Herbal mixture water extract of (Chrysanthemum morifolium, Portulaca oleracea, Sanguisorba officinalis, Sophora flavescens, Phellodendron amurense, Cnidium monnieri) which exhibit several beneficial effects including acne and skin diseases, was tested for anti-microbial activity and anti-inflammation effects. The herbal mixture extract showed antimicrobial activity against Stapylococcus epidermis, and Propionibacterium acne. The growth of Stapylococcus epidermis, and Propionibacterium acne was inhibited completely by addition of 1.0% of the extract. Also in the present study we examined the mixture extract on compound 48/80 induced allergy and LPS induced cyclooxygenase-2(COX-2) gene expression in RAW 264.7 macrophage. The results indicated the ear swelling and histamine release induced by compound 48/80 were dose-dependently reduced, ranging 28-60%, and 48-72% , respectively. Furthermore the extract inhibited the expression of LPS-induced COX-2 proteins and mRNAs without an appreciable cytotoxic effects on RAW 264.7 cells. The LPS-induced cytokine gene expression including IL-$1{\beta}$, TNF-$\alpha$, and IL-6 were dose-dependently suppressed by the mixture extract. Based on these results, it is concluded that the herbal mixture water extract can be applied to the acne and skin diseases therapy.
Acne vulgaris is a common dermatological condition affecting the pilosebaceous units and having a multifactorial etiology. In Unani terminology, acne vulgaris is referred as Busoor Labaniya, characterized by white lesions on the face, nose, and cheeks; on squeezing, release cheesy material. In conventional medicine, mild cases are best addressed with topical regimens, but more severe cases require systemic medications. Retinoids (retinoic acid, adapalene, isotretinoin, tazarotene), benzoyl peroxide, clindamycin, erythromycin, and azelaic acid are a few examples of topical medications, whereas systemic drugs include antibiotics (Doxycycline, minocycline, erythromycin, azithromycin). In the Unani System of Medicine, numerous single and compound drugs have been used to treat Busoor Labaniya. These drugs are considered harmless and do not have any major side effects. In Unani system of medicine, acne vulgaris is effectively managed with natural medicines as well as therapeutic regimens with minimal side effects even after long-term usage. Acne vulgaris is usually treated with systemic blood purifiers along with topical Unani drugs. Hence, clinical studies with proper scientific parameters are needed to be conducted to establish and validate their efficacy in the prevention and control of acne vulgaris.
The in vitro antibacterial activity against antibiotic-resistant Propionibacterium acnes of kaempferol isolated from the Impatiens balsamina alone and in combination with erythromycin or clindamycin antibiotics was investigated. The antibiotic combination effect against antibiotic-resistant P. acnes was studied by checkerboard test. Kaempferol and quercetin demonstrated antibacterial activities against P. acnes. Minimum inhibitory concentrations (MICs) for both compounds were ${\leq}32\;{\mu}g/ml\;and\;{\leq}64{\mu}g/ml$ for clindamycin-sensitive and -resistant P. acnes, respectively. The four combination formulations (kaempferol and either erythromycin or clindamycin; quercetin and either erythromycin or clindamycin) exhibited a synergic inhibition of P. acnes growth. The combination of kaempferol with quercetin showed an indifferent effect. The combination of clindamycin with kaempferol or quercetin showed a greater synergic effect than that of erythromycin with kaempferol or quercetin. Thus, these combinations demonstrated the potential to treat acne.
The intradermal injection of Propionibacterium acnes (ATCC 6919) into the ears of female Sprague-Dawley rats produced a chronic inflammation with the formation of acneiform lesions. Inflammation was characterized by more than four times of ear thickness and 2.8 times of ear weight at day 21. Histologically, massive infiltration of neutrophils, macrophage and lymphocytes, hyperplastic epidermis, comedones containing keratin mass and inflammatory materials were observed. Both ginseng saponin and extract from Korean red ginseng significantly redliced the ear thickness and their effects were similar to that of benzoyl peroxide. Ginseng samples and beneoyl peroxide modified lipid constituents of P. acnesinjected rat ear tissues. Even though no marked histological changes in inflammatory lesions were observed in ginseng-treated ear tissues, Korean red ginseng showed a possibility of reduce in the risk of acne development.
Leukocytes are reportedly the first line of the innate immune defense and essential for the control of common bacterial infections. Therefore, in this work, the antibacterial activity of crocodile leukocyte extract against Propionibacterium acnes was evaluated, and we also characterized the related activity of skin infection. The leukocyte extract showed the minimum inhibitory concentration to be $100{\mu}g/ml$ to P. acnes. SEM imaging demonstrated that the leukocyte extract adversely affected P. acnes cell permeability in a concentration-dependent manner. Furthermore, the crocodile leukocyte extract could significantly reduce proinflammatory markers and decrease inflammatory signs in infected mouse ears. The crude leukocyte extract was further purified using FPLC and RP-HPLC. The resulting fraction F5 was indicated as the anti-acne peptide-containing fraction. The molecular mass of the peptide contained in F5 was calculated to be 4,790.5 Da. N-Terminal sequencing revealed the amino acid sequence as GPEPVPAIYQ, which displays similarities to immunoglobulin A and leucine-rich repeat neuronal protein. This is the first reported amino acid sequence of a crocodile leukocyte extract that possesses anti-acne activity. To attempt to use it in a prototype cosmetic, an anti-acne gel containing crude crocodile leukocyte extract was formulated, resulting in seven gel formulations (G1, G2, G3, G4, G5, G6, and G7). The formulations G5, G6, and G7 exhibited 2-fold higher anti-acne activity than G1-G4. Investigation of accelerating stability studies of anti-acne gel formulations G5, G6, and G7 demonstrated that a low storage temperature ($4^{\circ}C$) is suitable for maintaining the physical properties and biological activity of the anti-acne gel products.
Propionibacterium acnes is the predominant organism in sebaceous regions of the skin and is thought to play an important role in the pathogenesis of inflamed lesions. Antibacterial compounds against P. acnes were isolated from the ethanol extract of Kaempferia pandurata Roxb. and identified as panduratin A and isopanduratin A. Minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of panduratin A for P. acnes were 2 and $4{\mu}g/mL$, respectively, while those of isopanduratin A were 4 and $8{\mu}g/mL$, respectively. The time-dependent killing effect showed that panduratin A and isopanduratin A completely inhibited the growth of P. acnes at 4 and $8{\mu}g/mL$ in 48 hr, respectively. Panduratin A and isopanduratin A demonstrated high antibacterial activities not only against P. acnes but also other skin microorganisms. The results suggest that panduratin A and isopanduratin A could be employed as natural antibacterial agents to inhibit the growth of acne and skin disease causing microorganisms.

The $C^{14}$ -labeled lipids were extracted and determined by liquid scintilation counting, Coptidis rhizoma markedly inhibited sebum production, 3. Propionibacterium acnes is the pharmacological target site of antiacne drugs. We have examined the antiacne activity of ninety seven natural products which have been used as Korean traditional medicines in various skin disorders. The antibacterial activity of extracts from the natural products were evaluated against P. acnes ATCC 9616 by disc method. Twelve natural products showed the potent antibacterial activity against P. acnes, and were, selected for the minimal inhibitory concentration(MC) against P. acnes. MICs of nine extracts were below 0.3% (w/v) and Sophora flavescens showed the most potent activity with a MIC of less than 0.008%(w/v) against P. acnes. Thus, the results suggest that nine natural products including S. flavescens can be developed as sources of promising potent antiacne agents.

Objective : Propionibacterium acnes (P. acnes) is a major pathogenic bacteria for acne vulgaris. This study was performed to evaluate the effects of Lithospermum erythrorhizon extracts on the inflammatory cytokines gene expression by P. acnes in human keratinocytes, HaCaT cell line. Methods : Anti-bacterial activity and cytotoxicity of LE extracts was analyzed by agar plate culture and XTT assay. The cytokines gene expressions were assessed by real time RT-PCR for IL-8, MCP-1 and TNF-${\alpha}$. During the cell culture and treatments, amounts of secreted TNF-${\alpha}$ were measured by ELISA. Translocation of transcription factor NF-${\kappa}B$ from cytoplasm into nucleus was observed by immunocytochemistry and confocal microscopy. Results : There were no anti-bacterial effects and cytotoxicity as high as $1,000{\mu}g/ml$ of LE extracts in XTT assay. Transcription levels of inflammatory cytokines, IL-8, MCP-1 and TNF-${\alpha}$ were increased by P. acnes in HaCaT. LE extracts decreased the upregulated gene transcription levels. However, amounts of secreted TNF-${\alpha}$ were similar in HaCaT cells with P. acnes and LE extracts. Translocation of NF-${\kappa}B$ into nucleus by P. acnes was significantly inhibited by LE extracts. Conclusions : From the results of this study, LE extracts have anti-inflammatory effects on HaCaT cells by P. acnes that decreased the mRNA expressions of IL-8, MCP-1 and TNF-${\alpha}$. This anti-inflammatory effects of LE extracts could provide the potential of therapeutic substance for acne vulgaris.
