A Study on Contents of Heavy and Trace Metal of the Agricultural Products around Mines Located in Chollanam-Do - with Yeongam, Boseong, Gokseong, Yeocheon Gun in the Center - (전라남도 광산 주변에서 수확한 농산물 충의 중금속 및 미량금속 함량 조사 - 영암, 보성, 곡성, 여천군을 중심으로 -)

  • To know a degree of pollution in agricultural products cultivated around 4 unworked mines located in Chollanam-Do, we investigated a concentration of heavy metal and trace metal to provide the basic data for its residual limits. 28 samples of seven kinds of agricultural products(lettuce, onion, potato, radish, pepper, pumpkin, bean) collected from 4 unworked mines located in Chollanam-Do in 2001 were analyzed by Mercury Analyzer for mercury and Atomic Absorption spectrophotometer for Pb, As, Cd, Cu, Mn, and Zn. Hg contents were detected N.D. ∼trace level(0.01 ppm and less) and As content were detected N.D. ∼ 0.029 lpm but most of same)leE were not detected. Cd contents were detected N.D∼0.124 ppm. The results of Hg, As, Pb and Cd content showed that for all the 7 species of agricultural products studied, none have accumulated levels dangerous enough to Pose health problems. The average contents of Cu were 3.070 ∼ 7.825 ppm in bean, the Mn were 3.688 ∼23.935 ppm in lettuce ailed the Zn were 5.690 ∼21.171 ppm in bean, respectively.

Incidence of Viral Diseases on Red Pepper in Yeongyang-Gun, Gyeongbuk Provice (경북 영양지역 노지고추의 바이러스병 발생양상)

  • Incidence of virus diseases in red pepper of open field in Yeongyang-Gun, Gyeongbuk Province was investigated by reverse transcription-polymerase chain reaction in 2012-2016. The infection rate of Cucumber mosaic virus (CMV), Broad bean wilt virus 2 (BBWV2), Pepper mottle virus, Potato virus Y, Pepper mild mottle virus and Tomato spotted wilt virus was 46.1%, 41.5%, 2.0%, 2.0%, 4.4% and 0.1% respectively. Incidence rate of single and mixed infection was 31.2% and 62.6%. Most of single infections were CMV and BBWV2. Among mixed infections, the incidence rate of CMV+BBWV2 mixed infection was the highest as 49.3% and most of mixed infections of triplex and tetraplex included CMV+BBWV2 mixed infection. CMV single infection caused mosaic, chlorosis, yellowing and vein necrosis and BBWV2 single infection induced cholosis and mosaic. CMV+BBWV2 mixed infection caused severe mosaic with chlorosis or malformation.

Antioxidant and Inhibition on Angiotensin Converting Enzyme Activity of Colored Potato Extracts (유색감자 추출물의 항산화 및 항고혈압 활성)

  • This experiment was conducted to enhance the colored potatoes utilization and to determine the biological activity of colored potato extracts. In order to understand the factors responsible for the potent antioxidant and antihypertensive ability of colored potatoes, it has been evaluated for anti-oxidative activity using Fenton's reagent/ethyl linoleate system and for free radical scavenging activity using the 1,1-diphenyl-2-picryl hydrazyl free radical generating system. There were significant differences of antioxidant activities in $50{\mu}g/mL$ extracts treatment among different colored potatoes. About two-fold higher radical scavenging activity was found in 'Daegwan 1-102', 'Daegwan 1-104' and 'Jasim' compared to that in 'Superior'. Based on the flesh color tested, potatoes with purple tuber showed higher radical scavenging activity than red potatoes, while white potato showed the lowest radical scavenging activity. The ability of 80% ethanol extracts from colored potatoes to influence the inhibitory activity of angiotensin converting enzyme(ACE) and xanthine oxidase(XOase) has also been investigated. Expect 'Jasim', the high levels of inhibition activity of xanthine oxidase in two colored potatoes such as 'Daegwan 1-102' and 'Daegwan 1-104' were highly correlated to $IC_{50}$ values of ACE inhibition activity. The various therapeutic benefit claims in the new functional medicinal usage of colored potatoes ascribed to the phenolic compounds and anthocyanin. This result revealed that the extracts of colored potatoes are expected to be good candidate for development into source of free radical scavengers and anti-hypertentive agent.

Occurrence and Distribution of Weed Species on Upland Fields in Chungnam Province (충남지역 밭작물 재배지 잡초발생 및 분포현황)

  • This study was conducted to provide basic information for weed control by surveying the occurrence of weed species. Surveys of weed species occurred in food crop (barley, maize, potato, soybean) fields were conducted in Chungnam province from April to September in 2014. Total 321 sites of food crop fields in 17 City / Gun in Chungnam province were investigated. From the result of this survey, 130 weed species in 36 families were identified and classified to 77 annuals, 27 biennials and 26 perennials. Based on the occurrence ratio, the most dominant weed species in Chungnam province food crop fields were Digitaria ciliaris (10.19%), followed by Conyza canadensis (7.82%), Portulaca oleracea (5.22%), Chenopodium album var. centrorubrum (5.21%), Capsella bursa-pastoris (3.47%) and Alopecurus aequalis (3.47%). Forty-four exotic weed species were identified. This information could be useful for estimation of future weed occurrence, weed population dynamics and establishment of weed control methods in food crop fields of Chungnam province.

Comparative Study on Biological Activities of Colored Potatoes, Hongyoung and Jayoung Cultivar (유색감자 홍영 및 자영 추출물의 생물학적 활성 비교)

  • This experiment was conducted to enhance the colored potatoes utilization and to determine the biological activity of colored potato extracts. In order to understand the factors responsible for the potent anti-oxidant ability of colored potatoes, it has been evaluated for anti-oxidative activity using oxygen radical absorbance capacity (ORAC) assay. 'Hongyoung' extract was significant anti-oxidant activities in ORAC assay. About two-fold higher radical absorbance capacity was found in 'Hongyoung' compared to that in 'Jayoung'. The ability of 80% ethanol extracts from colored potatoes to influence the inhibitory activity of nitric oxide (NO) and nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2) has also been investigated. The various therapeutic benefit claims in the new functional medicinal usage of colored potatoes ascribed to the phenolic compounds and anthocyanin. This result revealed that the extracts of colored potatoes are expected to be good candidate for development into sources of free radical scavenger or COX-2 inhibiting agents.

Stem Rot on Ligularia fischeri Caused by Sclerotium rolfsii in Korea (Sclerotium rolfsii에 의한 곰취 흰비단병)

  • In June 2012 and 2013, a destructive stem rot symptoms of Ligularia fischeri occurred sporadically in Hoengseong-gun and Pyeongchang-gun Gangwon-do, Korea. The typical symptom included water-soaking on the main stem, rotting, wilting and blighting, which eventually leads to death of the plant. White mycelial mats were spread over lesions and brown sclerotia were formed on stems and near soil surface. The sclerotia were white to brown, spherical or irregular, 1-3 mm in size on potato dextrose agar (PDA), The optimum temperature range of hyphal growth was $25-30^{\circ}C$ and the hyphal diameter was $4-10{\mu}m$. The typical clamp connections were observed in the hyphae of the fungus grown on PDA. The resulting sequence of 695 bp was deposited in GenBank. A BLAST search revealed that sequences of the this isolates showed >99% identity with those of Sclerotium rolfsii. On the basis of the morphological characteristics and phylogenetic analyses of molecular markers ITS rDNA, the fungi were identified as S. rolfsii. A pathogenicity test was carried out to fulfill Koch's postulates. To our knowledge, this is the first report of S. rolfsii on Ligularia fischeri in Korea.

Alternaria Leaf Spot Caused by Alternaria mali on Black Chokeberry in Korea (Alternaria mali에 의한 아로니아 점무늬낙엽병)

  • In early June 2014, leaf spot symptoms were observed on black chokeberry (Aronia melanocarpa) in Yesan-gun and Geumsan-gun, Chungcheongnam-do in Korea. The initial symptoms on leave surfaces were brown small-circular spots with a yellow halo lesion, and gradually the small spots were fused, all of infected leaves dropped eventually. A fungus were isolated from the initial lesion, and cultured on potato dextrose agar. Colony color on upper surface of plate varied from olive gray to charcoal gray. Size of conidia mostly extend to $19-50{\times}5-9{\mu}m$ in nature and $20-59{\times}8-13{\mu}m$ in culture, with 3-8 transverse septa and usually no longitudinal septum or only 1 longitudinal septum in 1-3 of the transverse compartments, and also have a short or long beak. Pathogenicity was investigated using wounded or unwounded black chokeberry and apple leaves. After 7 days of inoculation, leaf spots were similar to the symptoms naturally occurred in the field. On the basis of mycological characteristics, pathogenicity, and ITS rDNA sequence analysis, this fungus was identified as Alternaria mali. This is the first report of Alternaria leaf spot on black chokeberry caused by A. mali in Korea.

Characteristics of Unrecorded Yeasts, Rhodosporidium fluviale, Rhodosporidium paludigenum, Candida sp. 80-J-3 and Kluyveromyces thermotolerans Isolated from Wild Flowers in Korea (국내 야생화들로부터 국내 미기록 효모, Rhodosporidium fluviale, Rhodosporidium paludigenum과 Candida sp. 80-J-3, Kluyveromyces thermotolerans의 특성)

  • Rhodosporidium fluviale 201-C-1 and Kluyveromyces thermotolerans 150-JE-2(1) were isolated from wild flowers in Geumsan-gun, Chungcheongnam-do and Wando-gun, Jeollanam-do, respectively. Rhodosporidium paludigenum 86-J-1 and Candida sp. 80-J-3 were also isolated from flowers in Jeju island, Korea. Characteristics for these yeasts were not reported before. Therefore, morphological and physiological characteristics for these yeasts were investigated in this study. R. fluviale 201-C-1 was halophilic asporogenous yeast and formed pseudomycelium. It also grew at $30^{\circ}C$ and pH 4.0-6.0. K. thermotolerans 150-JE-2(1) was sugar-tolerant and halotolerant asporogenous yeast. It formed pseudomycelium and showed positive urease activity. Candida sp. 80-J-3 and R. paludigenum 86-J-1 were asporogenous yeasts, and grew well in yeast extract-peptone dextrose(YPD) medium, potato-dextrose medium as well as vitamin-free medium and YPD medium containing 5% NaCl.

Distribution of Habitats and Ecology of Weedy Melons (Cucumis melo var. agrestis Naud.) in Korea (우리나라 야생잡초 참외의 자생지 분포지역 및 생태)

  • Natural habitats of weedy melons were distributed on the islands along and on the west and south coasts of Korean peninsula including Boryeong, Seosan (Taean), Seocheon, Okgu, Buan, Gochang, Yeonggwang, Muan, Shinan, Haenam, Jindo, Wando, Goheung, Yeocheon, Hadong, Namhae, Goseong, Tongyeong, Geoje, and Jeju islands including Jeju city, Bukjeju-gun and Nam Jeju-gun. Weedy melons were found growing wildly in or around the cultivated lands in these regions. Natural habitats of weedy melons were in and around the cultivated lands. Weedy melon plants were found most often in soybean fields, followed by fields of mungbean, sweet potato, pepper, sesame, cotton, and peanuts. The plants were also found growing wild in foxtail millet fields, rice paddy levees along the streams, upland field edges, watermelon fields, corn fields, vegetable gardens near farmhouse, orange fields, compost piles, fallow fields, roadside and home gardens. They inhabited in sunny and a little dry spaces in relatively low-height crop plant fields in general. The time of fruit maturity was from early July to late October with the most frequency in September according to post survey answer. Fruits dropped off from the fruit stalk when matured. This phenomenon was thought beneficial for perpetuation in the wild. The fruits were being used commonly for food and toys for children. It was thought that weedy melons were perpetuating through the cycle of human and animal feeding of the fruits, human and animal droppings, often mixed in compost, and application of the compost to crop fields by human.

Gray Mold of Pisum sativum Caused by Botrytis cinerea in Korea (Botrytis cinerea에 의한 완두 잿빛곰팡이병)

  • In March of 2003, the gray mold occurred severely on pea (Pisum sativum) grown in the green houses and open fields in Changseon-myon, Namhae-gun, Gyeongnam Province, Korea. The symptoms started with water-soaking lesions in the leaves, stems and pods. The typical symptoms of the leaves, stems and pods became withered, discolored and blighted, and then eventually died. The mycelia, conidia and conidiophore of the pathogen appeared on leaves, stems and pods. The conidia were gray, 1-celled, mostly ellipsoid or ovoid in the shape and were $8{\sim}17{\times}5{\sim}13\;{\mu}m$ in size. Conidiophores were $14{\sim}30\;{\mu}m$ in size. The sclerotia were formed abundantly on potato dextrose agar. The optimum temperature for sclerotial formation was $20^{\circ}C$. Pathogenicity of the causal organism was proven according to Koch's postulate. The causal organism was identified as Botrytis cinerea based on mycological characteristics. This is the first report on gray mold of Pisum sativum caused by B. cinerea in Korea.
