Micropropagation and adventitious root production via the culture of Polygonatum odoratum were performed. Stem segments of seedlings of Polygonatum odoratum were the most efficient explants for adventitious shoot formation compared to leaf and root segments. Exogenous cytokinin treatment was required for adventitious shoot formation. Among the cytokinin (BA, Kinetin and Zeatin) tested, BA was most effective for shoot formation from stem segments. Auxin (NAA or IBA) in combination with cytokinin significantly enhanced adventitious shoot formation. Twenty five percent of explants produced adventitious shoots on medium with 2.0 mg/L BAP alone, while 83% of explants produced adventitious shoots on medium with the combination of 2.0 mg/L BAP and 0.1 mg/L IBA. Rooting of adventitious shoots was achieved after transferring to 112 MS medium supplemented with 0.1 mg/L IBA and 0.5 mg/L zeatin. When stem segments were cultured on MS medium with various kinds of auxin (IBA, NAA and 2,4-D), adventitious roots were formed from callus. frequency of adventitious root formation was highest in 2,4-D than NAA and IBA. When roots were in clusters together with parental stem segments, growth of roots actively occurred in hormone-free MS liquid medium. The above results represent that possible application for the mass production of roots and plantlets through in vitro culture system of Polygonatum odoratum.
Effects of cadmium on growth and nitrogen fixation activity of Melilotus suaveolens, a biennial legume plant dominating in the area of poor soil were quantitatively analyzed during the growing period. Cadmium treatments of 10, 30 and 100 ppm resulted in 12, 22 and 35% inhibition of plant height and 14, 25 nd 26% reduction of chlorophyll contents of leaves, respectively. The plant biomass reduced 51, 70 and 89% for leaves, 33, 50 and 59% for stems, and 42, 52 and 70% for roots, respectively by 10, 30 and 100 ppm Cd treatments. Cadmium contents of roots treated with 10, 30 and 100 ppm Cd were 62, 112 and 183 folds higher than that of the control, respectively. Cadmium contents of stems were about 1/2.2 of those of roots, but leaves contained only 1/27.8 of those of roots. Cadmium treatments resulted in increase of T/R ratios and decrease of F/C ratios significantly in the later growth period. Nodule formation was reduced to 8% in 42 days by the treatment of 100 ppm Cd. Specific nitrogen fixation activity of nodules attained 61.0, 24.0, 1.6 and 0.7 mol $C_2H_4{\cdot}g$ fr wt $nodule^{-1}{\cdot}h^{-1}$ on 42nd day, respectively for 0, 10. 30 and 100 ppm of Cd treatments. Total amount of nitrogen fixation per plant reduced by 73, 98 and 99% with the treatments of 10, 30 and 100 ppm Cd.
Adventitious root culture was established in the Jawahar variety of Withania somnifera using MS basal medium supplemented with 0.5 (mg/l) IAA and 2.0 (mg/l) IBA. Root tips from germinated seedlings, MS0 maintained plants and adventitious roots were maintained in suspension medium (1/2 MS basal medium supplemented with 3% sucrose) for a period of 1 to 6 months. The weight gain in roots was noted and the withanolides were extracted from the dry roots using solvents petroleum ether, 50% ethanol and chloroform. The withanolides in the chloroform fractions of all root samples analyzed were compared using thin layer chromatographic analysis. Withanolide content in adventitious root sample was found to be superior compared to other roots at any given point of time during the 6month growth period.HPLC analysis of in vitro adventitious roots showed the presence of a new compound.
The roots of grapevine in the field in which the crown gall was occurred severely in Chungbuk province were collected and Agrobacterium spp. were isolated from the roots using the selective media. The selected 13 isolates were identified as A. tumefaciens with fatty acid analysis using MIDI system, nucleotide sequence of 16S rDNA, biochemical characteristics, and PCR with the species-specific primers. A. vitis, a pathogen of crown gall disease of grapevine was not isolated from the roots. All of the isolates did not show pathogenicity on the tomato seedlings and the stem and root of grapevine. Eric-PCR showed that DNA band patterns of the root isolates were a little more similar to A. tumefaciens than A. vitis. However, overall similarity between the root isolates and the pathogenic strains of A. tumefaciens and A. vitis was low by rep-PCR. These results suggest that a pathogen causing crown gall in grapevine in Chungbuk province may transmitted through the seedlings rather than via soil or roots.

This experiment was carried out to determine the adequate machine peeling and retting time of peony roots, which are used as medicinal resources in Korea. When the roots were peeled for $10{\sim}30$ minitues in the peeling machine, the epidermis of root was removed, and higher yield was obtained and paeoniflorin content as compared with roots which were peeled for longer time. The paeoniflorin content of root varied with the different parts of root. The highest content was recorded in the upper part and the lowest in the middle part. The loss of paeoniflorin increased in the roots processed for expanded peeling and retting time. Lightness of peeled root was higher and ${\Delta}\;E$ was lower in the roots which were peeled by machine for longer time.

Ginseng(Panax ginseng C.A. Meyer) is important medicinal plant but requires 4-year cultivation for root harvest because of slow growth. In contrast, ginseng callus and hairy roots grow vigorously and may Produce the same or more biologically active compounds for human health than natural ginseng roots. Therefore, ginseng callus and hairy roots can be used for commercial purposes. Polyacetylene, one of anti-cancer compounds in ginseng, was not detected in the callus cultured on the medium containing 2, 4-B, but cells derived from the callus growth was excellent, The ginseng calli cultured on the medium containing 2mg11 CPA and 0.05mg/1 BA was grown vigorously and produced panaxydol, one of ginseng polyacetylene. The biosynthesis of polyacetylene in callus was not affected by addition of NAA and sucrose in media. The SH medium was better than the MS medium for ginseng callus growth and biosynthesis of panaxydol. Another ginseng anti-cancer compounds, ginsenoside-Rg$_3$, Rh$_1$and Rh$_2$ were detected in ginseng hairy roots by heat treatment. Those of Panax ginseng were obtained after root disks of three-year old roots were infected with Agrobacterium rhizogenes Rl000 $A_4$T in dark condition after one month of culture. The optimum growth of hairy roots was achieved in the culture of 1/2 MS liquid medium in dark(22$^{\circ}C$) under 60 rpm gyratory shaking. Hairy roots grew well in 5 ι Erlenmeyer flasks, 1ι roller drums, 10ι jar-fermenters, and especially in 20ι air-lift .culture vessels. All heat treatments had remarkably different ginsenoside contents. Eleven ginsenosides were determined in heat treatment, eight in freeze dried hairy roots. Contents of ginsenoside-Rbl , Rb2, Rc, Rd. Re, Rf, and Rg$_1$tested in all heat treatments were less than those of freeze dried hairy roots. Contents of glnsenoside-Rg$_2$ in heat treatment for 1 hour at 105$^{\circ}C$ was 4.92mg/g dry wt, 3.9 times higher than 1.27 mg/g dry wt of freeze dried hairy roots. The optimum condition of heat treatment for the production of ginsenoside-Rg$_3$and Rhl was 2 hours at 105$^{\circ}C$, and ginsenoside content was 2.58mg/g dry wt and 3.62mg/g dry wt, respectively. The production of ginsenoside-Rh2 was the highest in heat treatment for 2 hours at 105$^{\circ}C$ among treatments examined, and ginsenoside-Rh$_2$content was 1.08mg/g dry wt.
In order to survey anthraquinone productivity from in vitro root culture, transformed hairy roots of Rumex crispus were induced from leaf segments by infection with Agrobacterium rhizogenes strain $A_4$ and compared with adventitious roots. The optimum condition of adventitious root formation from leaf segments was 5 $\mu$M NAA added to MS medium. Mannopine was detected in the extract of hairy roots by paper electrophoresis, but not in adventitious roots. Secondary root tips of both adventitious roots and hairy roots elongated without lateral root branching in hormone free MS medium, but primary root tips showed more rapid growth with extensive lateral root branching. MS basal medium was the best for growth of the adventious roots and hairy roots for anthraquinone content. Adventitious root tips and hairy root tips cultured in liquid MS medium supplemented with 0.05 $\mu$M NAA and 0.1 $\mu$M kinetin (contained 5% sucrose) showed the maximal growth and anthraquinone content. Anthraquinone content of hairy roots was increased by the culture periods, but was reduced after 25 days of culture.
This work was carried out to establish adventitious root culture system in three Panax species (wild-grown P. ginseng, P. quinquefolium, and P. japonicum) to analyze their ginsenoside productivity. Adventitious roots were induced directly from segments of seedlings after cultured on MS(Murashige andSkoog 1962) solid medium containing 3.0 mg/l IBA. Omission of $NH_4NO_3$ from the medium greatly enhanced both the frequency of adventitious root formation and number of roots per explants in all the three Panax species. However, elongation of post-induced adventitious roots was enhanced on medium with $NH_4NO_3$. Two-step culture protocol: $NH_4NO_3$-free medium for first two weeks of culture, followed by $NH_4NO_3$ containing medium for further 4 weeks, greatly enhanced the fresh weight increase of adventitious roots in all the three ginseng species. The fresh weight of adventitious roots was high in P. quinquefolium and low in P. ginseng, followed by P. japonioum regardless of the composition of medium. Pattern and content of ginsenosides in adventitious roots differed among the three Panax species. Total ginsenoside content of adventitious roots in P. quinquefolium, P. ginseng, and p. japonicum was 8.03, 15.7 and 1.2 mg/g dry weight, respectively. Among the three speices, adventitious roots in P. quinquefolium produced hig-hamount of ginsenosides. The pattern of ginsenoside fractions between P. ginseng and P. quinquefolium was similar but the amount of ginsenoside differed between the two, While, in P japonicum, total ginsenoside content was very low and some ginsenosides such as ginsenoside Rb2 and Rf were not detected. Conclusively, we demonstrate that same culture condition was required for induction and elongation of adventitious roots of three ginseng species but growth of adventitious roots and their ginsenoside production were different among them.
Dendropanax morbifera (Araliaceae) is an endemic species in Korea and distributed in the southern part of Korea. The roots and stems of this plant have been used for folk medicine for the treatment of migraine headache, dysmenorrheal, and remove wind dampness and for Vanishes production. Production of adventitious roots in D. morbifera by in vitro cultures could be used as alternatives materials. Leaf, stem, and root segments from D. morbifera seedling were cultured on Murashige and Skoog (MS) medium supplemented with 3.0 mg/L IBA and 30 g/L sucrose. After 4 weeks of culture, the highest induction of adventitious roots was obtained from the leaf segment. Frequency of adventitious root formation on medium with various kinds of auxins (IAA, NAA, 2,4-D, and IBA) and various concentrations of IBA (0, 0.1, 0.5, 1.0, 3.0, and 5.0 mg/L) was tested. The maximum induction of adventitious root was obtained on medium with 1.0 mg/L IBA. In liquid culture, growth of root was best 1/2MS medium supplemented with 1.0 mg/L IBA and 30 g/L sucrose. Adventitious roots were cultured in 5 L bioreactor containing 1/2 MS medium supplemented with 1.0 mg/L IBA and 30 g/L sucrose and mass-production of adventitious roots was successfully achieved. This study demonstrated for the first time to produce adventitious roots in D. morbifera.
To study the effects of metal ions on the activity of anti-oxidase enzymes, the activity of superoxide dismutase (SOD) and peroxidase (POD) and isozyme patterns of Brassica juncea have been studied after treating with CD, Cu, Zn, and Al. The activity of SOD after treating with metal ions was higher than that of untreated control. SOD activity in leaves increased by treatment of 50 ppm of Zn and 500 ppm of Al. POD in stems gave highest activity after treating with 500 ppm of Cu. When the activity was compared by plant parts, lowest POD activity was observed in leaves in which protein content was higher than other tissues. When the activity was expressed as percentage of control, SOD activity was increased after treating with metal ions. SOD activity in leaves and roots of metal treated plant was significantly increased under the metal ions stress conditions. In the roots of 50 ppm of Zn treated plant, SOD activity was extremly high. POD activity was inhibited with Cd and Zn treatment in all parts of the plant. However, in leaves and stems, there was marked increase in activity after treating with Cu. The patterns of SOD isozyme after metal treatment show that two bands were stained in all metal ion treated and that no new band appeared. POD isozyme band intensity resulting from the treatment of metal ions was in order of roots > stems > leaves, but there was no significant difference.
