Recently, the recognition is changing about cultural heritage, and the various types of buildings or facilities of modern or contemporary times have been designated as cultural properties after that Registered Cultural Properties System is enforced. The purpose of this study is to survey how the newly born the historic buildings of modern or contemporary times through the remodeling process of the Studio 'Byeol'(the Training Center for Performers of Korean Traditional Music) in the National Theater of Korea so-called a microcosm of performing arts history. In the process, it will examine the merits and demerits of various alternatives and the direction of the remodeling etc., and propose an utilization as a basic data of post evaluation for the remodeling of a historic building. The result are as followings. Firstly, the remodeling that gave a new physical properties to a building can be used a method of conservation and reuse on a historic building. The remodeling of a historic building must be eclectically progress between the owner and the citizen or the economic value and the historicity conservation. And, the remodeling of historic buildings such as the Training Center for Performers of Korean Traditional Music must consider the conservation of the exterior walls in whole or in part at least. Secondly, an architect Lee Hee Tae(李喜泰) who had been to develop his own architectural vocabulary and to test based on the korean traditional architecture and the Training Center for Performers of Korean Traditional Music must be newly evaluated today. Lastly, the remodeling alternatives of the Training Center for Performers of Korean Traditional Music have been analyzed with three types, which is 'repairing only the interior which maintains the size and an appearance of present', 'extending the outer wall to the external column line', 'extending the basement'. And, it was analyzed with the appropriate final decision that it remodels only the interior in the current situation because of a historicity, a budget, a relevant law etc.

In this paper, an alternative reliability-based methodology is developed and implemented on the safety evaluation of structures subjected to seismic loading. To effectively elaborate the approach, structures are represented by finite elements and seismic loading is applied in time domain. The accuracy of the proposed reliability-based methodology is verified using Monte Carlo Simulation. It is confirmed that the presented approach provides adequate accuracy in calculating structural reliability. The efficiency and robustness in problems related to performance-based seismic design are verified. A structure designed by experts satisfying all post-Northridge seismic design requirements is studied. Rigidities related to beam-to-column connections are incorporated. The structure is excited by three suites of ground motions representing three performance levels: immediate occupancy, life safety, and collapse prevention. Using this methodology, it is demonstrated that only hundreds of deterministic finite element analyses are required for extracting reliability information. Several advantages are documented with respect to Monte Carlo Simulation. To showcase an applicability extension of the proposed reliability-based methodology, structural risk is calculated using simulated ground motions generated via the broadband platform developed by the Southern California Earthquake Center. It is validated the accuracy of the broadband platform in terms of structural reliability. Based on the results documented in this paper, a very solid, sound, and precise reliability-based methodology is proved to be acceptable for safety evaluation of structures excited by seismic loading.
In earthquake engineering, dynamic analyses are usually conducted by using a nonlinear analytical model of the entire building in order to identify the performance against earthquakes. At the same time, a large number of dynamic analyses are required to consider uncertainties on analytical models and ground motions. Therefore, it is necessary for the analytical model to be adequate, that is to say, the runtime should not be too long as the entire building is modeled to be in much detail, or the nonlinear model should not yield outputs very far from the actual ones by excluding important behaviors too much. The analytical model is usually developed based on experimental results, which have been already conducted for reinforced concrete columns with relaxed details. Therefore, this study aimed at making analytical models to be able to simulate the hysteretic behavior of the columns simply and easily. The analytical model utilizes a lumped hinge model to represent nonlinear moment-rotation hysteretic behavior of RC columns, which is feasible for nonlinear dynamic analyses usually conducted in earthquake engineering and for matching the analytical model to test results. For the analytical model, elements and material models provided by OpenSees are utilized. The analytical model can define the envelope curve, pinching, and unloading stiffness deterioration, but shortcoming of this model is not to be able to consider axial force-moment interaction directly and to simulate strength deterioration after post-capping completely. However, the analytical model can still represent test results well by considering that the goal of this study is to propose a general way to represent the hysteretic behavior of RC columns with relaxed details, not to provide parameters for a refined hysteretic model that can be just applied case by case.

Objective : Thoracoscopic spinal surgery provides minimally invasive approaches for effective vertebral decompression and reconstruction of the thoracic and thoracolumbar spine, while surgery related morbidity can be significantly lowered. This study analyzes clinical results of thoracoscopic spinal surgery performed at our institute. Methods : Twenty consecutive patients underwent video-assisted thoracosopic surgery (VATS) to treat various thoracic and thoracolumbar pathologies from April 2000 to July 2006. The lesions consisted of spinal trauma (13 cases), thoracic disc herniation (4 cases), tuberculous spondylitis (1 case), post-operative thoracolumbar kyphosis (1 case) and thoracic tumor (1 case). The level of operation included upper thoracic lesions (3 cases), midthoracic lesions (6 cases) and thoracolumbar lesions (11 cases). We classified the procedure into three groups: stand-alone thoracoscopic discectomy (3 cases), thoracoscopic fusion (11 cases) and video assisted mini-thoracotomy (6 cases). Results : Analysis on the Frankel performance scale in spinal trauma patients (13 cases), showed a total of 7 patients who had neurological impairment preoperatively : Grade D (2 cases), Grade C (2 cases), Grade B (1 case), and Grade A (2 cases). Four patients were neurologically improved postoperatively, two patients were improved from C to E, one improved from grade D to E and one improved from grade B to grade D. The preoperative Cobb's and kyphotic angle were measured in spinal trauma patients and were $18.9{\pm}4.4^{\circ}$ and $18.8{\pm}4.6^{\circ}$, respectively. Postoperatively, the angles showed statistically significant improvement, $15.1{\pm}3.7^{\circ}$ and $11.3{\pm}2.4^{\circ}$, respectively(P<0.001). Conclusion : Although VATS requires a steep learning curve, it is an effective and minimally invasive procedure which provides biomechanical stability in terms of anterior column decompression and reconstruction for anterior load bearing, and preservation of intercostal muscles and diaphragm.
Japanese encephalitis virus (JEV) envelope (E) protein holds great promise for use in the development of a recombinant vaccine. Purified recombinant E (rE) protein may be useful for numerous clinical applications; however, there are limitations in using the Escherichia coli expression system for producing high-quality rE protein. Therefore, in this study, the yeast expression system was used to generate the rE protein. For protein production using the yeast system, the full-length JEV E gene was cloned into Pichia pastoris. SDS-PAGE and immunoblotting analysis demonstrated that the rE protein had a molecular mass of 58 kDa and was glycosylated. The predicted size of the mature unmodified E protein is 53 kDa, suggesting that post-translational modifications resulted in the higher molecular mass. The rE protein was purified to greater than 95% purity using combined ammonium sulfate precipitation and a SP-Sepharose Fast Flow column. This purified rE protein was evaluated for immunogenicity and protective efficacy in mice. The survival rates of mice immunized with the rE protein were significantly increased over that of Hyphantria cunea nuclear polyhedrosis virus E protein (HcE). Our results indicate that the rE protein expressed in the P. pastoris expression system holds great promise for use in the development of a subunit vaccine against JEV.
A FOV (field-of-view) of the HV-MSC (high voltage multi-scan CCD, $1024{\times}1024$ pixels) camera mounted in the post-column HV-GIF (high voltage gatan image filter) has been drastically enlarged by the projection lens current adjustment. An imaging area of the HV-MSC camera obtained at the lowest magnification (2,000x) is $112{\mu}m^2$ which corresponds to the recording area of the film at the magnification of 8,800x, while the achievable recording area is only $0.43{\mu}m^2$ at the same magnification without this technique. Ignoring the image distortion of less than 5%, we have designed an on-site reference graph to estimate projection lens currents for microscope magnifications above 8,800x, where the recording area on the HVMSC is same as that on the film.
An experimental study was performed for thin-walled rectangular concrete-filled tubular (CFT) columns. The present study mainly focused on evaluation of the axial load-carrying capacity of concrete-filled tubular columns using high-strength steel and slender section. The test parameters were width-to-thickness ratio, concrete strength, steel yield strength, and the use of stiffeners. Five specimens were tested under monotonic axial loading. Although elastic local buckling occurred in the slender-section specimens with high-strength steel, the specimens exhibited considerable post-buckling reserve. The test results also satisfied the predictions of a current design code. The specimens strengthened with vertical stiffeners exhibited improved strength and ductility when compared with the un-stiffened specimens.
Phosphomannomutase (ManB) converts mannose-6-phosphate (M-6-P) to mannose-1-phosphate (M-1-P), which is a key metabolic precursor for the production of GDP-D-mannose used for production of glycoconjugates and post-translational modification of proteins. The aim of this study was to express the manB gene from Escherichia coli in Lactococcus lactis subsp. cremoris NZ9000 and to characterize the encoded enzyme. The manB gene from E. coli K12, of 1,371 bp and encoding 457 amino acids (52 kDa), was cloned and overexpressed in L. lactis NZ9000 using the nisin-controlled expression system. The enzyme was purified by Ni-NTA column chromatography and exhibited a specific activity of 5.34 units/mg, significantly higher than that of other previously reported ManB enzymes. The pH and temperature optima were 8.0 and $50^{\circ}C$, respectively. Interestingly, the ManB used in this study had two substrate specificity for both mannose-1-phosphate and glucose-1-phosphate, and the specific activity for glucose-1-phosphate was 3.76 units/mg showing 70% relative activity to that of mannose-1-phosphate. This is the first study on heterologous expression and characterization of ManB in lactic acid bacteria. The ManB expression system constructed in this study canbe used to synthesize rare sugars or glycoconjugates.
Our paper shows the results of 302 samples of herb medicines about fungal contamination at Yakyeang markets in Seoul. The sample medicines were treated VICAM pretreatment and analysed by post column derivatisation procedure(PHRED-HPLC) with a fluorescence detector. Aflatoxin B1 was founded from 50.3% of samples, aflatoxin B2 was 39.7%, aflatoxin G1 was 21.2% and aflatoxin G2 was 23.5%. The detected ranges of aflatoxin B1, B2, G1 and G2 were from 0.1 to $57.2\;{\mu}g/kg$, 0.1 to $42.6\;{\mu}g/kg$, 0.1 to $23.5\;{\mu}g/kg$ and 0.1 to $9.5\;{\mu}g/kg$ respectively. Among total samples, 26 samples contained aflatoxin B1 violated the regulation (less than 10 ug/kg) for aflatoxin B1 of KFDA. From the result, we could presumed that more than a half of samples were contaminated by aflatoxins. Therefore, it seems to be necessary that the new safety giudeline will be established aflatoxin B2, G1 and G2 from herb medicines as aflatoxin B1.
The investigation was carried out to identify the active constituent in yeast extract for fruit body formation of Lentinula edodes. The result suggests that free thiamin which is hewn as the active substance for the fruiting of L edodes, was detected but thiamin mono, di, three phosphates were not detected in the yeast extract produced by Difco Co.. Therefore, the thiamin content of the yeast extract was determined, the yeast extract was fractionated to five portion by the post-column fluorescence method. The content of thiamin in yeast extract( 1g) was 0.436mg as thiamin hydrochloride. It was found that 76% of the total thiamin(0.332mg) was contained in fraction II. About 20% of the total thiamin(0.087mg) was present in fraction I, but not in fractions III, IV and V. In accordance with the contents of thiamin in the fractions, the fruit body formation was the highest by the treatment of fraction II(100%) and followed by fraction I (60%), V(50%), III(30%). Thiamin did not influence for the vegetative mycelial growth of L. edodes, but be used for fruit body formation.
