  • Activation of the Wnt/β-catenin pathway plays a pathogenic role in age-related macular degeneration (AMD) and is thus a potential target for the development of therapeutics for this disease. Here, we demonstrated that Wnt5a antagonized β-catenin response transcription (CRT) induced with Wnt3a by promoting β-catenin phosphorylation at Ser33/Ser37/Thr41 and its subsequent degradation in human retinal pigment epithelial (RPE) cells. Wnt5a decreased the levels of vascular endothelial growth factor (VEGF), tumor necrosis factor-α(TNF-α), and nuclear factor-κB (NF-κB), which was up-regulated by Wnt3a. Furthermore, Wnt5a increased E-cadherin expression and decreased cell migration by down-regulating Snail expression, thereby abrogating the Wnt3a-induced epithelial-mesenchymal transition (EMT) in human RPE cells. Our findings suggest that Wnt5a suppresses the pathogenic effects of canonical Wnt signaling in human RPE cells by promoting β-catenin phosphorylation and degradation. Therefore, Wnt5a has significant therapeutic potential for the treatment of AMD. [BMB Reports 2015; 48(9): 525-530]

  • The present study investigated the sedative effects of Sophora flavescens (SF) and its bioactive compound, matrine through performing locomotor activity test and the electroencephalography (EEG) analysis in the rat. The underlying neural mechanism of their beneficial effects was determined by assessing c-Fos immunoreactivity and serotonin (5-HT) in the brain utilizing immunohistochemical method and enzyme-linked immunosorbent assay. The results showed that SF and matrine administration had an effect on normalization of caffeine-induced hyperactivity and promoting a shift toward non-rapid eye movement (NREM) sleep. c-Fos-immunoreactivity and 5-HT level in the ventrolateral preoptic nucleus (VLPO), a sleep promoting region, were increased in the both SF and matrine-injected groups. In conclusion, SF and its bioactive compound, matrine alleviated caffeine-induced hyperactivity and promoted NREM sleep by activating VLPO neurons and modulating serotonergic transmission. It is suggested that SF might be a useful natural alternatives for hypnotic medicine.

  • An auxin-producing bacteria (A-1) was isolated from soils of Oyster mushroom farmhouse in Daejeon city, South Korea. The strain A-1 was classified as a novel strain of Ochrobactrum anthropi based on a chemotaxanomic and phylogenetic analyses. The isolate was confirmed to produce indole-3-acetic acid (IAA), one of auxin hormones, by TLC and HPLC analyses. The maximum concentration of IAA, $5.6mg\;L^{-1}$ was detected from the culture broth of O. anthropi A-1 incubated for 24 h at $35^{\circ}C$ in R2A broth containing 0.1% L-tryptophan. To investigate the growth-promoting effects to the crops, the culture broth of O. anthropi A-1 was inoculated to water cultures and seed pots of mung bean as well as lettuce. In consequence, the adventitious root induction and root growth of mung bean and lettuce were 2.7 and 1.4 times higher than those of the non-inoculated, respectively.

  • This work is focused on the definition and the analysis of both complete and incomplete similitudes for the dynamic responses of thin shells. Previous numerical and experimental investigations on both structural and structural-acoustic systems motivated this further analysis, mainly centred on the incomplete (distorted) similitudes. These similitudes and the associated scaling laws are defined by using the classical modal approach (CMA) and by invoking also the Energy Distribution Approach (EDA) in order to take into account both the cinematic and energetic items. The whole procedure is named SAMSARA: Similitude and Asymptotic Models for Structural-Acoustic Research and Applications. A brief summary of the procedure is herein given and the attention is paid to the analytical models of thin stiffened and unstiffened cylindrical shells. By using the well-known smeared model, the stiffened cylinder equations are used as general framework to analyse the possibility to define exact (replicas) or distorted similitudes (avatars). Despite the extreme simplicity of the proposed models, the results are really encouraging. The final aim is to define equivalent models to be used in laboratory measurements.

  • Purpose: The purpose of this study was to assess the relationship between menopausal symptoms and decline in cognitive functioning of menopausal women with mediating effects of health promoting behavior. Methods: Using a convenience sampling, 140 menopausal women were recruited for the cross-sectional survey. Data were collected by using the Menopause Rating Scale, Health Promoting Lifestyle Profile, Everyday Cognition, and Korean Mini-Mental State Examination. Results: The mean scores for menopausal symptoms, health promotion behavior, and subjective cognitive decline were 14.40, 153.79, and 67.40 respectively. Health promotion behavior was directly affected by menopausal symptom ($R^2=8%$). Cognitive decline was directly affected by menopausal symptom ($R^2=11%$). Menopausal symptom (${\beta}=.33$, p<.001) and health promotion behavior (${\beta}=.21$, p=.014) were found to be predictive factors in subjective cognitive decline and explained 14%. Health promotion behavior had a partial mediating effect in the relationship between menopausal symptom and perceived cognitive decline (Sobel test: Z=2.05, p=.040). Conclusion: Based on the findings of this study, developing nursing intervention programs focusing on decreasing menopausal symptoms and encouraging health promotion behavior are recommended to improve cognitive decline in menopausal women.
