Identification of Cuts-specific Myogenic Marker Genes in Hanwoo by DNA Microarray (DNA Microarray 분석을 통한 한우 부위별 특이 마커 유전자의 발굴)

  • Myogenic satellite cells (MSCs) are mononuclear, multipotent progenitors of adult skeletal muscle possessing a capacity of forming adipocyte-like cells (ALC). To identify the skeletal muscle type-specific myogenic and adipogenic genes during MSCs differentiation, total RNA was extracted from bovine MSCs, myotube-formed cell (MFC), and ALC from each of Beef shank, Longissimus dorsi, Deep pectoral, and Semitendinosus. DNA microarray analysis (24,000 oligo chip) comparing MSCs with MFC and ALC, respectively, revealed 135 differentially expressed genes (> 4 fold) among four cuts. Real-time PCR confirmed expression of 29 genes. Furthermore, the whole tissue sample RNAs analysis showed 6 differentially expressed genes in Beef shank. Among which, 1 gene in MSCs, 4 in MFC, and 1 in ALCs were highly expressed. This study will provide an insight for better understanding the molecular mechanism of differentiation of skeletal muscle type-specific MSCs. The identified genes may be used as marker to distinguish skeletal muscle types.

Relationship Between Orthodontic Root Resorption and Asthma, Allergy, and Psychological Stress (교정치료와 관련된 치근흡수와 천식, 앨러지 및 심리적 스트레스와의 상관성에 관한 연구)

  • One aspect of undesirable outcomes in orthodontic treatment includes excessive resorption of dental roots with mechanotherapy. The aim of this study is to demonstrate whether diseases affecting the immune system adversely are prevalent in orthodontic patients who manifest excessive resorption of dental roots with orthodontic tooth movement. The records of 51 orthodontic patients (25 males and 26 females, $16.1{\pm}3.3$ yr old) and 51 pair-matched controls ($1.5.4{\pm}4.1$ yr old) were analyzed retrospectively. The pretreatment questionnaires and the treatment records disclosed that the incidence of asthma, allergy, and signs indicative of psychological stress, was significantly higher in the root resorption cohort. The immune system is either altered or adversely affected in all these conditions. Since the progenitors of osteoclasts and odontoclasts are derived from mononucleated cells of monocyte and macrophage lineage, which are prominent cellular members of the immune system, the study leaded to the conclusion that excessive root resorption may occur in orthodontic patients who are psychologically stressed, or who have asthma and allergy, or any other conditions that may adversely affect and modify the immune system, and a careful examination and interpretation of a patient's medical history may be beneficial to both patient and practioner.

  • SNe light curves have been used to understand the expansion history of the universe, and a lot of efforts have gone into understanding the overall shape of the radioactively powered light curve. However, we still have little direct observational evidence for the theorized SN progenitor systems. Recent studies suggest that the light curve of a supernova shortly after its explosion (< 1 day) contains valuable information about its progenitor system and can be used to set a limit on the progenitor size, R*. In order to catch the early light curve of SNe explosion and understand SNe progenitors, we are performing a ~8hr interval monitoring survey of nearby galaxies (d < 50 Mpc) with 1-m class telescopes around the world. Through this survey, we expect to catch the very early precursor emission as faint as R=21 mag (~0.1 Rsun for the progenitor). In this poster, we outline this project, and present a few scientific highlights, such as the early light curve of SN 2015F in NGC 2442.

Genetic Variation of Two Isolated Relict Populations of Vaccinium uliginosum L. in Korea (들쭉나무 격리잔존 2개 집단의 유전변이)

  • In order to provide the molecular genetic information necessary for conservation of bog whortleberry (Vaccinium uliginosum L), one of the rare species in Korea, I-SSR analysis was performed on two populations on Mt. Halla and Mt. Seorak. A total of 68 I-SSR products were observed, and higher level of genetic diversity was observed in Mt. Halla population (S.I.=0.539) than in the Mt. Seorak population (S.I,=0.401). Level of genetic diversity in this species was relatively higher than those in other rare species analysed with I-SSR marker. From the results of AMOVA, exceptionally large proportion of genetic diversity (33.5%) was resulted from genetic difference between two populations, and only 66.5% of the genetic variation was allocated in common among individuals within each population, compared with the results in other long-lived woody species. This remarkably high degree of genetic heterogeneity existed between Mt. Halla and Mt. Seorak populations might suggest that they might be originated from the independent progenitors before the post glacier ages, respectively, and/or that they undergone random genetic drift respectively due to geographical isolation resulted from dramatic changes in environmental conditions after the post glacier ages.

  • Comparing to mammals, male bird has the homozygote ZZ and female has the heterozygote n. Therefore, the sex of fertilized eggs is defined by female chromosome constitution. Although this cytological observation had been established, the molecular and cellular mechanism of germ cell differentiation are essentially unknown in aves. Especially, the differentiation of germ cells in mixed-sex chimeras has not yet been clearly elucidated. Primordial germ cells, which are the progenitors of sperm or egg after sexual maturity, firstly arise in the epiblast and migrate to embryonic gonads through the blood vessel. During the embryo development, these PGCs differentiate in the pathway of mate or female, respectively and develop the sperm or egg cells after sexual maturity. In this paper, we confirmed that the female PGCs could migrate into the recipient male gonads after transferring and differentiate into germ cells in the embryonic stages. The primordial germ cells were isolated from the female embryonic gonads of 5.5-day-old incubation and re-injected into the male recipient embryos of 2-day-old incubation, which produced mixed-sex chimera in the germline. The finding in this study demonstrated the ability of migration and differentiation of gonadal primordial germ cells in mixed-sex chicken.

Complete Mitochondrial Genome Sequence and Genetic Diversity of Duroc Breed (돼지 Duroc 품종에서 미토콘드리아 유전체 서열의 특성과 집단의 유전적 다양성)

  • Duroc is widely used to improve the meat quality and productivity. To elucidate the phylogenetic relation and the sequence specificity for the maternal property, the complete sequence of mitochondrial genome was determined and the population diversity of Duroc was investigated in this study. The length of mtDNA tested is 16,584-bp. There are several insertion/deletion mutations in the control region and coding regions for tRNA and rRNA, respectively, but not in peptide-coding regions. Four peptide-coding genes(COⅡ, COⅢ, ND3 and ND4) showed incomplete termination codon sequences such as T--, and two(ND2 and ND4L) did alternative initiation codons(AIC), respectively. Especially, the initiation codon sequences of ND2 gene were polymorphic in this population. Polymorphisms were detected in 11-bp duplication motif within control region as well as ND2 and CYTB. Variation patterns observed from the tests on three mtDNA regions were linked completely and then two haplotypes obtained from combining the data dividing this population. Duroc mtDNA is observed at the European pig cluster in the phylogenetic tree, however, the results from the population analyses supported previous opinions. This study suggests that the breed Duroc was mainly originated from the European pig lineage, and Asian lineage was also used to form the pig breed Duroc as maternal progenitors.

Increased Expression of Fas Antigen and Apoptosis in Aplastic Anemia Bone Marrow Cells (재생불량성 빈혈의 병태생리에서 Fas 항원과 Apoptosis의 역할)

  • Background: Clinical observations and laboratory studies have supported an immune basis for most acquired aplastic anemias, with the majority of patients responding to immunosuppressive therapy. Fas, a member of the tumor necrosis factor (TNF) receptor superfamily is a critical downregulator of cellular immune responses. Proinflammatory cytokines like interferon gamma (IFN-${\gamma}$) and TNF-${\alpha}$ can induce Fas expression and render hematopoietic progenitor cells susceptible to Fas-induced growth suppression and apoptosis. Methods: In order to investigate the involvement of apoptosis in the pathogenesis of aplastic anemia (AA), we measured the expression of Fas antigen and caspase-3 on bone marrow (BM) mononuclear cells (MNCs) of AA in the presence or absence of IFN-${\gamma}$, TNF-${\alpha}$, or macrophage inflammatory protein 1-${\alpha}$ (MIP-$1{\alpha}$). Results: We confirmed that AA BM MNCs were more apoptotic and highly expressed Fas antigen than normal donors. Stimulation by IFN-${\gamma}$, TNF-${\alpha}$, or MIP-$1{\alpha}$ increased Fas antigen and caspase-3 expression in AA BM MNCs than BM MNCs of normal donors. Anti-Fas monoclonal antibody enhanced IFN-${\gamma}$, TNF-${\alpha}$, or MIP$1{\alpha}$ mediated caspase-3 expression in BM MNCs of normal donors. Among these three cytokines, IFN-${\gamma}$ enhanced apoptosis most strongly via Fas-caspase-3 pathway. Conclusion: These results suggest that Fas signal pathway may play a role in the pathophysiology of aplastic anemia and negative hematopoietic regulators like IFN-${\gamma}$ can induce apoptosis of bone marrow progenitors in part by Fas induction.

Early-type Dwarf Galaxies in the Virgo Cluster: An Ultraviolet Perspective

  • Since the ultraviolet (UV) flux of an integrated population is a good tracer of recent star formation activities, UV observations provide an important constraint on star formation history (SFH) in galaxies. We present UV color-magnitude relations (CMRs) of early-type dwarf galaxies in the Virgo cluster, based on Galaxy Evolution Explorer (GALEX) UV data and the Extended Virgo Cluster Catalog (EVCC, Kim, S. in prep.). The EVCC covers an area 5.4 times larger (750 deg2) than the footprint of the classical Virgo cluster catalog by Binggeli and collaborators. We secure 1304 galaxies as members of the Virgo cluster and 526 galaxies of them are new objects not contained in the VCC. Morphological classification of galaxies in the EVCC is based on the optical image ("Primary Classification") and spectral feature ("Secondary Classification") of the SDSS data. We find that dwarf lenticular galaxies (dS0s) show a surprisingly distinct and tight locus separated from that of ordinary dwarf elliptical galaxies (dEs), which is not clearly seen in previous CMRs. The dS0s in UV CMRs follow a steeper sequence than dEs and show bluer UV-optical color at a given magnitude. Most early type dwarf galaxies with blue UV colors (FUV-r < 6 and NUV-r < 4) are identified as those showing spectroscopic hints of recent or ongoing star formation activities. We explore the observed CMRs with population models of a luminosity-dependent delayed exponential star formation history. The observed CMR of dS0s is well matched with models with relatively long delayed star formation. Our results suggest that dS0s are most likely transitional objects at the stage of subsequent transformation of late-type progenitors to ordinary red dEs in the cluster environment. In any case, UV photometry provides a powerful tool to disentangle the diverse subpopulations of early-type dwarf galaxies and uncover their evolutionary histories.

The Effect of Genetically Modified Lactobacillus plantarum Carrying Bone Morphogenetic Protein 2 Gene on an Ovariectomized Rat

  • Objective : Osteoporosis result from age-related decline in the number of osteoblast progenitors in the bone marrow. Probiotics have beneficial effects on the host, when administered in appropriate amounts. This study investigated the effects of probiotics expressing specific genes, especially the effects of genetically modified bone morphogenetic protein (BMP)-2-expressing Lactobacillus plantarum CJNU 3003 (LP) on ovariectomized rats. Methods : Twenty-eight female Wistar rats (250-300 g, 12 weeks old) were divided into four groups : the sham (control), the ovariectomy (OVX)-induced osteoporosis group (OVX), the OVX and LP (OVX/LP), OVX and genetically modified BMP-2-expressing LP (OVX/LP with BMP) groups. The three groups underwent bilateral OVX and two of these groups were administered two different types of LP via oral gavage daily. At 16 weeks post-OVX, blood was collected from the heart and the bilateral tibiae were extracted and were scanned by ex-vivo micro-computed tomography and stained with hematoxylin-and-eosin (H&E) and Masson's trichrome stain for pathological assessment. The serum levels of osteocalcin (OC), rat C-telopeptide of type I collagen (CTX-I), BMP-2, and receptor activator of nuclear factor-ĸB ligand (RANKL) were measured. Results : The 3D-micro-computed tomography images showed that the trabecular structure in the OVX/LP with BMP group was maintained compared with OVX and OVX/LP groups. No significant differences were detected in trabecular thickness (Tb.Th) between control and OVX/LP with BMP groups (p>0.05). Furthermore, a tendency toward increased BMD, trabecular bone volume, Tb.Th, and trabecular number and decreased trabecular separation was found in rats in the OVX/LP with BMP groups when compared with the OVX and OVX/LP groups (p>0.05). The H&E and Masson's trichrome stained sections showed a thicker trabecular bone in the OVX/LP with BMP group compared with the OVX and OVX/LP groups. There was no difference in serum levels of OC, CTX and RANKL control and OVX/LP with BMP groups (p>0.05). In contrast, significant differences were found in OC and CTX-1 levels between the OVX and OVX/LP with BMP groups (p<0.05). Conclusion : Our results showed that the expression of genetically modified BMP-2 showed inhibition effect for bone loss in a rat model of osteoporosis.

Immunocytochemistry of Metallothionein Expression in Developing Rat Liver (발생중인 흰쥐 간에서의 Metallothionein 발현에 관한 면역세포화학적 연구)

  • Metallothionein (MT) is a family of ubiquitous, low molecular weight (6-7 kDa), cysteine-rich protein with a high affinity to metal ions and has no aromatic amino acids and histidine. Some of the known functions of MT include detoxification of heavy metals and alkylating agents and neutralization of free radicals. Also, this protein may affect a number of cellular processes including gene expression, apoptosis, proliferation and differentiation. But, its actual functions are still not clear. The present study was undertaken to examine immunocytochemically the localization of MT in developing rat liver. On the day 11 of gestation, the fetal rat liver has already been formed and contained numerous oval cells with high nuclear cytoplasmic ratio, which were the progenitors of hepatic parenchymal cells, but no reaction products of MT were detected at this time. And then, positive reactions against MT started to appear predominantly in the parenchymal cells of liver from the 13th day after gestation. Reaction products, immunogold particles or brown coloration, were localized at both the nucleus and the cytoplasm of the parenchymal cells, except mitochondria. The intensity of this reaction gradually increased, and exhibited the strongest at birth. The intensity of MT staining and immunogold labelling diminished with growth, and by the 15th day after birth weak positive reaction was observed in the cells. In brief, positive reactions for MT were observed in the oval cells and the parenchymal cells during fetal stage, meanwhile they were present only in the parenchymal cells after birth. The present results suggest that MT possibly involves parechymal cell proliferation and differentiation through the storage or the supply of various metal ions in the developing rat liver.
