Novel porous composite ceramic bone scaffolds composed of biodegradable PHBV(polyhydroxybutyrate-co-hydroxyvalerate) and TA(toothapatite) have been fabricated for bone tissue engineering by a modified solvent casting and particulate leach-ing method with salt-contained heat compression technique. The results of this study suggest that the PHBV-TA composite scaffold, especially the scaffold containing 30 weight$\%$ of TA may be a good candidate far bone tissue engineering of non-load bearing area in oral and maxillofacial region.
Predicting the mechanical properties of the 3-D scaffold using finite element method (FEM) simulation is important to the practical application of tissue engineering. However, the porous structure of the scaffold complicates computer simulations, and calculating scaffold models at the pore level is time-consuming. In some cases, the demands of the procedure are too high for a computer to run the standard code. To address this problem, the representative volume element (RVE) theory was introduced, but studies on RVE modeling applied to the 3-D scaffold model have not been focused. In this paper, we propose an improved FEM-based RVE modeling strategy to better predict the mechanical properties of the scaffold prior to fabrication. To improve the precision of RVE modeling, we evaluated various RVE models of newly designed 3-D scaffolds using FEM simulation. The scaffolds were then constructed using microstereolithography technology, and their mechanical properties were measured for comparison.
Extraordinary properties of nanocomposites make them a primary replacement for many conventional materials. Anterior cruciate ligament (ACL) reconstruction, which is a frequent surgery in sport activities, is one of the fields in which nanocomposites could be utilized. In the present study, the mechanical properties of different porous scaffolds made from graphene nano-composites are presented ad load bearing capacity of these materials is calculated using finite element method. The numerical results are further compared with experimental published data. In addition, several geometrical and material parameters are analyzed to find the best configuration of nanocomposite scaffolds in reconstruction of ACL. Moreover, coating of detoxification chemicals are extremely easier on the nano-structured materials than conventional one. Detoxification potential of nano-composites in the injured body are also discussed in detail. The results indicated that nano-composite could be successfully used in place of auto- and allografts and also instead of conventional metallic screws in reconstruction of ACL.

Background To produce patient-specific nasal implants, it is necessary to harvest and grow autologous cartilage. It is crucial to the proliferation and growth of these cells for scaffolds similar to the extracellular matrix to be prepared. The pore size of the scaffold is critical to cell growth and interaction. Thus, the goal of this study was to determine the optimal pore size for the growth of chondrocytes and fibroblasts. Methods Porous disc-shaped scaffolds with 100-, 200-, 300-, and 400-㎛ pores were produced using polycaprolactone (PCL). Chondrocytes and fibroblasts were cultured after seeding the scaffolds with these cells, and morphologic evaluation was performed on days 2, 14, 28, and 56 after cell seeding. On each of those days, the number of viable cells was evaluated quantitatively using an MTT assay. Results The number of cells had moderately increased by day 28. This increase was noteworthy for the 300- and 400-㎛ pore sizes for fibroblasts; otherwise, no remarkable difference was observed at any size except the 100-㎛ pore size for chondrocytes. By day 56, the number of cells was observed to increase with pore size, and the number of chondrocytes had markedly increased at the 400-㎛ pore size. The findings of the morphologic evaluation were consistent with those of the quantitative evaluation. Conclusions Experiments using disc-type PCL scaffolds showed (via both morphologic and quantitative analysis) that chondrocytes and fibroblasts proliferated most extensively at the 400-㎛ pore size in 56 days of culture.

Tissue engineering techniques require the use of a porous biodegradable/bioresorbable scaffold, which server as a three-dimensional template for initial cell attachment and subsequent tissue formation in both in vitro and in vivo. Small intestinal submucosa (SIS) has been investigated as a source of collagenous tissue with the potential to be used as biomaterials because of its inherent strength and biocompatibility. SIS-loaded poly(L-lactide-co-glicolide)(PLGA) scaffolds were prepared by solvent casting/particle leaching. Characterizations of SIS/PLGA scaffold were carried out by SEM, mercury porosimeter, and so on. Muscle-derived stem cells can be differentiated in culture into osteoblasts, chondrocytes, and even myoblasts by the controlling the culture environment. Cellular viability and proliferation were assayed by 3-(4,5-dimethylthiazole-2-yl)-2,5-diphenyltetrazolium-bromide(MTT) test. Osteogenic differential cells were analyzed by alkaline phosphatase(ALP) activity. SIS/PLGA scaffolds were implanted into the back of athymic nude mouse to observe the effect of SIS on the osteoinduction compared with controlled PLGA scaffolds. Thin sections were cut from paraffin embedded tissues and histological sections were conducted hematoxylin and eosin (H&E), Trichrome, and von Kossa. We observed that bone formatioin of SIS/PLGA hybrid scaffold as natural/synthetic scaffold was better thean that of only PLGA scaffold. It canb be explained that SIS contains various kinds of bioactive molecules for osteoinduction.

Macro porous ceramics possessing controlled microstructures and chemical compositions have increasingly proven useful in the industrial sphere. Their sintered structures have found application in both established and emerging, areas such as thermal insulation in buildings, filtration of liquids and molten materials, refractory insulation, bone scaffolds and tissue engineering. Stable ceramic foams can be formed by wet chemical methods using inorganic particles(e.g., $Al_2O_3$ or $SiO_2$). The wet foams are dried and sintered with improved porosity and mechanical properties. This review examines the different techniques used to prepare porous ceramics from ceramic foams, focusing on the explanation of this versatile method of direct foaming from the past to the present. Comparisons of the processes and the processing parameters are explained with the produced microstructures.
In order to improve the bioactivity and mechanical properties of hydroxyapatite (HAp), chitosan (Chi) was in situ combined into HAp to fabricate a composite scaffold by a sublimation-assisted compression method. A highly porous film with sufficient mechanical strength was prepared and the bioactivity was investigated by examining the apatite formed on the scaffolds incubated in simulated body fluid. In addition, the cytotoxicity of the HAp/Chi composite was studied by evaluating the viability of murine fibroblasts (L-929 cells) exposed to diluted extracts of the composite films. The apatite layer was assessed using scanning electronic microscopy, inductively coupled plasma-optical emission spectrometry and weight measurement. Composite analysis showed that a layer of micro-sized, needle-like crystals was formed on the surface of the composite film. Additionally, the WST-8 assay after L-929 cells were exposed to diluted extracts of the composite indicated that the HAp/Chi scaffold has good in vitro cytocompatibility. The results indicated that HAp/Chi composites with porous structure are promising scaffolding materials for bone-patch engineering because their porous morphology can provide an environment conductive to attachment and growth of osteoblasts and osteogenic cells.

Porous ceramics are promising materials for a number of functional and structural applications that include thermal insulation, filters, bio-scaffolds for tissue engineering, and preforms for composite fabrication. These applications take advantage of the special characteristics of porous ceramics, such as low thermal mass, low thermal conductivity, high surface area, controlled permeability, and low density. In this review, we emphasize the direct foaming method, a simple and versatile approach that allows the fabrication of porous ceramics with tailored microstructure, along with distinctive properties. The wet foam stability is achieved under the controlled addition of amphiphiles to the colloidal suspension, which induce in situ hydrophobization, allowing the wet foam to resist coarsening and Ostwald ripening upon drying and sintering. Different components, like contact angle, adsorption free energy, air content, bubble size, and Laplace pressure, play vital roles in the stabilization of the particle stabilized wet foam to the porous ceramics. The mechanical behavior of the load-displacements curves of sintered samples was investigated using Herzian indentations testes. From the collected results, we found that microporous structures with pore sizes from 30 ㎛ to 570 ㎛ and the porosity within the range from 70% to 85%.

Ternary blends of poly(L-lactic acid) (PLLA), poly(${\varepsilon}$-caprolactone) (PCL) and polyethylene oxide (PEO) were produced with different concentrations of components via melt blending. By leaching the PEO from the samples by water, porous materials were obtained with potential application for bio scaffolds. Sample porosity was evaluated by calculating the ratio of porous scaffold density (${\rho}^*$) to the non-porous material density (${\rho}_s$). Highest porosity (51.42%) was related to the samples containing 50 wt%. of PEO. Scanning electron microscopy (SEM) studies showed the best porosity resulted by decreasing PLLA/PCL ratio at constant concentration of PEO. Crystallization behavior of the ternary blend samples was studied using differential scanning calorimetry (DSC). Results revealed that the crystallinity of PLLA was improved by addition of PEO and PCL to the samples. The porosity plays a key role in governing the compression properties. Mechanical properties are presented by Gibson-Ashby model.
Considerable effort has been directed toward the use of silk fibroin as a biotechnological material in biomedical applications on account of its excellent biodegradability, biocompatibility, and unique mechanical properties. For use in tissue engineering, it is very important to design and control the pore architecture of polymeric scaffolds, which provide the vital framework for seeded cells to organize into functioning tissue. In the present study, a silk fibroin scaffold with controlled interconnectivity and pore size was prepared using an electrospinning method with poly(ethylene oxide).
