Use of East Deep Sea Water for the Increase of Functional Components of Ginseng (Panax ginseng C.A. Meyer) and Tomato (Lycopersicon eculentum L.) (인삼과 토마토의 기능성 성분 증진을 위한 동해 해양심층수의 이용)

  • This experiment was conducted to investigate the effect of deep sea water on fruit quality and yield of tomato. In the deep sea water treatments, fruit growth and weight were decreased as the concentration of deep sea water increased. Especially, the fresh weight of second truss was decreased significantly than first truss. Soluble solid content was increased significantly in higher concentration treatment especially at 30mM and 40mM treatment. That was increased more in the first than in the second truss fruits. Most of hexose in fruits were glucose and fructose. The reason of increased glucose and fructose contents was the decline of growth because of salinity stress by deep sea water treatment. however deep sea water treatment increased the lycopene content, especially in 20mM treatment. It is assumed that deep sea water treatment cause induction and promotion of ethylene. The higher concentration of deep sea water to the solution, the eater fruit quality improvement was noticed. However, proportional yield reduction accompanied concentration, 20mM deep sea water improved fruit quality without a significant yield reduction. The Re content was the highest among ginsenosides in all treatments. The contents total of ginsenosides in all treatments, except EC 8 treatment, was higher than those in the controlled treatment. The PT/PD value was 1.31 of the lowest in the EC 8 treatment and was 2.52 of the highest in the EC6 treatment. Rf contents in high increase was detected at all treated ginseng roots.

Reduction of Stem Inside-cavity and Improvement of Flower Quality in Chrysanthemum 'Baekma' by pH Stabilization and Foliar Spray of Ethephon (양액 pH 안정화와 에세폰 살포에 의한 국화 '백마'의 줄기동공 경감과 절화품질 향상)

  • This study was conducted to investigate effects of nutrient solution pH control agent and foliar spray of ethephon on the reduction of stem inside-cavity and improvement of flower quality in chrysanthemum 'Baekma'. Changes of pH in nutrient solution as affected by the kind of pH control agent showed more settled pattern in the plot of KOH treatment than in others. Plant growth and development such as cut flower length, leaf numbers, cut flower weight and petal number were the greatest in the plot of KOH. However, the size and area of stem cavity was large in KOH rather than $KHCO_3$. As changes in the absorbing patterns of mineral elements as affected by nutrient solution pH control agent in closed system, contents of total nitrate, phosphorus, potassium and calcium by $KHCO_3$ was absorbed into the plant less than KOH, so that it remained a lot of mineral element residues rather than KOH in closed system. Plant growth as affected by the foliar spray of ethephon showed growth retardation effect in the plot of solution diluted to 1 : 500 and growth promotion effect in the plot of solution diluted to 1 : 1,000 or 1 : 2,000. The number of petals was the best in the plot of foliar spray of ethephon solution diluted to 1 : 1,000 before flowering at 45 days showing 331 petals compared to control showing 302 petals. The size and area of stem cavity as affected the foliar spray of ethephon was smaller 1 mm and 7%, respectively, in the plot of solution diluted to 1 : 2,000 before flowering at 30 days than in control. Therefore, treating pH stabilization using KOH after floral initiation stage with the foliar spray of ethephon solution diluted to 1 : 2,000 before flowering at 30 days would help to reduce stem cavity size and improve flower quality in hydroponically grown chrysanthemum 'Baekma'.

Phosphate solubilizing effect by two paraburkholderia bacteria Isolated from button mushroom medium (양송이배지로부터 분리한 두 Paraburkholderia 속 세균에 의한 인산가용화 효과)

  • The present study was conducted to investigate the synergistic effects caused by single and co-inoculation of the phosphate solubilizing bacteria (PSB), Paraburkholderia phenazinium YH3 and Paraburkholderia metrosideri YH4. Phosphate solubilization was assessed by measuring the phosphorus contents for 7 days in a single and co-inoculation medium. Co-inoculation of the two strains was found to release the highest content of soluble phosphorus ($1,250{\mu}g\;mL^{-1}$) into the medium, followed by the single inoculation of P. metrosideri YH4 ($1196.59{\mu}g\;mL^{-1}$) and P. phenazinium YH3 ($994.34{\mu}g\;mL^{-1}$). The highest pH reduction, organic acid production and glucose consumption was also observed in the co-inoculation medium of the two strains. A plant growth promotion bioassay revealed that co-inoculation with the two strains enhanced the growth of romaine lettuce more than single inoculation with either of the two strains (28.5% for leaf and 16.6% for root). Although there was no significant difference between single and co-inoculation of bacterial strains in terms of phosphorous release and plant growth, the synergistic effects of co-inoculation with PSB could be beneficial for crop growth.

Effect of NaCl Stress on the Growth, Antioxidant Materials, and Inorganic Ion Content in Head Lettuce Seedlings (양상추 유묘의 생육, 항산화물질 및 무기이온의 함량에 미치는 NaCl 스트레스의 영향)

  • In head lettuce seedlings, NaCl stress was treated with hydroponic culture containing 0, 50, or with 100 mM NaCl in the seedling grown up to two leaf stages. Our focus was on the effect of NaCl on fresh and dry weights, antioxidant materials, and inorganic ion level. Fresh and dry weights of head lettuce seedlings increased with the increase in salinity while the optimal growth occured at 50 mM NaCl. The chlorophyll a (Chl a), total Chl and Chl a/b ratio increased 6 days after treatment with 100 mM NaCI. However, the Chl b content decreased. Total glutathione increased only in the root of head lettuce seedlings, whereas significant increase of total arcorbate content was observed in both shoot and root after the treatment with 100 mM NaCl. In addition, the NaCl treatment resulted in the decreased level of spermidine content, and a increased spermine content. Furthermore, $Na^+$ content in shoot and root increased significantly while $K^+$, $Ca^{2+}$, and $Mg^{2+}$ content decreased. The alteration of inorganic ion level after treatment with NaCl caused the reduction of $K^+/Na^+$, $Ca^{2+}/Na^+$, and $Mg^{2+}/Na^+$ ratio with the increase of NaCl concentration. Taken together, these findings indicate that the treatment of NaCl causes the induction of oxidative stress, and results in the alteration of metabolic mechanism in head lettuce seedlings.

Bacterial core community in soybean rhizosphere (콩 근권의 핵심 세균 군집)

  • Soybean is well known to be originated from Korea and far-east Asian countries, and studies of many root nodule bacteria associated with soybean have mainly-focused on nitrogen fixation, but much less study was carried out on bacterial community in the rhizosphere of soybean. In this study, we analyzed the bacterial community in rhizosphere of Korean soybean, Daepungkong using the pyrosequencing method based on the 16S rRNA gene to characterize the change of the rhizosphere community structure according to the growth stages of soybeans and to elucidate bacterial core community in rhizosphere of soybean. Our results revealed that bacterial community of rhizosphere soil differed from that of bulk soil and was composed of a total of 21 bacterial phyla. The predominant phylum in the rhizosphere of soybean was Proteobacteria (36.6-42.5%) and followed by Acidobacteria (8.6-9.4%), Bacteroidetes (6.1-10.9%), Actinobacteria (6.4-9.8%), and Firmicutes (5.7-6.3%). The bacterial core community in soybean rhizosphere was mainly composed of the operational taxonomic units (OTUs) belonging to the phylum Proteobacteria throughout all growth stages. The OTU00006 belonged to the genus Bradyrhizobium had the highest abundance and Steroidobacter, Streptomyces, Devosia were followed. These results show that bacterial core community in soybean rhizosphere was mainly composed of OTUs associated with plant growth promotion and nutrient cycles.

Phyto-restoration Potential of Soil Properties using Secale cereale for Recycle of Soils with Residual TPHs (Total Petroleum Hydrocarbons) after Off-site Treatment (잔류유분 함유 반출처리토 재활용을 위한 호밀 식재 식물상 토성회복 가능성)

  • The amount of TPH contaminated soil treated at off-site remediation facilities is ever increasing. For the recycle of the treated-soil on farmlands, it is necessary to restore biological and physico-chemical soil characteristics and to remove residual TPH in the soil by an economic polishing treatment method such as phytoremediation. In this study, a series of experiments was performed to select suitable plant species and to devise a proper planting method for the phyto-restoration of TPH-treated soil. Rye (Secale cereale) was selected as test species through a germination test, among 5 other plants. Five 7-day-old rye seedlings were planted in a plastic pot, 20 cm in height and 15 cm in diameter. The pot was filled with TPH-treated soil (residual TPH of 1,118 mg/kg) up to 15 cm, and upper 5 cm was filled with horticulture soil to prevent TPH toxic effects and to act as root growth zone. The planted pot was cultivated in a greenhouse for 38 days along with the control that rye planted in a normal soil and the blank with no plants. After 38 days, the above-ground biomass of rye in the TPH-treated soil was 30.6% less than that in the control, however, the photosynthetic activity of the leaf remained equal on both treatments. Soil DHA (dehydrogenase activity) increased 186 times in the rye treatment compared to 10.8 times in the blank. The gross TPH removal (%) in the planted soil and the blank soil was 34.5% and 18.4%, respectively, resulting in 16.1% increase of net TPH removal. Promotion of microbial activity by root exudate, increase in soil permeability and air ventilation as well as direct uptake and degradation by planted rye may have contributed to the higher TPH removal rate. Therefore, planting rye on the TPH-treated soil with the root growth zone method showed both the potential of restoring biological soil properties and the possibility of residual TPH removal that may allow the recycle of the treated soil to farmlands.

Physiological Response of Chinese Cabbage to Salt Stress (염 스트레스에 대한 배추의 생리학적 반응)

  • In order to understand the plant responses to salt stress (0, 50, and 100 mM NaCl), Chinese cabbage seedlings grown up to two leaf stages by hydroponic culture were used. Fresh and dry weight, chlorophyll (Chl), antioxidant materials, polyamine content, antioxidant enzyme activities, and inorganic ion level were evaluated. Fresh and dry weights of Chinese cabbage increased with the increase in salinity while the optimal growth occurred at 50 mM NaCl. The Chl a, total Chl, carotenoid content, and Chl a/b ratio increased by the 6 days after treatment with 100 mM NaCI; however, the Chl b content decreased. Glutathione increased in the root of Chinese cabbage for 6 days. Dehydroascorbate increased remarkably by day 6 caused by the salt stress in both leaf and the root. While ascorbate peroxidase increased, the activity of catalase and glutathione reductase decreased gradually in the first leaf for 6 days. The $Na^+$ content increased by 12.5-fold in the 3 days after treatment with 100 mM NaCI in the shoot, whereas the $Ca^{2+}$, $K^+$, and $Mg^{2+}$ content measured in the same treatment decreased by 43 to 57%. Spermidine content decreased as salinity increased, but spermine content increased. The growth promotion, glutathione and ascorbic acid content in Chinese cabbage were increased by low salt stress, and shortening of the cultivation period for growth increase of Chinese cabbage is expected.

Soaking Effects of Brassinosteroids on Germination of Rice Under Simulated Acid Rain (인공산성비에 의한 볍씨발아 장해경감에 대한 Brassinosteroids의 처리효과)

  • This study measured the soaking effects of two brassinosteroids(BRs), homobrassinolide(hBR : 225, 23S-homobrassinolide) and epibrassinolide(epiBR : 22R, 23R. 24R-$2{\alpha}$, $3{\alpha}$, 22, 23-Tetrahydroxy-B-homo-7-oxa-$5{\alpha}$-ergostan-6-one) on the germination of two rice cultivars, 'Milyang 23' and 'Naepungbyeo' exposed to simulated acid rain(SAR). The results suggested that promoting effect of two BRs on alleviating damage to SAR would be attributed to the promotion of plant growth, root elongation, and particularly germination rate of both cultivars. Eight organic acids were determined by HPLC. Succinic acid acetic acid in 'Milyang 23' under SAR were highest among eight organic acids.

Isolation and Characterization of Fungal Diversity from Crop Field Soils of Nigeria

  • In order to find indigenous beneficial fungal species from crop field soils of Nigeria, 23 soil samples were collected from various places of Nigeria in June, 2013 and fungi were isolated through serial dilution technique. Isolated fungi were purified and differentiated according to their morphological and microscopic characteristics. In total, 38 different representative isolates were recovered and the genomic DNA of each isolates was extracted using QIAGEN$^{(R)}$ Plasmid Mini Kit (QIAGEN Sciences, USA) and the identification of fungi was carried out by sequence analysis of internal transcribed spacer (ITS) region of the 18S ribosomal DNA (18S rDNA). Recovered isolates belonged to 9 fungal genera comprising Fusarium, Aspergillus, Chaetomium, Coniothyrium, Dipodascaceae, Myrothecium, Neosartorya, Penicillium and Trichoderma. Aspergillus spp., Penicillium spp. and Trichoderma spp. were the most dominant taxa in this study. The antagonistic potentiality of species belonged to Trichoderma against 10 phytopathogenic fungi (F. oxysporum, C. gloesporoides, P. cytrophthora, A. alternata, A. solani, S. rolfsii, F. solani, R. solani, S. sclerotiorum and P. nicotiana) was assessed in vitro using dual culture assay. The dual culture assay results showed varied degree of antagonism against the tested phytopathogens. The potential Trichoderma spp. will be further evaluated for their antagonistic and plant growth promotion potentiality under in vivo conditions.

According to the this study tried to provide basic information for vitalizing participation in the renewable energy business by analyzing the factors affecting the new energy business participation roadmap using the multi-layered model. As a result of this study, it can be seen that in order to achieve an effective new energy business participation roadmap, it is necessary to re-establish the new and renewable energy business participation factors by industry. Specifically, it is necessary to prepare appropriate countermeasures such as the leading organization of the project, profit distribution method, green industry financial support, project implementation method, power plant location, and comprehensive long-term strategy, and there is an institutional approach that can promote the development of related industries. found to be necessary. Above all, it suggested that in order to achieve an effective new energy project participation roadmap, an institutional approach is needed that can expand the supply of new and renewable energy while minimizing the burden on the national economy and promote the development of related industries.
