Study on Pharmaceutical Byproducts and Sludges for Practical Application as Raw materials of organic compost (제약업종 부산물 및 오니의 퇴비원료로 활용 가능성 연구)

  • This study was conducted to investigate use possibility of pharmaceutical byproducts(process sludge) and waste water sludges as raw materials of organic compost at Fertilizer Official Regulation of Fertilizer Management Law in 2002. All pharmaceutical byproducts were satisfied the standard levels of raw material regulated in organic compost, some waste water sludges were deficient in the level. The content of n-hexane extractable material(HEM) was in byproduct higher than in waste water sludge. This was presumed that the sludge with containing a lot of organic matter was high in HEM content. Of the whole microorganism flora, bacteria was mainly detected, and yeast and filamentous fungi took up less population which was fluctuated depending on the source of sludges. Most dominated bacteria were identified into Genus of Pseudomonas. Pseudomonas syringae and Rathayibacter bathayi was classified as plant pathogenic bacteria.

BIOLOGICALLY-BASED DOSE-RESPONSE MODEL FOR NEUROTOXICITY RISK ASSESSMENT

  • The regulation of neurotoxicants has usually been based upon setting reference doses by dividing a no observed adverse effect level (NOAEL) by uncertainty factors that theoretically account for interspecies and intraspecies extraploation of experimental results in animals to humans. Recently, we have proposed a four-step alternative procedure which provides quantitative estimates of risk as a function of dose. The first step is to establish a mathematical relationship between a biological effect or biomarker and the dose of chemical administered. The second step is to determine the distribution (variability) of individual measurements of biological effects or their biomarkers about the dose response curve. The third step is to define an adverse or abnormal level of a biological effect or biomarker in an untreated population. The fourth and final step is to combine the information from the first three steps to estimate the risk (proportion of individuals exceeding on adverse or abnormal level of a biological effect or biomarker) as a function of dose. The primary purpose of this report is to enhance the certainty of the first step of this procedure by improving our understanding of the relationship between a biomarker and dose of administered chemical. Several factors which need to be considered include: 1) the pharmacokinetics of the parent chemical, 2) the target tissue concentrations of the parent chemical or its bioactivated proximate toxicant, 3) the uptake kinetics of the parent chemical or metabolite into the target cell(s) and/or membrane interactions, and 4) the interaction of the chemical or metabolite with presumed receptor site(s). Because these theoretical factors each contain a saturable step due to definitive amounts of required enzyme, reuptake or receptor site(s), a nonlinear, saturable dose-response curve would be predicted. In order to exemplify this process, effects of the neurotoxicant, methlenedioxymethamphetamine (MDMA), were reviewed and analyzed. Our results and those of others indicate that: 1) peak concentrations of MDMA and metabolites are ochieved in rat brain by 30 min and are negligible by 24 hr, 2) a metabolite of MDMA is probably responsible for its neurotoxic effects, and 3) pretreatment with monoamine uptake blockers prevents MDMA neurotoxicity. When data generated from rats administerde MDMA were plotted as bilolgical effect (decreases in hippocampal serotonin concentrations) versus dose, a saturation curve best described the observed relationship. These results support the hypothesis that at least one saturable step is involved in MDMA neurotoxicity. We conclude that the mathematical relationship between biological effect and dose of MDMA, the first step of our quantitative neurotoxicity risk assessment procedure, should reflect this biological model information generated from the whole of the dose-response curve.

β-lapachone-Induced Apoptosis of Human Gastric Carcinoma AGS Cells Is Caspase-Dependent and Regulated by the PI3K/Akt Pathway

  • ${\beta}$-lapachone is a naturally occurring quinone that selectively induces apoptotic cell death in a variety of human cancer cells in vitro and in vivo; however, its mechanism of action needs to be further elaborated. In this study, we investigated the effects of ${\beta}$-lapachone on the induction of apoptosis in human gastric carcinoma AGS cells. ${\beta}$-lapachone significantly inhibited cellular proliferation, and some typical apoptotic characteristics such as chromatin condensation and an increase in the population of sub-G1 hypodiploid cells were observed in ${\beta}$-lapachone-treated AGS cells. Treatment with ${\beta}$-lapachone caused mitochondrial transmembrane potential dissipation, stimulated the mitochondria-mediated intrinsic apoptotic pathway, as indicated by caspase-9 activation, cytochrome c release, Bcl-2 downregulation and Bax upregulation, as well as death receptor-mediated extrinsic apoptotic pathway, as indicated by activation of caspase-8 and truncation of Bid. This process was accompanied by activation of caspase-3 and concomitant with cleavage of poly(ADP-ribose) polymerase. The general caspase inhibitor, z-VAD-fmk, significantly abolished ${\beta}$-lapachone-induced cell death and inhibited growth. Further analysis demonstrated that the induction of apoptosis by ${\beta}$-lapachone was accompanied by inactivation of the phosphatidylinositol 3-kinase (PI3K)/Akt signaling pathway. The PI3K inhibitor LY29004 significantly increased ${\beta}$-lapachone-induced apoptosis and growth inhibition. Taken together, these findings indicate that the apoptotic activity of ${\beta}$-lapachone is probably regulated by a caspase-dependent cascade through activation of both intrinsic and extrinsic signaling pathways, and that inhibition of the PI3K/Akt signaling may contribute to ${\beta}$-lapachone-mediated AGS cell growth inhibition and apoptosis induction.

Density Effect and Diversity of Fish in Water System at Both Reservoirs in the Youngsan-ri, Goseong-gun (고성군 용산리의 두 저수 수계에서 어류의 다양성과 밀도 효과)

  • Four sites and one site were used to analyze fish diversity at the water systems of the Sineun reservoir and the Jeonchon reservoir, respectively. The field experiments were conducted to test the density dependence that could lead to population regulation by artificial inferences and environmental changes. We examined the effects of environmental factors on fish densities using SMATR freeware. It was estimated to be reduced to the density effect at four sites in 2012. Shannon-Weaver indices of the diversity (H’) of the Sineun reservoir were similar to those of the Jeonchon reservoir. Species diversity was in a range of 0.645 to 2.105. The H’ value of the upper region was higher than those of middle and low regions were, and values of richness were lower in downstream than upstream. Using the maximum likelihood solution for the removal estimators of two low regions of the river stations, the estimated migration probabilities from the resident fish to the migrated fish for five species (Cyprinus cuvieri, Carassius auratus, Pseudorasbora parva, Misgurnus mizolepis, and Oryzias latipes) had a mean of 0.623. Especially, migration probabilities from the Jeonchon reservoir to the Sineun reservoir for five species were high (a mean of 0.681). The period of migration was suggested to be about one month because of short geographical distances (50 m). We found no significant difference between the three categories in the distribution of the other four species, indicating the species probability was similar among stations.

  • Fusarium graminearum (teleomorph Gibberella zeae) is an important plant pathogen that causes head blight of major cereal crops such as wheat, barley, and rice, as well as causing ear and stalk rot on maize worldwide. Plant diseases caused by this fungus lead to severe yield losses and accumulation of harmful mycotoxins in infected cereals [1]. Fungi utilize spore production as a mean to rapidly avoid unfavorable environmental conditions and to amplify their population. Spores are produced sexually and asexually and their production is precisely controlled. Upstream developmental activators consist of fluffy genes have been known to orchestrate early induction of condiogenesis in a model filamentous fungus Aspergillus nidulans. To understand the molecular mechanisms underlying conidiogenesis in F. graminearum, we characterized functions of the F. graminearum fluffy gene homologs [2]. We found that FlbD is conserved regulatory function for conidiogenesis in both A. nidulans and F. graminearum among five fluffy gene homologs. flbD deletion abolished conidia and perithecia production, suggesting that FlbD have global roles in hyphal differentiation processes in F. graminearum. We further identified and functionally characterized the ortholog of AbaA, which is involved in differentiation from vegetative hyphae to conidia and known to be absent in F. graminearum [3]. Deletion of abaA did not affect vegetative growth, sexual development, or virulence, but conidium production was completely abolished and thin hyphae grew from abnormally shaped phialides in abaA deletion mutants. Overexpression of abaA resulted in pleiotropic defects such as impaired sexual and asexual development, retarded conidium germination, and reduced trichothecene production. AbaA localized to the nuclei of phialides and terminal cells of mature conidia. Successful interspecies complementation using A. nidulans AbaA and the conserved AbaA-WetA pathway demonstrated that the molecular mechanisms responsible for AbaA activity are conserved in F. graminearum as they are in A. nidulans. F. graminearum ortholog of Aspergillus nidulans wetA has been shown to be involved in conidiogenesis and conidium maturation [4]. Deletion of F. graminearum wetA did not alter mycelial growth, sexual development, or virulence, but the wetA deletion mutants produced longer conidia with fewer septa, and the conidia were sensitive to acute stresses, such as oxidative stress and heat stress. Furthermore, the survival rate of aged conidia from the F. graminearum wetA deletion mutants was reduced. The wetA deletion resulted in vigorous generation of single-celled conidia through autophagy-dependent microcycle conidiation, indicating that WetA functions to maintain conidia dormancy by suppressing microcycle conidiation in F. graminearum. In A. nidulans, FlbB physically interacts with FlbD and FlbE, and the resulting FlbB/FlbE and FlbB/FlbD complexes induce the expression of flbD and brlA, respectively. BrlA is an activator of the AbaA-WetA pathway. AbaA and WetA are required for phialide formation and conidia maturation, respectively [5]. In F. graminearum, the AbaA-WetA pathway is similar to that of A. nidulans, except a brlA ortholog does not exist. Amongst the fluffy genes, only fgflbD has a conserved role for regulation of the AbaA-WetA pathway.

Interactions between Pine Needle Gall Midge, Thecodiplosis japonensis (Diptera: Cecidomyiidae), and its Parasitoids in Newly Invaded Areas (솔잎혹파리 확산지역에서 솔잎혹파리와 기생봉의 상호작용)

  • The pine needle gall midge (PNGM), Thecodiplosis japonensis Uchida et Inouye, is a serious pest in pine forests in Korea. Since the first report of PNGM infestation in Korea in 1929, the infestation area has been expanded gradually. In 1975 the distribution of PNGM and its parasitoids were surveyed throughout several infested areas in Korea. Annual survey has been made for the rates of gall formation by PNGM and parasitism by its parasitoids at the monitoring sites including newly infested area in 1975 since 1980. These data were used to examine the relationship between PNGM and its parasitoids for newly invaded areas. The gall forming rate of PNGM and the parasitism by the parasitoid were 34.8 and 1.9% in 1975, respectively, while the gall forming rate and the parasitism were 20.7 and 8.9% in 1985, respectively. The relationships of densities between PNGM and its parsitoid were weak in the early stage of dispersion, but the density of parasitoid was with an asymptotic increase along with PNGMs density increase during the observation period for 10 years.

Expression of Neurotrophic Factors and Their Receptors in Rat Posterior Taste Bud Cells

  • Taste is an important sense in survival and growth of animals. The growth and maintenance of taste buds, the receptor organs of taste sense, are under the regulation of various neurotrophic factors. But the distribution aspect of neurotrophic factors and their receptors in distinct taste cell types are not clearly known. The present research was designed to characterize mRNA expression pattern of neurotrophic factors and their receptors in distinct type of taste cells. In male 45-60 day-old Sprague-Dawley rats, epithelial tissues with and without circumvallate and folliate papillaes were dissected and homogenized, and mRNA expressions for neurotrophic factors and their receptors were determined by RT-PCR. The mRNA expressions of brain-derived neurotrophic factor (BDNF), neurotrophin-3 (NT3), receptor tyrosine kinase B (TrkB), exclusion of nerve growth factor (NGF), neurotrophin-4/5 (NT4/5), receptor tyrosine kinase A (TrkA), receptor tyrosine kinase C (TrkC), and p75NGFR were observed in some population of taste cell. In support of this result and to characterize which types of taste cells express NT3, BDNF, or TrkB, we examined mRNA expressions of NT3, BDNF, or TrkB in the $PLC{\beta}2$ (a marker of Type II cell)-and/or SNAP25 (a marker of Type III cell)-positive taste cells by a single taste cell RT-PCR and found that the ratio of positively stained cell numbers were 17.4, 6.5, 84.1, 70.3, and 1.4 % for $PLC{\beta}2$, SNAP25, NT3, BDNF, and TrkB, respectively. In addition, all of $PLC{\beta}2$-and SNAP25-positive taste cells expressed NT3 mRNA, except for one taste bud cell. The ratios of NT3 mRNA expressions were 100% and 91.7% in the SNAP25-and $PLC{\beta}2$-positive taste cells, respectively. However, two TrkB-positive taste cells co-expressed neither $PLC{\beta}2$ nor SNAP 25. The results suggest that the most of type II or type III cells express BDNF and NT3 mRNA, but the expression is shown to be less in type I taste cells.

  • Era of multi-channel digital multimedia on the Internet, cable, satellite and IPTV and the growth of new media such as terrestrial broadcast advertising with the poor rating has also dropped sharply. In this situation, terrestrial TV suggesting the introduction for total amount system appearing comments. In this study, the introduction of these intermediate advertising the public and professional awareness about the issue by comparing the introduction for total amount advertising feasibility was done in the purpose. The results were as follows. First, Studies on terrestrial TV suggesting the introduction of total amount system and public opinion about the differences in perception between experts is high. Second, Both the public and professionals about the total amount system preferred application stage were in favor of gradual for/limited permit cross-media. Third, the general population was higher than the positive response from experts about the benefits of the total amount system. Finally, the general public about the disadvantages of the total amount system they were more positive response than the experts. The introduction for total amount system validity of these data for future total amouny system to provide a basis for enforcement is expected to give.

Role of Trophobolast in Implantation and Placenta Development (착상 및 태반 발달과정에 따른 영양막세포의 역할)

  • The placenta, which is a temporary organ derived from the fetus during pregnancy, is critical to support fetus development via optimal regulation between mother and fetus. Trophoblast as a major cell population of the placenta is one of the earliest to differentiate and shows an extensive proliferation or/and differentiation up to the formation of the placenta. The role of the trophoblast show dynamic changes from early embryo implantation to placentation during pregnancy. Implantation of the blastocyst into the endometrium of the maternal uterus is mediated by invasion of the differentiated trophoblast (e.g. syncytiotrophoblast) from the trophectoderm. During pregnancy, the unique role of the trophoblast is to invasion, eroding, and metastasizing in the placenta as well as to ensure appropriate bidirectional nutrient or waste flow required for growth and maturation of the embryo. The dysfunction of the trophoblast during pregnancy can result in several gynecological diseases including preeclampsia and congenital malformation in neonatal medicine. Therefore, trophoblasts act as a conclusive factor in placental and fetal development. This brief review outlines the classification of trophoblast and its function in the placenta during pregnancy. Also, we introduce the latest research in trophoblast for implantation and the placenta development, and the application potential of trophoblast for infertility and obstetrical diseases.

A Preliminary Survey of Attitude Towards IVF Surrogacy (체외수정을 통한 대리모 출산에 대한 인식 조사)

  • Objectives: The aim of this study was to find out the attitude of Korean women towards IVF surrogacy which is not yet regulated by the law. Methods: We evaluated with a questionnaire, in how far IVF surrogacy was accepted in different groups of Korean women. Total 300 questionnaires were sent out to women who visited our infertility clinic, and to the nurses and office workers of our hospital. Among them, 246 questionnaires were returned, and 211 of them (85.7%) were complete and analyzed. The responders were 58 unmarried women and 153 married women including 60 infertile patients. Results: Only 17 (8.1%) women believed that IVF surrogacy was an acceptable treatment for infertile women without uterus (eg. MRKH or hysterectomized patients), and 125 (59.2%) women chose the adoption. There were no significant differences in response amongst different age group, marital status, or in relation to income level or educational level, or infertility status. A significantly higher level of acceptance was noted only in religious groups, especially in Christians. Most of responders (66.5%) chose unrelated and unknown women as a desirable host, and 28.4% of responders chose the sister or sister-in-law. Conclusions: Overall the different groups were critical towards IVF Surrogacy in Korea, irrespective of the infertile patient group. Despite the established clinical ability to have her own genetic child in women without uterus, this survey reveals that only minority supports the IVF surrogacy. So we need the survey with large, randomized population and then make a regulation about the IVF surrogacy in law.
