  • Rhizomes of Polygonatum species belong to Liliaceae are important herbal drugs in the traditional medical practice of Asian region. Two representative Chinese drugs derived from this genus are Hwangjeong and Okjuk. Though botanical origins of these drugs are officially listed as P. falcatum, P. sibiricum and P. kingianum for Hwangjeong and P. odoratum var. pluriflorum for Okjuk in the Korean Pharmacopoeia and Korean Herbal Pharmacopoeia, respectively, they are often sold as a mixture of several different species in the market. 둥굴레 분획물과 Selenium이 Streptozotocin 유발 당뇨 흰쥐의 혈당수준과 지질과산화에 미치는 영향 (The Effect of BuOH Fraction of Polygonatum odoratum with Selenium on Blood Glucose Level and Lipid Peroxidation in Streptozotocin Induced Diabetic Rats)

  • The purpose of this study was to investigate the effect of butanol(BuOH) fraction of Polygonatum odoratum with selenium tr-eatment on blood glucose levels and lipid peroxidations in streptozotocin(STZ) induced diabetric rats. Male Sprague-Dawly rats weighing(180-200g) were divided into five groups : normal STZ-control and three expreimental groups(P, odoratum group P, odo-Se group and Se group) Diabetes mellitus was induced by injection STZ in the tail vein at the dose of 45mg/kg B.W The BuOH fraction of Polygonatum odoratum(500mg/kg. B,W) given orally administered for 14 days. The Se treated group were fed a AIN-76 recommendation diet mixed with Na2Seo3(2mg/kg diet). Diabetic rats showed the lower weight gain compared to the normal rats. the plasma glucose levels of the P. odo-Se group were significantly lower than the other experimental groups. The plasma cholesterol levels were higher in STZ-control and Se groups compared toP.odoratum and P. odo-Se groups and HDL-cholesterol levels were increased in the diabetic experimental groups fed on BuOH fraction of P. odoratum with Se supplementation. The liver and muscle glycogen levels were not significantly differ among all groups. The plasma free fatty acid levels were lower in diabetic experimental groups fed on BuOh fraction of P. odoratum or Se sup-plementation than STZ-control and Se groups. Diabetics rats showed the higher levels of triglyceride in plasma andlower levels in liver compared with the normal group. Supplementation with Se decreased significantly the liver triglyceride level. The MDA levels in liver and kidney were significantly reduced in all the experimental groups. In conclusion administration of BuOH fraction of Polygonatuum odoratum with selenium supplementation reduced blood glucose levels and peroxdative tissue damage in STZ induced diabetic rats showing the possibility of preventiave and therapeutic use of the wild edible plant to the diabetes mellitus.

둥굴레(Polygonatum Odoratum var. Pluriflorum Ohwi)추출물의 당뇨 유발 흰쥐에 대한 혈당강화 효과 (Hypoglycemic Effect of Polygonatum Odoratum var. Pluriflorum Ohwi Extract in Streptozotocin-Induced Diabetic Rats)

  • The hypoglycemic effect of Polygonatum odoratum var. Pluriflorum Ohwi was investigated after extracted with methanol. The methanol extract was fractionated into 4 layers ; hexane, chloroform, butanol and aqueous. Fifty male Sprague-Dawley rats(200-300g) were induced diabetes mellitus by the streptozotocin injection(45mg/kg B.W) into the tail vein and were divided into 5 groups ; diabetic control and 4 experimental groups. All groups of the rats were fed on a AIN-76 diet, and the 4 experimental groups were orally administered with each fraction(500mg/kg B.W) for 12 days and the diabetic control group was orally administered CMC. The body weight gain was monitered and the blood levels of glucose and cholesterol were measured. Levels of protein, triglyceride, and free fatty acid in plasma were analysed. The contents of protein, triglyceride and glycogen in liver and of protein and glycogen in muscle were also determined. The body weight gain was shown significantly higher in the H2O fraction group than that in the diabetic control group and the heart weight was greater in the CHCl3 group. The extents of blood glucose decrement were greater in the BuOH and H2O groups than that in the control group. The urinary glucose excretion was shown relatively small amount in the BuOH and H2O groups. The plasma cholesterol and protein levels were not influenced by these four fractions in diabetic rats. The liver glycogen level was significantly higher in the BuOH group. The results suggest that the orally administered BuOH and H2O fraction of Polygonatum odoratum var. Pluriflorum Ohwi exhibited hypoglycemic effect in streptozotocin induced diabetic rats.

층층갈고리둥굴레 (Polygonatum sibiricum Redoute) 뿌리줄기와 잎의 증숙 시간과 증숙 온도에 따른 생리활성 변이 연구 (Changes of Biological Activities of Rhizome and Leaves of Polygonatum sibiricum Redoute according to Steaming Time and Temperature)

  • Background: The aim of this study was to analyze the total phenol and total flavonoid contents and antioxidant activity of steam-treated leaves and rhizomes of Polygonatum sibiricum Redoute. In addition, we aimed to confirm their potential use as cosmetic materials by investigating their anti-aging and skin-whitening activity. Methods and Results: The leaves and rhizomes of P. sibiricum were treated with steam at different temperatures for different durations, and the antioxidant activity (DPPH and ABTS radical scavenging activity) and total phenol and total flavonoid contents of each sample were tested. The steam temperature and treatment duration siginificantly affected the antioxidant activity and, total phenol and total flavonoid content of the leaves and rhizome of P. sibiricum. Treating the P. sibiricum samples with steam at 120℃ for 12 h, yielded higher total phenol and total flavonoid contents. Comparatively, the samples treated with steam at 120℃ for 12 to 24 h showed significantly higher antioxidant activity. Further, the steamed samples of P. sibiricum demonstrated collagenase and tyrosinase inhibition activity, which indicated their anti-aging and skin-whitening properties. The samples steamed at 120℃ for 12 h, exhibited higher collagenase and tyrosinase inhibition activity. Conclusions: Leaves and rhizomes of P. sibiricum steamed at 120℃ for 12 h, showed highest antioxidant activity and, total phenol and total flavonoid contents than all other samples. Our results indicate the potential of using P. sibiricum as a cosmetic material by confirming its excellent anti-aging and whitening activity.

  • 현재 농가에서 소규모 재배가 이루어 지고 있는 둥굴레의 적정 재식 밀도를 구명하여 종근의 알맞는 소요량을 구명하고 생산력을 높임으로서 안정적인 공급을 기할 수 있도록 알맞은 재식거리를 구명하기 위하여 시험하여 얻은 결과를 요약하면 다음과 같다. 1. 휴간 30, 주간 10 cm(반당 33,000본)으로 재배함으로서 지상부 및 지하부 생육이 모두 주간거리 20, 30cm에 비하여 양호하였다. 2. 근경수량은 수확본수가 많고 본당 생중이 무거운 $30{\times}10\;cm$에서 건근경중 1,593 kg/10a로 가장 높았다.

둥굴레 근경의 가열조건에 따른 갈변반응 특성의 모니터링 (Monitoring of Maillard Reaction Characteristics under Various Roasting Conditions of Polygonatum odoratum Root)

  • 반응표면분석법을 이용하여 둥굴레 근경의 가열조건에 따 라 갈변 반응기질과 관능적 특성을 모니터링하였다. 총 유리당은 중심점에서 최소점을 나타 내었고 그 전후로 증가하는 경향을 나타내었고, 총 유리아미노산은 볶음온도가 증가하고 볶 음시간이 길어질수록 감소하는 경향을 나타내었다. 그 중 threonine, glycine 및 serine의 함 량응ㄴ 시간이 경\ulcorner마에 따라 초기함량의 91~94%가 감소함을 보였다. 또한 갈색도는 볶음 시간 18분까지 볶음온도와 볶음시간이 증가할수록 증가하다가 그 이후로 감소하는 경향을 나타내었다. 둥굴레의 맛, 색, 향에 대한 관능적 평가는 5% 이내에서 유의차가 인정되었다. 전반적 기호도는 볶음온도는 13$0^{\circ}C$에서 볶음시간 15분과 25분에서 높은 것으로 나타났다. 향에 대한 관능적 품질과 유리아미노산은 갈변반응의 다른 기질에 비하여 높은 수준에서의 상관관계를 나타내었다.

  • 경상남도농업기술원 함양약초시험장 포장에 발생한 병든 둥굴레로부터 병원균을 분리하여 병원균의 특성과 병징을 토대로 Sclerotium rolfsii에 의한 둥굴레 흰비단병을 동정하였다. 이 병의 병징은 지제부가 암갈색으로 변하고 그 위에 흰색의 곰팡이가 솜털처럼 밀생하고, 병반부위에 갈색의 둥근 균핵을 많이 형성하며 지제부 부근의 토양 표면에도 형성되었다. 병원균은 PDA 배지 상에서 왕성하게 생장하였으며 균사생장 중에 특유의 Clamp connection이 관찰되었고 균사가 가장 잘 자랄수 있는 온도는 $30^{\circ}C$이였다. PDA 배지에서 시간이 경과되면 갈색을 띤 구형 또는 부정형의 많은 균핵을 형성하였는데 균핵의 크기는 $0.9{\sim}6.8{\times}0.6{\sim}5.2\;mm$ (평균 $2.7{\sim}2.3\;mm$) 이었다.

둥굴레차의 고품질화를 위한 볶음조건의 최적화 (Optimization of Roasting Conditions for High-Quality Polygonatum odoratum Tea)

  • 전통적인 볶음방법으로 제조되고 있는 둥굴레차의 고품질화를 위하여 반응표면분석법에 의한 볶음조건을 최적화하였다. 둥굴레차의 품질지표로서 수용성 고형분, 갈색도, 페놀성 성분 및 전자공여 작용은 $145^{\circ}C$와 55분에서 나타나는 능선까지는 볶음온도와 시간이 증가함에 따라 증가하였으나 그 이상의 온도와 시간에서는 다시 줄어들었다. 둥굴레차의 관능적 품질로서 전반적인 기호도는 최대 관능평점을 나타낸 $146^{\circ}C$와 52분까지는 볶음온도와 시간이 증가할수록 높아졌으나, 최대점을 지나서는 온도가 높아지고 시간이 경과함에 따라 점점 줄어들었다. 이상과 같이 각각의 볶음조건에 따른 둥굴레차의 품질 특성을 종합하여 superimposed contour map을 작성한 결과 볶음온도 $135{\sim}140^{\circ}C$, 볶음시간 $58{\sim}64$분이 둥굴레차의 품질을 극대화시킬 수 있는 최적 볶음조건 범위로 나타났으며, 최적조건 범위내의 주어진 볶음조건$(137^{\circ}C,\;60\;min)$에서의 실험치는 예측치와 일치하였다.

둥굴레, 어성초, 구기자 혼합 음료의 배합비 최적화 및 항산화 효과 (Optimization of Production and Antioxidant Effects of Beverage Prepared using Hot-water Extracts of Polygonatum odoratum, Houttuynia cordata, and Lycium chinensis)

  • 본 연구에서는 당류 등 별도의 첨가물질을 전혀 사용하지 않고 오직 둥굴레, 어성초 및 구기자로부터 추출한 열수추출액만을 혼합하여 기호도가 높으면서도 혈당 및 지질대사 개선 효능을 가진 건강음료를 제조하는 연구의 일환으로, 관능평가와 반응표면분석에 의하여 이들 재료의 혼합비율을 최적화하고 최적 비율로 혼합 제조한 음료의 항산화능을 조사하였다. 그 결과, 음료 제조 시 열수추출액들의 최적 혼합비율은 둥굴레 열수추출액의 첨가비율 2.15%, 어성초 열수추출액의 첨가비율 1.74%, 구기자 열수추출액의 첨가비율 0.19%인 것으로 예측되었다. 제조한 음료의 항산화능을 알아보고자 조사한 총 폴리페놀, 전자공여능 및 환원력은 각각 3.42 mg/mL, 23.78% 및 0.26이었다.

렛 근육세포 L6에서 둥굴레 추출물의 인슐린저항성 개선 (Attenuation of insulin resistance using steamed Polygonatum odoratum var pluriflorum extract in rat skeletal muscle cells L6 myoblast)

  • Objectives : The purpose of this study was to investigate inhibitory effects of steamed Polygonatum odoratum extract (POE) on insulin resistance in rat skeletal muscle cells, L6 cells.Methods : Polygonatum odoratum (P. odoratum) extract was extracted with ethyl acetate. Activity of α-glucosidase in POE was measured for blood glucose regulation. MTT assay was examined for cell toxicity. Western blot analysis for measurement of adiponectine, peroxisome proliferator-activated receptorγ (PPARγ), insulin receptor substrate (IRS), glucose transporter 4 (Glut-4) and phosphorylation of serine/threonine-specific protein kinase (Akt) expressions were performed. Akt signaling pathway were analyzed with LY294002, which is a specific PI3K/Akt inhibitor.Results : The results revealed that POE inhibited α-glucosidase activity. Treatment of POE in L6 cells inhibited the differentiation of L6 cells compared to those of vehicl control. Additionally, protein expressions of adiponectine, PPARγ, IRS and Glut-4 were significantly regulated compared to those of vehicle control (p < 0.05), respectively. Futhermore, phosphorylation of Akt was increased in L6 cells treated with POE compared to that of vehicle control (p < 0.05). pAkt expression was significantly accentuated with Akt inhibitor (LY294002).Conclusions : These results suggest that POE may have potential as a natural agent for prevention/improvement of diabetes, especially, regulation of blood glucose. Therefore, further additional study should be conducted to elucidate in depth the pharmaceutical efficacy of these.
