Ethanol Induces Cell Death by Activating Caspase-3 in the Rat Cerebral Cortex

  • Ethanol has long been implicated in triggering apoptotic neurodegeneration. We examined the effects of ethanol on the rat brain during synaptogenesis when a spurt in brain growth occurs. This period corresponds to the first 2 postnatal weeks in rats and is very sensitive to ethanol exposure. Ethanol was administered subcutaneously to 7-day- postnatal rat pups by a dosing regimen of 3 g/kg at 0 h and again at 2 h. Blood ethanol levels peaked ($677{\pm}16.4mg/dl$) at 4 h after the first ethanol administration. The cerebral cortexes of the ethanol-treated group showed several typical symptoms of apoptosis such as chromosome condensation and disintegration of cell bodies. Activated caspase-3 positive cells were found in the cortex within 2 h of the first injection, and reached a peak at 12 h. In addition, TUNEL staining revealed DNA fragmentation in the same regions. These results demonstrate that acute ethanol administration causes neuronal cell death via a caspase-3-dependent pathway within 24 h, suggesting that activation of caspase-3 is a marker of the developmental neurotoxicity of ethanol.

$Telazol^{\circledR}$ 투여가 임신 Rat의 생식에 미치는 영향 (The Effect of Exogenous $Telazol^{\circledR}$ on the Reproduction in Pregnant Rat)

  • The reproductive effect of $Telazol^{\circledR}$ was studied in pregnant rats. Two different doses (20 or 40 mg/kg/day) were injected on days one of 3, 5 or 8 of pregnancy and 3 consecutive days of 3~5 or 5~7 in pregnant rats. On days 8 or 21, the rats were anesthetized, the cesarean section was performed, the uterus was carefully examined, and the numbers of corpora lutea, implantation sites and viable fetuses, fetal and placental weights and pre-and post-implantation losses in rats were surveyed. Simultaneously the concentrations of sodium and potassium of the plasma, uterine and amniotic fluids were determined. And gestation period in pregnant rats, postnatal offspring survival and body weight gains were evaluated. Administration of $Telazol^{\circledR}$ did not exert a bad influence upon body weight gains in pregnant rats. Reproductive indices were normal. The concentrations of sodium and potassium of the plasma, uterine and amniotic fluids in pregnant rats remained unchanged. Gestation period of rats, postnatal offspring survival and body weight gains were normal. It is concluded that $Telazol^{\circledR}$ is devoided of significant adverse reproductive effects in pregnant rats.

  • 본 연구는 생후 발달 과정 동안 monocarboxylate transporter(MCT) isoform과 MCT의 발현 조절 단백질로 알려진 basigin(Bsg)과 embigin의 mRNA 발현을 흰쥐의 부정소에서 부위별로 real-time PCR 방법을 사용하여 알아보았으며, 에스트로젠과 에스트로젠 수용체 α의 작용에 의해 MCT1 발현이 조절되는지를 알아보기 위해 estrogen receptor α knockout(αERKO) 마우스를 이용하여 immunohistochemistry 방법을 통해 탐구하였다. 본 연구 결과는 다양한 MCT isoform(MCT1, 2, 3, 4와 8), Bsg과 embigin의 mRNA 발현이 부정소의 부위별로 연령에 따라 다르게 나타나며, 부정소에서 MCT1 단백질 발현은 corpus와 caudal 부위에서 apical 지역에 한정되어 나타나는 것을 보여 주었다. 또한 부정소에서 MCT1 단백질 발현은 에스트로젠 수용체 α의 존재 여부와 상관 없음이 보여졌다. 따라서, 본 연구는 MCT가 남성 생식기관인 부정소에서 정자 성숙과 저장을 위한 적절한 환경을 형성함으로써 남성 생식력의 유지에 관여 할 수 있음을 시사한다. 흰쥐의 Nucleus Raphe Magnus로부터 분리된 신경세포의 전기생리학적 성질 (Electrophysiological Properties of the Neurons Dissociated from the Nucleus Raphe Magnus in Postnatal Rats)

  • Neurons in the nucleus raphe magnus are involved in descending modulation of nociceptive transmission. In this study, we attempted to investigate electrophysiological properties of the NRM neurons dissociated from the postnatal rat medulla. The NRM neurons in the coronal slices of and the dissociated neurons from the postnatal rat medullae were immunohistochemically identified using antibody against serotonin. Relatively small number of neurons were positively stained in both preparations. The positively stained neurons displayed large cell body with double or multiple neurites. Using whole-cell patch clamp configuration ionic currents were recorded from the dissociated NRM-like neurons selected by criteria such as size and shape of cell body and cell population. Two types, high- and low-threshold, of voltage-dependent calcium currents were recorded from the dissociated NRM-like neurons. Some neurons displayed both types of calcium currents, whereas others displayed only high-threshold calcium current. Voltage-dependent potassium currents were also recorded from the dissociated NRM neurons. Some neurons displayed both transient outward and delayed rectifier currents but others showed only delayed rectifier current. These results suggest that there are at least two types of calcium currents and two types of potassium currents in the dissociated NRM neurons.

SD rat 망막신경절세포의 생후 성숙기간에 따른 빛 자극 반응 특성 (Characteristics of Light-evoked Retinal Ganglion Cell Activity with Postnatal Maturation in SD Rat)

  • 한국형 인공시각장치 개발 목적으로 토끼망막을 사용하여 차후 개발될 인공시각장치에 인가하기 위한 적절한 전기자극 파라미터를 추출하는 실험을 수행하였다. 유전적 망막변성모델인 RD mouse (rd/rd (C3H/HeJ))를 망막질환모델로 사용하기 앞서 본 연구에서는 SD rat을 사용하여 설치류의 생후성숙기간 동안 변화하는 망막신경절세포의 빛 자극에 대한 변화 양상을 전반적으로 파악하는 control 실험을 하였다. 망막신경절세포의 흥분파는 8${\times}$8의 MEA (multi- electrode array)로 기록하였다. 개안 이전시기(pre-eye opening period)인 생후 15일까지는 망막신경절세포의 자발적 집단발사현상 (moving spontaneous bursts)이 옮겨 다니는 것을 발견하였다. 이 시기의 쥐들은 시각기능이 완성되지 않아 빛 자극에 의해 유발되는 흥분파를 보이지 않으며 오로지 이들의 촉각(tactile sense)에 의하여서만 이동하는 것을 관찰하였다. 그러나 생후 2주(post-eye opening period)가 지나면, 빛 자극에 의해 ON, OFF, ON/OFF 반응이 유발됨을 확인할 수 있었다. 전체적인 ON, OFF, 그리고 ON/OFF 망막신경절세포의 양상은 각각 $40\%,\;50\%,\;5\%$였으며, 이들 세포가 확인된 안구에서의 해부학적인 위치는 등쪽 관자방향(dorso-temporal)이 $50\%$, 배쪽 방향(ventral)이 $37.5\%$, 그리고 등쪽 코 방향(dorso-nasal)이 $12.5\%$로 나타났다. 설치류 망막을 대상으로 빛 자극에 의한 반응을 알아보는 실험을 할 때 생후 2${\~}$3주령이 가장 적합한 시기임을 확인하였다. 것이 바람직하다고 판단되었다.의한 발아촉진 효과는 종피의 탈색과 부식으로 인한 광흡수의 증대에서 기인되는 것으로 추측된다.3월 8일과 3월 15일 피복처리구의 그린업이 가장 효과적이었다. 한국잔디 및 한지형 잔디의 비닐 피복으로 인한 초봄 그린업 촉진시 유의할 점은 충분한 수분 유지를 위해 비닐 피복전에 관수를 하거나 비온 후에 비닐 피복하는 것이 좋으며, 비닐 제거시 잔디의 일소현상의 피해를 줄이기 위해 흐린날 비닐을 제거하는 것이 좋을 것으로 판단되었다.유발식이 급여로 $524\%$가 증가된 동맥경화지수는 질경이 에틸아세테이트 분획 병합투여로 대조군에 비하여 $40.2\%,\;51.2\%$가 감소됨으로써 동맥경화 발병 위험율이 대조군에 비하여 감소되었다. HDL-콜레스테롤 함량/총콜레스테롤 함량 비는 고콜레스테롤혈증 유발 식이 급여로 정상군보다 $73.9\%$가 감소됐으나 질경이 에틸아세테이트 분획 병합투여로 대조군에 비하여 유의한 증가를 나타냈다. 26두가 발정 반응($96.3\%$)을 나타내어 2.75${\~}$3.5의 경우가 2.50 이하의 경우에 비하여 높은 발정반응을 나타내었다.osterone과 0.40의 정의 상관이었고 근내지방도와 creatinine 농도간에는 -0.55의 비교적 높은 음의 상관계수가 추정되었으나 유의성은 인정되지 않았다. 6. 한우 비거세우의 도살시 혈청 성분 농도와 도체형질과의 상관에서 육량지수는 연령에 대해 보정한 HDLC 농도와 -0.71의 음의 상관이었으며, 도체율은 globulin과 0.70의 높은 정의 상관이었고, 등지방두께는 연령으로 보정된 HDLC와 0.69로 정의 상관관계를 보였다. 도체중은 triglyceride와 0.51의 정의 상관, 배최장근단면적은 testosterone과 -0.91,

Maternal caffeine consumption has irreversible effects on reproductive parameters and fertility in male offspring rats

  • Objective: Concerns are growing about the decrease in male reproductive health. Caffeine is one of the popular nutrients that has been implicated as a risk factor for infertility. In the present study, we examined whether in utero and lactational exposure to caffeine affects the reproductive function of the offspring of rats. Methods: Pregnant rats received caffeine via drinking water during gestation (26 and 45 mg/kg) and lactation (25 and 35 mg/kg). Body and reproductive organ weight, seminiferous tubule diameter, germinal epithelium height, sperm parameters, fertility rate, number of implantations, and testosterone level of the offspring were assessed from birth to adulthood. Results: Significant dose-related decreases were observed in the body and reproductive organ weight, seminiferous tubule diameter, and germinal epithelium height of the offspring. Sperm density had declined significantly in offspring of the low-dose and high-dose groups, by 8.81% and 19.97%, respectively, by postnatal day 150. The number of viable fetuses had decreased significantly in females mated with male offspring of the high-dose group at postnatal days 60, 90, 120, and 150. There were also significant reductions in testosterone levels of high-dose group offspring from birth to postnatal day 150. Conclusion: It is concluded that maternal caffeine consumption impairs gonadal development and has long-term adverse effects on the reproductive efficiency of male offspring rats.

신문혈 자침이 어린 백서 해마의 단백질 발현에 미치는 영향 (Proteomic Changes by Acupuncture Stimulation at HT7 in the Hippocampus of Rat Pups)

  • Objectives : Hippocampus, a region of temporal lobe, plays an important role in the pathogenic mechanisms of brain diseases such as Alzheimer's disease, depression and temporal lobe epilepsy. This research is designed to investigate hippocampal changes after acupuncture stimulation at Shinmun(HT7) using 2-dimensional gel electrophoresis(2-DE). Methods : On postnatal-day 15, rat pups were randomly devided into Normal(NOR) or HT7 group. All of Pups kept with their mothers for 7 days, but pups in HT7 group received acupuncture stimulation at HT7 daily. On postnatal-day 21, hippocampus of each rat pup was dissceted 30 minutes after last acupuncture stimulation and the protein expressions were investigated using 2-DE. Results : After acupuncture stimulation at HT7, expression of 20 proteins were significantly increased. Succinate semialdehyde dehydrogenase, glyceraldehyde-3-phosphate dehydrogenase-like, transketolase, aconitate hydratase and phosphoglucomutase-1 were related to glucose methabolism. Eukaryotic initiation factor(eIF) 4A-II, eIF 4A-III, mitochondrial Tu translation elongation factor and chain A of crystal structure of the 70-Kda heat shock cognate protein involve in the protein synthesis in ribosome. Tubulin ${\beta}$-4 chain, tubulin T ${\beta}$-15 and tubulin ${\alpha}$-1B chain comprise cytoskeleton. Glutathione S-transferase(GST) ${\omega}$-1, GST P and GST Yb-3 can reduce oxidative stress. ${\beta}$-soluble N-ethylmaleimide-sensitive fusion protein attachment protein is required for vesicular transport between the endoplasmic reticulum and the Golgi apparatus, glycerol-3-phosphate dehydrogenase plays a major role in lipid biosynthesis, creatine kinase U-type catalyses the conversion of creatine and consumes adenosine triphosphate to create phosphocreatine and adenosine diphosphate. Platelet-activating factor acetylhydrolase IB subunit alpha and voltage depedent anion-selective channel protein 2 were also increased. Conclusions : The results suggest that acupuncture stimulation at HT7 may enhance glucose and lipid metabolism, protein synthesis, cytoskeletal substance and anti-oxidative stress in hippocampus.

기관형 배양에서 흰쥐 태자 폐상피세포의 분화 (Differentiation of the Fetal Rat Pulmonary Epithelial Cells in Organotypic Culture)

  • In order to study the differentiation of the epithelial cells during the development of fetal rat lung tissue, histological changeB in organotypic culture and in vivo were examined. Light microscopy and scanning electron microscopy were used to analvre the histological change in rat lung from the 15th nary of gestation to the 111th nary after birth. In organotypic culture system, the pulmonary epithelial cell differentiation was studied by scanning electron microscopy. The results obtained from this study were as follows. 1. During deveiopment of lung, the glandular stage lasted from the Isth day to the lsth naut of gestation; the canalicular stage from the 17th nay to the 19th naut of gestation; the saccuiar stage from 20th nary to the birth. Alveolar stage was observed at the 3rd nary of postnatal rat lung. 2. In organotvpic culture of fetal rat lung cells organized alveolar-like structures resembling those of in uiuo state were observed on the gelatin matrix. In contrast with in vivo state, fetal lung cells formed group of type ll pneumocytes predominently along the contours of the matrix. These cells have large apical surface, short microvilli and secreted materials which may be sunactant. These results suggested that an orsanotypic culture retaining epithelial- -mesenchvmal relationships is appropriate culture model to study the pulmonary epithelial cell (especially type ll pneumocvte) differentation.

The Expression Patterns of Connexin Isoforms in the Rat Caput Epididymis During Postnatal Development

  • Intercellular interactions are important for the proper development and regulation of tissue function. This is especially necessary in the epididymis, a part of male reproductive tract where sperm become mature and acquire their fertilizing capacity. The caput region of the epididymis consists of several types of cells, including principal, basal, and apical cells. Direct intercellular communication is thus required to precisely regulate the functions of the caput epididymis. In this regard, connexin (Cx) is a molecule that forms channels, which allow the direct exchange of small molecules between cells, enabling intercellular communication. In this study, the expression of Cx isoforms in the caput epididymis at different postnatal ages was determined by using quantitative real-time polymerase chain reaction analysis. Nine of 13 Cx isoforms were detected. The transcript levels of Cx30.3, 31, 31.1, 32, and 40 were highest at 45 days of age, while the expression of Cx43 and 45 gradually decreased with age. A substantial fluctuation of Cx26 expression was detected, with significant decreases before and during puberty, followed by a transient increase at adult-hood and rapid decreases at an old age. A significant increase in Cx37 transcript was observed at 25 days of age, followed by gradual decreases at adult and old ages. These results indicate the significant differential expression of various Cx isoforms in the caput epididymis during postnatal development. It further suggests that the functional regulation and developmental maturation of the caput epididymis are highly related to the postnatal age-related differential expression of Cx isoforms.

Expressional Analysis of Glucose Transporter Isoforms in the Efferent Ductules of Male Sprague Dawley Rat during Postnatal Development

  • A cell frequently utilizes glucose as a fuel of energy and a major substrate of lipid and protein syntheses. A regulation of glucose movement into and out of the cells is precisely controlled by cooperative works of passive and sodium-dependent active processes. At least 13 glucose cotransporter (Slc2a, GLUT) isoforms involve in passive movement of glucose in cells. The efferent ductules (EDs) play in a number of important functions for maintenance of male fertility. In the present study, using real-time PCR analysis, we determined gene expression of five Slc2a isoforms in the EDs. In addition, we compared expression levels of these Slc2a isoforms according to postnatal development ages, 1 week, 2 weeks, 1 month, and 3 months. Results from the current study showed that expression of Slc2a1, Slc2a3, and Slc2a5 mRNAs reached the highest levels at 1 month of age, followed by a transient decrease at 3 months of age. In addition, the level of Slc2a4 mRNA reminded at steady until 1 month of age and was significantly reduced at 3 months of age, whereas the highest level of Slc2a 8 mRNA was detected at 2 weeks of age. Data from the present study indicate a differential expression of various Slc2a isoforms in the ED according to postnatal ages. Thus, it is believed that glucose movement through the epithelial cells in the ED would be regulated by the coordinated manner among Slc2a isoforms expressed at a given age.
