Identification and Functional Analysis of Mating Type Loci in the Pleurotus eryngii

  • Pleurotus eryngii has recently become a major cultivated mushroom; it uses tetrapolar heterothallism as a part of its reproductive process. Sexual development progresses only when the A and B mating types are compatible. Such mating incompatibility occasionally limits the efficiency of breeding programs in which crossing within loci-shared strains or backcrossing strategies are employed. Therefore, understanding the mating system in edible mushroom fungi will help provide a short cut in the development of new strains. We isolated and identified pheromone and receptor genes in the B3 locus of P. eryngii and performed a functional analysis of the genes in the mating process by transformation. A genomic DNA library was constructed to map the entire mating-type locus. The B3 locus was found to contain four pheromone precursor genes and four receptor genes. Remarkably, receptor PESTE3.3.1 has just 34 amino acid residues in its C-terminal cytoplasmic region; therefore, it seems likely to be a receptor-like gene. Real-time quantitative RT-PCR (real-time qRT-PCR) revealed that most pheromone and receptor genes showed significantly higher expression in monokaryotic cells than dikaryotic cells. The pheromone genes PEphb3.1 and PEphb3.3 and the receptor gene PESTE3.3.1 were transformed into P5 (A3B4). The transformants were mated with a tester strain (A4B4), and the progeny showed clamp connections and a normal fruiting body, which indicates the proposed role of these genes in mating and fruiting processes. This result also confirms that PESTE3.3.1 is a receptor gene. In this study, we identified pheromone and receptor genes in the B3 locus of P. eryngii and found that some of those genes appear to play a role in the mating and fruiting processes. These results might help elucidate the mechanism of fruiting differentiation and improve breeding efficiency.

Effect of LED Light on Primordium Formation, Morphological Properties, Ergosterol Content and Antioxidant Activity of Fruit Body in Pleurotus eryngii (LED광원이 큰느타리버섯 자실체의 발생, 생육, 에르고스테롤 함량 및 항산화활성에 미치는 영향)

  • Light wavelength is the major factor of fruit body development associated with mushroom cultivation, but its wavelength range in Pleurotus eryngii is poorly understood. Using four kinds of light emitting diode (LED) including blue (475 nm), green (525 nm), yellowed (590 nm) and red (660 nm), we investigated to elucidate suitable light wavelength during primordium formation and fruit body development of P. eryngii on bottle cultivation. Primordia formation did not occur in blue light and red light. The morphological properties of fruit body in fluorescent lamp and blue light irradiation were showed thicker and larger pileus than those in other LEDs. However, length of stipe in fluorescent lamp and blue light was shorter than that of other LEDs. The DPPH radical was high in blue light, green light, and yellow light except for red light, and the polyphenol was high in four kinds of LED sources. And ergosterol was the highest in the green light. Thus, the high-quality mushroom production of P. eryngii is possible to green light condition considering productivity and functional materials.

Applicability of Spent Mushroom Media as Horticultural Nursery Media (버섯재배 후 탈병배지의 원예용 상토재료 이용성 검토)

  • This study was carried out to investigate applicability of Spent Mushroom Media(SMM) as horticultural nursery media. After the mushroom has been harvested, the SMM contains a lot of organic material, different microorganism and high density of mushroom hypha. The pH, phosphate and exchangeable cation concentrations of SMM of Flammulina velutipes were higher than those of any other treatment. The CEC and $NH_4-N$ were the highest in SMM of bottle-cultivated oyster mushroom (Pleurotus ostreatus). Bacteria and fungi showed the highest density in SMM of Flammulina velutipes. Most dominant bacteria were Microbacterium sp., Rhodococcus sp. and Agrobacterium sp. in SMM of Flammulina velutipes and Bacillus sp., Pseudomonas sp., Curtobacterium sp. and Microbacterium sp. in that of Pleurotus eryngii. The SMM contained high density of mushroom hypha that inhibited germination of seed and growth of young seedlings. Therefore, composting process of the SMM is indispensible to decline of vitality of mushroom hypha. The SMM of Flammulina velutipes with 0~30% vermiculite showed high germination rate in red pepper and chinese cabbage seeds. SMM of Pleurotus eryngii with 20% vermiculite showed 100% germination rate in red pepper seeds, but chinese cabbage seeds nearly failed to germinate with 30% vermiculite. The growth of red pepper was increased according to increasing mixture ratio of vermiculite. Accordingly, we concluded that SMM of Flammulina velutipes contained 0~30% of vermiculite can be used to horticultural growth bed for red pepper.

Phylogenetic relationships of Pleurotus species based on RAPD analysis

  • Oyster mushrooms including of P. ostreatus, P. eryngii, P. pulmonarius and P. cornucopiae are one of the famous mushrooms for foods in Korea. RAPD were carried out using 14 of oligoprimers to analyze the phylogenetic relationship among 57 strains of 32 Pleurotus species. Most of species formed the minimum clade with strains within species and was divided respectively species. Therefore clade was separated well in accordance species. Pleurotus species formed again clade to be added in close related to other species, and were discriminated by sixteen clades with each representative species including high similarity groups. Sixteen clades were composed representative species according to each clade. There were clade I of P. pulmonarius(P. sajor-caju, P. opuntiae, P. sapidus), clade II of P. eryngii(P. fuscus var. ferulae, P. fossulatus), clade III of P. ostreatus(P. ostreatus var. columbinus, P. spodoleucus, P. floridanus), clade IV of P. florida, clade V of P. djamor(P. flabellatus, P. incarnates, P. salmoneo-stramineus), clade VII of P. populinus(P. subareolatus), clade VIII of P. cystidiosus(P. cystidiosus var. formosensis), clade X of P. dryinus(P. dryinus var. pometi), clade XIV of P. cornucopiae(P. citrinopilieatus, P. euosmus), and clade XV of P. australis. These species were representative species each clades. Five species, P. ulmarius(clade VI), P. griseus(clade IX), P. calyptratus(clade XI), P. lampas(clade XII), P. smithii(clade XIII)and P. serotinus (clade XVI) were used each one strain in analysis, so they were clustered other groups.

Physicochemical Characteristics of Ethanol Extracts from Each Part of the Pleurotus eryngii (새송이버섯(Pleurotus eryngii) 부위별 추출물의 이화학적 특성)

  • This study has examined the physicochemical properties of the Pleurotus eryngii, including their proximate components, amount mineral content, total dietary fiber, total sugar, reducing sugar and free sugar. Additionally, it measured the P. egii ethanol extracts and the total amounts of polyphenol compounds as well as its electron donating ability (EDA) of the substance fraction (SF). The P. eryngii powder's moisture content was 9.0% and each of the other element content such as carbohydrate, crude protein, crude ash and crude fat was found to be 63.06%, 20.70%, 5.20% and 2.0% respectively. Potassium (K) was shown to be the greatest inorganic content and manganese (Mn) was the lowest. Furthermore fructose, galactose, glucose lactose and maltose free sugar content was found in this order. 387 mg% of the total amounts of polyphenol was found in the P. eryngii whole body ethanol extract, 158 mg% in the stipe extract, 593 mg% in the pileus extract and 607 mg% in the substance fraction (SF). Electron donating ability (EDA) was highest at 91.12% in the whole body extract and lowest at 62.90% in the stipe extract. Additionally, the EDA for substance fraction (SF) 0.02%-0.1% was found to be 57.78-77.33%, which was lower than the 0.02%-tocopherol (93.92%) and BHT (96.72%). From these results, it can be assumed that P. eryngii offers superior antioxidative effects with its high content of fiber, inorganic materials and total amounts of polyphenol as well as high electron donating ability (EDA), thereby making it ideal for use in functional foods and industrial materials.

Methods of Agrocybe cylindracea simultaneous cultivation for small scale bottle cultivation farmers of Pleurotus eryngii (소규모 큰느타리 병재배 농가에서 버들송이 동시재배 방법)

  • Small scale mushroom growers take a lot of the costs in the bottle cultivation than the large scale growers. Therefore, they are not competitive in the market. Mushroom cultivation in small scale mushroom farm is labor-intensive and this experiment was carried out to establish the method for the cultivation of various kinds of mushrooms together under the similar conditions in the production system. As a result, the common medium were selected based on the growing conditions of Pleurotus eryngii and Agrocybe cylindracea, and the level of medium moisture content were studied. The results were shown below. When the medium in the input stage for bottle cultivation was filled by using automatic machine, the range of the three state rates in the bottle is different depending on ingredients and the mixing ratio. The optimum moisture content of the medium for some mushroom production was a different trend based on the medium cluster of the raw materials. The optimum moisture content for P. eryngii in the medium was 70% (douglas fir sawdust, rice bran or sawdust, dried bean curd refuse). In the medium containing douglas fir sawdust, wheat bran was 65%, and the medium including douglas fir sawdust, wheat bran, dried bean curd refuse was 67%. The optimum moisture content of the above three media for A. cylindracea was 70%. The suitable medium for the bottle cultivation of P. eryngii was selected as douglas fir sawdust 75%, wheat bran 20%, dried bean curd refuse 5%, and moisture content 67%. The medium of A. cylindracea was selected as douglas fir sawdust 75%, wheat bran 25%, and moisture content 70%. The suitable medium for bottle cultivation of P. eryngii and A. cylindracea was shown as douglas fir sawdust 75%, rice bran20%, dried bean curd refuse 5%, and 70% moisture content to be used as a common medium of the growing. The incubation period, primordial formation, and growth environment conditions of P. eryngii and A. cylindracea were a similar trend. Therefore, the small scale farmers of P. eryngii can cultivate A. cylindracea together with P. eryngii.

Analysis of Environment Factors in eryngii Cultivation House (새송이 버섯 재배사의 환경요인 계측)

  • Pleurotus eryngii is one of the most promising mushrooms produced on the domestic farms. The quality as well as quantity of Eryngii is sensitively affected by micro climate factors such as temperature, relative humidity, $CO_2$ concentration, and light intensity. At the commercial mushroom cultivation houses, this study was carried out to find out reasonable range of each environmental factor and yield together with economic and safe structures influencing on the optimal productivity of Eryngii. This experiment was conducted from Jan. 26, 2003 to Aug. 2, 2003. Ambient temperature during the experiment period was not predominantly different from that of a normal year. The capacity of the hot water boiler and the piping systems were not enough. Maximum temperature difference between the upper and lower growth stage during a heating time zone was about $6^{\circ}C$. The max. and min. relative humidity were ranged approximately $42{\sim}100%$. The $CO_2$ concentration and the illuminance were lowly maintained during growing period. The average yield per bottle was about $54{\sim}102g$.

Analysis of Environment Factors in Pleurotus eryngii Cultivation House (새송이버섯 재배사의 환경요인 분석)

  • Pleurotus eryngii(King oyster) is one of the most promising mushrooms produced on the domestic farms. The quality as well as quantity of Eryngii is sensitively affected by micro climate factors such as temperature, relative humidity, $CO_2$ concentration, and light intensity. To safely produce high-quality Eryngii all the year round, it is required that the environmental factors be carefully controlled by well designed structures equipped with various facilities and control systems. At the commercial mushroom cultivation house(A,B), this study was carried out to find out reasonable range of each environmental factor and yield together with economic and safe structures influencing on the optimal productivity of Eryngii. this experiment was conducted for about two-month from Nov. 11, 2002 to Dec. 30, 2002 in Eryngii. cultivation house-A, B. Ambient temperature during the experiment period was not predominantly different from that of a normal year. The capacity of the hot water boiler and the piping systems were not enough. Maximum air temperature difference between the upper and lower growth stage during a heating time zone was about 2~3$^{\circ}C$. The max. and min. relative humidity were ranged approximately 60~100%, and average relative humidity was ranged approximately 80~100%. And $CO_2$concentration increased until maximum 1,600~1,800 ppm with the passing growing period. The illuminance in cultivation house was widely distributed from 20lx to 160 lx in accordance with position, and it was maintained lower than the recommended illuminance range 100~200 lx. The average yield per bottle was about 67~85g. But the optimal productivity will be evaluated by considering the quality and quantity of mushroom production, energy requirements, facility construction and management cost, etc.

Changes of freshness in Pleurotus eryngii according to oxygen permeability of packaging film and net weight (포장재 산소투과도 및 포장량에 따른 큰느타리버섯 신선도 변화)

  • Recently, Pleurotus eryngii, King oyster mushroom is the main export item in South Korea mushroom industry. For appropriate transportation to Europe and USA markets, it is necessary to elucidate suitable conditions of storage and treatment of post-harvest during long time shipping. Whole mushrooms were packed with polypropylene anti-fog film (30 um) without trimming. The range of package weight and oxygen permeability of film were 200 g~400 g and 2000~5000 $cc/m^2$, 24 h,atm, respectively. The weight loss ratio was increased with increasing storage time. There were no big differences in weight loss ratio according to package weight and oxygen permeability of film at low temperature storage($4^{\circ}C$). In case of package weight is 400 g and oxygen permeability of film is 3000 $cc/m^2$, 24 h, atm. As a results of freshness, the optimum packing weight and oxygen permeability of film of P. eryngii were 400 g and below 2,000 $cc/m^2$, 24 h, atm, respectively.
