  • Quercetin-3-O-${\beta}$-D glucofuranoside, mp 243-$6^{\circ}$, was isolate dfrom leaves of Polygonum sachalinense Fr. Schm and characterized on the basis of its spectral data. This is the second report of its occurrence in nature.

  • In Korea, two species of Ho-chang-Ho-chang $(Polygonum\;cuspidatum \;S_{IEB}.\;et\;Z_{UCC}.)$ and Wang-Ho-chang $(Polygonum \;sachalinense\;F_R.\;S_{CHM}.)$-had been identified, but the components of the leaves of Polygonum species growing in Korea had never been reported. The authors examined the anatomical characteristics of Korean Polygonum species, identified the flavonoids contained in their leaves and obtained the following results: Korean Polygonum species should be divided into three species: $Polygonum\;cuspidatum\;S_{IEB}.\;et\;Z_{UCC}.,\;P. \;sachalinense\;F_R.\;S_{CHM}\;and\;P.\;elliptica\;M_{IGO}$. The last one was illustrated in the report. P. cuspidatum contained reynoutrin and quercitrin, but no isoquercitrin could be identified. P. sachalinense also contained both reynoutrin and quercitrin. P. elliptica, which had never been studied, included the same flavonoids.

Anthraquinones from the Rhizome of Polygonum sachalinense (왕호장근의 성분 연구)

  • The rhizome of Polygonum sachalinense Fr. Schm. (=Reynoutria sachalinensis Nakai, Polygonaceae) has been used as "Polygoni Rhizoma" (Hu Zhang) in the Orient as laxatives, diuretics and for treatment of suppurative dermatitis, gonorrhoea, favus and athlete's foot. From the methanolic extract of the dried rhizome physcion, emodin, emodin-8-O-$\beta$-D-glucoside as anthraquinone derivatives and .betha.-sitosterol glucoside were isolated and identified. Stilbene derivatives which have antibacterial and antifungal activities were also isolated.

  • Anthraquinones, physcion (I), mp $204{\sim}205^{\circ}$ and emodin (II), mp $254{\sim}255^{\circ}$, and $emodin-8-O-{\beta}-{_D}-glucoside$ (IV), mp $191{\sim}192^{\circ}$ together with ${\beta}-sitosterol$ glucoside (III), mp $280{\sim}282^{\circ}$ were isolated from the roots of Polygonum ellipticum Migo and P. sachalinense Fr. Schm. (Polygonaccae). Stilbene derivatives, 3,5,4'-trihydroxystibene (V), mp $258{\sim}260^{\circ}$ and $3,5,4'-trihydroxystilbene-3-O-{\beta}-{_D}-glucoside$ (VI), mp $142{\sim}144^{\circ}$ were also isolated.

  • Free amino acids and mineral substance in radix of Polygonum cuspidatum, Polygonum elliptica, and Polygonum sachalinense were analyzed by amino acid autoanalyzer and atomic absorption spectrophotometer. Gross contents of free amino acids in root of P. cuspidatum and P. elliptica are 12.99mg/g and 9.58mg/g respectively. The analysis of inorganic constituents of Polygonum sp. showed that it generally contains copper, iron, manganese, zinc, potassium and potassium salt.

  • Three species of genus Polygonum, namely P. cuspidatum, P. sachalinense and P. ellipticum are distributed in Korea. Polygonum ellipticum Migo is a perennial herb in Polygonaceous plants. The root of the plant (Polygoni Rhizoma, 'Ho-jang') have been used as laxative, diuretic and for the treatment suppurative dermatitis in the oriental medicine. As the part of the study for the comparison of the three species in their components, the authors attempted to isolate the anthraquinones and stilbene derivative from the rhizome of P. ellipticum. The methanolic extract of dried rhizome of this plant was fractionated into ether soluble and insoluble fraction and each fraction was applied to column chromatography to isolate above mentioned components. Anthraquinone derivatives were isolated first; comp. I, mp $204{\sim}205^{\circ}$ (physcion), comp. II, mp $254{\sim}255^{\circ}$ (emodin), comp. IV, mp $191{\sim}192^{\circ}$ $(emodin-8-O-{\beta}-D-glucoside)$ and comp. III, mp $280{\sim}282^{\circ}$ $({\beta}-sitosterol-glucoside)$. They were identified by chemical properties and UV, IR and NMR spectra and by the direct comparison with authentic samples. Stilbene derivative was isolated secondly; comp. V, mp $255{\sim}256^{\circ}$ which was reported to possess antibacterial and antifungal activities.

  • Polygonum cuspidatum $S_{IEB}$. et $Z_{UCC}$. ('Ho-Jang'), P. sachalinense $F_R$. $S_{CHM}$. ('Wang Ho-Jang') and P. ellipticum $M_{IGO} ('Dung Geun Ib Ho-Jang') are the three species growing in Korea. Their rhizomes can be classified into two types: horizontal rhizome type ('Ho-Jang') and erect type ('Wang Ho-Jang' and 'Dung Geun Ib Ho-Jang'). The most of 'Ho-Jang' on Korean markets are the rhizome of 'Wang Ho-Jang' and only limited quantity of the rhizome of 'Ho-Jang' is used. 'Dung Geun Ib Ho-Jang' is a species transplanted from the China continent and is not collected for medicinal use. It is suggested that the name 'Ho-Jang Geun' (Reynoutriae Radix) should be corrected to 'Ho-Jang' (Reynoutriae Rhizoma=Polygoni Rhizoma).

  • The rhizomes of Polygonum cuspidatum, P. sachalinense and P. ellipticum which grow in Korea and which can be used for medicinal purpose were collected monthly from April to October and were analyzed for the contents of the free and combined derivatives of oxyanthraquinones. On the basis of the total oxyanthraquinone contents, the suitable month for collecting the rhizome of the former can be April or May, since it contained 0.46% in May and 0.05% in August. The rhizomes of the latter two species should be collected in June or July, since they contained $0.72{\sim}3%$ and $0.31{\sim}2%$ in April.
