Effects of Extracellular Proteins on the Recovery of Embryogenic Potential in Long-term Cultures of Daucus carota (세포외 단백질을 이용한 장기 배양 식물세포(Daucus carota)에서의 Embryo 생성에 관한 연구)

  • A declining tendency in embryogenic capability was seen during 6 months culture period during which embryo production decreased from 1000 embryos/ml to 500 embryos/ml. The presence of extracellular factors extracted from newly established embryo cultures restored the embryogenic capability and even enhanced the embryo production up to 5 times (2500 embryos/ml) for old carrot suspension cultures compared with that of control cultures. The stimulating effect on the embryo production indicates that the enhancing effect comes from extracellular compounds that are probably protein molecules.

  • This study was conducted to develop pig-housings based on the forecasting models of swine production, the weather conditions, and so on in Korea. The Korean pig-housings were developed according to the following basis : 1. They should be suitable to domestic weather conditions. 2. They should be designed based on the forecasting models of swine production of farrow to finish operation among the forecasting models of swine production in Korea. 3. Proper environments should be offered to pigs according to the growth. 4. The environmental control, the treatment of swine wastewater, and so on should be interrelated. 5. Manual energy should be saved by effective arrangements of pig-housings. In the future, performance test of the Korean pig-housings and development of facility automation systems which are suitable to these should be accomplished.

Recent researches on Sapstaining Fungi Colonizing Pines

  • During last decade there has been noticeable progress in the research of the biology of sapstaining fungi that cause considerable economic losses to forest product industry. The researches generated broad ranges of knowledge on sapstaining fungi regarding their occurrence on conifer wood, taxonomy, nutrient physiology, pigmentation biochemistry and molecular biology, and biological control. Major problematic groups in the sapstain production are Ophiostoma, Ceratocystis, and Leptographium genera. With Ophiostoma as a model, it is found that the type of carbon source is important in the growth and pigment production of sapstaining fungi. The operation of dihydroxy naphthalene (DHN) melanin pathway for black to bluish pigment production has been confirmed in those cosmetic fungi both at biochemical and molecular levels. The development of albino technology using nutrition competition has been shown to be promising as an environmentally friendly biological control method for sapstain control.

  • The purpose of this study is to establish the direction for creating the beautiful and lively space of the reli-gious architecture as a consquence of trying to under-stand the forms of control of the light and the symbolic meaning of the production in the interior space of the re-ligious architecture. The scope of this study has been limited to the natural light and to the establishment of theoretical background by analyzing the essence and method of space formation by the light in a side view of development and psychology and establishing the concept related to the architectur-al space formation of the light as well as researching and analyzing the meaning and the method of production of the light. And at the same time the conclusion has been induced as a guide of verifying theoretical background by analyz-ing the practical use related to the control and the pro-duction of the light through the spot research and the referential materials in a way of case research into 5 do-mestic religious architectures as the subject of research.

  • This paper considers a supply chain consisting of a component manufacturer and a product manufacturer. The component manufacturer provides components for the product manufacturer based on a vendor-managed inventory type of supply contract, and also faces demands from the market with the option of to accept or reject each incoming demand. Using the Markov decision process model, we examine the structure of the optimal production control and inventory rationing policy. Two types of heuristics are presented. One is the fixed-buffer policy and the other uses two linear functions. We implement a computational study and present managerial insights based on the observations.

Effects of Propylene Glycol on Milk Production, Serum Metabolites and Reproductive Performance during the Transition Period of Dairy Cows

  • The objective of this study was to investigate the effects of an oral drench of propylene glycol (PG) on milk production, serum metabolites and reproductive performance during the transition period of animals. Twenty-four 2-3 multiparous Holstein cows (average body weight 565 kg, body condition score about 3.6, at the $9^{th}$ month of gestation) were selected, blocked, and then randomly assigned into a PG and a control group. The control and the PG group cows were orally drenched with water or 50 ml sugarcane molasses mixed with 500 ml PG from 7 days pre-partum to 30 days post-partum, respectively. Experimental results indicated that the oral drench PG had no effect on dry matter intake (DMI). The milk yield of the PG group was significantly higher than that of the control group (p<0.05), whereas milk fat content, milk protein and somatic cell counts (SCC) were not significantly different between groups. Concentration of plasma glucose in the PG group was significantly higher than that of the control group (p<0.05). Conversely, the concentrations of non-esterified fatty acids (NEFA), and blood urea nitrogen (BUN) in the PG group were lower than those of the control group (p<0.05). Concentrations of insulin and ketone bodies were not significantly difference between groups. Body condition score (BCS) in the PG group was significantly higher than that of the control group (p<0.05). In reproductive performance there was no difference between groups. The experimental results indicate that supplementation of PG during the transition period of dairy cows can supply energy rapidly, resulting in reduced catabolism of body tissue and increased milk yield.

The Utilization of Rumen Content-Barley Meal in Diets of Growing Lambs

  • The nutritive value of rumen contents and barley mixture (4:1 w/w; RCB) was evaluated and the effect of their feeding on growth performances in Najdi lambs was studied. A metabolism trial was conducted with 16 rams divided into four dietary groups. The diets were: a whole-mixed control diet and three diets where RCB was incorporated at the rates of 25, 50 and 100%, replacing an equal amount of control diet. The results showed that there was a depression (p<0.05) in DM digestibility for the 100% RCB diet in comparison with other diets. The digestibility of CP was higher(p<0.05) for the 25% RCB diet as compared to the control diet; there was a trend for a small (p>0.05) decrease in digestibility as level of RCB increased. Lambs in all studied diets were in positive nitrogen balance; the differences between diets were not significant. A total of 45 lambs were allotted into three groups and used to evaluate the effect of dietary inclusion of RCB (0, 25 and 50%) on growth performance and carcass traits. Daily DM intake, final body weight, carcass weight and dressing percentage were not different among treatments. Average weight gain and ether extract (EE) in 9-11th control joint were higher (p<0.05) in lambs fed control diet than those fed RCB diets. The substitution of RCB for 50% of control diet exhibited 11.8% reduction in feeding cost for each kg of body weight gain.

Effect of golden needle mushroom (Flammulina velutipes) stem waste on laying performance, calcium utilization, immune response and serum immunity at early phase of production

  • Objective: This experiment was conducted to evaluate the effects of golden needle mushroom (Flammulina velutipes) stem waste (FVW), on organic eggs production, calcium utilization, antibody response, serum immunoglobulin, and serum cytokine concentration at early phase of production in laying hens. Methods: A total 210, 19 weeks old aged ISA Brown layers were randomly assigned into 5 equal treatment groups, with 7 replications of 6 hens each. Dietary treatment included a standard basal diet as control; antibiotic (0.05% flavomycin); 2% FVW; 4% FVW; and 6% FVW. The experimental duration was 10 weeks. Results: There was no significant differences (p>0.05) on hen day egg production, egg weight, egg mass, feed intake, and feed conversion ratio (FCR) among experimental groups. Unmarketable eggs were significantly lower (p<0.05) both in 4% FVW and 6% FVW fed groups than control group. The calcium retention and calcium in egg shell deposition were significantly higher (p<0.05) in FVW inclusion groups than control and antibiotic groups. Antibody titers against Newcastle diseases were significantly higher (p<0.05) in 6% FVW fed group (except combined with 4% FVW at day 147) and infectious bronchitis were significantly higher (p<0.05) in FVW fed groups (except 2% FVW and 4% FVW at day 161) than control and antibiotic groups. Serum immunoglobulin sIgA was significantly higher (p<0.05) in all levels of FVW and IgG was significantly higher (p<0.05) in 4% FVW than control and antibiotic groups. Serum cytokine concentration interleukin-2 (IL-2) was significantly higher (p<0.05) in 6% FVW; IL-6 and tumor necrotic $factor-{\alpha}$ were significantly higher (p<0.05) both in 4% FVW and 6% FVW than control and antibiotic groups; IL-4 was significantly higher (p<0.05) in antibiotic, 2% FVW and 4% FVW fed groups than control. Conclusion: F. velutipes mushroom waste can be used as a novel substitute for antibiotic for organic egg production and sound health status in laying hens.

Reproductive Functions in Nili-Ravi Buffaloes after Short Term Treatment with Recombinant Bovine Somatotropin Hormone

  • Effects of short-term treatment with somidobove (recombinantly produced bovine somatotropin, BST) on estrous cyclicity and fertility were studied in dairy buffaloes. Twenty buffaloes of Nili-Ravi breed calving during the same season were assigned to either control (n=8) or treated group (n=12). The buffaloes of treated group received single infection (prolonged release) of 320 mg of somidobove on day-60 postpartum. The mean values for interval to first postpartum estrus, first service conception rate, services per conception, service period and calving interval for the treated group were 96.4 days, 66.7%, 1.70, 164 days and 473 days, respectively. The corresponding values for the control group were 92.5 days, 62.5%, 1.87, 135 days and 439 days. Means of all variables did not differ between control and treated group (p > 0.05). Three buffaloes of the control and four buffaloes of the treated group did not conceive at first service. Out of these, two buffaloes of control and one buffalo of treated group exhibited normal estrous cycles. It is concluded from these data that short term BST-treatment has no adverse effect on reproductive functions of dairy buffaloes.

The Improvement of the Sanitary Production and Distribution Practices for Packaged Meals (Kim Pab) Marketed in Convenience Stores Using Hazard Analysis Critical Control Point (HACCP) system (편의점 판매용 김밥 도시락 생산 및 유통과정의 품질개선을 위한 연구)

  • Time-temperature relationship and microbiological quality were assessed and critical control points were identified through hazard analysis during the phases of production in two different packaged meals (Dosirak) manufacturing establishments (A, B:Kim Pab). Microbiological tests on foods, equipments and utensils were done according to standard procedures and included total plate count, coliforms and fecal coliforms. The results of the study are summarized as follows : time-temperature control management was needed because time-temperature abuse more than 8 hours at dangerous temperature zone (5-6$0^{\circ}C$) was observed from pre-preperation to distribution phase; Poor sanitary practices of employees were observed in hand washing and using disposable gloves; Microbiological analysis results of equirpments and utensils showed possible cross-contamination risks when foods were contacted with them; Kim Pab needed thorough quality control because it included various mixed ingredients of cooked and uncooked and had many apportunities of cross-contamination either by equipments or hands through whole production processes.
