Species belonging to the dinoflagellate genus Prorocentrum are known to cause red tides or harmful algal blooms. To understand the dynamics of a Prorocentrum sp., its growth and mortality due to predation need to be assessed. However, there are only a few Prorocentrum spp. for which heterotrophic protist predators have been reported. We explored feeding by the common heterotrophic dinoflagellates Gyrodinium dominans, Oxyrrhis marina, Pfiesteria piscicida, Oblea rotunda, and Polykrikos kofoidii and the naked ciliate Strombidinopsis sp. (approx. 90 ㎛ cell length) on the planktonic species Prorocentrum triestinum, P. cordatum, P. donghaiense, P. rhathymum, and P. micans as well as the benthic species P. lima and P. hoffmannianum. All heterotrophic protists tested were able to feed on the planktonic prey species. However, O. marina and O. rotunda did not feed on P. lima and P. hoffmannianum, while G. dominans, P. kofoidii, and Strombidinopsis sp. did. The growth and ingestion rates of G. dominans and P. kofoidii on one of the seven Prorocentrum spp. were significantly different from those on other prey species. G. dominans showed the top three highest growth rates when it fed on P. triestinum, P. cordatum, and P. donghaiense, however, P. kofoidii had negative growth rates when fed on these three prey species. In contrast, P. kofoidii had a positive growth rate only when fed on P. hoffmannianum. This differential feeding on Prorocentrum spp. between G. dominans and P. kofoidii may provide different ecological niches and reduce competition between these two common heterotrophic protist predators.
The dynamics and seasonal succession of dinoflagellate community, and their correlations with environmental parameters were investigated during the period from April, 1986 to March, 1987 at 6 selected stations in Masan Bay, Korea. The dinoflagellate standing crops varied extensively with months and stations: ranging from 44 to 2,789,900 cella/ l at surface layer and from 52 to 165,714 cells/ l at bottom layer, respectively. The distribution of standing crops by size class of dinoflagellate species showed that a group of 20-um size class was predominant throughout the present survey, since the most of dominant species have belonged to the size category. Among dominant species, Gyrodinium fissum was most dominant throughout the year in all sampling stations, particulary in June. Scripsiella trochoidea was predominant in May, Protoperidinium bipes and Prorocentrum minimum from June to September, Prorocentrum triestinum, P micans and Ceratium fusus from September to December, and Heterocapsa triquetra from December to April. The stepwise multiple regression analysis between dinoflagellate and environmental parameters revealed that salinity, nitrate-N, pH, and transparency were correlated with the variations of standing crops of dinoflagellate.
This study presents a molecular phylogenetic analysis of the harmful dinoflagellate Cochlodinium polykrikoides, by use of partial sequence of small subunit (SSU) rRNA gene from most of the major taxa(24 species) in dinoflagellates. The class Dinophyceae clade formed a strong monophyletic relationship with C. polykrikoides and several taxa. On the basis of deeper nodes, the phylogenetic relationships placed C. polykrikoides closer to the order Prorocentrales rather than to the order Gymnodiniales, which was supported by a strong bootstrap value (100%) in the analyses of Neighbor-Joining and Parsimony methods. There is strong support for C. polykrikoides being placed in the same branch as Gymnodiniaceae and being connected in a clade with Prororcentrum micans among Prorocentrales. Morphological data show that C. polykrikoides is well associated with the genus Gyrodinium; however, this species is genetically closer to Gymnodinium than to Gyrodinium. The placement of C. polykrikoides always formed an independent branch separated from other dinoflagellates. In conclusion, planktonic P. micans plays an important role as an ancestor of Gymnodinium, whereas C. polykrikoides appears to be used an intermediate position between P. micans and Gymnodinium based on evolution.

This study was carried out to assess the feasibility of the electron beam irradiation as a mean of red tide control in coastal water. Prorocentrum minimum, Prorocentrum micans, Cochlodinium polykrikoides, Heterosigma akashiwo, Alexnadrium catenella were selected and cultured for experiments, and red tide occurring in Tongyeong(2007. 8. 15) was also tested under the same conditions. The irradiation dose were 1 kGy, 2 kGy, 4 kGy and 8 kGy. The result showed 50~65% extinction in red tide cells was observed right after irradiation dose of 1 kGy and 86~97% within 1 day after irradiation, compared with control. Chlorophyll-a concentration of red tide was reduced by 50~64% immediately and it was drastically reduced up to 86~97% 1 day after irradiation. When the culture was irradiated at 1 kGy, 28~47% of s-protein was released immediately, and 77~138% was released 1day after irradiation. 77~212% of s-carbohydrate was excreted after 1 day while 16~45% of s-carbohydrate was excreted immediately. A transmission electron microscope(TEM) observation for the irradiated red tide revealed that the cell was destroyed and intracellular biopolymeric substance was leached out from the damaged cell as a result of electron beam irradiation. These results imply that electron beam irradiation is enable to control red tide by flocculation with extracellular biopolymer. The paralytic shellfish poisoning(PSP) toxin contents produced by Alexandrium catenella was decreased 48% by 1 kGy of electron beam irradiation compared with the unirradiated cells. As a result, electron beam irradiation was effective for detoxication as well as destruction of red tide.
A series of experiments was conducted to identify the potential of five phytoplankton species as standard test species for marine ecotoxicological tests. The candidate phytoplankton species are Skeletonema costatum, Heterosigma akashiwo, Prorocentrum micans, Isochrysis galbana, and Tetraselmis suecica. Salinity tolerance and sensitivity on potassium dichromate as a reference material were identified. Toxicity of eleven ocean dumped sewage sludges and four red tide expellent extracts were estimated by the inhibition of population growth rates (PGR) of marine diatom S. costatum, While most species revealed relatively weak tolerance on salinity, T. suecica demonstrated the highest salinity tolerance ranged from $5\~35$ psu and the others $15\~35$ psu. H. akashiwo revealed the highest sensitivity as 72h $IC_{50}$=0.76mg/L and T. suecica the lowest as 72h $IC_{50}$=8.89mg/L on potassium dichromate. Sludge extracts from industrial waste, domestic sewage and livestock farm waste sludge showed high toxicity as 72h $IC_{50}$<$2\%$ and lowest toxicity from filtration bed sludge as 72h $IC_{50}$=$30.50\%$ NOEC (No Observed Effective Concentration) of sludge extract ranged from <$0.4\%$ to $1.6\%$ and this indicated high phytotoxicity of ocean dumped sewage sludge. The test sensitivity of phytoplankton PGR inhibition was much higher than those of marine rotifer Brachionus plicatilis mortality test and bioluminescent inhibition test by marine bacteria Vibrio fischeri, and comparable with the sea urchin (Strongylocentrotus intermedius) fertilization test. As a result the phytotoxicity test using phytoplankton PGR inhibition ($IC_{50}$) must be a useful tool for marine phyto-toxicological evaluation of ocean dumped materials.

To provide essential information of the role of marine bacteria on the dinoflagellate blooms, distribution of marine bacterial flora and dinoflagellate species was investigated in Chinhae Bay located in southern part of Korea from August 1989 to April 1990. Two hundred and fifty one strains of marine bacteria were isolated from seawater samples collected from the study area. Among them, Flavobacterium spp. and Acinetobacter spp. were the most dominant in bacterial flora. Another 32 strains which comprised 13 percent of total strains were Erythrobacter spp.. Based on the physiological character, Erythrobacter spp. were identified as Erythrobacter longus, Erythrobacter sp.(J-2) and Erythrobacter sp. (J-8). From the phytoplanktonic community, fourteen genera and twenty nine taxa of dinoflagellate species were identified. Based on the spatio-temporal frequence and abundance Gymnodinium sanguneum, Prorocentrum micans and Prorocentrum minimum were the aestival dominent species. However, Heterocapsa triquetra was appeared as predominant species in April. Cell density of about 2,000 cells/ml was prevailed in the bloom of August, but it developed into more intensive bloom of above 500 cells/ml in September. The water quality showed eutrophic or hypereutrophic condition, which was proved by high concentration of dissolved inorganic nitrogen, ortho-phosphate and chemical oxygen demand. Oxygen deficient water mass was found in the bottom overlying waters in August and September. High relationship between abundant bacterial flora and persistent dinoflagellate blooms in eutrophic condition would be approvable.

A series of experiments were conducted to establish a marine ecotoxicological standard method using marine primary producers, Skeletonema costatum, Heterosigma akashiwo, Prorocentrum micans, Isochrysis galbana, Tetraselmis suecica as candidate test species. Marine diatom, S. costatum was selected as standard test species in terms of the ecological roles and international uses as test species. Test methods and test acceptability criteria are as follows; $72{\sim}96$ hr population growth inhibition $EC_{50}$ as endpoint. Static non-renewal method, and population growth rates over 0.04/hr in control as test acceptability criterium. S. costatum is widely distributed in the world ocean and used for standard species of marine toxicity test method by ISO (International Standardization Organization). Possible salinity ranges for this test method are $20{\sim}35\;psu$, and reproducibility and interlaboratory test results were consistent through the calibration tests. Sensitivity of the test method was comparable or better than other toxicity tests such as rotifer neonate mortality, bioluminescent bacterial inhibition, seaweed sporulation and sea urchin fertilization tests.

To develop an easy and rapid method of quantifying lipid contents of marine dinoflagellates, we quantified lipid contents of common dinoflagellate species using a colorimetric method based on the sulpho-phospho-vanillin reaction. In this method, the optical density measured using a spectrophotometer was significantly positively correlated with the known lipid content of a standard oil (Canola oil). When using this method, the lipid content of each of the dinoflagellates Alexandrium minutum, Prorocentrum micans, P. minimum, and Lingulodinium polyedrum was also significantly positively correlated with the optical density and equivalent intensity of color. Thus, when comparing the color intensity or the optical density of a sample of a microalgal species with known color intensities or optical density, the lipid content of the target species could be rapidly quantified. Furthermore, the results of the sensitivity tests showed that only $1-3{\times}10^5cells$ of P. minimum and A. minutum, $10^4cells$ of P. micans, and $10^3cells$ of L. polyedrum (approximately 1-5 mL of dense cultures) were needed to determine the lipid content per cell. When the lipid content per cell of 9 dinoflagellates, a diatom, and a chlorophyte was analyzed using this method, the lipid content per cell of these microalgae, with the exception of the diatom, were significantly positively correlated with cell size, however, volume specific lipid content per cell was negatively correlated with cell size. Thus, this sulpho-phospho-vanillin method is an easy and rapid method of quantifying the lipid content of autotrophic, mixotrophic, and heterotrophic dinoflagellate species.
In this study, the PHYTO-PAM-fluorometric method was used to evaluate the ETR$_{max}$ in terms of sensitivity to DIN/DIP against 14 microalgae: Prorocentrum micans, Heterocapsa triquetra, Gymnodinium impudicum, Cymnodinium catenatum, Amphidinium caterae, Chlorella vulgaris, Chroococcus minutus, Microcystis aeruginosa, Chlorella ellipsoidea, Nannochloris oculata, Oocystis lacustris, Chroomonas salina, Gloeocystis gigas, and Prymnessium parvum. We found that P. micans, H. triquetra, and A. caterae exposed to the maximum level of DIN/DIP were significantly smaller in the ETR$_{max}$ than that of the minimum and moderate mixture. Unlikely the ETR$_{max}$, the initial slope alpha was not significantly different at the level of 60 DIN/DIP. In G. catenatum, the moderate levels of 15 and 20 in DIN/DIP were found to be significantly different from the ETR$_{max}$ at Chl-Ch4. Gymnodinium impudicum had a higher value than that of the ETR$_{max}$ than that of dinoflagellates used in this study, ranging from 306.1 (Ch4, DIN/DIP: 10) to 520.1 (Ch4, DIN/DIP: 30). The ETR$_{max}$ value obtained from other microalgae was similar to C. impudicum at any of the ratios of DIN/DIP and channels. Consequently, the influence of offshore water current assures us of the suppression of photosynthesis and electron transport rate in dinoflagellates. Gymnodinium impudicum has not been researched in the area of red tides in Korea, but it will be enough to creat the massive algal blooms in the future because of higher potential photochemical availability.
This survey was carried out to assess important species of phytoplankton in the ocean and bay in Korea from 32 references. The number of important species assessed from 50 genera 116 species. Of them, Bacillariophycea(diatoms) 34 genera 89 speceis(76.6%), Dinophyceae(dinoflagellates) 11 genera 22 species(18.9%), Chrysophyceae 2 genera 2 species(1.7%), Cyanophycea(blue-green algae), Raphidophyceae, Euglenophyceae(euglenoids) 1 genera 1 species(0.9%) respectively. By ecological characters, the number of dominant species were 79 species including Actinoptychus seranius, 50 species including Coscinodiscus centralis were recorded as frequently apperaing species and 36 species including Cochlodinium polykrikoides were recorded as red-tide causative organism. Also, 11 species including Prorocentrum micans were surveyed as the indicator including all ecological characters.
