Potato (Solanum tuberosum) is one of the most important food crops worldwide and yields of potato can be affected by virus infection. While more than 40 viruses have been found in potato, only nine viruses (potato leafroll virus, potato viruses A, M, S, V, X and Y, potato moptop virus and tobacco rattle virus) and one viroid (potato spindle tuber viroid) have a significant economic impact on potato, worldwide. This review describes the geographical distribution of the most important viruses infecting potato and the genes for resistance or tolerance that have been identified against these various infectious agents. In some cases such resistance genes have been found only in other Solanum species. Few genes for resistance to the vectors of these viruses have been obtained and even fewer have been deployed successfully. However, transgenic resistance in potato has been achieved against seven of these disease agents.
To classify the inheritance of resistance to potato virus Y, crosses between susceptible flue-cured tobacco variety NC 95 and resistant variety McNair 30 were conducted. The parents, $F_1$ plants, $F_2$ populations, and haploid plants derived from anthers of $F_1$ plants were screened for a resistance of two potato virus Y strains (PVY-VB and PVY-VN) isolated in Korea. The Chi-square values for the $F_3$ populations and haploids of $F_1$ fitted 1 :3 and 1 :1 ratios of resistant to susceptible for two strains, respectively. Therefore, it was found that the resistance of McNair 30 for the potato virus Y was controlled by a single recessive gene. Moreover the resistance to two strains screened was inherited dependently.
A program was initiated to transfer potato virus Y vein-necrosis strain resistance from N. africana to N. tabacum The Fl plants between the above species were self-sterile, but all amphidiploid plants from the Fl plants and backcrossed flowers, that is, the N. tabacum flowers crossed with amphidiploid were self-fertile. The parent, amphidiploid plants of Fl, F2 population of the amphidiploid and the backcrossed generation were screened for a resistance of potato virus Y vein-necrosis strain isolated in Korea. The Chi-square values for the F2 population of the amphidiploid and the backcrossed generation fitted 35: 1 and 5 : 1 ratios of resistant to susceptible for the potato virus Y vein-necrosis strain, respectively. Therefore, it was found that the resistance of N. tabacum for the potato virus Y vein-necrosis strain was controlled by a single dominant gene.
Potato virus X (PVX) resistance in potato is one of the best-characterized resistance models, however little is known in pepper. To evaluate the resistance to PVX in Capsicum annuum, a total of eleven pepper accessions were used for resistance screening against two PVX strains, USA and UK3. None of them were resistant against strain UK3, whereas four resistant genotypes were found against strain USA, three of which were further characterized. Two unlinked dominant genes were identified for both genotypes Bukang and Perennial; resistance in the genotype CV3 seemed to be conferred by two complementary dominant genes. These results demonstrated that the resistance to PVX in C. annuum is different from that in potato. This is the first report on genetic analysis of PVX resistance in C. annuum.
The coat protein mediated resistance to potato virus Y is assessed here in transgenic potato plants (Solanum tuberosum L., cv Claustar). Therefore, the corresponding cDNA from tunisian isolate of the virus was cloned into Agrobacterium tumefaciens binary vector. The transgenic lines were subsequently analysed for the presence and expression of the transgene. The CP cDNA copy number was determined for kanamycin resistant plants. Three selected transgenic lines and their S1 progeny resulting from tuber germination showed a high protection level against the virus. These data appear to support the hypothesis that the virus resistance is mediated by the translated viral coat protein expressed in transgenic potato lines.
Viral diseases have been a major limiting factor threating sustainable potato (Solanum tuberosum L.) production in Pakistan. Surveys were conducted to serologically quantify the incidence of RNA viruses infecting potato; Potato virus X (PVX), Potato virus Y (PVY), Potato virus S (PVS), Potato virus A (PVA), Potato virus M (PVM) and Potato leaf roll virus (PLRV) in two major potato cultivars (Desiree and Cardinal). The results suggest the prevalence of multiple viruses in all surveyed areas with PVY, PVS and PVX dominantly widespread with infection levels of up to 50% in some regions. Co-infections were detected with the highest incidence (15.5%) for PVX and PVS. Additionally the data showed a positive correlation between co-infecting viruses with significant increase in absorbance value (virus titre) for at least one of the virus in an infected plant and suggested a synergistic interaction. To test this hypothesis, glasshouse grown potato plants were challenged with multiple viruses and analyzed for systemic infections and symptomology studies. The results obtained conclude that multiple viral infections dramatically increase disease epidemics as compared to single infection and an effective resistance strategy in targeting multiple RNA viruses is required to save potato crop.

The complementary DNA (cDNA) of potato virus Y- vein necrosis strain (PVY-VN) replicase gene (Nlb) was transformed into tobacco (Nicotiana tabacum cv. Burley 21) plants. Out of 25 putative transformants regenerated, 3 were resistant to PVY-VN, one highly resistant plant with no symptom until seed harvest time and the other two with mild chlorotic spot symptoms at late stages after infection. No symptom was observed in the highly resistant plant, while mild vein necrotic symptoms were developed on suckers of the moderately resistant plants after seed harvest time, In the first generation (T1) via self fertilization, resistance to susceptibility frequency in transgenic plants from the highly resistant transformant was about 3 : 1, while it was lowered much (about 1:2 and 1:19) in T1 of the moderately resistant transformants. In the second generation (T2) of the highly resistant plant, resistance frequencies were similar to T1, but resistance levels varied greatly and appeared to be decreased. Since the demonstration that the transgenic plants expressing tobacco mosaic virus(TMV) coat protein(CP) gene showed resistance to TMV infection, there have been numerous attempts to produce virus-resistant plant by introducing of a part of or modified viral genome. This study was conducted to investigate the characterization and variability of disease outbreak of transgenic potato(T-potato) with the CP gene of potato leaf roll virus(PLRV) in an isolated field from 2000 to 2002. In the field inspection, incidence of PLRV on T-potato showed only 3.5%, while non-transgenic potato(N-potato) revealed 13.4%. Infection rate of PLRV was considerably low on T-potato with 4.2% compared to 15.4% of N-potato in ELISA tests. Those of potato virus M, potato virus Y and potato virus X on both potatoes were not statistically different. Infection of potato virus A was not observed on both potatoes. Incidence of potato late blight caused by Phytopkhora infestans on T-potato and N-potato did not differ each other with 52.7%, and 50.8%, respectively, Mating type of the causal fungus isolated from both potatoes was all Al types. Results indicates that the CP gene of PLRV affects specifically to the virus in the transgenic potato.
Potato Virus Y(PVY), vein necrosis strain, in Korea causes severe symptoms on burley tobacco(Nicotiana tabacum L.). As the results, programs to incorporate PVY resistance into commercial cultivars were initiatEd. But the development of the homozygous fertile line resistant to PVY is time consumming. This study was conducted whether the Fl hybrid could be used to reduce the yield losses caused by PVY. Four F1 hybrids were made between male - sterile(ms) NC 107 and KB 107 as maternal parent, and TC 612 and TC 613 as Pollen donor, respectively, and were evaluated for their PVY resistance and negatively associated traits. (ms NC 107 X TC 612) F1, named as KB 109, Ivas applied to yield trial and compared with commercial cultivars for the level of disease resistance, agronomic characteristics, chemical contents and physical properties. All Fl hybrid could be used commercially as the PVY resistant cultivar. Especially KB 109 have the resistance against PVY, tobacco mosaic virus and black shank(Phytophthora parasitica var. nicotianae). It had wider leaves, flowered one day later, and yield of acceptable quality was higher than that of Burley 21, standard cultivar in Korea.

Efficacy of plant growth promoting rhizobacteria(PGPR) Bacillus vallismortis strain EXTN-1 has been proved in eliciting induced systemic resistance(ISR) in several crops. The present paper described the beneficial effects of EXTN-1 in potato as increase in yield and chlorophyll content, and plant protection against Potato Virus Y and X(PVY & PVX). EXTN-1 induced systemic resistance to the plants resulting in significant disease suppression in the field. Also the plants under treatment with EXTN-1 had higher chlorophyll content. The bacterized plants had significantly higher yields over the untreated control plants. The strain induced activation of defense genes, PR-1a and PDF 1.2 in transgenic tobacco model, which indicated the possible role of both SA, and JA pathways in EXTN-1 mediated plant protection against crop diseases.
