Selection of Carbon, Nitrogen Source and Carrier for Mass Production of Beauveria bassiana (Beauveria bassiana 대량배양을 위한 탄소원, 질소원 및 고체 기질 선발)

  • For mass production of entomopathogenic fungus Beauveria bassiana 149, isolated from moth larva, by two-phase fermentation, we performed selection of carbon and nitrogen sources for liquid culture and examined solid fermentation on carrier, ingredient, temperature, and water content. Spore production with rice powder, corn powder, and starch from sweet potato was higher than that of sucrose and dissolvable starch for liquid fermentation as first-phase fermentation. As a nitrogen source, addition of peptone and yeast powder showed higher spore production than $NaNO_3$, fish powder, and soybean powder. The isolate produced more conidia in sawdust + wheat bran + corn powder, sawdust + wheat bran and rice shell + wheat bran as carrier and ingredient than vermiculite as carrier. Conidia production of B. bassiana 149 in solid-phase fermentation was twice higher at 30 than 20. Conidia yield was higher at 60% and 70% water content ($26.9{\times}10^8$ and $38.6{\times}10^8conidia/g$) than 40% and 50% ($13.9{times}10^8 $and $11.6{\times}10^8conidia/g$), respectively.

Production Conditions and Properties of Glucose Isomerase from Streptomyces griseolus (Streptomyces griseolus기원의 포도당 이성화효소의 생성 조건과 성질)

  • Cultural characteristics of Strptomyces griseolus isolated from the soil were investigated. This strain was disclosed to utilize D-xylose, and D-glactose in preference order as a carbon source with the formation of glucose isomerase. The addition of sweet potato starch also proved effective promoting the total enzyme activity measured at 29% higher than the control. Corn cob, one of waste agricultural resources, was hydrolyzed in 2~3% $H_2SO_4$ solution at $100^{\circ}C$, 3~5 hours to produce a xylose syrup which gave rise to the recovery of 19.9% in a batch system and 28.2% in a repeated system. By the addition of both 2% of xylose syrup(Be'28) prepared by and us 65% of corn steep liquor (total nitrogen 1.2%), enzyme induction was maximized. The enzyme activity was stimulated by the xylose and the cell growth by the C.S.L. Also, remarkable increase of enzyme activity was noticed by the addition of protein acid hydrolysate 86.2% higher than the control. $QO_2$ of the biomass cultured in 30L capacity jarfermentor recorded low oxygen requirement of 251.2 1/hr. Maximum activity of glucose isomerase was observed noted at the 9th hour after inoculation which is 2 hours faster than the stationery was observed noted at the 9th hour after inoculation which is 2 hours faster than the stationery phase of the biomass growth. Glucose isomerase from the strain was activated by adding the $Co^{++}\;and\;Mg^{++}$ with optimum temperature of $73^{\circ}C$ and pH of 7.2. Conversion ratio of 60% glucose to frutose was 42.5% after 70 hours reaction.

Elementary School Teachers에 Conceptions on Cell (초등학교 교사들의 세포에 대한 개념)

  • The purpose of this study is to investigate degree of the concept formation on cell in elementary school teachers and to clarify the patterns of their misconceptions. Data were collected by interview with 120 elementary school teachers, ranged from twenties to fifties in age, working in Taegu city. The instrument was developed by researchers and was categorized into four: cell as the basic units of life, morphology of cell, function of cell, growth of individual from the viewpoint of cell. The results are as follows: First, about 80% of teachers had two perspective outcome of cell: Cells are the basic units of life on earth. But the potato and meat, which we used to eat as food, are not constructed of cells but aggregated of nutrients. Second, most of elementary school teachers recognized that shapes of the cell in organisms were diverse, but some of them only could present several kinds of shape on cell. The 35% of teachers had misconception that shape of cell is all the same in a individual. It shows that this result is caused by lack of opportunity to observe the various kinds of cells. Third, most teachers understood tell can be seen only through a microscope. Yet in comparison with relative size, it was revealed that misconception of cell size was induced by the term of 'basic unit'. In addition, they thought that large organisms are built from large cells, and small organisms from small cells. Fourth, Elementary school teachers used to confuse the terms between chloroplast and chlorophyll, and believed that the genetic material was contained only in the reproductive cells and seeds. It was also revealed that they thought the nutrients such as starch and fat are located at intercellular space. Fifth, the 60% of teachers conceived correctly that growth of individual depends on mitosis and increase in cell number. The rest of them, however, misunderstood that it is due to the increase of tell volume.

Effects of pumpkin seed oil and saw palmetto oil in Korean men with symptomatic benign prostatic hyperplasia

  • This study was to investigate the role of complementary and alternative medicine in the prevention and treatment of benign prostatic hyperplasia. For this purpose, a randomized, double-blind, placebo-controlled trial was performed over 12 months on 47 benign prostatic hyperplasia patients with average age of 53.3 years and international prostate symptom score over 8. Subjects received either sweet potato starch (group A, placebo, 320 mg/day), pumpkin seed oil (group B, 320 mg/day), saw palmetto oil (group C, 320 mg/day) or pumpkin seed oil plus saw palmetto oil (group D, each 320 mg/day). International prostate symptom score, quality of life, serum prostate specific antigen, prostate volume and maximal urinary flow rate were measured. In groups B, C and D, the international prostate symptom score were reduced by 3 months. Quality of life score was improved after 6 months in group D, while those of groups B and C were improved after 3 months, compared to the baseline value. Serum prostate specific antigen was reduced only in group D after 3 months, but no difference was observed in prostate volume in all treatment groups. Maximal urinary flow rate were gradually improved in groups B and C, with statistical significance after 6 months in group B and after 12 months in group C. None of the parameters were significantly improved by combined treatment with pumpkin seed oil and saw palmetto oil. From these results, it is suggested that administrations of pumpkin seed oil and saw palmetto oil are clinically safe and may be effective as complementary and alternative medicine treatments for benign prostatic hyperplasia.

Sensory and Mechanical Characteristics of the Lemon Red Ginseng-pyun prepared by different ratio of red ginseng (레몬홍삼과편의 홍삼 배합비에 따른 관능적.텍스쳐 특성)

  • This study was performed to determine the sensory evaluation and mechanical characteristics of lemon red ginseng-pyun. Lemon red ginseng-pyun was made with lemon juice(21.4%), sweet potato starch(6.7%), water(53.3%), sugar(13.3%), honey(5.3%) and various concentrations of red ginseng powder(0, 2, 4, 6, 8, 10%). The quality characteristics of the sample were estimated in terms of pH, color difference, texture profile analysis and sensory evaluation. The pH of lemon red ginseng-pyun showed no significant difference among the different ratios of red ginseng. With increasing red ginseng powder addition, the lightness was significantly(p<0.05) lower, and a value and b values were significantly(p<0.05) higher. In sensory evaluation, with increasing red ginseng powder addition, the color and bitter taste of lemon red ginseng-pyun were stronger, while clarity of the 10% added red ginseng was significantly(p<0.05) lower than that of the 0% added red ginseng. Elasticity, hardness, chewiness, softness. sweet taste and overall acceptability were not significantly different in any group. In texture profile analysis, chewiness, cohesiveness, gumminess and hardness were significantly(p<0.05) increased in the 10% added red ginseng but springiness was not significantly different in any group. Overall acceptability was related to texture, appearance, overall taste and sweet taste of lemon red ginseng-pyun. Therefore, development of lemon red ginseng-pyun with no sugar or artificial sweeteners is encouraged as a healthy diet for diabetes and hypertension patients, because red ginseng is defined as a functional foods.

The Development of the Dietary Fiber Food Frequency Questionnaire and the Dietary Fiber Intake of Middle School Students in the Chungbuk Province (식이섬유용 식품섭취빈도조사지 개발 및 이를 이용한 충북지역 중학생의 식이섬유 섭취 실태)

  • In this study, the FFQ-50 questionnaire which composed of 50 food items frequently eaten as sources of dietary fiber, was developed and used to estimate the dietary fiber intake of middle school students in the Chungbuk province. According to the survey, the average daily intakes of dietary fiber for the boys and girls were $23.3{\pm}12.3$ g (93.2%) and $20.8{\pm}11.5$ g (104.0%), respectively, showing a significant difference between the gender. The proportions of boys and girls who did not reach to the sufficient dietary fiber intake were 66.2% and 53.9% respectively. The major sources of dietary fiber were grain foods and vegetables, which measured at 77.24%, followed by fruits, seaweeds, tofu and tofu products, potato and starch, and mushrooms. The cooked white rice contributed the highest (17.81%), followed by baechu-kimchi, cooked brown rice, ramyeon (instant noodle), tangerine, sesame leaf jangajji (pickled sesame leaf), topokki (spicy rice cake stir-fry), blanched and seasoned bean sprouts, bread, and radish kimchi. The study showed that the cooked white rice was the major source of dietary fiber in the boys' diet followed by baechu-kimchi, ramyeon, cooked brown rice, tangerine, bread, strawberry, blanched and seasoned bean sprouts, sesame leaf jangajji, radish kimchi. Girls were taking in dietary fiber mostly from boiled white rice followed by baechu-kimchi, boiled brown rice, tangerine, ramyeon, sesame leaf jangajji, topokki, blanched and seasoned bean sprouts, radish kimchi, and boiled barley.

Application of Thermotolerant Yeast at High Temperature in Jar-fermentor Scale.

  • We investigated the possibility of industrial application and economit process of high temperature fermentation by thermotolerant alcohol producing yeasts as previously reported. From the 20% glucose media, the RA-74-2 produced 11.8% (v/v) ethanol at $32^{\circ}C$ (0.5% inoculum) and 10.6% (v/v) ethanol at $40^{\circ}C$ (3% inoculum), respectively. Also, 11.3% (v/v) ethanol was produced for 96 hours in the temperature-gradient fermentation. These results suggest that the RA-74-2 could isuccessfully be applied to save the cooling water and energy in industrial scale without re-investment or modification of established fermentation systems. When potato starch was used as the substrate for the RA-74-2, high temperature fermentation above $40^{\circ}C$ was more appropriate for industrial utilization because organic nitrogen was not necessary to economical fermentation. As the naked barley media just prior to industrial inoculation, taken from the Poongkuk alcohol industry Co., were used, 9.6% (v/v) ethanol was produced at $40^{\circ}C$ for 48 hours in jar-fermentor scale (actually, 9.5-9.8% (v/v) ethanol was produced at 30~$32^{\circ}C$ for 100 hours in industrial scale). The ethanol productivity was increased by the high glucoamylase activity as well as the high metabolic ratio at $40^{\circ}C$ Therefore, if the thermotolerant yeast RA-74-2 would be used in industrial scale, we could obtain a high productivity and saving of the cooling water and energy. Meanwhile, the RA-912 produced 6%(v/v) ethanol in 10% glucose media at $45^{\circ}C$ and showed the less ethanol-tolerance compared with industrial strains. As the produced alcohol was recovered by the vacuum evaporator at $45^{\circ}C$ in 15% glucose media, the final fermentation ratio was enhanced (76% of theoretical yields). This suggest that a hyperproductive process could be achieved by a continuous input of the substrate and continuous recovery of the product under vacuum in high cell-density culture.

Production and Chracteristics oil Antifungal agents from Bacteria (세균으로부터 항진균성 물질의 생산 및 특성)

  • For the production of antifungal compound, strain B-1 was used as a strong producing strain among bacteria isolated from various soil samples. The optimum medium for the production of antifungal compound was PDB (potato starch 0.4%, dextrose 2%, pH5.1). The optimum conditions for the production of antifungal compound didn't affect on the carbon and nitrogen sources. The produced compound showed broad antimicrobial activity to the tested strains such as five fungi and four bacteria. The optimum pH and temperature of the production antifungal compound were pH 5.0 and 28$^{\circ}C$, respectively. Ether extrct (1$\mu\textrm{g}$/${\mu}\ell$) of culture broth was confirmed inhibitory zone by the thin layer chromatography and plate assay. The antimicrobial compound was unstabled after heat (121$^{\circ}C$) trsatment. Strain B-1 was mass cultured in a 5-liter tormentor, containing 3 liters of PDB medium at 28$^{\circ}C$, pH 5.0, 120 (pm with aeration (1L/min).

Effect of Conidial Number and Nutrition on the Germination of Conidia in Septoria glycines (분생포자수 및 영양상태가 대두갈색무늬병균의 분생포자 발아에 미치는 영향)

  • Conidial germination of Septoria glycines Hemmi, brown spot fungus of soybean, was studied by slide germination test. Poor conidial germination of S. glycines was observed on sterile distilled water, but potato dextrose agar(PDA) and distilled water floated with soybean leaf disc furnished a satisfactory medium for conidial germination. Exogenous supply of carbon source was essential for conidial germination, while phosphorous and potassium were not evident as that for carbon. Soluble starch was the most suitable as a carbon source for conidial germination and followed by D-glucose, D-galactose and lactose in that order. Maximum germination was attained in the $5\times10^{-2}mol$. concentration of glucose. Germination was decreased with increment of conidial concentration and was almost completely suppressed in the density of 10,000 conidia per $mm^2$. It suggested existing a self-inhibitor(s). Non-washed conidia germinated more than washed conidia and this was obvious when the conidia density was over $2\times10^3$ conidia per $mm^2$ on the dry agar block.

Effect on Mycelial Growth and Fruit Body Development According to Supplementary Nutrition in Bag Cultivation of Neolentinus lepideus (잣버섯 봉지재배시 영양원이 균사배양 및 자실체 생육에 미치는 영향)

  • This study was carried out to elucidate suitable supplementary nutrition for Neolentinus lepideus cultivation. The main substrate for cultivation was douglas fir sawdust, and supplementary materials for experiment were corn meal, corn hull, corncob, beet pulp, and potato starch. The higher amount of total nitrogen source was showed in corn meal (0.45%) than any other nutritive materials. Incubation ratio and cultivation period showed the best 97% and the shortest 43 days on medium with corn meal substrate. The fruit body yield and biological efficiency on medium with 10% corn meal were highest 122 g/1 kg and 35%, respectively. Factor analyses concerning fruiting yield showed total nitrogen source (r = 0.95) and crude fat (r = 0.65). These results are recommended that N. lepideus cultivation is corn meal material as the source of the supplementary nutrition and total nitrogen source as the factor of fruiting body yield on the Douglas fir sawdust medium.
