Pilot Plant Scale Extraction and Concentration of Purple-Fleshed Sweet Potato Anthocyanin Pigment (자색고구마 anthocynin 색소의 대량추출 및 농축)

  • Performance of pilot plant scale extraction and concentration of purple-fleshed sweet potato anthocyanin pigment was tested and the characteristics of pigment extracts and concentrates were investigated. Fifty kilograms of purple-fleshed sweet potato was extracted with 500 L of 1% citric acid in 20% ethanol. As a whole, extraction pattern of the large scale extraction was similar to that of the laboratory scale extraction. The extracted pigment solution was filtered twice with a bag filter and a winding type microfilter and the filtrate was concentrated by a large scale vacuum evaporator at $40^{\circ}C$ and 600 mmHg vac. The mean values of total optical density (TOD) of the extract and the concentrate were 6.53 and 120.45, respectively. Browning index (BI) and Degradation index (DI) of extract were 5.86 and 1.55 and those of concentrate were 5.89 and 1.56, respectively, which indicated that the pigments were not changed or degraded through the extraction and concentration process.

  • Natural gas liquefaction plant and LNG carrier needs large capital investment. Therefore a lot of small or middle scale natural gas fields aren't developed due to poor profitability. If natural gas is made to gas hydrate instead of liquefaction, developing small-scale natural gas field can be profitable because building cost of gas hydrate plant and carrier are economical. Because the process of making gas hydrate consumes much energy, the gas hydrate formation process has to be optimized for energy consumption. In this study, gas hydrate formation process was investigated experimentally. Experimental apparatus consists of reactor, pressure regulator, chiller, and magnetic stirrer. 99.95% methane was used to make gas hydrate. Tests were conducted at variable pressure and temperature condition.

High Frequency Bulblet Formation in Scale and Stem Thin Cell Layer Explant Cultures of Lilium Oriental Hybrids (오리엔탈 백합의 인편 및 줄기의 박판 세포층 절편으로부터 고빈도 자구형성)

  • An efficient system for in vitro bulblet formation of Lilium oriental hybrids(cvs). Casa Blanca and Siberia is described. Transverse thin cell layer(tTCL)(1mm thick) explants of 'Casa Blanca' formed bulblets at a frequency of 97.7% when cultured on Murashige and Skoog (MS) medium supplemented with 1mg/L 2,4 dichlorophenoxyacetic acid(2,4-D) (On average 15.6 bulblets were formed per explant). The frequency of bulblet formation was drastically reduced when the explant ghickness was thinner than 1 mm. Explants from the outermost layer of bulb scale produced greater frequency of bulblet formation than middle or innermost layer. Among auxins supplemented to culture medium at 1 mg/L, 2,4-D led to greater frequency of bulblet formation on explants than dicamba, picamba, or phenylacetic acid(PAA). tTCL explants from the middle region of the outermost layer bulb scale yielded greater frequency of bulblet formation than the upper or lower region. tTCL stem explants of 'Siberia' formed bulblets at a frequency of 95.3% when cultured on MS medium with 1 mg/L 2,4-D(On average 9.1 bulblets were formed per explant). The system estabilished in this study will be useful for in vitro rapid propagation and genetic transformation of Lilium Oriental hybrids.

  • To develop a plant safety training system using virtual reality technology, we constructed a training system for a large scale plant. Compared with safety training for small plants or unit equipment, many system configurations such as virtual plant model, in-process data processing, work instruction, etc. are required and integrated system works have been carried out. The target plant, RDS process, is a high-risk process(high-temperature, high-pressure) that takes into account the training scenarios that can be taken in the event of a leaking fire in the range and refer to the actual shutdown procedure. The proposed safety training integration system can be used in similar situations that can occur in the RDS process and can be a platform for safety training using virtual reality in a large plant.

  • Recent desalination plant system are being operated by applying one of the technologies as like MSF, MED and, or RO. The production cost of pure water from sea water by the desalination plant was U$9/m$^3$ in 1960, but now has been reduced to U$1/m$^3$ at present. The Power and Desalination Plant Project in Al Cobar of Saudi Arabia gave good chance to us export the similar plant systems actively to the world market. 1Hidd IWPP Project in Bahrain, Taweelah B Extension Project in Abu Dhabi of UAE and Ras Laffan B IWPP of Qatar are the recent large scale desalination projects under construction.

Large-scale Culture of Plant Cell and Tissue by Bioreactor System

  • Large-scale cultures of plant cell, tissue, and organ have been achieved by using BTBB. When different sized BTBBs (5 L, 20 L, 100 L, 300 L, and 500 L) were tested for the culture of yew cells (Taxus cuspidata Sieb. et Zucc.), cell growth increment reached to 94.5% in SCV after 24 days of culture with 30% of inoculation cell density. However, there were some variations in the production of taxol and its derivatives among the BTBBs of different size. Approximate 4 ㎎/l of taxol and 84 ㎎/l of total taxanes were obtained by using a 500L BTBB after 6 weeks of culture. With a 20L BTBB, about 20,000 cuttings of virus-free potatoes (cv. Dejima) could be obtained by inoculating 128 explants and maintaining 8 weeks under 16 hr light illumination. The frequency of ex vitro rooting of the cuttings revealed as more than 99% under 30% shade. By incorporating two-stage culture process consisting of multiple bulblet formation in solid medium and bulblet development in liquid medium, mass propagation of lily through bioreactor seemed to be possible. In the case of 'Marcopolo', the growth of mini-bulblets in BTBB was nearly 10 folds faster than that of the solid medium. Time course study revealed that maximum MAR yield of ginseng (Panax ginseng C. A. Meyer) in a 5 L and 20 L BTBB after 8 weeks of culture was 500 g and 2.2 ㎏, respectively. By cutting the MAR once and/or twice during the culture, the yield of root biomass could be increased more than 50% in fresh weight at the time of harvest. With initial inoculum of 500 g of sliced MAR in a 500 L BTBB, 74.8 ㎏ of adventitious root mass was obtained after 8 weeks of culture. The average content of total ginseng saponin obtained from small-scale and/or pilotscale BTBBs was approximately 1% per gram dry weight. Based on our results, we suggest that large-scale cultures of plant cell, tissue, and organ using BTBB system should be quite a feasible approach when compared with conventional method of tissue culture.

Efficient aerobic denitrification in the treatment of leather industry wastewater containing high nitrogen concentration

  • To treat leather industry wastewater (LIW) containing high nitrogen concentration, eight aerobic denitrifiers were isolated from sludge existing in an LIW-treatment aeration tank. Among them, one strain named as KH8 had showed the great ability in denitrification under an aerobic condition, and it was identified as Pseudomonas aeruginosa R12. The aerobic denitrification ability of the strain KH8 was almost comparable to its anaerobic denitrification ability. In lab-scale aerobic denitrifications performed in 1-L five-neck flasks for 48 hr, denitrification efficiency was found to be much improved as the strain KH8 held a great majority in the seeded cells. From the nitrogen balance at the cell-combination ratio of 10:1 (the strain KH8 to the other seven isolates) within the seeded cells, the percentage of nitrogen loss during the aerobic denitrification process was estimated to be 58.4, which was presumed to be converted to $N_2$ gas. When these seeded cells with lactose were applied to plant-scale aeration tank for 56 day to treat high-strength nitrogen in LIW, the removal efficiencies of $COD_{Cr}$ and TN were achieved to be 97.0% and 89.8%, respectively. Under this treatment, the final water quality of the effluent leaving the treatment plant was good enough to meet the water-quality standards. Consequently, the isolated aerobic denitrifiers could be suitable for the additional requirement of nitrogen removal in a limited aeration-tank capacity. To the best of our knowledge, this is the first report of aerobic denitrifiers applied to plant-scale LIW treatment.

Removal of VOC compounds in the vent of a pharmaceutical plant using a pilot-scale biofilter (Pilot-scale 바이오필터를 이용한 제약공정 배출가스의 처리)

  • A pilot-plant biofilter ($1750\;m\;W{\times}2750\;mm\;L{\times}2000\;mm\;H$) packed with polyurethane foam ($20\;m\;W{\times}20\;mm\;L{\times}20\;mm\;H$) was installed in an pharmaceutical plant emitting gas streams containing n-hexane and alcohols. The biofilter was successfully operated for 74 days under highly fluctuating incoming concentrations at a residence time of 12.8-24.8 sec. Alcohols and n-hexane were removed by more than 90% from 5 and 20 days after start up, respectively. Malodor was also removed more than 95% from 20 days after start up.
