Drought resistance and salt resistance of seedlings were compared with the polyethylene glycol(P.E.G) and NaCl solutions of the same osmotic potential($\Psi_\pi$=-10 bar). Plant height, seedling dry weight, chlorophyll content and leaf water potential decreased while the free proline content increased more in the P.E.G. than in the NaCl solution. Free amino acids increased 2.6 times in the P.E.G. solution and 3.6 times in the NaCl solution more than in the untreated(Hoagland's solution). Free proline occupied 66% and 61% of the content of total amino acids under water stress and salt stress, respectively. Besides free proline, phenylalanine in the P.E.G. solution and phenylalanine, alanine and asparagine in the NaCl solution increased distinctly. In short, it was shown that water and salt stress responses in seedling stage were relatively similar.
Bacillus sp. YBL-7 which had been isolated from ginseng root-rot suppressive soil was able to antagonize Fusarium solani causing ginseng root-rot by their antibiotic substance. In order to develop multifunctional antagonist on Bacillus sp. YBL-7 as a biocontrol agent against Fusarium salam', optimal conditions for protoplast transformation system of Bacillus sp.YBL-7 by the vector plasmid pE194 were investigated. The protoplasts of Bacillus sp. YBL-7 were obtained at best efficiency by treatment with 200${\mu}g$/ml of lysozyme in the pH 7.0 of SMM buffer for 90 minutes at $40^{\circ}C$. The cell wall of the protoplast was regenerated on the agar plate containing 1.2% agar and 0.7 M mannitol. Under the best condition for protoplast formation and regeneration, the optimal transformation was achieved with 40% polyethylene glycol (M.W. 4000) treatment for 10minutes. The vector plasmid pE194 showed the best transformation frequency at 5$\mu$g/ml of final concentration. The pE194 was very stable over 80% in the transformants.
A series of polyethers containing the thiazole or oxazole subcyclic moiety have been synthesized. Reaction of 2-aryl-4-hydroxymethylthiazole with tetra- and pentaethylene glycol di-p-tosylate in THF provided corresponding α,ω-bis[2'-aryl-4'-methylthiazole]polyethylene glycol in good yields. Similar treatment of 2-phenyl-4-hydroxymethyloxazole 7 and 2-phenyl-5-hydroxymethyloxazole 8 with tetraethylene glycol di-p-tosylate yielded the corresponding 1,13-bis [2'-phenyl-4'-methyloxazole]tetraethylene glycol 16 and 1,13-bis[2'-phenyl-5'-methyloxazole]tetraethylene glycol 17 in 69 and 43% yields in respectively. The potentiometric properties of PVC-based ion selective membranes containing 66 wt% o-nitrophenyloctyl ether (NPOE) and 4 wt% polyethers 9-17 have been examined. The membranes containing thiazole and oxazole polyether derivatives exhibited high selectivity toward silver (I) ion. It was observed that the response slopes of the electrodes to silver ion vary with the length of polyether chain linking two thiazole subcyclic moiety. Potentiometric data suggest that the number of ether units, CH2OCH2, for phenylthiazole derivatives be greater than 5 to result in near-Nernstian response. However, the response behaviors of the membrane electrodes based on phenyloxazole podands 16 and 17, which have different orientation, were correspondingly similar to those of the electrodes based on phenylthiazole podands 9 and 10. On the other hand, the ISEs based on thiazole polyether derivatives with different terminal substituents, e.g., phenyl 10, naphtyl 14, and thienyl 15, except that with pyridyl 12, exhibited little difference in their potentiometric properties.
Polyvinyl chloride (PVC) ultrafiltration membrane was prepared by blending 12 wt.% of PVC in N, N-dimethylacetimide (DMAc) with polyethylene glycol 400 (PEG400) as an additive. The influence of PEG400 concentration on the PVC membrane morphology, permeability, fouling and rejection were investigated. Fouling and rejection of the PVC membrane were characterized by dextran T-100 filtration. The results showed that membrane water flux was increased up to $682Lm^{-2}h^{-1}$ when 28 wt.% of PEG400 was added into the PVC membrane solution. The best membrane performance with a low fouling and a high selectivity was achieved by adding 12 wt.% concentration of PEG400, which resulted in 90% rejection of dextran and 90% of flux recovery ratio. At further addition of PEG400 concentration, irreversible fouling was starting to increase. A 90% of irreversible fouling was formed in the PVC membrane when more than 22 wt.% of PEG400 is added.
Effect of extraction pH on mechanical properties such as tensile strength (TS) and elongation (E) and on water vapor permeability (WVP) of soybean protein isolate (SPI) edible films extracted from soybean meal was investigated. Five pHs, acidic range (pH 2.0 and pH 3.0), neutral range (pH 7.0) and alkalic range (pH 10.0 and 12.0), were used to extract SPI. TS of the film extracted at pH 7.0 was the lowest, and WVP of $SPI_3$ (SPI extracted at pH 3) film was the lowest value among the films. The WVP of $SPI_3$ films was $3.349\;{\times}\;10^{-10}\;g{\cdot}m/m^2{\cdot}s{\cdot}Pa$ and increased to $3.871\;{\times}\;10^{-10}\;g{\cdot}m/m^2{\cdot}s{\cdot}Pa$ as film thickness increased from $55\;{\mu}m$ to $72\;{\mu}m$ thickness. Three different plasticizers (glycerol, polyethylene glycol and propylene glycol) were used for $SPI_2$ (SPI extracted at pH 2) film. TS of $SPI_2$ films was 12.297 MPa and decreased to 1.356 MPa for glycerol and showed the same trend in other two plasticizers. The SPI films extracted at acidic range were shown higher mechanical properties and lower water vapor permeabilities than those of extracted at neutral and alkalic ranges. The difference of SPI film properties seemed to be attributed by 11S/7S ratio as well as protein content.
- To investigate the possibility of genetic development for a multi-purpose strain of Bacillus subtilis YBL-7 against Fusat-iurn solani causing root rot of many impotant corps, the plasmid pGU66 inserting urease gene of Bacillus pasteurii had been introduced into Bacillus subtilis YBL-7 by PEG-induced protoplast (PIP) transformation system. Protoplasts of B. subtilis YBL-7 were prepared by treating the cells with lysozyme (200 $\mu g$/ml) in hypertonic buffer (SMMP). The highest transformation frequency was achieved when cells of the strain with lysozyme at $42^{\circ}C$ for 90 minutes. Optimal transformation was obtained using polyethylene glycol (MW 4000) at final concentration of 30% (V/V). The transformation frequency was increased proportionally to 1.2 $\mu g$ of plasmid DNA. At best condition, the transformation frequency (transformants/ regenerants/$\mu g$ of DNA) for pGU66 was appoximately $4 \times 10^{-3}$. Also, the urease gene was strongly expressed in the transformants of B. subtilis YBL-7 and maintained steadily. The antifungal ability of transformant was very similar to that of B. ssubtilis YBL-7.

Low levels of virus contamination and naturally occurring reverse transcription-polymerase chain reaction (RT-PCR) inhibitors restrain virus detection in oysters. A rapid and efficient oyster-processing procedure that can be used for sensitive virus detection in oysters was developed. Poliovirus type 1 Sabin strain was used to evaluate the efficacy of virus recovery. The procedure included (a) acid-adsorption and elution with buffers (0.25M glycine-0.14 M NaCl, pH 7.5; 0.25M threonine-0.14M NaCl, pH 7.5); (b) polyethylene glycol (PEG) precipitation; (c) resuspension in Tween 80/Tris solution and chloroform extraction; (d) the second PEG precipitation; (e) viral RNA extraction with TRIzol and isopropanol precipitation; and (f) RT-PCR combined with semi-nested PCR. The overall recovery of elution/concentration was 19.5% with poliovirus. The whole procedure usually takes 19 hr. The overall detection sensitivity was 4 RT-PCR units of genogroup I norovirus (NoV) and 6.4 RT-PCR units of genogroup II Nov/25 g of oysters initially seeded. The virus-detecting method developed in this study should facilitate the detection of low levels of NoV in oysters.
In this report the practical case of Viking ship's conservation in Denmarke specially among the Eurpoean nations is introduced. The contents of it are summarized as follows :From 1957 to 1962 the Danish National Museum Salvaged five Viking ships from the bottom of Roskilde Fjord, Which were composed of the pieces of timber whose surface was soft because they had lain on the sea bed for about a thousand years. Excavation had been carried out in the same way as in the field by driving down a sheet piling around the wrecks and pumping the water out. These pieces of the wreck ships were packed in airtight plastic bags one by one to be transported for Brede and then immidiately had to go through the treatment for conservation. The conservation treatment process for the pieces includes three steps ; the preliminary process prior to the hardening treatment, the hardening and the assemble of the ships. In the first step ; the preliminary process, all remains of mud and shells from the fjord bed are washed off, and measuring followed ; every single piece of wreckage was drawn so that the form and size of the piece, nail holes, and breaks were registered before conservation. In the second ; the hardening treatment step, the pieces of the woreckage were filled with P.E.G. This Polyethylene Glycol method was the best to handle in the subsequent mounting of the ships in the museum. In the final, the Glycol-treated pieces were pieced together to spips with support of a system of reinforcements. They were to fit in place after corrections of the form were made several times.
Biodegradable edible films were prepared from rice protein concentrates (RPC) made from rice wine meal by alkaline extraction and isoelectric precipitation. The effect of film forming solution pH and plasticizers were studied, and cross-linkers were added to improve mechanical properties and water vapor permeabilities (WVP) of films. Films could be formed within pH $8{\sim}11$ with tensile strength (TS) of 4.3{\sim}5.7\;MPa$. Films produced under pH 11 had the highest TS (5.7 MPa) and the lowest WVP $(0.44\;ng{\cdot}m/m^2{\cdot}s{\cdot}Pa)$. Added glycerol, polyethylene glycol 200 (PEG) and its mixture (GLY:PEG=50:50) as plasticizers also affected the mechanical properties and WVP of films. TS and elongation at break (E) of films at various plasticizer levels were $5.5{\sim}1.0\;MPa$ and $3.6{\sim}24.3%$, respectively. At the same plasticizer concentration, the highest TS was observed when glycerol was used whereas the highest E was measured when mixture was used as plasticizer. WVPs of films with thickness of $60\;{\mu}m$ were $0.39{\sim}0.54\;ng{\cdot}m/m^2{\cdot}s{\cdot}Pa$. WVP of films decreased as the ratio of glycerol/PEG 200 was decreased, and WVP increased as the total amount of plasticizer added to the films increased. Film strength was improved by the addition of small amount of sodium hydrogen sulfate, succinic anhydride, ascorbic acid and citric acid, whereas TS of films containing $0.5{\sim}2.0%$ of NaCl and $CaCl_2$ were lower than those without the salts. The highest TS (6.3 MPa) was achieved with films containing 0.1% of succinic anhydride.
The present study evaluated effect of egg yolk immunoglobulin (IgY) from the hen immunized with Edwardsiella tarda. The purification of anti-E. tarda IgY was performed by polyethylene glycol (PEG). Purified IgY had heavy chain of 64 kDa and light chain of 27 kDa size. The IgY was instable against olive flounders digestive factors and artificially modulated pH 2 and 3. Nevertheless, some activity of IgY appeared in intestine. IgY was orally administered with viable E. tarda to the olive flounders and the efficacy of protection against E. tarda infection was evaluated. Orally administered IgY at a dose of 20 mg/fish delayed infection period of E. tarda cannulated at $10^{6{\sim}8}CFU/fish$ to small size (30~40 g) and middle size (110~120 g) flounder. Moreover, orally administered IgY at dose of 20 mg/fish inhibited the penetration of E. tarda cannulated at $10^8CFU/fish$ into the liver, kidney, spleen and gill via intestine. The fish orally administered with IgY showed increased survival rate. These results suggest that egg yolk containing anti-E. tarda IgY is effective in preventing edwardsiellosis.
