Response of Soil Microbial Communities to Different Cultivation Systems in Controlled Horticultural Land

  • Ester-linked fatty acid methyl ester (EL-FAME) profiles were used to describe differences in soil microbial communities influenced by conventional farming system (CFS), and organic farming system (OFS) in controlled horticultural land. Soil physicochemical properties and soil microbial communities were determined in the experimental fields. Higher organic matter content in OFS reduced soil bulk density which in turn increased the soil porosity. Generally, soil chemical properties in OFS were higher than those of CFS, but EC value in OFS was significantly lower than that of CFS. With the exception of Fe content, other macronutrient contents and pH in both farming system decreased with the soil depth. Soil microbial biomass of OFS was approximately 1.3 times in topsoil and 1.8 times in subsoil higher than those of CFS. Lower ratios of cy17:0 to $16:1{\omega}7c$ and cy19:0 to $18:1{\omega}7c$ were found in the CFS soils than the OFS soils, indicating that microbial stress decreased. The ratio of MUFA to SFA was higher in OFS due to organic input to the soil. In principal components analysis (PCA), the first variable accounted for 54.3%, while the second for 27.3%, respectively. The PC1 of the PCA separated the samples from CFS and OFS, while the PC2 of the PCA separated the samples from topsoil and subsoil. EL-FAMEs with the positive eigenvector coefficients for PC1 were cy17: 0 to $16:1{\omega}7c$ ratio, cy19:0 to $18:1{\omega}7c$ ratio, soil pH, soil organic matter, and soil $NO_3$-N content. Our findings suggest that the shifting cy19:0 to $18:1{\omega}7c$ ratio should be considered as potential factors responsible for the clear microbial community differentiation observed between different cultivation systems and soil depth in controlled horticultural land.

Discrimination of the drinking water taste by potentiometric electronic tongue and multivariate analysis (전자혀 및 다변량 분석법을 활용한 먹는물의 구별 방법)

  • Organoleptic parameters such as color, odor, and flavor influence consumer perception of drinking water quality. This study aims to evaluate the taste of the selected bottled and tap water samples using an electronic tongue (E-tongue) instead of a sensory test. Bottled and tap water's mineral components are related to the overall preference for water taste. Contrary to the sensory test, the potentiometric E-tongue method presented in this study distinguishes taste by measuring the mineral components in water, and the data obtained can be statistically analyzed. Eleven bottled water products from various brands and one tap water from I city in Korea were evaluated. The E-tongue data were statistically analyzed using multivariate statistical tools such as hierarchical clustering analysis (HCA), principal component analysis (PCA), and partial least squares discriminant analysis (PLS-DA). The results show that the E-tongue method can clearly distinguish taste discrimination in drinking water differing in water quality based on the ion-related water quality parameters. The water quality parameters that affect taste discrimination were found to be total dissolved solids (TDS), sodium (Na+), calcium (Ca2+), magnesium (Mg2+), sulfate (SO42-), chloride (Cl-), potassium (K+) and pH. The distance calculation of HCA was used to quantify the differences between 12 different types of drinking water. The proposed E-tongue method is a practical tool to quantitatively evaluate the differences between samples in water quality items related to the ionic components. It can be helpful in quality control of drinking water.

Analysis of Morphological Characteristics for Normal Maize Inbred Lines (종실옥수수 자식계통들에 대한 형태적 특성 연구)

  • We evaluated the morphological characteristics of 156 maize inbred lines, which were developed to breeding normal maize variety at Maize Experiment Station, Gangwon Agricultural Research and Extension Services, by examining 11 quantitative and three qualitative characteristics. On the evaluation of three qualitative traits for 156 maize inbred lines, most inbred lines showed yellow (85 and 84 inbred lines) at tassel color (QL1) and silk color (QL2), and showed semi erect (105 inbred lines) at plant type (QL3). While, the evaluation of 11 quantitative traits for 156 maize inbred lines, they showed the morphological variation in days of tasseling (QN1, 56.5 to 76.0 days), days of silking (QN2, 59.0 to 85.5 days), stem thickness (QN3, 12.7 to 42.9 mm), plant height (QN4, 111.8 to 239.8 cm), ear height (QN5, 48.2 to 126.5 cm), 100 kernel weight (QN6, 14.9 to 36.4 g), ear length (QN7, 10.0 to 79.0 cm), setted kernel length (QN8, 8.0 to 70.5 cm), ear thickness (QN9, 4.0 to 22.0 cm), total kernel weight (QN10, 22.0 to 490.0 kg) and water content (QN11, 9.3 to 11.9%), respectively. As a result, 11 inbred lines (00hf3, 00hf19, 00hf30, 00hf36, 02S8069, 02S8072, 02S8090, 02S8099, 05S10011, 06S8085-6, 07S8011) in the 156 normal maize inbred lines have showed comparatively high values. While, the results of PCA (principal component analysis) indicated that the ear length (QN7), setted kernel length (QN8), ear thickness (QN9) and total kernel weight (QN10) greatly contributed in positive direction on the first principal components. And also, days of tasseling (QN1), days of silking (QN2), plant height (QN4) and ear height (QN5) contributed in negative direction on the second principal component. Thus these morphological characters, which were greatly contributed in the first and second principal components, might be considered to be useful for discrimination among 156 normal inbred lines. Specifically, this study's assessment of morphological characteristics of 156 normal inbred lines will be helpful useful for normal maize breeding programs such activities as planning crosses for hybrid and line development at Maize Experiment Station, Gangwon Agricultural Research and Extension Services.

Physicochemical and sensory characteristics of commercial RTD cold brew coffees (RTD (Ready-to-drink) 콜드브루커피의 이화학 및 감각특성 분석)

  • The sensory characteristics of eight commercial ready-to-drink (RTD) cold brew coffees were compared by descriptive analysis. The cold brew coffee samples were analyzed for hunter color values, brown color, turbidity, pH, titratable acidity, soluble solids, total phenolic content, and chlorogenic acid content. Three appearances, nine aroma, five flavor/taste, and four texture/mouth-feel related sensory attributes were evaluated by a panel of nine judges. The results of three-way analysis of variance of descriptive data showed that all sensory attributes except "fruit" aroma, "dark chocolate" aroma, "bitter" aroma, and "sweet" taste had significant differences among the samples (p<0.05). Based on the principal component analysis (PCA) of the descriptive data, the samples were primarily separated by first and second principal components, which accounted for 81.78% of the total variance among the samples with high intensities of "nutty aroma", "grain aroma", and "grain taste" versus "earth aroma", "sour aroma", "sour taste", "astringent", "smoothness", and "residual". In the correlation analysis of sensory terms and physicochemical parameters, titratable acidity and soluble solids showed significant positive correlations with earth aroma and smoothness characteristics.

Systematic Study on the Aconitum alboviolaceum Complex (Ranunculaceae) in Korea (한국산 줄바꽃 종집단의 분류학적 연구)

  • The Aconitum alboviolaceum Kom. complex includes four controversial species described from Korea; A. albouiolaceum, A. pseudolaeue, A. longecassidatum, and A. quelpaertense. The main objective of this study was to determine the taxonomic identities and systematic relationships among the species in the A. albouiolaceum complex based on morphology, numerical analyses and DNA sequence analysis. In the present study, variations in the principal morphological characters and chloroplast DNA noncoding region sequences (psbA-trnH IGS, trnL intron, and trnL-trnF IGS) were examined for 95 individuals from 20 populations. Also, neighbor-joining analysis was adopted to infer their relationships. Morphological variation appeared to be considerably high but not to be related to geographic distribution. These morphological results suggest that reevaluation of key morphological characters are needed for the proper taxonomic treatment of the complex. The length of psbA-trnH IGS region ranged from 241 to 250 bp, that of the trnL intron from 526 to 532 bp, and that of the trnL-trnF IGS region from 466 to 472 by in all taxa. Nine haplotypes were recognized from the analysis. Seven populations shared more than two haplotypes, while other thirteen populations shared only one haplotype. In the phylogenetic analysis, the nine haplotypes formed four groups, separated A. sibiricum, one of the sister groups of the complex. There also was no distinct grouping pattern supporting the species and populations observed. These results suggest that introgression or speciation might have been involved in the formation of taxa of A. alboviolaceum complex.

Comparison of Isoflavone Content and Composition in Soybean (Glycine max L. (Merr)) Germplasm

  • Soybean is known as to have a several healthy ingredients. Among them, isoflavones are effective in reducing obesity, menopausal symptom. Isoflavones consist of 12 isomers, including Aglycon, Glucoside, Malonyl glucoside, Acetyl glucoside, and are usually found in soybean seeds. The content is determined by the sum of 12 isomers, and the content value difference between the varieties is huge. In this study, we investigated the agronomic traits, 12 isomer of isoflavone content and composition for 49 soybean germplasms. This germplasms were selected from the 23,000 germplasms with the highest total content of isoflavones possessed by the National Agrobiodiversity Center. Seed samples were cultivated in experimental field located in Jeonju City on April 04, 2019. Matured seeds were harvested and portions of each seed samples were oven-dried, pulverized, and analyzed for their isoflavone compositions using HPLC-DAD. The soybean samples showed distinction in their agronomic traits, isoflavone compositions and contents. The days to flowering ranged between 38 and 69 days while the days to maturity ranged between 103 and 156 days. The seed coat color of soybean germplasms was 24 in black, 10 in yellow, 2 in green, 5 in yellowish green, 4 in green with black spot, 4 in pale yellow. The germplasm with the highest total content of isoflavones was the IT178054(1257.61±7.98 ㎍/g), but the germplasms containing the largest number of isoflavone isomers were IT274592, IT275005, both germplasms had 11 isoflavone isomers excluding Malonyl glycitin. The largest source of Aglycon, the most easily absorbed isoflavone form in the human body, was IT274592(DZ: 8.83±0.30 ㎍/g, GL: 11.14±0.81 ㎍/g, GE: 8.16±0.26 ㎍/g), while only IT274592, IT275005, IT308619 contained all three components of Aglycon. In Principal Component Analysis(PCA), the first two principal components showed more than 3.5 Eigen value and accounted for 58.2% of variability. The total content value had strong relationship with Malonyl genistin content value. Acetyl isomers had strong relationship, but Malonyl isomers were only related to isomers except Malonyl glycitin. These results will help in research on soybean varieties to enhance isoflavone ingredients.

THE ANALYSIS OF THE FT-NIR SPECTRA OF WATER ON THE BASIS OF TWO-STATE MODEL

  • Robinson with ${coworkers}^{1}$ have introduced two-state outer-neighbor bonding model to explain the anomalies of water. The studies on the properties of water as a function of temperature and pressure revealed that, unlike other ideas, all $H_2O$ molecules in liquid are tetrabonded. On the average they are forming two different bonding types. One type is the regular tetrahedral water-water bonding similar to that found in the ordinary ice Ih, whereas the other is a more dense nonregular tetrahedral bonding similar to that appearing in the ice II. The transformation between these two bonding forms is evidenced by FT-NIR experiment. The FT-NIR measurements were done for liquid water in the temperature range from $20^{\circ}C$ up to $80^{\circ}C$ in a wide extent of frequencies: 12 000 - 4000 $cm^{-1}$ /. Temperature dependent variations in the volume fraction of these two structures are directly related to the spectral changes. The absorbance variations are explored by means of the two-dimensional correlation spectroscopy (2DCOS), principal component analysis (PCA), curve fitting and second derivatives. The presence of the isosbestic points in a range of the combination and overtone transitions indicates that the experimental spectra are a superposition of two temperature independent components. One component of diminishing intensity with temperature increase, is assigned to a stronger hydrogen bonds occurred in the Ih type, whereas the second component showing an opposite behavior, one can attribute to a weaker H-bonds characteristic for the II type. The understanding of the hydrogen bonding network in the liquid water is very important in interpretation of the interaction between water and protein chain. The two-state model of water surrounding the protein surface could advance an understanding of the hydration process.

Safety Knowledge and Changing Behavior in Agricultural Workers: an Assessment Model Applied in Central Italy

  • Background: In recent years, the interest in health and safety in the workplace has increased. Agriculture is one of the human work activities with the highest risk indexes. Studies on risk perception of agricultural workers are often referred to as specific risk factors (especially pesticides), but the risk perception plays an important role in preventing every kind of accident and occupational disease. Methods: The aim of this research is to test a new method for understanding the relation between risk perception among farmers and the main risk factors to which they are exposed. A secondary aim is to investigate the influence of training in risk perception in agriculture. The data collection was realized using a questionnaire designed to investigate the risk perception; the questionnaire was given to a sample of 119 agricultural workers in central Italy. Through the use of the "principal components analysis" it was possible to highlight and verify the latent dimensions underlying the collected data in comparison with scales of attitudes. Results: Results show that the highest percentage of strong negative attitude is among the people who have worked for more years, while farmers who have worked for fewer years have a marked positive attitude. Conclusion: The analysis of the questionnaires through the synthetic index method (Rizzi index) showed that agricultural workers involved, in particular the elderly workers, have a negative attitude towards safety; workers are hostile to safety measures if they have not attended special training courses.

Genetic Diversity and Population Genetic Structure of Cephalotaxus koreana in South Korea

  • The Korean plum yew (Cephalotaxus koreana Nakai) is a shade-tolerant, coniferous shrub. The seeds have been used as a folk medicine in Korea, and an alkaloid extract (HTT) is known to have anticancer properties. We estimated the genetic diversity of 429 trees in 16 populations in South Korea using 194 polymorphic amplicons from seven combinations of AFLP primer-restriction enzymes. The average number of effective alleles and the percentage of polymorphic loci were 1.37 and 79.4%, respectively. Shannon's diversity index and the expected heterozygosity were 0.344 and 0.244, respectively. We divided 16 populations into four groups on the UPGMA dendrogram and the PCA biplot. The first two principal components explained 84% of the total genetic variation. Genetic differentiation between populations explained 14% of total genetic variation, and the remaining 86% came from difference between individuals within populations, as determined by an analysis of molecular variance (AMOVA). However, the genetic differentiation did not correlate with the geographic distance between populations from the Mantel test. The Bayesian statistics, which are comparable to Wright's $F_{ST}$ and Nei's $G_{ST}$, were ${\theta}^I=0.406$ and ${\theta}^{II}=0.172$, respectively. The population genetic diversity was slightly lower, and the strength of genetic differentiation was much weaker, than the average of those plants having similar life histories, as assessed using arbitrary marker systems. We discuss strategies for the genetic conservation of the plum yew in Korea.

A reappraisal of Sambucus pendula Nakai on Ulleung Island and its allies (울릉도 말오줌나무와 근연종의 재검토)

  • Sambucus pendula Nakai, which is an endemic on Ulleung Island of Korea, is characterized by a large pendulous inflorescence and small fruit. A set of 256 individuals were used to investigate the patterns of intraspecific variation of S. racemosa subsp. kamtchatica, S. racemosa subsp. sieboldiana, and S. williamsii including S. pendula using PCA (principal components analysis). This analysis showed that S. pendula was distinct based on its large inflorescence and long peduncle. Our data showed a morphological distinction between subsp. kamtchatica from northeastern areas and subsp. sieboldiana from Jeju Island, but the two taxa overlap in the southern and eastern parts of Korea in terms of chromosome number, color of style, and allozyme data. Our study agrees that S. pendula on Ulleung Island may be the result of genetic drift that occurred during isolation since the Quarternary period. This has been suggested as a reason for the genetic differences observed between two taxa and would explain the unique variation patterns of S. pendula. However, the morphological differentiation between the S. racemosa complex and S. pendula is not considered sufficient to warrant recognition of specific status. Therefore, we recommend that only one polymorphic species of S. racemosa in Eurasia be recognized and that S. pendula be considered a subspecies of S. racemosa.
