Detection of Mycobacterium leprae by Real-time PCR Targeting Mycobacterium leprae-Specific Repetitive Element Sequence

  • Mycobacterium leprae detection is difficult even with molecular biological techniques due to the low sensitivity of current methodologies. In this report, real-time PCR targeting the M. leprae-specific repetitive element (RLEP) sequence was developed as a new diagnostic tool and evaluated using clinical specimens. For this, M. leprae DNAs were extracted from skin biopsy specimens from 80 patients and analyzed by real-time PCR using TaqMan probe. Then, the detection efficiency of the real-time PCR was compared with that of standard PCR. In brief, the rate of positive detection by the standard PCR and real-time PCR was 32.50% and 66.25%, respectively. The results seemed to clearly show that the TaqMan real-time PCR developed in this study may be a useful tool for sensitive detection of M. leprae from clinical specimens.

Droop Control to Compensate Load Voltage Unbalance for Inverter-based Distributed Generations with Unequal Impedance Lines (불균등 임피던스 선로를 갖는 인버터기반 분산전원의 부하전압 불평형을 보상하는 드룹 제어)

  • This paper proposes a droop control scheme to compensate the unbalanced line-to-line voltage of unbalanced 3-phase load which is coupled with two inverter-based distributed generations through unequal impedance lines. Unbalanced line-to-line load voltages occur due to using single-phase loads, which brings about bad effects on the coupled inverters and the distributed generations. In order to compensate the unbalanced line-to-line voltages, a positive sequence voltage control was used for sharing the active and reactive power and a negative sequence control was used for reducing the negative sequence voltage. The feasibility of the proposed scheme was first verified by computer simulations, and then experiments with a hardware set-up built in the lab. The experimental results were compared with the simulation results to confirm the feasibility of the proposed scheme.

Comprehensive Coordinated Control Strategy of Virtual Synchronous Generators under Unbalanced Power Grid

  • When grid voltage is unbalanced, the grid-connected output current and power of Virtual Synchronous Generators (VSGs) are distorted and quadratic. In order to improve the power quality of a grid connected to a VSG when the grid voltage is unbalanced, a comprehensive coordinated control strategy is proposed. The strategy uses the positive sequence current reference command obtained by a VSG in the balanced current control mode to establish a unified negative sequence current reference command analytical expression for the three objectives of current balance, active power constant and reactive power constant. In addition, based on the relative value of each target's volatility, a comprehensive wave function expression is established. By deriving the comprehensive wave function, the corresponding negative sequence current reference value is obtained. Therefore, the VSG can achieve the minimum comprehensive fluctuation under the premise that the three targets meet the requirements of grid connection, and the output power quality is improved. The effectiveness of the proposed control strategy is verified by simulation and experimental results.

Unbalanced Power Sharing for Islanded Droop-Controlled Microgrids

  • Studying the control strategy of a microgrid under the load unbalanced state helps to improve the stability of the system. The magnitude of the power fluctuation, which occurs between the power supply and the load, is generated in a microgrid under the load unbalanced state is called negative sequence reactive power $Q^-$. Traditional power distribution methods such as P-f, Q-E droop control can only distribute power with positive sequence current information. However, they have no effect on $Q^-$ with negative sequence current information. In this paper, a stationary-frame control method for power sharing and voltage unbalance compensation in islanded microgrids is proposed. This method is based on the proper output impedance control of distributed generation unit (DG unit) interface converters. The control system of a DG unit mainly consists of an active-power-frequency and reactive-power-voltage droop controller, an output impedance controller, and voltage and current controllers. The proposed method allows for the sharing of imbalance current among the DG unit and it can compensate voltage unbalance at the same time. The design approach of the control system is discussed in detail. Simulation and experimental results are presented. These results demonstrate that the proposed method is effective in the compensation of voltage unbalance and the power distribution.

  • Parvimonas micra is Gram-positive, strict anaerobic, non-motile, and non-spore forming coccus. It is a member of oral flora and is related to oral infectious diseases as well as systemic diseases. P. micra KCOM 1037 (= ChDC B276) was isolated from human postoperative maxillary cyst lesion. Here, we present the complete genome sequence of P. micra KCOM 1037.

Development of a Quantitative Real-time Nucleic Acid Sequence based Amplification (NASBA) Assay for Early Detection of Apple scar skin viroid

  • An assay for detecting Apple scar skin viroid (ASSVd) was developed based on nucleic acid sequence based amplification (NASBA) in combination with realtime detection during the amplification process using molecular beacon. The ASSVd specific primers for amplification of the viroid RNA and molecular beacon for detecting the viroid were designed based on highly conserved regions of several ASSVd sequences including Korean isolate. The assay had a detection range of $1{\times}10^4$ to $1{\times}10^{12}$ ASSVd RNA $copies/{\mu}l$ with reproducibility and precision. Following the construction of standard curves based on time to positive (TTP) value for the serial dilutions ranging from $1{\times}10^7$ to $1{\times}10^{12}$ copies of the recombinant plasmid, a standard regression line was constructed by plotting the TTP values versus the logarithm of the starting ASSVd RNA copy number of 10-fold dilutions each. Compared to the established RT-PCR methods, our method was more sensitive for detecting ASSVd. The real-time quantitative NASBA method will be fast, sensitive, and reliable for routine diagnosis and selection of viroid-free stock materials. Furthermore, real-time quantitative NASBA may be especially useful for detecting low levels in apple trees with early viroid-infection stage and for monitoring the influence on tree growth.

  • The rate of convergence for an almost surely convergent series $S_n={\Sigma^n}_{i-1}X_i$ of independent random variables is studied in this paper. More specifically, when S$_{n}$ converges almost surely to a random variable S, the tail series $T_n{\equiv}$ S - S_{n-1} = {\Sigma^\infty}_{i-n} X_i$ is a well-defined sequence of random variables with T$_{n}$ $\rightarrow$ 0 almost surely. Conditions are provided so that for a given positive sequence {$b_n, n {\geq$ 1}, the limit law sup$_{\kappa}\geqn | T_{\kappa}|/b_n \rightarrow$ 0 holds. This result generalizes a result of Nam and Rosalsky [4].

  • Scott transformers have widely used to convert three phases into two phases and compensate the unbalance. Theoretically, the loads of the two secondary phases are same, no unbalance appears in the PCC(point of common coupling). But Due to the uncertainty of traction load, the unbalance are generally presented at the PCC. In this paper The amount of the voltage unbalance is expressed in the ratio of the negative sequence voltages to the positive sequence voltage. We tried to compensate the unbalance using SVC(Static Var Compensator)in an unbalance traction loads state by modeling. The SVC are installed and controlled to provide different amounts of reactive power compensation.

  • This paper presents a speed control method for the capacitor-run induction motor. The equivalent circuit of the motor is analyzed using the forward(Positive sequence) and backward(negative sequence) components, and simple circuit equations are obtained. Simulations for the speed control are performed by adjusting the voltage magnitude of the auxiliary winding. A prototype system has been implemented which consists of an inverter and a controller with TMS320C31 digital signal processor. The experimental results using 1/4hp capacitor-run induction motor show a good agreement with analyses.
