Agrocybe cylindracea로부터 추출한 다당류의 면역증강활성 (Immunopotentiating Effect of Polysaccharides Extracted from Agrocybe cylindracea)

  • Agrocybe cylindracea으로부터 분리 배양한 균사체 배양액을 이용하여 균사체내 다당류(IP)와 균사체외 다당류(EP)를 분리하고, 동물실험을 통한 각 다당류의 면역증강활성에 관해 실험을 실시하였다. 탐식능에 대한 실험결과, IP와 EP 모두는 복강 침출 세포(PEC)에서 대조군에 비해 6.6-50%, 말초 임파구(ML)에서 67-167%, 비장(SP)에서 16-90%의 전반적으로 높은 탐식능을 나타냈다. 또한 항체 생성능 측정을 위한 plaque forming cell(PFC)와 rosette forming cell(RFC)의 실험결과에서 볼 때, PFC형성능은 대조군에 비해 EP투여 II군에서 46-50%, IP투여 I군에서 49-70%의 활성증가를 나타내었다. 또한 RFC형성능은 대조군에 비해 IP투여 I군에서 91%의 증가를, EP투여 I군에서 33%, EP투여 II군에서 43%의 활성증가를 나타내어 높은 항체생성이 기대되었다. Mitogen 활성은 농도 의존적으로 세포 증식능을 보였으며, IP와 EP를 비교할 때, EP는 IP에 비해 약2.5배의 세포증식 활성을 보였다.

Chitosan이 마우스의 정상 및 cyclophosphamide로 억제된 일차 체액성 면역반응에 미치는 영향 (Effects of Chitosan on the Normal and Cyclophosphamide-suppressed Primary Humoral Immune Response in Mice)

  • In order to investigate the effects of chitosan on the normal and cyclophosphamide (CY)-suppressed primary humoral immune response in mice, chitosan was orally administered alone (single dose of 62.5, 250 mg/kg) or with CY (20 mg/kg, i.p.) to female ICR mice on the 2nd day before or after immunization with SRBC-antigen. When chitosan alone was administered before antigenic challenge, splenic IgM plaque forming cells (PFC) and splenic cellularity were slightly increased and serum IgM was not changed when compared with control group. However, chitosan significantly enhanced PFC, serum IgM and splenic cellularity when administered after antigenic challenge. The PFC numbers, serum IgM and splenic cellularity were significantly decreased by the treatment of CY, whereas those values were slightly increased by the concomitant treament of CY and chitosan when compared with CY alone-administration. These results indicate that chitosan is able to increase normal humoral immunity (HI) and to slightly inhibit the suppressive effects of CY on HI.

Effect of Kp, an Antitumor Protein-Polysaccharide from Mycelial Culture of Phellinus Linteus on the Humoral lammune Response of Tumor-Bearing ICR Mice to Sheep Red Blood Cells

  • The immunomodulating activity of Kp, an antitumor protein-polysacchanide preparation from the shake-cultured mycelia of Phellinus linteus, was investigated in ICR mice subcutaneously implanted wit $1\times10^6$ cells of sarcoma 180. The mice were intraperitoneally administered with Kp at a does of 100 mg/kg once daily for five consecutive days starting from 24 hrs after the tumor implantation. Ten days after the last injection, the mice were immunized with $1\times10^7$ or $4\times10^8$ sheep red blood cells (SRBC) and five days later, the antibody-forming immune response were assessed by direct hemolytic plaque assay. To an immunization does of $1\times10^7$ SRBC, the Kp-treated mice elicied a successful humoral immune response despite the turmor-burden and produced $259\times10^3$ plaque-forming cells (PFC)/spleen, while the corresponding tumor-bearing control mice showed virtually no reponse $(2.0\times10^3$ PFC/spleen) (the stimulation index=129.5). However, to an immunization dose of $4\times10^8$ SRBC, both of the control mice and Kp-treated mice showed almost the same level of strong humoral immune response. From these data it is clear that Kp effectively restores the humoral immune response of the turmor-bearing ICR mice.

  • Effects of squalene on humoral immune system in mice were investigated. Squalene exhibited significant increases in the circulating leukocyte counts and relative spleen and thymus weights of the mice. However, the relative liver weight was slightly decreased. Hemagglutination titers (HA) were signficantly enhanced by squalene while Arthus reaction was not affected. Splenic plaque forming cells (PFC) were also greatly increased by squalene, especially at doses of 50 and 100 mg/kg of it.

  • Effects of Compound-A, a phenylpropanoid isolated from Arctium lappa fruit, on sheep red blood cells (sRBC)-induced arthus reaction (AR) were studied in ICR male mice and determined the plaque forming cells (PFC) numbers and hemagglutinin (HA) titer. Two weeks after sensitization of i.p. injection of sRBC (4x10$^{8}$ cells), ICR male mice were challenged by i.p. injection of sRBC (2$\times{10}^8$ cells).   • To determine whether or not bisphenol A affects the Immune system, female ICR mice were treated bisphenol A (BPA) orally at the doses of 100, 500 and 1,000 mg/kg for 30 consecutive days. Four days before enumerating Plaque -forming cells (PFCs) mice were immunized intraperitoneally with sheep red blood cells (SRBCs). The spleen cellularity and PFC/spleen were significantly reduced by 30-day exposure to BPA (1,000 mg/kg/day), but the PFC/10$\^$6/ spleen cells was slightly decreased.. When splenocytes isolated from the mice exposed to BPA for 30 days were cultured in the presence of LPS, Con A or PHA with IL-2, the lymphocyte proliferation ex vivo was not significantly suppressed by BPA. Our present results indicated that 30-day exposure of mice to BPA might have mild immunotoxic potential.

Lymphoblastosis Inhibition and Plaque-forming Cell Response of Several Anti-inflammatory Steroids in Mice

  • Anti-inflammatory glucocorticoid (GC) derivatives have been clinically used in immune-malfunctional diseases for their immunosuppressive activity. However, there is still a lack of knowledge on the relationship between anti-inflammatory and immunosuppressive activities. In order to compare immunosuppressive activities with the known anti-inflammatory activities of the GC derivatives, eight clinically used GC derivatives including hydrocortisone, prednislone, 6$\alpha$-methyl prednisolone, triamcinolone, dexamethasone, betamethasone, triamcinolone acetonide and fluocinolone acetonide were selected, and lymphoblastosis inhibition and plaque-forming cell (PFC) response in mice were studied as immunological parameters. In Con A-induced lymphoblatosis inhibition invitro, all derivatives showed potent inhibition $IC_{50}$ values of the derivaties except methyl prednisolone and triamcinolone were less than $10^{-7}$M and good dose dependency was obtained. This result was well correlated with that of their anti-inflammatory potencies obtained. This result was well correlated with that of their anti-inflammatory potencies and their receptor binding affinities. However, in PFC response, consistent result were not obtained. Total numbers of PFCs per spleen were decreased by some derivatives, but numbers of PFCs per $10^6$ cells were not decreased by systemic administration of but numbers of PFCs per $10^6$ cells were not decreased by systemic administration of GC at the dose of 0.05 mg/mouse. Furthermore, at the dose of 0.1 mg/mouse, numbers of PFCs per $10^6$ cells were found to be increased, although total PFCs per spleen were decreased.

Effects of Swainsonine on the Humoral Immune Response of Lipopolysaccharide

  • Effects of swainsonine (SW;8${\alpha}$, ${\beta}$-indolizidine-${\alpha}$, $2{\alpha}$8-triol from Locoweed) on the humoral immune responses of lipopolysaccharide (LPS) wer studied in ICR mice. Mice were divided into 4 groups (10 mice/group), and LPS was given to each mouse 1 hr after i.p. injection with 3.7 mg/kg of swainsonine, by i.p. injection twice a week for 14 days at a dose of 2 mg/kg. Humoral immune responses were evaluated by hemagglutination (HA) titer and splenic plaque forming cells (PFC). The results of this study were summarized as follows: Mice administrated each of LPS and SW showed significant enhancement of the weight ratios of spleen to body, HA titer, 2-mercaptoethanol-resistant HA(MER-HA) titer and PFC compared with those in controls. However, the LPS plus SW treatment decreased HA titer, MER-HA titer and PFC corresponding to humoral immunity, as compared with those in the mice treated with LPS alone. These findings indicated that LPS significantly enhanced humoral immune responses, but their enhancement effects were lowered somewhat by SW.

  • Phellinus linteus (PL)-hot water extract (PL-W) or- methanol extract (PL-M) was orally administered alone (single dose of 400, 800, 1600 mg/kg; 800 mg/kg/day for 5 days) or with cyclophosphamide (CY, 20 mg/kg, i.p.) to female ICR mice. Within PL alone-treated group, WBC and plaque forming cells (PFC) to SRBC were slightly and significantly enhanced when compared with control group. The relative thymus and spleen weights, WBC and PFC numbers were significantly decreased by the treatment of CY, whereas those values were markedly increased by the concomitant treatment of CY and PL when compared with CY administration alone. To assess the effects of PL and/or CY on the mitogen response of splenocytes io LPS, mouse splenocytes were stimulated with or without LPS in the presence of various concentration of PL and/or CY in vitro and splenocytes proliferation (SP) was measured by MTT assay. PL alone increased both SP and LPS- stimulated SP. Moreover, SP and LPS-induced SP suppressed by the treatment of CY alone were significantly restored by PL-treatment. These activities were higher by PL-M than by PL-W, These results indicated that PL was able to increase humoral immunity and to inhibit immunotoxicity induced by CY.

  • Effects of Astragali radix extract (AG) on the humoral immunotoxic responses of zinc chloride (Zn) were studied in ICR mice. Mice were divided into 4 groups (10 mice/group), and Zn was given to the mice, 1 hr after i.p. injection with 0.5 g/kg of AG, by i.p. injection daily for 10 days at a dose of 25 mg/kg. Mice were immunized and challenged with sheep red blood cells (SRBC). Zn treatment increased the relative weight of spleen compared with those in controls, but decreased the hemagglutination (HA) titer 2-mercaptoethanol-resistant HA (MER-HA) titer and splenic plaque forming cells (PFC). AG treatment significantly increased the relative weight of spleen, HA titer and PFC compared with those in controls. Combination of Zn and AG significantly increased the relative weight of spleen compared with those in controls, but decreased the HA titer, MER-HA titer and PFC. The relative weight of spleen was significantly increased in the Zn and AG combination group than those in Zn alone treatment group. But the HA titer, MER-HA titer and PFC were slightly increased in the group of combination. These results suggest that AG might slightly restore humoral immune responses lowered by an excess of zinc.
