Process Development of Alcohol Production by Extractive Fermentation(III) -An Optimum Composition of PEG/Dx for Extractive Alcohol Fermentation- (추출발효에 의한 알코올.제조공정 개발(III) -추출 알코올 발효에 최적인 PEG/Dx의 조성-)

  • Extractive fermentations with the extract of Jerusalem artichoke in an aqueous-two-phase-system of polyethyleneglycol(PEG) and dextran(Dx) were investigated to obtain the effects of composition of PEG and Dx on both fermentation ,characteristics and partition ratio of alcohol. The specific growth rate of K. Fraglis CBS 1555 increased with a decrease of concentration of PEG and Dx. It augmented along with concentration of initial sugar up to 80g/l but decreased thereafter. The specific production rate of alcohol showed a rising tendency up to 100g/lof initial sugar, whereafter represented a decreasing trend. The partition ratio of alcohol between two phases augmented according to decrease of Dx comic. and increase of PEG cone. regardless of initial sugar concentrations. The ratio, however, decreased with Increment of initial sugar concentration at constant composition of PEG and Dx. The partition coefficient of alcohol had an ascending effect to the increase of PEG cone, but it had little effect on the changes of concentrations of Dx and initial sugar. The present study suggests that the optimum composition of PEG and Dx in the aqueous-two-phase-system by extractive fermentation were around 6.5%(w/v) of PEG and 3%(w/v) of Dx in considerations of emulsion state, sedimentation and separation of two phases, alcohol partition ratio, and specific growth rate.

Dehydration and pore swelling effects on the transfer of PEG through NF membranes

  • In order to investigate the significance of "salting-out" and "pore swelling" effects on the nanofiltration of neutral solutes, rejection properties of two NF ceramic and polymeric membranes were studied with single polyethyleneglycol (PEG) solution and mixed PEG/inorganic electrolyte solutions. For both membranes, the rejection rate of PEG was found to decrease significantly in the presence of ions. In the case of the ceramic membrane (rigid pores), this phenomenon was imputed to the sole partial dehydration of PEG molecules induced by the surrounding ions. This assumption was confirmed by the lowering of the PEG rejection rates which followed the Hofmeister series. Experimental data were used to compute the resulting decrease in the Stokes radius of PEG molecules in the presence of the various salts. Concerning the polymeric membrane, the decrease in the rejection rate was found to be systematically higher than for the ceramic membrane. The additional decrease was then ascribed to the swelling of the pores. The experimental data of rejection rates were then used to compute the variation in the mean pore radius in the presence of the various salts. The pore swelling phenomenon due to accumulation of counterions inside pores was supported by electrokinetic charge density measurements.

Local hydrogel patterning and microcantilever fabrication using dynamic mask lithography (동적 마스크 리소그래피를 이용한 하이드로젤 국소 패터닝 기법과 캔틸레버 제작)

  • We report a new method for highly controllable local patterning of a hydrogel on microfabricated cantilevers and fabrication of all hydrogel microcantilevers. We constructed a dynamic mask based photolithography setup using a commercial beam projector, a 3-axis microstage and other optical components. Dynamic masks generated from the beam projector controlled the shape, size, and position of hydrogel patterns while the 3-axis microstage mainly controlled the thickness of hydrogel patterns and hydrogel microcantilevers. Using the constructed setup, polyethyleneglycol diacrylate (PEGDA) was patterned on microfabricated cantilevers in a highly controlled manner. Currently, the smallest PEGDA patternable is a 5-${\mu}m$-diameter circle with a thickness of ~$10{\mu}m$. To confirm thicknesses of patterned PEGDAs on silicon microcantilevers, resonance frequencies of microcantilevers were measured before and after each PEGDA patterning. Thicknesses extracted from resonance measurements showed good agreement with measurements using an optical microscope. In addition, PEGDA microcantilevers with various dimensions and thicknesses were fabricated on glass and silicon substrates. Surfaces of fabricated all hydrogel microcantilevers were flat enough to facilitate other post processing and to be used for various sensing applications.

  • PEG phase transfer catalyzed reduction of nitrobenzene with $Fe(CO)_5$ afforded aniline up to over 50% yield al room temperature. The reaction rate of nitrobenzene was dependent on the chain length and number of binding sites of PEG, and it was increased with the concentration of NaOH and solvent polarity. But the yield of aniline was maximum) with 2N NaOH solution and it was decreased at higher concentrations.

Effect of Polycaprolactone on the Mechanical Properties of PEG/HMX Propellant (Polycaprolactone이 PEG/HMX 추진제의 기계적 성질에 미치는 영향)

  • Mechanical properties of polyethyleneglycol(PEG)/cyclo-tetramethylene tetranitramine(HMX) propellant were studied by adding polycaprolactone(PCP) and silane derivatives as coupling agent. Mechanical properties of the propellant were enhanced by increasing the content of PCP which was partially replaced with PEG. PCP/HMX propellant showed 17% increase in maximum tensile strength and 59% increase in elongation compared to those of PEG/HMX propellant. However, as the content of PCP was increased, the hydrolytic stability of this propellant was found to be deteriorated due to the ester group of PCP by measuring hardness drop of cured propellant and swell ratio difference of PEG/PCP binder stored at $40^{\cire}C$ and 90% relative humidity. Hardness of the PEG/PCP/HMX propellant was decreased with increasing triethoxysilyl propionitrile(TESPN) and dinitrosalicylic acid (DNSA) content from accelerated aging at $60^{\cire}C$.

  • We have studied on the surface and adhesion properties for acrylic terpolymers, having both hydrophobic and hydrophilic side chains, synthesized via solution polymerization. In order to develop a waterborne material. we tried to synthesize these terpolymers via emulsion polymerization. The polymeric emulsion synthesized was mainly composed of methyl methacrylate (MMA), methoxy-polyethyleneglycol methacrylate (MPEGMA) having hydrophilic side chains and methoxypolypropyleneglycol methacrylate (MPEGMA) having hydrophobic side chains. The viscosities of this series increased with an increase in the content of the co-monomer such as MPEGMA and (MPEGMA). This behavior resulted in the increase in the diameter and heterogeneity of the emulsion particle via AFM observation. Furthermore. the tensile adhesion strength and 90-degree peel strength of the adhesive of these polymeric emulsions were measured. In the case of polymeric emulsion composed of the same content of both hydrophilic and hydrophobic component, the adhesion property showed the highest value. However, since the adhesion properties as a practical applicable adhesive were poor, some improvements were required. When the composition above was modified with butyl acrylate (BA), the improvement effect on adhesion strength was accepted. In particular, 90-degree peel strength increased up to a maximum of 400% of the original value.

Heterogeneity of Mammalian Plasma Albumin (포유류 혈장알부민의 이질성)

  • Plasma albumin was purified from the fresh bovine blood using a minor modification of the polyethyleneglycol and ethanol procedure. The resulting protein solution was tested for its purity by both electrophoretic and immunochemical methods and found to contain only the albumin molecules. Each of the four thiol reagents, maleate, iodoacetate, iodoacetamide and glutathione, was incubated with the purified plasma albumin. The electrophoresis on cellulose acetate of those complexes in various buffers with different component and pH demonstrated that the albumin-glutathione complex was separated into two zones in all buffers used except the barbital and sodium acetate buffers, that the complexes of albumin-iodoacetate and albumin-iosoacetamide also into two zones only in pH 4.8 citrate buffer and in pH 4.8 succinate buffer and that the new zone had more positive net charge compared to the native protein in any case. These results might suggest a possibility that the electrophoretic albumin fraction is composed of at least two molecular species with different conformation.

  • The polycarboxylic (PC) type concrete superplasticizer was synthesized. The effects of ethylene oxide group number and its molecular weight on the properties of the polycarboxylic type concrete superplasticizer and the concrete motar properties were studied. To investigated of the interfacial properties of the premixed-concrete with the superplasticizer, the type and the amount of polyethylene glycol, meta acrylate added, and type of the initialization agent were studied. Also the interfacial properties of the superplasticizer aqueous phase, the wettability on the cement particle, the fluidity of the cement mortar, and the strength properties of the concrete were measured. For a high fluidity of the cement mortar and a high strength of concrete, a low value of the surface tension and contact angle were required for PC. To have a good performance for PC, the reaction condition of 1.3 mol ratio of MA against to MPEG was suitable with KSP initiator.

Development of L-Lysine Producing Strains from Cellulosic Substrate by the Intergeneric Protoplast Fusion - Conditions for Fusion and Properties of Fusants- (속간 원형질체 융합에 의한 섬유질 기질로부터 L-Lysine 생산균주 개발 -융합조건 및 융합체의 성질 -)

  • To produce L-lysine from cellulosic substrate, the intergeneric protoplast fusion between Cellulomonas flavigena and Corynebacterium glutamicum, Cellulomonas flavigena and Brevibacterium flavum was performed. The fusion frequencies were 1.9$\times$10$^{-6}$ to 2.1$\times$10$^{-6}$ for the regenerated protoplasts when two parental strains were treated with 30% of polyethyleneglycol (M.W.6000) containing 5 mM EDTA at 3$0^{\circ}C$ for 30 min. Two fusants, FCB3 and FCC 19 were finally selected by comparision of their genetic stability and L-lysine productivity. The properties of fusants-DNA con-tent, G+C content and L-lysine productivity-were investigated. The DNA content of fusants was greater than those of the parental strain and their G+C contents are equal to half of total G+C con-tent of two parental strains. The fusants showed high productivity of L-lysine from carboxy methyl cellulose as substrate.

A Study on the UV-cut Properties of Cotton Fabrics Treated with UV-absorber (자외선 흡수제 처리 면직물의 소비성능 개선(제1보) - 자외선 차단성능에 관한 연구 -)

  • The influence of ultraviolet(UV)-ray in sun light on human skin has been noted. Textiles can provide protection against harmful UV-radiation. Normally UV-absorbing finishes are used to get better protection. The purpose of this study is to evaluate the UV-cut properties of cotton fabrics treated with UV-absorber. 2,2-dihydroxy-4,4-dimethoxbenzophenone, as UV-absorber was applied to 100% cotton fabric. Reagents added in finishing solution were Triton X-100, polyethylene glycol 400, and $MgCl_2{\cdot}6H_2O$, and C.I. Direct Red 81. Both untreated and treated cotton fabrics were exposed to a xenon arc lamp for 20 and 80 hours. UV absorption spectra of finishing solutions and UV transmission spectra of fabrics were measured by the UV/VIS spectrophotometer. The results of this study can be summarized as follows. The results of this study can be summarized as follows. Absorption and the related transmission spectra were modified in a controlled way with UV-absorber. Absorption effect of UV-absorber was improved by adding Triton X-100, PEG 400, and $MgCl_2{\cdot}6H_2O$ in finishing solution. The UV absorption of finishing solution was in the following order: U/D/T/P/M>D/T/P/M> D/T> D/P, D>U/T/P/M>U/T>T/P/M>T. The UV transmittance of cotton fabrics was remarkably decreased by the application of UV-absorber and additives. The UV-cut properties were most improved by the application of U/D/T/P/M.
