Effects of Sediment Elutriates on the Early Reproductive Outputs in the Pacific oyster, Crassostrea gigas

  • This is a subsequent study to our previous finding that Pacific oysters, Crassostrea gigas, gained a so-called upper plateau concentration, around 30,000 ng/g dry weight digestive gland for benzo(a)pyrene, showed reproductive behavior but produced their ensuing reproductive outputs damaged. A serial dilution of sediment elutriates from Jinhae Bay, Korea, where pollution was progressive, were exposed to gametes of the Pacific oyster for 0, 5, 10, 20, 30, and 60 min to detail the pollutant effects on very young specimens. There was an apparent critical dilution over which adverse effects are evident. This was $10\%$ of the present sediment elutriate, corresponding to 0.3 ng/g on the basis of total polycyclic aromatic hydrocarbons (PAHs) for the oyster. Within the dilution the embryonic development was not influenced by the duration of exposure to its gamete stage. At higher dilutions over the critical dilution, occurrence of abnormality increased dependent on the pollutant dilution and the duration of exposure. Similar trends were also found in larval mortality. However, overall, the chemical toxicity was more significant to morphogenesis than to survival, suggesting a potential recruitment of the pollutants-induced abnormal larvae in the wild population to threaten the population integrity.

Protein Binding of [S]-Perillyl Alcohol in HSA using High-Performance Frontal Analysis (HPFA를 이용한 HSA와 [S]-Perillyl Acohol의 단백질 결합력)

  • An on-line frontal analysis HPLC system was developed to determine the unbound concentration of (S)-perillyl alcohol, an potential anti-cancer agent, in human serum albumin (HSA) solution, The analysis was performed on a Develosil 100 Diol 5 (10 cm x 4.6 mm I.D.) high-performance frontal analysis (HPFA) column. Sodium phosphate solution was used as the mobile phase (pH 7.4, ionic strength 0.17) at a flow rate of 1 $m\ell$/min. UV wavelength was set at 205 nm. A injection volume of 600${mu}ell$ was chosen to ensure the compound eluted formed a zonal peak with a plateau. By Scatchard analysis, it was found that the binding constant(K) and binding number(n) of (S)-perillyl alcohol to molecular HSA were 2.05 x $10^6$ [$M{-1}$], 0.00428, respectively.

The Features of Asian Dust Events Originated in Manchuria (만주에서 발원한 황사현상 (II) -2001년 이후 사례를 중심으로-)

  • The northeast part of China(hereafter Manchuria) is one of Asian dust source regions along with Gobi, Inner Mongolia and Loess Plateau. In this study, a geographical survey over the area was carried out to determine its soil characteristics in June 2009. It revealed that some parts of the area, especially near Keerchin desert, consist of alkali clay soil mixed up with sand. Manchuria, where is a vast cornfield, can be a potential source region of Asian dust from fall to following spring after harvesting. The frequency of Asian dust over the region from 1996 to 2009 was examined using 3-hourly GTS SYNOP data and it showed that the occurrence of Asian dust over the region is high in the springtime. It was also revealed that snow cover is the key parameter affecting on the frequency through the analysis of NCEP reanalysis data. To scrutinize the path and structure of Asian dust from Manchuria, the event on 3~4 April 2008 and 25 January 2010 were intensively investigated with regard to features of synoptic weather patterns, satellite imagery, airstream, naked eye-observations, concentrations of PM10, 2.5 and 1.0. For this case, the Asian dust from the area reached to Korea less than a day. However, the duration time of the dust in Korea was short (< 7 hours). The average of hourly PM10 reached up to $340{\mu}g/m^{3}$ at Baengnyeondo during the period. The high PM2.5 and PM1.0 concentrations were also observed at several sites in Korea, indicating that air pollutants could be transported along with the dust.

  • Lithium cobalt oxide cathode for thin-film rechargeable lithium batteries were fablicated by electron-beam evaporation. Annealed lithium cobalt oxide, which was deposited on to stainless steel substrate, showed well-developed (003) planes of the hexagonal structure and potential plateau at $\~3.9 V$. Lithium cobalt oxide thin films had the stoichiometric Li/co ratio at high deposition rates and exhibited high discharge capacity at $15{\AA}/s$. As the annealing temperature increased, discharge capacity increased with maximum value at $700^{\circ}C$, but showed low capacity as a result of reaction with substrate above $700^{\circ}C$. Unuiformity of the lithium and cobalt in the depth profile gave initial capacity loss with charge/discharge performance.

Biochemical Characterization and Genetic Diversity of Pongamia pinnata (L.) Pierre in Eastern India

  • Biochemical characteristics of 24 Pongamia pinnata genotypes (candidate plus trees) from three agroclimatic zones were estimated and molecular characterization through RAPD markers was done. Various biochemical characters viz. seed oil, total carbohydrates, protein, acid value and Iodine number recorded significant variation among different genotypes. The highest seed oil content was 41.87% while seeds of 14 genotypes recorded above average (32.11%) for the trait. Seed oil and protein content exhibited a significant positive correlation and moderate heritability. Out of the initially selected twenty-five random primers, twenty-two RAPD primers were found to be highly reproducible and produced a total of 183 loci of which 147 (80.32%) loci were polymorphic. Percentage of polymorphism varied from 44% to 100% with an average of 80.62%. High level of genetic variation was found among different genotypes of P. pinnata. Both molecular and oil content (biochemical) markers appeared useful in analyzing the extent of genetic diversity in Pongamia and the result of these analyses will help to better understand the genetic diversity and relationship among populations. Overall, the Pongamia genotypes included in the study showed a correlation with their geographical origins such that genotypes from the same region tend to have higher genetic similarity as compared to those from different regions. However, in UPGMA based Nei's analysis, some genotypes were found not to be grouped based on geographical origins possibly due to the exchange of germplasm over time between farmers across the regions. The results from oil content analyses showed that several genotypes in 'Central and Western Plateau' agroclimatic zone of Jharkhand displayed a good potential for high oil content. The study provides insight about P. pinnata populations in Jharkhand (India) and constitutes a set of useful background information that can be used as a basis for future breeding strategy and improvement of the species.

Effect of thermal regime on the seismic response of a dry bridge in a permafrost region along the Qinghai-Tibet Railway

  • Dry bridges have been widely applied in the Qinghai-Tibet Railway (QTR) to minimize the thermal disturbance of engineering to the permafrost. However, because the Qinghai-Tibet Plateau is an area with a high potential occurrence of earthquakes, seismic action can easily destroy the dry bridges. Therefore, a three-dimensional numerical model, with consideration of the soil-pile interactions, is established to investigate the thermal characteristics and their impact on the seismic response of the dry bridge in permafrost region along the QTR. The numerical results indicate that there exist significant differences in the lateral displacement, shear force, and bending moment of the piles in different thermal conditions under seismic action. When the active layer become from unfrozen to frozen state, the maximum displacement of the bridge pile reduces, and the locations of the zero and peak values of the shear force and bending moment also change. It is found that although the higher stiffness of frozen soil confines the lateral displacement of the pile, compared with unfrozen soil, it has an adverse effect on the earthquake energy dissipation capacity.

  • Annual epidemics of seasonal influenza occur during autumn and winter in temperate regions and have imposed substantial public health and economic burdens. At the global level, these epidemics cause about 3-5 million severe cases of illness and about 0.25-0.5 million deaths each year. Although annual vaccination is the most effective way to prevent the disease and its severe outcomes, influenza vaccination coverage rates have been at suboptimal levels in many countries. For instance, the coverage rates among the elderly in 20 developed nations in 2008 ranged from 21% to 78% (median 65%). In the U.S., influenza vaccination levels among elderly population appeared to reach a "plateau" of about 70% after the late 1990s, and levels among child populations have remained at less than 50%. In addition, disparities in the coverage rates across subpopulations within a country present another important public health issue. New approaches are needed for countries striving both to improve their overall coverage rates and to eliminate disparities. This review article aims to describe a broad conceptual framework of vaccination, and to illustrate four potential determinants of influenza vaccination based on empirical analyses of U.S. nationally representative populations. These determinants include the ongoing influenza epidemic level, mass media reporting on influenza-related topics, reimbursement rate for providers to administer influenza vaccination, and vaccine supply. It additionally proposes specific policy implications, derived from these empirical analyses, to improve the influenza vaccination coverage rate and associated disparities in the U.S., which could be generalizable to other countries.

Effects of Bay K, cAMP and Isoprenaline on the Na-Ca Exchange Current of Single Rabbit Atrial Cells (토끼 심방근에서 Na-Ca 교환 전류에 대한 Bay K, cAMP, Isoprenaline 효과)

  • Ca movements during the late plateau phase in rabbit atrium implicate Na-Ca exchange. In single atrial cells isolated from the rabbit the properties of the inward current of Na-Ca exchange were investigated using the whole cell voltage clamp technique. The inward currents were recorded during repolarization following brief 2 ms depolarizing pulse to +40 mV from a holding potential of -70 mV. Followings are the results obtained: 1) When stimulated every 30 sec, the inward currents were activated and reached peak values $6{\sim}12\;ms$ after the beginning of depolarizing pulse. The mean current amplitude was 342 pA/cell. 2) The current decayed spontaneously from the peak activation and the timecourse of the relaxation showed two different phases: fast and slow phase. 3) The recovery of the inward current was tested by paired pulse of various interval. The peak current recovered exponentialy with a time course similar to that of Ca current recovery. 4) Relaxation timecourse was also affected by pulse interval and time constant was reduced almost linearly according to the decrease of pulse interval between 30 sec and 1 sec. 5) The peak inward current was increased by long prepulse stimulation, Bay K, isoprenaline or c-AMP. 6) The relaxation time constant of the inward current was prolonged by Bay K or c-AMP, and shortened by isoprenaline. From the above results, it could be concluded that increase of the calcium current potentiates and prolongs intracellular calcium transients, while shortening of the timecourse by isoprenaline or short interval stimulations might be due to the facilitation of Ca uptake by SR.

  • Taebaek city is located in the mountainous plateau area therefore the major industry was coal industry. According to dramatically declining of the major industry, Taebaek city is need to alternative industry which associated with considering the geographical characteristics of natural tourism resources for increasing the local economy. On the basis of these reasons, this study can be suggested ecological planning and mitigation of deterioration technique about the these study site. That is the reserved area for mountainous experience theme park. As the results of environmental ecology assessment are following as; The natural ecosystem areas, multi-layer structure forest such as forest of Pinus densiflora, forest of Quercus mongolica and deciduous forest have a high value of nature, diversity and potential. In addition, wild bird habitats were important area as a inhabitation, breeding, feeding and hiding. Therefore, on these areas should be preserved. Also, it needs to conserve on there such as more than three types of wild bird inhabitate areas, the fringe of high biological diversity, the wetland that have got good vegetation condition and the function of amphibia, reptiles crossing. In addition, inhabitation, the waterway of wetland form that have got wide waterside width needs to conserve. In conclusion, on the basis of analysis results such as conditions of plan, environment, ecological assessments, survey informations are able to suggest the connectivity of the axis of forest and management plan. Moreover, in the part of forest restoration plan, we suggest the plan of transplant for a compensation of damaged forest by land use.

Effect of Drought Stress on the Concentration of Nitrogen Metabolites in White Clover

  • To investigate the changes in the composition and pool size of nitrogen metabolites under drought stress, white clover (Trifolium repens L.) were exposed to -0.04MPa (well-watered, control) or to -0.12MPa (drought-stressed) of soil water potential during 28 days. Dry weight of leaves in drought-stressed plants was remarkably decreased by 45% and 74% within 14 days and 28 days, respectively, compared with control. For nitrate concentration after 28 days of treatment, a significant difference (1.6 times higher in drought-stressed plants) was observed only in stolon. NH$_3$-NH$_4$$^{+}$ concentration in all three organs of drought-stressed plants linearly increased to more than 1.6 times higher level at 28 day when compared to the initial level (day 0), while the increasing rate in control was much less than that of drought-stressed plants. Proline concentrations in drought-stressed plants remarkably increased and reached to 7, 13 and 17 times higher level at 28 day compared to control. Protein concentration in leaves of drought-stressed plants tended to decrease, while it slightly increased during the first 14 days and reached a plateau afterward in control. There was not significant difference in the proteins concentration of stolon and roots throughout experimental period. On SDS-PAGE analysis, two major proteins specifically induced by drought stress (16-kD and 18-kD) were detected in stolon.n.
