Quality Changes of Radish Juice by Mixed Culture with Lactic Acid Bacteria during Storage (유산균의 혼합배양에 의한 무주스의 저장중 품질변화)

  • The radish juice inoculated with mixed culture of Leuconostoc mesenteroides and Lactobacillus brevis was fermented and stored. Radish juice was heat-treated for 30 min at 65$^{\circ}C$ and 15 min at 80$^{\circ}C$, and stored for 8 weeks at 25$^{\circ}C$, 15$^{\circ}C$, and 5$^{\circ}C$, respectively. When the radish juice was heat-treated, any bacteria were not detected and any quality changes were not occured. On the other hand non-heated radish Juice, pH was decreased and acidity was increased during storage. The turbidity of the heat-treated radish juice was higher than that of the non-heated juice when it stored at 5'E. From the sensory evaluation, the pasteurization condition of 15 min at 80$^{\circ}C$ was not appropriate because the fresh smell of Dongchimi was decreased and the cooked smell was increased. The radish juice stored at 5$^{\circ}C$ without heat-treatment was evaluated to have the best quality. The storage period of the non-heated radish juice was shown to be extended over 8 weeks at 5$^{\circ}C$.

Quality change of sliced ginseng depending on different drying methods (건조방법에 따른 홍삼 절편의 품질 변화)

  • This study investigated quality change of ginseng depending on far-infrared (FIR) drying methods. The ginseng was cut into 2 mm slices and measured moisture content (MC), color, total polyphenol content (TPC), total flavonoid content (TFC) and DPPH radical scavenging activity (DPPH). The color values (L, a, and b) were decreased after far-infrared (FIR) drying. As increase in drying time, color was changed. Considering quality parameters including MC, TPC, TFC, and DPPH, the optimum condition for drying was set at $65^{\circ}C$ for 60 min. Furthermore, we compared color and functional compounds depending on drying process including FIR, freeze-, vacuum, and hot-air drying. The color values of ginseng were decreased when dried using vacuum and hot-air. While the color values of ginseng were increased after freeze drying. Comparison with another drying method, FIR drying presented the highest color difference. FIR and vacuum-dried ginseng indicate higher contents of TPC and TFC than freeze- and hot air-dried ginseng. And hot-air-dried ginseng present the highest DPPH when compared to another drying methods. On the other hand, freeze dried ginseng show the lowest TPC, TFC, and DPPH.

Antioxidant activity and metabolic regulation of sodium salicylate on goat sperm at low temperature

  • Objective: The purpose of this study was to explore the effect of sodium salicylate (SS) on semen preservation and metabolic regulation in goats. Methods: Under the condition of low temperature, SS was added to goat semen diluent to detect goat sperm motility, plasma membrane, acrosome, antioxidant capacity, mitochondrial membrane potential (MMP) and metabonomics. Results: The results show that at the 8th day of low-temperature storage, the sperm motility of the 20 μM SS group was 66.64%, and the integrity rates of the plasma membrane and acrosome were both above 60%, significantly higher than those of the other groups. The activities of catalase and superoxide dismutase in the sperm of the 20 μM SS group were significantly higher than those of the control group, the contents of reactive oxygen species and malondialdehyde were significantly lower than those in the control group, the MMP was significantly higher than that in the control group, and the contents of Ca2+ and total cholesterol were significantly higher than those in the control group. Through metabonomics analysis, there were significant metabolic differences between the control group and the 20 μM SS group. Twenty of the most significant metabolic markers were screened, mainly involving five metabolic pathways, of which nicotinic acid and nicotinamide metabolic pathways were the most significant. Conclusion: The results indicate that SS can effectively improve the low-temperature preservation quality of goat sperm.

The Cultural Characteristics of Paecilomyces tenuipes (눈꽃동충하초의 배양적 특성)

  • This study was conducted to investigate the cultural characteristics of Paecilomyces tenuipes PJ-1 separated from the nature. We investigated the effect of media, pH and temperature in growth of Paecilomyces tenuipes PJ-1 on solid culture media and the effect of inoculum size, glass beads in liquid culture condition. Paecilomyces tenuipes showed the most favorable growth on PDA among 5 different media. The optimum growth pH and temperature were at pH 6.0 and $25^{\circ}C$ on solid culture. Mycelial growth of P. tenuipes decreased rapidly above $35^{\circ}C$ and under $15^{\circ}C$. In liquid culture, the optimum inoculum size was 10.0% and the SED and PMV value were increased with adding glass bead and glass bead size in the range of glass bead 0~50 ea and size 3, 5 mm.

Optimization of Preparation Condition on Oriental Melon Jam by Response Surface Methodology (반응표면 분석에 의한 참외잼의 제조조건 최적화)

  • This study was carried out to establish the preparation condition of muskmelon jam. A central composite design was applied to investigate effects of muskmelon paste content(40, 45, 50, 55, 60 g), fructose ratio of sugar(20, 35, 50, 65, $80\%$) and pectin addition(l, 2, 3, 4, 5 g). The maximum sugar content was 61.48 $^{\circ}$Brix in 41.04 g of muskmelon paste content, $48.10\%$ of fructose ratio of sugar and 2.12 g of pectin content. The maximum value of softness was 2.71 g in 45.06 g of muskmelon paste content, $79.46\%$ of fructose ratio of sugar and 2.71 g of pectin addition. The minimum value of jelly strength was $0.04\;g{\cdot}cm$ in 47.80 g of muskmelon paste content, $63.0\%$ of fructose ratio of sugar and 1.99 g of pectin addition. The maximum value of organoleptic overall palatability was 5.89 in 55.65 g of muskmelon paste content, $73.19\%$ of fructose ratio of sugar and 2.42 g of pectin addition. The optimum conditions predicted for each corresponding physicochemical and organoleptic properties of muskmelon jam were 55.2 g(muskmelon paste content), $76.3\%$(fructose ratio of sugar) and 2.5 g(pectin addition).

Rearrangement of the Designated Area and Modification of Features of Buryeongsa Valley as a Scenic Site (불영사계곡의 명승 지정구역 조정 및 현상변경 방안 연구)

  • Since ancient times, Korea has been called a land of beauty. Scenic sites under the Cultural Properties Protection Act include picturesque places that are famous for their natural scenic beauty as well as their historical and cultural value. Scenic sites are managed as natural assets to promote their preservation and use. However, the management of scenic sites can produce adverse effects on regional development and ownership rights. Moreover, the purpose of their designation as cultural assets is not fully understood because scenic sites are managed by focusing on restraint on users' act the same as was applied to historic sites. Therefore, the purpose of this study is to protect inhabitants' rights of ownership by arranging the boundaries of designated areas and by providing standard permission for condition changes in the Buryeongsa Valley, which was designated as a Scenic Site in 1979. The results of this study can be summarized as follows: First, arranging the boundaries of the designated area includes the arrangement of the edge lines standardized on the visual range of the mountain ridge, preventing the loss of landscape beauty in the designated district; the internal clearing district focuses on the existing settlement. Gearing the designated areas after the arrangement of the boundaries results in $11,928,932m^2$, 38.6% compared to the existing designated areas. Second, it establishes a 500m buffer zone inside the radius of the boundary of the cultural asset as a standard for condition changes that seriously affect landscape preservation. Third, the standards for permission on building 'height regulations' are divided into flat and gable, according to the roof shape. The adopted standard is 8m high for 2 story flat roofs, and 12m high for 2 story gable roofs.

Changes in physicochemical characteristics of porcine blood under various conditions of enzyme hydrolysis (효소분해조건에 따른 돈혈의 식품학적 품질 특성 변화)

  • The aim of this study was to investigate physicochemical properties of porcine blood hydrolyzed by proteases under various conditions for utilization as a food source. Five kinds of proteases (Alcalase, Neutrase, Protex-40L, PTPF-1430, and KMFP-15) were tested at different concentrations (0.1, 0.2, and 0.3%, w/v) during hydrolysis at 55 for 4 hr. Hydrolysis with $^{\circ}C$ KMFP-15 showed the lowest pH by 7.3. The highest soluble solid ($24.3^{\circ}Brix$) and free amino acid (4,944 mg%) contents were obtained by hydrolysis with KMFP-15 (w/v) at 0.2% addition level, which was not significantly different from the sample hydrolyzed at 0.3% level. Under the optimal condition of KMFP-15 at 0.2%, porcine blood was hydrolyzed at 60 up to 8 hr. The $^{\circ}C$ free amino acid content reached the highest at 4 hr, and then decreased with longer hydrolysis time. Under the optimal hydrolysis conditions, porcine blood hydrolysis powder had plenty of crude proteins, amino acids, and minerals, including iron, potassium, and zinc. The results showed that porcine blood could be utilized as an useful source of food supplement. The optimum conditions of hydrolyzing porcine blood, using 0.2 KMFP at $60^{\circ}C$ for 4 hr, can be used in the commercial production of protein supplements, amino acid sources, and iron fortifying agents.

Physiological Properties of Oak Mistletoe (Loranthus yadoriki) Extracts by Microwave Extraction Condition (마이크로웨이브 추출조건에 따른 참나무 겨우살이 추출물의 생리활성)

  • The physiological properties of oak mistletoe slice extracts were investigated. The total polyphenol content of mistletoe extracts was 50.28-109-69 mg%, depending on extraction conditions. The polyphenol content increased as extract concentration rose (25 mL/g>50 mL/g>100 mL/g). The electron-donating ability (EDA) of the 60% (v/v) ethanolic extract was 68.88% at 25 mL/g; this was the highest value of all extracts tested (p<0.05). The SOD-like activities of water, 30%, 60%, and 90% (all v/v) ethanolic extracts were 15.24-30.84% lower than those of 1.0% and 0.1% (both w/v) L-ascorbate solutions. Tyrosinase inhibitory activities of all samples were 3.40-30.92% lower than those of 1.0% and 0.1% (both w/v) L-ascorbate solutions. Nitrite-scavenging abilities measured at pH 1.2 and pH 3.0 were highest when the water, and the 30%, 60% and 90% (all v/v) ethanolic extracts, were tested. The results expand our understanding of the physiological properties of oak mistletoe slice extracts.

Optimized Methods to Maintain Motility and Viability in Normozoospermic Males (정자 운동성 및 수명 보존을 위한 최적 배양에 관한 연구)

  • Objectives: To determinate the optimal culture condition to maintain lifespan in human sperm, we evaluated the effect of different temperature on sperm motility and viability up to 5 days in normal specimens. Methods: Ejaculated semen samples with normal semen parameters were gently washed in HEPES buffered Tyrod's-Albumin-Lactate-Pyruvate (HTALP) media. Each 5 ml of HTALP + 0.3% albumin with $1{\times}10^6$ sperm/ml was incubated for 5 days in $37^{\circ}C$, $22^{\circ}C$, and $4^{\circ}C$. The sperm motility and kinematics were analyzed using computer assisted sperm analysis (CASA), membrane integrity was assessed by hypoosmotic swelling test (HOST), and capacitation status was evaluated by chlorotetracycline (CTC) fluorescence pattern. Each parameter was measured on day 1, 3, and 5, respectively. Results: The motility, viability and live/uncapacitated pattern were demonstrated significantly in temperature- and time-dependent difference (p<0.05). While the sperm cultured for 1 day in each temperature was not significantly different, the sperm cell kept in $22^{\circ}C$ after 3 days were preserved sperm motility, viability, membrane integrity, and F pattern better than in other culture temperatures. Conclusions: HTALP can be used a basic medium for culture and longevity preservation, and sperm cell kept at $22^{\circ}C$ is beneficial for assisted reproductive techniques.

Conservation of Minutes of the Cabinet Meeting held by the National Archives of Korea Measures to Improve the Binding Methods for Bulky Bound Records (국가기록원 소장 국무회의록 보존처리 -거대 편철 기록물의 보존성 향상을 위한 편철 개선-)

  • Minutes of cabinet meetings (hereafter referred to as "minutes") offer records on important activities of the government of the Republic of Korea. The National Archives of Korea has implemented a series of preservation for these minutes such as condition survey, deacidification, microfilming, digitization, making boxes, etc. Conservation treatment plans were further established in 2018 to pursue more preservation of over 2,000 volumes of minutes based on the result of the condition survey of the minutes. Among ninety-seven volumes that require conservation treatments, six volumes have been treated so far. A major concern of conservation treatment of minutes was found that a single volume of minutes contains 800 pages, leading to heavier and bulkier than most of other bound volumes. This paper presents the measures taken to solve the problems resulting from this bulky structure of the bound minutes. It is expected that the study will offer an opportunity to consider effective conservation methods for records on paper of which the physical shapes are yet to be determined.
