented soy protein containing multienzyme). Through the entire experimental period, backfat loss and return-to-estrus intervals were not affected by the treatments(P>0.05). Nitrogen digestibility was increased significantly(P<0.05) in FSPM treatment compared to CON treatment. Blood urea nitrogen(BUN) concentration was increased significantly (P<0.05) in FSPM treatment compared to CON treatment. At the initial  period, total protein content of milk was higher significantly(P<0.05) in FSPM treatment compared to CGLT treatment and at the final period, total fat content of milk was higher significantly(P<0.05) in FM treatment compared to CON treatment. Rectal temperature showed similar tendency of change among treatments. The final piglet body weight, weight gain and ADG were higher significantly in FSPM treatment compared to CON treatment. On diarrhea rate in piglet, just one piglet occurred in CGLT treatement. In conclusion, 2.5% dietary plant protein source containing multienzyme suppelmentation improved N digestibility, BUN concentration, fat and protein contents in milk and weight gain in piglet.
