Pharmacognostical Studies on Ha-Su-O(Polygoni Radix) (하수오류(何首烏類)의 생약학적 연구)

  • Experiments were carried out to evaluate the pharmacognostical aspects of Polygoni Radix which consists of the roots of Polygonum multiflorum $T_{HUNBERG}$(Polygonaceae), Cynanchum wilfordii $H_{EMSLEY}$ (Asclepiadaceae) and Polygonum ciliinerve $(N_{AKAI})O_{WI}$ (Polygonaceae), since it was known to be effective as tonic, nutrient, etc. in oriental medicine. The histological features of the above three plants of Polygoni Radix were compared in view point of external and internal morphologies and their habitats. Also their constituents were compared on the basis of anthraquinone group. From the methanol extract of Polygonum ciliinerve $(N_{AKAI})O_{WI}$, furthermore, a specific product of this country, two anthraquinone derivatives were isolated. One of the substances was identified as emodin $(C_{15}H_{10}O_5,\;m.p.\;268-269^{\circ})$ through comparison of the authentic compound and spectroscopy.

  • Melanin biosynthesis inhibitors are useful not only for the materials used in cosmetics as skin-whitening agents but also for the remedy of hyperpigmentation. In order to find the new skin-whitening compounds from the natural products. screening of tyrosinase and melanin biosynthesis inhibitory activities in vitro has been carried out. The MeOH extracts and/or fractions of Polygoni multiflori Radix, Dalbergiae odoriferae Lignum. Solnani nigri Herba. Polygoni cuspidati Radix. POlygoni multiflori Ramulus. Salviae Radix showed tyrosinase inhibitory effects. Four methanolic extracts also showed melanin biosynthesis inhibitory effects in B-16 melanoma cell line.

Antibiotic Effects of blood-activating stasis-dispelling medicinals on Vaginal Microorganisms (활혈거어약(活血祛瘀藥)이 질내(膣內) 미생물(微生物)에 미치는 영향(影響))

  • Purpose : The aim of this study is to investigate the antibiotic effects of 14 herbs among blood-activating stasis-dispelling medicinals on vaginal microorganisms. Methods : Staphylococcus aureus, Methicillin-resistant Staphylococcus aureus, Candida albicans, and Gardnerella vaginalis were used for vaginal pathogenic microorganisms. Lactobacillus gasseri, Streptococcus spp. and Escherichia coli HB101 were used for vaginal normal flora. The blood-activating stasis-dispelling medicinals, Mucunae Caulis, Salviae Radix, Persicae Semen, Myrrha, Zedoariae Rhizoma, Achuranthis Radix, Leonuri Herba, Melandrii Herba, Gleditsiae Spina, Lycopi Herba, Scirpi Rhizoma, Caesalpiniae Lignum, Corydlais Tuber and Polygoni Cuspidati Radix were used in this study. In vitro antibiotic activities were observed by optical density and colony test. Results : The optical density and colony test showed that Gleditsiae Spina, Scirpi Rhizoma, Corydlais Tuber, Polygoni Cuspidati Radix and Melandrii Herba of herbs among blood-activating stasis-dispelling medicinals had antimicrobial effect. Gleditsiae Spina had antimicrobial susceptibility and selective toxicity in Gardnerella vaginalis and MRSA. Scirpi Rhizoma had antimicrobial susceptibility and selective toxicity in Staphylococcus aureus and MRSA. Corydlais Tuber had antimicrobial susceptibility and selective toxicity in MRSA. Polygoni Cuspidati Radix had antimicrobial susceptibility and selective toxicity in Gardnerella vaginalis, Staphylococcus aureus and MRSA. Melandrii Herba had antimicrobial susceptibility and selective toxicity in Gardnerella vaginalis. Conclusion : According to the above results, we could suggest that Gleditsiae Spina, Scirpi Rhizoma, Corydlais Tuber, Polygoni Cuspidati Radix and Melandrii Herba of herbs among blood-activating stasis-dispelling medicinals be available to antimicrobial agent of vaginal pathogenic microbial species in vitro.

  • Polygonum cuspidatum has been used as treatments of dermatitis, inflammation, hyperlipidemia and diuretics in folk remedies. In order to evaluate the urinary effect of Polygoni cuspidati Radix, its MeOH extract was administerd in rats. We determined the total urin volume, chemical parameters(urea nitrogen, creatinine, uric acid), electrolytes(natrium, potassium, chloride) ) in serum and urin. Polygoni cuspidati Radix showed increase in urin volume and electrolytes.

The Biochemical and Histological Studies of the Oriental Medicine Extract on Hair Growth Effect

  • The hair growth effect of the oriental medicine extract which is composed of Polygoni multiflori Radix, Angelica gigantis Radix and Lycii Fructus was studied biochemically and histologically. The study was conducted dividelly into three groups, control, propecia and oriental medicine extract and three groups were compared each other in skin androgen, testosterone (T) and dihydrotestosterone (DHT), content biochemically and the number of hair follicular unit and hair follicle histologically. The oriental medicine extract and propecia decreased skin DHT conetent and increased skin total (T+DHT) content compared with control. In transverse sections of skin specimens the oriental medicine extract and propecia increased total counts of follicular units and follicles compared with control. On the basis of the result, the oriental medicine extract has same hair growth effect as propecia and it is suggested that the oriental medicne extract is capable of a therapeutic agent of alopecia.

Effects of YongdamSaganTangGamibang(YSTG) on the Inflammatory Reactions. (용담사간탕가미방이 염증치료 및 예방에 미치는 영향에 대한 연구)

  • This study was carried out to investigate the effects YSTG on the skin inflammatory reactions. Among the herbal ingredients of YSTG, ethanol extracts of Cinnamoni Ramulus, Polygoni Avicularis Herba and Scutellariae Radix showed potent radical scavenging activity, more than 90$\%$ at a concnentration of 0.01$\%$, tested by DPPH(I,1-diphenyl-2-picryl-hyrazyl) method. And ethanol extract of Saussureae Radix inhibited the lipopolysaccharide-induced release of nitric oxide(NO), 86$\%$ at 50㎍/㎖, by the macrophage RAW 246.7 cells. In addition, the ethanol extracts of Cinnamoni Ramulus and Polygoni Avicularis Herba showed antibacterial activities on Staphylococcus aureus that was reported to cause impetigo and atopic disease, Cinnamoni Ramulus, Saussureae Radix and Plantaginis Semen on Propionibacterium acnes that causes acne, and Cinnamoni Ramulus on Candida albicans that causes cadidiasis, Saussureae Radix on Corynebacterium xerosis that cause osmidrosis axillae. Taken together, I expect that YSTG may be used as an effective drug for treatment on skin inflammation.

Beneficial effect of Polygoni Multiflori Radix in high fructose diet-induced metabolic syndrome rat model (고과당식이 랫드모델에서 적하수오 투여에 의한 대사증후군 개선효과)

  • Objectives : Polygoni Multiflori Radix (Jeokhasuo in Korean) is a Oriental traditional herbs widely used in East Asian countries. Overconsumption of fructose results in hypertension, dyslipidemia, obesity and impaired glucose tolerance which have documented as a risk of cardiovascular diseases. This experimental study was designed to investigate the beneficial effects of an ethanol extract from Polygoni Multiflori Radix (PMR) in high-fructose (HF) diet-induced metabolic syndrome rat model. Methods : Sprague-Dawley (SD) rats were divided into three groups; Control group, receiving regular diet and tap water, HF group, and HF + PMR group both receiving supplemented with 65% fructose (n=10), respectively. The HF + PMR group initially received HF diet with PMR (100 mg/kg/day) for 8 weeks. Results : PMR significantly prevented the metabolic disturbances such as hyperlipidemia, hypertension and impaired glucose tolerance. Chronic treatment with PMR significantly decreased body weight, fat weight and adipocyte size, suggesting a role of anti-obesity effect. PMR led to improve the hyperlipidemia through the increase in HDL cholesterol level as well as the decrease in triglyceride and LDL cholesterol level. In addition, PMR suppressed adhesion molecules and endothelin-1 (ET-1) expression in aorta resulting in the decrease of hypertension. In muscle tissue, PMR significantly recovered the HF-induced insulin resistance through increase of insulin receptor substrate-1 (IRS-1), p-$AMPK{\alpha}1/2$, and p-Akt expression. PMR improved HF-induced metabolic disorders and its action was caused by energy metabolism-mediated insulin signaling activation. Conclusions : These results demonstrate that PMR may be a beneficial therapeutic for metabolic syndrome through the improvement of hyperlipidemia, obesity, insulin resistance and hypertension.

Antioxidant activity of aqua-acupuncture of Polygoni Radix rubra in rats treated with AAPH (적하수오(赤何首烏) 약침(藥針)의 AAPH처리된 흰쥐에 대한 항산화작용)

  • This study was done to elucidate the antioxidant activities of Polygoni Radix rubra(赤何首烏) by way of aqua-acupuncture methods. After 10% & 20% concentrations of liquid extract of Polygoni Radix rubra(PRR) were acupunctured on Joksamri(足三里) of rats with acute and chronic experimental oxidation by AAPH(2, 2' -azobis (aminoidinopropane), hydrochloride), various kinds of experiments were performed. The results were obtained as follows: 1. In acute oxidant experiment, the amount of TBARS was significantly increased from 0.5 hr to 1.5 hr after AAPH treatment 2. In acute oxidant experiment by AAPH, uric acid as significantly increased while the others didn't show any difference such as total protein, albumin, bilirubin. 3. In chronic oxidant experiment by AAPH, PRR significantly increased the body weight in comparison with control group, whereas liver and spleen tended to increase, lung and kidneys tended to decrease. 4. In chronic oxidant experiment by AAPH, PRR significantly decreased the GPT, hemolysis of erythrocytes, TBA and BUN, where GOT was significantly reduced in 20% PRR treated group, and TG was significantly in 10% PRR treated group. These results suggest that PRR has potent antioxidant activity and needs more studies on microsomal fraction of liver and kidney.

Study on the Effect of Radix polygoni Multiflori on Cultured Mouse Cerebral Neurons Damaged by Organic Mercury (하수오가 유기수은으로 손상된 생쥐의 배양대뇌신경세포에 미치는 영향에 관한 연구)

  • To investigate the neurotoxic effect of organic chloride on cultured mouse cerebral neurons, cytotoxic effect was measured by MTT assay after cultured cerebral neurons were incubated with various concentrations of methyl mercuric chloride(MMC) for 24 hours. The protective effect of Radix Polygoni Multiflori(RPM) on MMC-induced neurotoxicity was also examined in these cultures. MMC decreased cell viability of cultured mouse cerebral neurons remarkably in a dose- and time-dependent manners. In protective effect of RPM it was remarkably effective in blocking the neuroxicity induced by MMC. From aboved the results, it is suggested that MMC induce neurotoxicity, and the herba extract, RPM is very effective in preventing MMC-induced cytotoxicity on cultured mouse cerebral neurons.

Effect of Radix Polygoni Multiflori on Spinal Motor Neurons Damaged by Methylmercuric Chloride (메틸수은으로 손상된 척수신경세포에 대한 하수오의 영향)

  • In order to evaluate the cytotoxicity of methylmercuric chloride(MMC) in cultured spinal motor neurons of neonatal mouse, cell viability was measured by MTT assay in spinal motor neurons treated with 1-30 μM MMC for 48 hours. And also, the protective effect of Radix Polygoni Multiflori(RPM) was examined by cell viability in these cultures. Cell viability was significantly decreased in dose-dependent manner after cultured cells were exposured to 20 μM MMC for 48 hours. Protective effect of RPM on MMC-mediated toxicity was very effective in these cultures. From above the results, it suggests that MMC has toxic effect in cultured mouse spinal motor neurons and herb extract such as RPM is very effective in blocking the neurotoxicity induced by MMC.
