A total of 163 virus infected pepper plants(Capsicum annuum L.) collected from various pepper growing regions in Korea were investigated on the presence of tobacco mosaic virus (TMV), cucumber mosaic virus (CMV), potato virus X(PVX), potato virus Y(PVY) and alfalfa mosaic virus (AMV) by serological methods. Van Slogteren's microprecipitin test was applied for the testing of TMV, PVX and PVY from infected plants, and Ouchterlony agar double diffusion test was used for CMV and AMV. Results obtained are as follows: 1. TMV, CMV, PVX, PVY and AMV were found to occur on the pepper plants growing in Korea. 2. The prevalence of each of these viruses among the 163 pepper plants investigated was in the order of CMV: 93 plants(57.0%)>TMV: 91 plants (55.8%)>AMV: 58 plants (35.6%)>PVY: 40 plants (24.5%)> PVX:6 plants(3.7%). 3. Among the 163 plants investigated, 72 plants (44%) showed infection with one kind of virus and 91 plants (56%) showed mixed infection with more than two different viruses. In general, heavier damage of the plants was observed from mixed infection. 4. The results of serological identification of pepper viruses coincided with those results obtained by sap inoculation experiment conducted at the Horticultural Experiment Station along with present investigation. Thus the serological techniques applied in this experiment proved to be very reliable for the identification of TMV, CMV, PVX, PVY and AMV from pepper plants infected with these viruses.
To classify the inheritance of resistance to potato virus Y, crosses between susceptible flue-cured tobacco variety NC 95 and resistant variety McNair 30 were conducted. The parents, $F_1$ plants, $F_2$ populations, and haploid plants derived from anthers of $F_1$ plants were screened for a resistance of two potato virus Y strains (PVY-VB and PVY-VN) isolated in Korea. The Chi-square values for the $F_3$ populations and haploids of $F_1$ fitted 1 :3 and 1 :1 ratios of resistant to susceptible for two strains, respectively. Therefore, it was found that the resistance of McNair 30 for the potato virus Y was controlled by a single recessive gene. Moreover the resistance to two strains screened was inherited dependently.
Potato plants carrying the Ry gene are extremely resistance to a number of potyviruses, but it is not known which variety expressed the resistance. In this investigation, combined classical and molecular techniques were used to identify virus resistance potatoes. Mechanical inoculation of 32 varieties of Korean potato cultivars, with potato virus Y (PVY), induced various symptoms, such as mosaic, yellowing, necrosis, mottle, vein clearing and vein bending. Different virus spreading patterns were observed, such as highly sensitive, moderate and resistant to $PVY^o$ inoculated leaves in different cultivars. From the results of double antibody sandwich-enzyme links immunosorbant assays (DAS-ELISA), coupled with reverse transcription polymerase chain reaction (RT-PCR), Winter valley and Golden valley were found to be highly susceptible and resistant cultivars to $PVY^o$ respectively. TEM was used as a complementary method to conform the localization of the virus in leaf tissues. TEM detect virus particles in Golden valley, where, ELISA and RT-PCR were unable to detect the CP gene. However, the interior part of the tissues was severely deformed in $PVY^o$ infected Winter valley, than Golden valley The Ry gene is involved in an induced response in $PVY^o$ infected Golden valley plants. The methods described in this study could be applied for the screening and development of antiviral potatoes.
A mutant of Potato vims Y (PVY) was occurred in PVY resistance flue-cured tobacco breeding line KF0402 $(TC1146{\times}KF117)$ showing vein necrosis at Suwon in Korea. This isolate, PVY-SWM, was differentiated from other PVY based on biological properties and nucleotide sequence analyses of coat protein gene. PVY-SWM caused typical symptoms on 21 indicator plants as compared to the PVY-TOJC37. Remarkably, the PVY-SWM induced distinctly different symptom of systemic vein necrosis on tobacco cultivars V.SCR, PBD6, TN86, TN90, Virgin A Mutant (VAM), Wislica, NC744, KB108 and KB111, which were reported to have the recessive potyvirus resistance gene va. In RT-PCR assays with specific primers for detection of PVY, a single band of about 800bp in length was produced. The amplified DNA was cloned and the nucleotide sequence was determined. The coat protein gene of PVY-SWM showed 88.4%-99.0% and 92.5%-98.5% identities to the 12 different PVY isolates of Genbank Database at the nucleotide and amino acidi respectively. Multiple alignments as well as cluster dendrograms of PVY-SWM isolate revealed close phylogenetic relationship to the $PVY^{NTN}$ subgroup.
한국을 비롯한 5개국에서 잎담배로부터 8계통, 감자로부터 분리된 1계통 등 모두 9계통의 PVY에 대한 잎담배 품종별 내병성을 온실내에서 조사하였다. 조사된 품종중 Dixie Bright 244-2, McNair 30, 및 Golden Stock Penish 등의 품종은 7가지의 PVY 계통에 대해 높은 내병성 반응을 보였다. 이외 Chile 및 US-NN계통에 대해서도 감수성품종에 비해 내병성을 나타냈다. 품종별 병징의 심, 경에 따른 엽중 바이러스의 농도는 접종된 바이러스 계통과 품종에 다라 다르게 나타나 엽중 바이러스 농도에 따른 내병성 정도 구분은 할 수 없었다.
Potato virus Y (PVY) is an important viral disease in burley tobacco in Korea and is mostly transmitted by green peach aphid, Myzus penicae, in nature. Effects of barrier nets on the immigration of the aphid population into tobacco fields and on the control of PVY were investigated in 3 tobacco fields with 1.8 m - height polyethylene (PE) nets on their outer - sides in Iseo - Myeon, Wanju - Kun, Cheonbuk. Immigration of aphids to the tobacco yields started at late April and reached maximum at early June. The immigrating aphid population was apparently blocked by the barrier nets to be reduced by 54-73%. PVY severity was also reduced by the barrier nets. However, the control value was variable, ranging 24-67%, which suggests that effect of the PE net barrier on the prevention of aphid-borne virus might be variable depending on the location and slope of the fields.
The cDNAs derived from the coat protein (CP) genes of six plant RNA viruses, tobacco mosaic virus-pepper strains (TMV-P) and -ordinary strain (TMV-OM), potato virus Y (PVY), turnip mosaic virus (TuMV), cucumber mosaic virus (CMV) and potato leafroll virus (PLRV), were subcloned into the transcription vector, pSPT18, containing SP6 and T7 promoters. The digoxigenin (DIG)-labeled RNA polymerase after linearlization of the cloned pSPTs with XbaI or SacI, and were tested for their sensitivities for the detection of the six viruses. In slot-blot hybridization, dilution end points for the detection of TMV-P and TMV-OM were 10-4, while those of PVY, TuMV and CMV were 10-3. PLRV was detected at the dilution of 10-2. When each RNA probe was applied for the detection of the viruses in the preparations from the leaf disks (8 mm in diameter, and 12 to 15 mg in weight) of infected natural host plants, TMV-P, TMV-OM and TuMV could be detected from one disk, while PVY from 1 or 2 disks. CMV was detected in the preparation from two disks, and PLRV from three disks. With DIG-labeled RNA probe, PVY was detected at 5 days after inoculation, but with ELISA the virus was detected at 8 days after inoculation to tobacco (Nicotiana tabacum cv. Xanthi nc) plants on which symptoms appeared at 9 days after inoculation. No difference was observed in cross reaction between the RNA probes for the detection of TMV-P and TMV-OM.
This study was conducted to develop a resistant tobarro against Potato virus Y (PVY) by transformation of the plants with genetically engineered viral genes. The complimentary DNAs (cDNAS) of potato virus Y-necrosis strain (PVY-Vn) replicase mutant genes (3'-deleted, 5'-deleted and ADD-mutant Nlbs) were synthesized through RT-PCR by using purified PVY-VN RNA and synthesized primers, and cloned in the sense orientation into a plant expression vector (pMBPI), The cDNAS of the genes were transferred into Agrobacterium tumefaciens LBA 4404, and then transformed into tobacco (Nicotiana tabacum cv. Burley 21) plants. Regenerated plants were tested for PVY resistance by inoculation test; 13 transgenic plants including 7 for 3'-deleted Nlb, 3 for 5'-deleted Nlb, and 3 for ADD-mutant Nlb appeared to be resistant at 4 weeks after inoculation with PVY-VN. Among the 13 transgenic tobacco plants, 8 plants had no symptom up to 14 weeks after inoculation. The progenies ($T_1$) from self-fertilization of the transgenic lines varied 0.0% to 81.2% in their resistance (% of resistant plants). The analysis of Nlb-31deleted, -5'deleted and -ADD mutant in the $T_1$ plants by polymerase chain reaction (PCR) showed that Nlb-3'deleted, -5'deleted and -ADD mutants were detected in all of the resistant plants. These results suggest that the PVY resistance was inherited in the $T_1$ generation.

ND860-2, Norchip, Russet Norkotah, Goldrush 및 Norgueen Russet 등 5개 품종의 감자 잎 조직을 감자 바이러스 Y(PVY)의 외피단백질 (CP) 유전자를 운반하는 Agrobacterium tumefaciens을 이용하여 형질전환시켰다. ND860-2와 Norchip품종과 같은 흰색 감자에서 재분화율과 형질전환율이 높게 나타났다. 형질전환체를 선발한 후 CP의 생성을 western blot으로 확인한 결과 30 kD의 밴드가 나타났다. 온실에서 재배된 형질전환체는 한 개체를 제외하고는 표현형에 있어서는 기존의 품종과 다르지 않았다. 인위적으로 감자 바이러스 Y를 감염시킨 15일 후의 검사결과 control구는 80%의 감염율을 보인 반면 형질전환체들은 38-58%의 감염율을 보였다. 30일 후의 검사 결과는 모든 5품종의 control구에서 100%의 감염을 보였으나 형질전환체는 7개체가 저항성을 보였다. 마지막 85일 후의 검사 결과 4개의 저항개체를 발견하였으며 이 개체들은 현재 계속 연구가 진행 중에 있다.
잎담배 4품종 (NC2326, NC95, NC744, Havana)과 2종의 식물 (Nicotiana repanda Wild., Physalis floridana Rydb.)을 이용하여 감자 또는 잎담배에서 분리된 10가지의 감자바이러스 Y (PVY) 계통 판별을 시도하였다. 10가지의 PVY 계통들을 이 식물들에 접종했을 때 괴저, 엽맥녹대 또는 mottling, 무병징등을 나타내면서 서로 다른 기주반응을 보여 쉽게 구분할 수 있었다. 조사된 계통들 중에 PVY-VN, PVY-N, PVY-NSNR, PVY-Chile 및 PVY-Argentina 등은 잎담배 품종 NC2326에서 괴저병징을 나타냈으며 이외 계통들은 엽맥녹대 또는 mottling 병징을 나타냈다. NC95에서는 PVY-MSMR에 의해 괴저병징을 이 외 계통들에 의해서는 엽맥녹대 또는 mottling 병징을 나타냈다. Havana에서는 PVY-NSNR, PVY-MSNR, PVY-VB 등에 의해 nettling 병징을, PVY-Chile 및 PVY-Argentina에 의해 괴저병징을 나타냈으나 이 외의 계통들에 의해서는 병징이 나타나지 않았다. NC744에서는 PVY-MSNR, PVY-MSMR에 의해 mottling 병징을, PVY-Chi 및 PVY-Argentina에 의해 괴저병징을 나타냈으며 이 외의 계통들에 의해서는 병징을 나타내지 않았다. N. repanda에서는 PVY-Argentina에 의해 괴저 반응을, PVY-VN 및 PVY-C에 의해서는 병징을 나타내지 않았으며 이 외의 계통들에 의해서는 mottling 병징을 나타냈다.
