  • The volcanic ash soils from the Jeju province have low available phosphate because of the high phosphate fixing power of the soils. Phosphorus fertilizerwas dressed after cultivating green manure crops and before cultivating potatoes in order to investigate potato yields, the available phosphate contents, and phosphorus balance in the soils. Green manure crops cultivated in this study were hairy vetch alone and hairy vetch and rye. During potato cultivation, four treatments were conducted in the experiments; NF (non fertilizer), P fertilizer ($N-P-K=0-25-0kg\;10a^{-1}$), NK fertilizer ($N-P-K=18-0-19kg\;10a^{-1}$), and NPK fertilizer ($N-P-K=18-25-19kg\;10a^{-1}$). There were no differences in the yields of potato stem and tuber from the cultivation plots of different green manure crops. However, in the plots with single-sown hairy vetch and mixed-sown hairy vetch and rye, the yields of potato stem and tuber were higher in the P fertilizer plot than in NF plot. The yield of tuber among the treatments with mixed-sown hairy vetch and rye was the highest in the NPK fertilizer plot. The available phosphate content in soils initially increased with time in all plots, but began to decrease gradually after Oct. 18. The available phosphate contents were high in the plots for phosphate fertilization, and the difference in available phosphate content between non-phosphorus fertilizer plots and phosphorus fertilizer plots increased with time. In the single-sown hairy vetch and mixed-sown hairy vetch and rye plot, the phosphorus balances in NF plot and NK fertilizer plot were very low, while those in the P fertilizer plot and NPK fertilizer plot were high. In conclusion, available phosphate contents in soil and the potato yields were increased by phosphorus fertilization when potatoes planted after cultivating hairy vetch and rye together, compared to hairy vetch alone.

  • This study was conducted to investigate the effect of solution culture systems on growth and mini-tuber yields of hydroponlcally grown potato (Solanum tuberosun L. cv. Dejima). The growth of stem and foliage at 40 and 60 days after planting were better in aeroponics system than the other systems, and stem length and number of stems per plant at 90 days after planting were the greatest as of 65.4 cm, 7.3 in aeroponics and the lowest as of 49.5 cm, 3.2 in scoria media system. Stolon length was the longest as of 30.4 cm in aeroponics at 90 days after planting, and number of stolons was the greatest as of 10.5 in NFT. Number of mini-tubers per plant at 90 days after planting was 67.1, 62. 5, 20.1 and 18.0 in aeroponics, NFT, perlite and scoria media, respectively. The perlite media system made the fewest enlargement of lenticels of mini-tubers. The results indicate that aeroponic system can be used effectively for mini-tuber potato production.

  • "Early Valley", is an early maturing potato cultivar with high yield potential. "Early Valley" is a clonal selection resulting from the cross between 'Suncrisp' and 'A87109-10'. It has medium plant height and light green foliage. "Early Valley" has medium flowering habit and white flowers. Tubers are smooth, yellow skin, light yellow flesh, round tuber shape, medium eye depth, and medium dormancy and good keeping quality. It has stable yield under wide range of climatic conditions. "Early Valley" is resistance to late blight, but moderately susceptible to common scab and hollow heart. This cultivar is also resistant to potato rotting at harvesting during the raining season. "Early Valley" has high level of antioxidant activity (about three times higher) and vitamin C (higher by 40%) than the 'Superior'. This cultivar has high level of tuber uniformity and capable of yielding 36.56 t/ha which is 17.07% higher than the control potato cultivar 'Superior' under optimum agronomical practices.

  • This work aimed to evaluate 11 potato families for insect resistance horticultural traits. The families were derived from crosses between introduced insect resistant and adapted genotypes. A randomized complete block design, with three replications was used. The plot consisted of 25 genotypes of a family, summing up to 75 genotypes tested per family. Two susceptible cultivars were included in the experiment as checks. The genotypes were evaluated for insect resistance, tuber yield traits, tuber appearance and tuber skin smoothness. The genetic value for the intensity of insect attack in the leaves and in the tubers was lower(higher resistance) in the 11 families than in the checks. For yield traits, there was no predominant effect among the families regarding the origin of the resistant donor species(Solanum berthaultii and/or S. chacoense). However, there was a large contribution of the C-1485-16-87 recurrent parent in crosses with resistant genotypes for tuber yield and tuber number. In relation to tuber appearance, only the family derived form the C-1485-16-87/ ND140(S. berthaultii) cross did not differ from the checks. had genetic value tuber skin smoothness, it was identified that families derived from S. berthaultii resistant parents showed higher genetic values than the check cultivars. For tuber skin smoothness, the genetic values of the families neither differed significantly from each other nor from the checks.

  • A computer vision system to measure low flow rate of solid particles was developed and tested to examine its performance with various sized 7 kinds of seeds, perilla, mung bean, paddy, small red bean, black soybean, Cuba bean and small potato tuber. The test was performed for two types of particle flow, continuous and discontinuous. For the continuous flow tested with perilla, mung bean and paddy, the tests resulted correlation coefficients for the flow rates measured by the computer vision and direct method about 0.98. Average errors of the computer vision measurement were in a range of 6∼9%. For the discontinuous flow tested with small red bean, black soybean, Cuba bean and small potato tuber, the tests resulted correlation coefficients for the flow rates measured by the computer vision and direct method 0.98∼0.99. Average errors of the computer vision measurement were in a range of 5∼10%. Performance of the computer vision system was compared with that of the conventional optical sensor to count particles in discontinuous flow. The comparison was done with black soybean, Cuba bean and small potato tuber, and resulted that the computer vision has much better performance than the optical sensor in a sense of precision of the measurement.

  • Since the discovery of generation of $O_2^-$ in plant, many evidence for the oxidative burst (OXB) has been accumulated in various combinations of plant and pathogen or elicitor systems. $O_2^-$ generating system responsible for the OXB was coupled with oxidation of reduced nicotinamide adenine dinucleotide phosphate (NADPH) in microsomal fraction isolated from sliced aged potato tuber slices which were treated by hyphal wall components elicitor from Phytophthora infestans (HWC). We developed new assay method for quantitative measurement of oxygen radical $O_2^-$ by using electron spin resonance (ESR) analysis during elicitorinduced OXB on the surface of plant tissues. The ESR analysis using an $O_2^-$ trapper, Tiron (1,2-dihydroxy-3,5benzenedisulfonic acid), provided a convenient assay for detecting only $O_2^-$ during elicitor-induced OXB producing various active oxygen species (AOS) on plant tissue surface. Tiron was oxidized to Tiron semiquinon radical by $O_2^-$. Quantity of the radical signal was measured by specific spectra on ESR spectroscopy. The level of $O_2^-$ was high in from surface of potato tuber tissue treated with hyphal cell wall elicitor (HWC) from Phytophthora infestans. There was no secondary OXB induction by $H_2O_2$ treatment in plant.

  • Hairy root induced by A. rhizogenes from potato tuber (Solanm tuberosum L.) synthesized the agropine and mannopine which were demonstrated with paper electrophoresis. And the starch contents in hairy root were increased gradually following the developmental stage. But protein contents were decreased. The activity of ${\beta}-glucan$ synthetase II(GSII) which is related to the cell wall biosynthesis was stimulated in hairy root on the developmental stage. And chloropromazine did not influence the activity of GS II while verapamil inhibited about 60% of the activity GS II. Therefore, these results showed $Ca^{2+}$ to be effective factor in the cell wall formation. Isozyme pattern of peroxidase was investigated in the callus and hairy root induced from potato tuber.

  • This study was to evaluate whether ionizing gamma radiation could be applied to break the dormancy of a potato minituber. The respiration rate of the minitubers was significantly affected by the storage temperature and a low dose gamma radiation. Ionizing radiation of 8 Gy enhanced the respiration rate of the potato tuber stored at $10^{\circ}C$ for 20 days. The potato tuber subjected to 4 and 8 Gy after 40 days storage at 10 and $20^{\circ}C$ exhibited higher respiration rates compared to the control (non-irradiated), but not at st. However, the ionizing radiation did not exhibit on significant effect on the respiration rate of the potato tuber stored for 60 days. It was observed that minitubers stored for 20 days had significant response to the storage temperature in terms of the total sugar content the higher the storage temperature, the lower the total sugar content. It was measured that the reducing sugar content was increased under the storage conditions both 5 and $10^{\circ}C$ for 40 days, but not to $20^{\circ}C$. The total sugar contents in the minituber stored for 60 days were similar to those stored for 40 days. The data was discussed on the relationships among the storage duration, temperature and ionizing radiation.

  • Recently, with increasing concern for sustainable agriculture and safe agricultural products, organic farming has become widely adapted as an alternative to conventional farming. This study was conducted to investigate the effects of earthworm casts (EWC) with $100\%$ organic compost on the growth and yield characteristics of fall-cropping potato (Solanum tuberosum L. cv. Dejima) in Jeju. The treatments consisted of seven plots: 2-, 4-, 6-, 8-, and 10-ton $ha^{-1}$ EWC-treated plots, 1.2-ton $ha^{-1}$ complex fertilizer $(N-P_2O_5-K_2O,\;10-10-14)-for-potato$ (CEP)-treated plot as conventional practices, and a control plot. The plant heights were greater in the plots where EWC and CFP were applied than in the control plot. Tuber diameter, number of stems per plant, and chlorophyll level tended to increase in the plots where 8-10 tons $ha^{-1}$ of EWC were applied. The application of CFP and EWC showed an increment in the average tuber weight per plant, but there was minimal significant difference. The application of 8-10 tons $ha^{-1}$ of EWC resulted in an increase in the total tuber yield (21.61-21.87 tons $ha^{-1}$) as compared to the other plots. The highest yield of market able tubers was $69.8\%$ of the total yield from the 10-ton $ha^{-1}$ EWC-treated plot. Consequently, with regard to the growth and yield characteristics of fall-cropping potato, the effects of EWC application were more favorable than the effects of the application of a chemical fertilizer.

  • Erwinia rhapontici causes soft-rot disease in a number of plants such as rhubarb, onion, hyacinth and garlic. Pectate lyase (Pel) depolymerizes pectin and other polygalacturonates, which is though to play a role in bacterial invasion of plants. Pel activity was not detected in E. rhapontici cultured in a minimal salts medium containing glycerol, polygalacturonate, or citrus pectin as a carbon source. However, when sterilized potato tuber and Chinese cabbage slices were added to minimal salts polygalacturonate (0.5%) medium, E. rhapontici produced pectate lyase enzyme. Also Pel activity was consistently detected from macerated potato tubers, Chinese cabbage leaves, lettuce leaves and celery petioles tissue. Pel in the extract of macerated Chinese cabbage caused by E. rhapontici strain 1, resulted in electrolyte loss, tissue maceration and cell death of potato tuber tissue. These results indicate that E. rhapontici produces pectate lyase only in the presence of non-diffusible plant components, and that this enzyme probably contributes to its pathogenicity.
